Annona cherimola Mill., cherimoya, is a tropical plantation crop of interest in fruit culture. Micropropagation techniques have been developed due to the need to increase productivity through clonal selection. Because of the mycorrhizal dependence exhibited by this crop for optimal growth and the recognized role of mycorrhiza establishment for the survival and development of most of the plants produced in vitro, the effect of mycorrhiza inoculation on the development of micropropagated plants of Annona cherimola was investigated. Mycorrhizal inoculation was assayed at two different stages of the micropropagation process: (i) immediately after the in vitro phase, before starting the acclimatization period, and (ii) after the acclimatization phase, before starting the post-acclimatization period under greenhouse conditions. Plantlet survival was about 50 % after the acclimatization period. Plant growth and development profited remarkably from mycorrhiza establishment. Most of the arbuscular mycorrhizal fungi (AMF) assayed greatly increased shoot and root biomass and leaf area. Micropropagated Annona plants seem to be more dependent on mycorrhiza formation for optimal growth than plants derived from seeds. The greatest effects of AMF on plant growth were observed when they were introduced after the acclimatization period.

The present study is, to the best of our knowledge, the first to investigate the use of the fractal dimension (FD) to quantify the growth and development of undisturbed, fully functional arbuscular mycorrhizal (AM) hyphae developing in vitro. The majority of the work focused on the model AM fungus Glomus intraradices DAOM 181602. The time course study and final measurements of an intact mature extraradical mycelium allowed us to compare the development of the mycelium and the FD value. The final FD value of 1.62 for the mature mycelium is similar to that obtained for highly branched root systems and tree crowns. The FD method was used to characterize the morphology of germinative and presymbiotic hyphae in the presence of stimulatory (strigolactone GR-24, 0.1 µmol·L–1 and bisphenol A, 10 µmol·L–1) and inhibitory (NaCl, 80 mmol·L–1) molecules, and the extraradical phase in the presence of an inhibitory molecule (NaCl, 80 mmol·L–1). Where possible, results were compared with those obtained using the traditional grid-line (GL) technique. The FD approach allowed treatment effects to be accurately quantified, both in germinative and extraradical phases. In the second case, this technique provided a single quantitative value of extraradical hyphal growth that included runner hyphae (RH) networks, and fine-branching (FB) ramifications. This is in contrast to the GL technique, which provides a value for the estimation of RH, but which is not suitable for accurately measuring FB hyphae. Given the ease with which the FD values can be calculated, and the fact that this method can provide a single value for the quantification of extraradical hyphal growth and development, we suggest that this method is useful for in vitro studies. Furthermore under certain situations of germinative or presymbiotic growth, it may be used in concert with the GL method to provide a greater degree of information about hyphal morphology. The usefulness and limits of the FD method at different stages of the AM fungal growth cycle are discussed.

Inoculum of arbuscular mycorrhizal fungi, with growing use in horticulture, is produced mainly in two technically different cultivation systems: in vivo culture in symbiosis with living host plants or in vitro culture in which the fungus life cycle develops in association with transformed roots. To evaluate the effectiveness and the infectivity of a defined isolate obtained by both production methods, a replicated comparative evaluation experiment was designed using different propagules of Rhizophagus irregularis produced in vivo on leek plants or in vitro in monoxenic culture on transformed carrot roots. The size of the spores obtained under both cultivation methods was first assessed and bulk inoculum, spores, sievings, and mycorrhizal root fragments were used to inoculate leek plantlets. Spores produced in vitro were significantly smaller than those produced in vivo. Although all mycorrhizal propagules used as a source of inoculum were able to colonize plants, in all cases, leek plants inoculated with propagules obtained in vivo achieved significantly higher mycorrhizal colonization rates than plants inoculated with in vitro inocula. Inoculation with in vivo bulk inoculum and in vivo mycorrhizal root fragments were the only treatments increasing plant growth. These results indicate that the production system of arbuscular mycorrhizal fungi itself can have implications in the stimulation of plant growth and in experimental results.

