Abstract The antioxidant capacity is the combined free radical scavenging effect of all antioxidant compounds found in the studied system. There is a growing need for accurate, numerical determination of this capacity (for easier comparison), so there are many analytical procedures, methods, and measurement systems available to researchers. Neither one is able to model the totality of real, naturally occurring reactions; therefore, conclusions about the antioxidant power of the studied sample can be drawn only after using several methods. In this work, the total phenolic content (TPC) of blackcurrant leaves and buds was determined, and the antioxidant capacity was tested using the DPPH and FRAP assays. 80% methanol was the most effective in the extraction of phenolics followed by 80% ethanol, while for the antioxidant capacity the acetone (50%)/water/acetic acid (2%) mixture proved to be the best. Significant differences were observed between cultivars and sampling dates, but the pattern of variation during the harvest period was similar for all cultivars.

The leaves from Camellia sinensis are used from ancient times for preparation of tea but also as raw material for different extracts which are used in food industry as well as in pharmaceutical or cosmetic products. Due to the increasing interest in tea health benefits, the aim of the present study was to characterize several brands of green and black tea found on local market, regarding their content in total phenolic compounds, flavonoids and antioxidant capacity. Total phenolics and flavonoids were determined spectrophotometrically using a modified Folin-Ciocalteu method, respectively a chromogenic system of NaNO2–Al(NO3)3–NaOH based method. The antioxidant capacity of each tea sample was assessed through the evaluation of free radical scavenging effect on 2,2-diphenyl-1-picrylhydrazyl. The results obtained for the green and black tea samples varied widely, depending on the tea variety. The antioxidant capacity of the analyzed teas ranged between 12.10 and 40.03%RSA, while the total phenolic content was within 2090 and 6080 mg GA/ 100g. The concentrantion in flavonoids was between 9.04 and 15.34 g/100g of tea.

Spent Coffee Ground (SCG) comprises a range of functional components with high antioxidant potential and health benefits. Although SCG has many advantages, antioxidant values obtained using enzymes is still scarce. Several studies reported that the use of enzyme-assisted extraction techniques could improve the detection of antioxidant compounds in samples of by-products. In this study, SCG was extracted using enzymeassisted techniques for different periods (0, 20, 40, 60, 80 and 100 mins). The total antioxidant capacity was measured using Peroxidase, Catalase, Ferric reducing antioxidant power (FRAP), Superoxide radical scavenging assay (SRSA) and 2,2-diphenyl-2- picrylhydrazyl (DPPH) analysis. 60 mins extraction was found to exhibit antioxidant activity in DPPH, SRSA, Catalase and Peroxidase of 84.18±7.01%, 32.83±6.75%, 76.65±5.52% and 75.01±7.67%, respectively. Besides that, the extraction also gave a high amount of content with 267.17±26.69 mg TAE/mL and 58.30±2.36 mg QE/g in total phenolic content and total flavonoid content respectively. Flavonoids such as quercetin, kaempferol, rutin, gallic acid, catechin, epigallocatechin, p-Coumaric acid and myricetin were found to be present in SCG extract. A high amount of catechin was found in all periods of extraction, whereby 60 mins had the highest concentration of 1741.65 mg/L, while 40 mins had the lowest concentration which was 389.85 mg/L. In conclusion, 60 mins extraction using a pectinase-assisted method was the best extraction period. Additionally, the individual flavonoid catechin in SCG showed that it has potential as an antioxidant. Finally, there is a strong correlation between antioxidative activity and both phenolic and flavonoid content

This study was aimed to evaluate the cytotoxicity of methanolic extract (ME) and aqueous extract (AE) of the rhizomes of Bergenia purpurascens through the Brine-shrimp bioassay, which revealed that ME possessed cytotoxicity with LC50 = 181.97 ìg/ml value. The plant extracts were also evaluated in vitro for antioxidant activity using different redox [Folin-Ciocalteu (FC) and Ferric Reducing Antioxidant Power (FRAP)] and radical scavenging [(2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2’-azinobis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS)] assays. Both ME and AE possessed antioxidant activity and ME displayed comparatively a greater potential. The total phenolic contents in ME and AE was 1.27 and 0.06 g gallic acid equivalent (GAE)/100 g dry mass (DM), respectively. The FRAP values determined for ME and AE were 15.82 and 2.58 ìmol Fe(II)/100 g DM, respectively. The DPPH radical scavenging effect (IC50) was for ME 19.86 ìg/ml and 295.91 ìg/ml for AE. At 100 ìg/ml concentration, up to 94% inhibition of ABTS+ was observed in 6 min incubation. Many plants with high antioxidant activity are great of significant in the pharmaceutical and cosmetic industries. Oxidative stress plays a major part in the development of many diseases including cancer, which is known imbalance free radicals and antioxidants. Herein, new natural antioxidant compounds have great interest in the scientific research. The genus Silene is a major group in the Caryophyllaceae family. In Turkey, Silene species have been used for several medicinal purposes such as skin softening, asthma, bronchitis. In our study, the antioxidant capacity of three Silene species (S. conoidea, S. dichotoma and S. italica) were evaluated by different in vitro assays, including free radical scavenging, reducing power, metal chelating, and phosphomolybdenum. In addition, total phenolic and flavonoid contents were analyzed spectrophotometrically. The water extracts contained higher total phenolic content than ethyl acetate extracts. All extracts showed antioxidant capacity. This data indicated that Silene species could potentially be used as antioxidant sources in pharmaceutical and cosmetic areas.