Nowadays, microorganisms that display plant growth promoting properties are significantly interesting for their potential role in reducing the use of chemical fertilizers. This research study proposed the isolation of the actinobacteria associated with arbuscular mycorrhizal fungi (AMF) spores and the investigation of their plant growth promoting properties in the in vitro assay. Three actinobacterial strains were obtained and identified to the genus Streptomyces (GETU-1 and GIG-1) and Amycolatopsis (GLM-2). The results indicated that all actinobacterial strains produced indole-3-acetic acid (IAA) and were positive in terms of siderophore, endoglucanase, and ammonia productions. In the in vitro assay, all strains were grown in the presence of water activity within a range of 0.897 to 0.998, pH values within a range of 5–11, and in the presence of 2.5% NaCl for the investigation of drought, pH, and salt tolerances, respectively. Additionally, all strains were able to tolerate commercial insecticides (propargite and methomyl) and fungicides (captan) at the recommended dosages for field applications. Only, Amycolatopsis sp. GLM-2 showed tolerance to benomyl at the recommended dose. All the obtained actinobacteria were characterized as plant growth promoting strains by improving the growth of chili plants (Capsicum flutescens L.). Moreover, the co-inoculation treatment of the obtained actinobacteria and AMF (Claroideoglomus etunicatum) spores could significantly increase plant growth, contribute to the chlorophyll index, and enhance fruit production in chili plants. Additionally, the highest value of AMF spore production and the greatest percentage of root colonization were observed in the treatment that had been co-inoculated with Streptomyces sp. GETU-1.

Walnuts are an economically important forest tree used for timber and nut production, and the nut of fruits is rich in various nutrients, becoming one of the four important nuts in the world. Walnuts have deep roots, which can be colonized by either ectomycorrhizal fungi or arbuscular mycorrhizal fungi in the soil. These mycorrhizal fungi form beneficial symbioses in roots of walnut. A large number of ectomycorrhizal fungi have been identified, whilst Boletus edulis, Calvatia uiacina, and Cantharelles cibarius isolated from walnut orchards stimulated plant growth and gave the capacity of stress tolerance in walnut. Moreover, Carya illinoensis is a very good host plant for commercial production of truffles, especially Tuber indicum. In addition, ectomycorrhizal fungi accelerate plant growth and enhance potential stress tolerance of walnuts. Inoculation with arbuscular mycorrhizal fungi also showed the improvement of plant growth and nutrient acquisition of walnut, the enhancement of drought tolerance in walnut, nutrient redistribution under walnut interplanting patterns, and the delivery of juglone by mycorrhizal hyphae. A culturable in vitro arbuscular mycorrhizal like fungus Piriformospora indica also enhanced salt tolerance of walnut plants. In this mini-review, the physiological roles of mycorrhizal fungi, including arbuscular mycorrhizal fungi, ectomycorrhizal fungi and arbuscular mycorrhizal like fungus (P. indica) on walnut plants are summarized, and future outlooks in the field are proposed.

Effects of inoculation with arbuscular endomycorrhizal fungi (Acaulospora scrobiculata and Glomus mosseae) on acclimatization and growth of chrysanthemum (Dendrathema glandiflora Tzevelev) plants, propagated in vitro, under different conditions of fertilization (0, 20, and 40 mg·L-1 of NPK) were studied. Mycorrhizal colonization did not influence surviving percentage of chrysanthemum plantlets during the acclimatization stage; however, we could colonize the developing roots and reduce the amount of inoculum needed and beneficial effects on plant growth were obtained during early stages of colonizing. Plant growth in greenhouse was regulated by synergism between the effect of endomycorrhizal fungus type and soil fertilization with N, P, and K. Effects of A. scrobiculata were observed as an increasein number of leaves, leaf area, stem diameter, root volume and fresh and dry weight of leaves, stem and root. The G. mosseae fungus improved N, P, Mg, and Zn content in leaves; P, K, Ca, Mg, and Zinc in stem and Ca content in root. On the other hand, A. scrobiculata only increased N content in leaves, stem and roots; P content in leaves and roots, and Ca content in stem. Percentage of mycorrhizal colonization on roots was affected by adding N, P, and K to soil. The highest values were obtained with fertilization doses of 20 mg·L-1. The number of spores of mycorrhizal fungi was increased by adding fertilizer to soil (40 mg·L-1 of NPK).