Background: Consumption of traditional herbal beverages has been generally increased in the last decades, Terebinth coffee, known as ‘‘menengic coffee’’ in Turkish, is one of the most consumed herbal coffees in Turkey, turpentine tree is one of the components of the Mediterranean bush, particularly in Algeria, known as Betoum el Kiffan is largely used as food and in traditional medicine. Aims: In this study, Total phenol, flavonoid content, and antioxidant activity of three extracts of Pistacia trebinthus fruit growing in Algeria was measured using radical scavenging activity tests and metal-related tests including, ferric-reducing antioxidant power (FRAP). The chemical composition profile of the fruits and the coffee brands was identified by thin-layer chromatography, the effects of roasting method of this fruit was rivaled also. Materials and Methods: The total phenolic content of the extracts was determined using the Folin-Ciocalteu method. All extracts of the terebinth fruits and coffee brands displayed a high DPPH scavenging effect. Results: The results of the ferric-reducing antioxidant power show that the reduction capacity is proportional to the increase in the concentration of the samples. All the extracts of the plant exhibit antioxidant activities lower than those of the reference product besides the infusion extract of the P. terebinthus roasted coffee, which is the most active with an optical density of 1.68 nm at a concentration of 400 μg/mL. The chromatography results show that the various extracts of Pistacia terebinthus fruit carry a large number of polyphenols, in particular the carboxylic acids phenols. Conclusions: The plant can be considered as a coffee substitute and opens up promising avenues for the food and pharmaceutical industry in Algeria. Green leafy vegetables (GLVs) are abundant in bioactive compounds and constitute a crucial part of a balanced diet. Sri Lankan green leafy vegetables which are edible and available for consumption have not been thoroughly investigated, whilst their consumption can deflate the risk of arising several degenerative diseases, such as cancer and cardiovascular diseases. The present study was aimed at evaluating the antioxidant capacities of the leafy vegetables, Gymnema lactiferum (Kurignan), Wattakaka volubilis (Aguna), and Argyreia populifolia (Girithilla), with different thermal treatments (70°C, 120°C, and 170°C) which are used in domestic cooking processes. Heat treatments significantly affect the antioxidant capacity and polyphenolic content of most vegetables analyzed, either increasing or decreasing the concentration of these compounds. In the presence of thermal treatments, Gymnema lactiferum (14.52-20.28 mg GAE/g DW) and Wattakaka volubilis (19.75-27.13 mg GAE/g DW) showed a significantly higher ( p < 0.05 ) total phenolic content. The temperature treatment did not alter the ABTS radical scavenging activity in Gymnema lactiferum. In contrast, an increment of ferric reducing antioxidant power (41.26-54.85 mg TE/g DW) and DPPH radical scavenging activity (0.11-0.26 mg TE/g DW) was observed. Wattakaka volubilis treated at 170°C appeared to have a significantly higher amount ( 104.93 ± 1.41   mg TE/g DW) of ferric reducing antioxidant power compared to its raw form. All cooking processes with their corresponding thermal treatments caused a significantly lower ( p < 0 ) amount of antioxidant capacity in Argyreia populifolia.

This study evaluated the biological activity (antioxidant assay) of Polygonum minus extracted using Supercritical Fluid Extraction (SFE) added with different types of co-solvents. The seven co-solvents employed were water, methanol, ethanol, 50% methanol, 50% ethanol, 70% methanol and 70% ethanol for selection of the best co-solvent prior to optimization of SFE. 70% methanol produced the highest total yield of extract (33.1%) compared to other co-solvents. The antioxidant capacity was then evaluated using four different assays: the total phenolic content (TP), the total flavonoid content (TF), the ferric reducing/antioxidant power (FRAP) and the free radical-scavenging capacity of 2,2-diphenyl-1-picrylhydrazyl (DPPH). The highest TP and TF were from 70% methanol extract (11.2 ± 0.15 mg GAE/g sample (mg GAE/g) and 11.9 ± 0.03 mg CAE/g sample (mg CEQ/g) respectively). 70% metanol extract also showed the highest FRAP value (346.7 ± 0.66 µmol Fe (II)/g sample) and the highest percentage of DPPH radical inhibition was also shown by 70% methanol extract (88.7 ± 0.40%). There was a positive correlation between the antioxidant capacity (FRAP and DPPH) with those of TP and TF contents. Therefore, the best co-solvent chosen for further optimization of SFE is 70% methanol.