AbstractPlant hormones play important roles in controlling how plants grow and develop. While metabolism provides the energy needed for plant survival, hormones regulate the pace of plant growth. Strigolactones (SLs) were recently defined as new phytohormones that regulate plant metabolism and, in turn, plant growth and development. This group of phytohormones is derived from carotenoids and has been implicated in a wide range of physiological functions including regulation of plant architecture (inhibition of bud outgrowth and shoot branching), photomorphogenesis, seed germination, nodulation, and physiological reactions to abiotic factors. SLs also induce hyphal branching in germinating spores of arbuscular mycorrhizal fungi (AMF), a process that is important for initiating the connection between host plant roots and AMF. This review outlines the physiological roles of SLs and discusses the significance of interactions between SLs and other phytohormones to plant metabolic responses.

This study aimed to characterize the halotolerant capability, in vitro, of selected actinomycetes strains and to evaluate their competence in promoting halo stress tolerance in durum wheat in a greenhouse experiment. Fourteen isolates were tested for phosphate solubilization, indole acetic acid, hydrocyanic acid, and ammonia production under different salt concentrations (i.e., 0, 0.25, 0.5, 0.75, 1, 1.25, and 1.5 M NaCl). The presence of 1-aminocyclopropane-1-carboxylate deaminase activity was also investigated. Salinity tolerance was evaluated in durum wheat through plant growth and development parameters: shoot and root length, dry and ash-free dry weight, and the total chlorophyll content, as well as proline accumulation. In vitro assays have shown that the strains can solubilize inorganic phosphate and produce indole acetic acid, hydrocyanic acid, and ammonia under different salt concentrations. Most of the strains (86%) had 1-aminocyclopropane-1-carboxylate deaminase activity, with significant amounts of α-ketobutyric acid. In the greenhouse experiment, inoculation with actinomycetes strains improved the morpho-biochemical parameters of durum wheat plants, which also recorded significantly higher content of chlorophylls and proline than those uninoculated, both under normal and stressed conditions. Our results suggest that inoculation of halotolerant actinomycetes can mitigate the negative effects of salt stress and allow normal growth and development of durum wheat plants.

Coffee is an important, high-value crop because its roasted beans are used to produce popular beverages that are consumed worldwide. Coffee plantations exist in over 70 countries and constitute the main economic activity of approximately 125 million people. Currently, there is global concern regarding the excessive use of agrochemicals and pesticides in agriculture, including coffee crops. This situation has motivated researchers, administrators, and farmers to seek ecologically friendly alternatives to decrease the use of synthetic fertilizers and pesticides. In the last decades, multiple studies of the rhizosphere, at the chemical, physical and biological levels, have improved our understanding of the importance of beneficial microorganisms to plant health and growth. This review aims to summarize the state of the use of plant growth-promoting microorganisms (PGPM) in coffee production, where the most extensively studied microorganisms are beneficial plant growth-promoting rhizobacteria (PGPR) and arbuscular mycorrhizal fungi (AMF). This review also contains information on PGPM, in regard to plantations at different latitudes, isolation techniques, mass multiplication, formulation methods, and the application of PGPM in nurseries, monoculture, and coffee agroforestry systems. Finally, this review focuses on relevant research performed during the last decade that can help us improve sustainable coffee production.