Fruit peels are increasingly being used as functional foods nowadays. Peelings of twelve varieties of Persea americana fruits consumed in Cameroon were investigated for their phenolic compounds (polyphenols and flavonoids) using three solvents systems, water, ethanol: water (50 : 50 v / v ), and ethanol, and antioxidant activity using total antioxidant capacity (TAC), ferric reducing antioxidant power (FRAP), and 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging methods. Total polyphenol, flavonoids, and antioxidant potential of the peels significantly varied with P. americana variety and also with the extraction solvents in the order ethanol > ethanol: water > water. Total phenolic content varied from 2407 (Fuerte florid) to 673 (Semil) mg GAE/g DM, respectively, while flavonoids varied from 986 to 119 mg QE/g DM for Fuerte florid and Hickson varieties, respectively. TAC, respectively, varied between 132.87 and 126.85 mg AAE/g DM with Hass and Semil varieties, respectively. The highest DPPH scavenging capacity was recorded for the ethanolic extract with Lula (86.33%) and the least for the aqueous extract with the Semil (30.11%) variety. With FRAP, the highest capacity was obtained with hydroethanolic extract of Fuerte florid (0.43 mg AAE/g DM) and the least for aqueous extract with the Semil (0.269 mg AAE/g DM) variety. In conclusion, varieties of avocado peels are a good source of antioxidants. Solvent extraction significantly affected the concentration of bioactive compounds but not the potency of the antioxidants. A weakly positive correlation but not significant between the quantity of polyphenol, flavonoid, and antioxidant capacity of avocado peelings was obtained in this study.

Objective: To develop vacuum dried (VD) Ambarella fruit powder and Ambarella fruit incorporated soup mix from large and miniature Ambarella and investigation of antioxidants and anti-diabetic properties. Methods: Ethanolic extracts of two Ambarella varieties were analyzed for antioxidant potential in terms of total phenolic content (TPC), total flavonoid content (TFC), ferrous reducing antioxidant power (FRAP), radical scavenging activities of DPPH and ABTS and α-amylase inhibitory activity. Results: Fresh dwarf Ambarella showed a significantly (p<0.05) high TPC (3.35±0.10 mgGAE/g) while fresh large Ambarella showed a significantly (p<0.05) high FRAP (0.71±0.13 mgTE/g) and DPPH (3.57±0.31 mg TE/g). In comparison of ethanolic extracts of VD Ambarella powders, a significantly (p<0.05) higher antioxidant potential in terms of FRAP (4.19±0.06 mgTE/g) exhibited in dwarf variety while significantly (p<0.05) higher ABTS (4.03±0.27 mgTE/g) and DPPH (3.00±0.49mgTE/g) exhibited in large Ambarella. Alpha amylase inhibition activities of fresh fruits of large and dwarf were 46.30±4.07% and 49.55±3.18% where as in VD powders were 27.59±5.03% and 15.58±5.86% respectively. An instant soup mixture was developed incorporating 20% of VD powder from large variety due its abundance. The antioxidant potential of ethanolic extract of the soup mixture in terms of TPC, TFC, ABTS, and FRAP were 0.55±0.00 mgGAE/g, 0.04±0.00 mgQE/g, 1.65±0.06 mgTE/g, and 0.04±0.15 mgTE/g. Alpha amylase inhibition activity of soup mixture was 39.49±0.29%. Conclusion: The both types of fresh fruits exhibited higher antioxidant potential except FRAP and higher anti-amylase inhibition than VD Ambarella powder.

Hungarian sour cherries (SC) are excellent source of anthocyanin (concentrations (100–300 mg in 100 g fresh fruit) and melatonin (0.15 mg in 100 g fresh fruit), but other flavonoid derivatives also can be isolated by aqueous alcoholic extraction. We have developed a new process for extracting non-extractable procyanidines bound to the membrane, proteins, and fibers. These compounds were seperated with UHPLC-MS methods, and the structure of individual components were identified on the basis of their mass fragmentation spectra. The antioxidant capacity of soluble and non-soluble antioxidants were measured with ferric reducing antioxidant power (FRAP), 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity (DPPH), trolox equivalent antioxidant capacity (TEAC) assays, and compared to the new measurement methods of water-soluble antioxidant capacity (ACW), lipid-soluble antioxidant capacity (ACL). Furthermore, total phenolic content (TPC) and total procyanidin content (PAC) were determinated. As a result of our investigation, we found that the solvent combination, where in the first step is water–ethanol (1:1), then 100% ethanol were suitable for the extraction of the extractable antioxidants. However, the chemiluminescence method that is based on the elimination of the superoxide radical is more accurate than other colorimetric methods which measure antioxidant capacity.
