Chemical composition and antimicrobial activity of Erodium species: E. ciconium L., E. cicutarium L., and E. absinthoides Willd. (Geraniaceae)

AbstractThe present study gives results of chemical composition analyses and antimicrobial activity testing of three Erodium species: E. ciconium L., E. cicutarium L., and E. absinthoides Willd. Essential oils were obtained by hydro-distillation from air-dried entire plants and analyzed by GC and GC-MS. A total of 209 different compounds were identified: 162 for E. cicutarium, 107 for E. ciconium, and 79 for E. absinthoides. Antimicrobial activity (broth microdilution method) of the oils was screened against a panel of Gram positive and Gram negative bacteria and a number of fungi. Moderate susceptibility of all tested strains was observed. Determined MIC values were 0.156–5 mg mL−1 (bacterial strains) and 0.039–0.325 mg mL−1 (fungal strains). Major component of the most active oil, palmitic acid, was also tested for activity together with stearic and myristic acids.

The subject of the study was the investigation of the chemical composition and antimicrobial activity of the essential oils (EOs) isolated from Calamintha sylvatica, C. vardarensis, C. nepeta and C. glandulosa, as well as their antibacterial activity in combination with antibiotics. The quantitative and qualitative analysis of EOs was performed using the GC/FID and GC/MS methods. The antimicrobial activity of EOs against six standard bacterial strains and one strain of yeast was tested using the broth microdilution method, while the antimicrobial activity of a combination of essential oils and gentamicin/ciprofloxacin was tested by the checkerboard method. The dominant components (> 10%) of the essential oils were: cis-piperitone epoxide and menthone ( C. sylvatica), pulegone and menthone ( C. vardarensis), pulegone and piperitenone ( C. nepeta), pulegone, piperitenone, menthone and piperitone ( C. glandulosa). EOs did not exhibit significant antimicrobial activity except the essential oil of C. vardarensis which was selectively active against Staphylococcus aureus (MIC - 21.25 μg/mL). The overall effect of essential oil-antibiotic combinations varied from synergistic (FICI ≤ 0.5) to antagonistic (FICI ≥ 2) depending on the bacterial strain tested.

AbstractFoot odour (bromodosis) is an embarrassing and perplexing condition mostly caused by bacteria of the Brevibacterium species. Essential oils are a credible option as an affordable treatment of odour and contribute towards antimicrobial efficacy. Therefore, this study sets out to investigate the antimicrobial activity of essential oil combinations against odour-causing bacteria. The broth microdilution method was used to investigate the antimicrobial activity of 119 essential oil combinations, and the fractional inhibitory index was calculated to determine the interactive profile. Combinations that resulted in synergy in 1 : 1 ratios were further evaluated in different concentrations, and isobolograms were plotted to determine the influence of the ratio on overall activity. Numerous combinations could be identified as having synergistic interactions against the Brevibacterium spp. and no antagonism was observed. The combination of Juniperus virginiana (juniper) and Styrax benzoin (benzoin) demonstrated synergy against all three Brevibacterium spp. tested and J. virginiana was the essential oil responsible for the majority of the synergistic interactions. The results reported here confirm the promising potential of the majority of these oils and selected combinations in treating and controlling bromodosis.

The principal objective of this study was to determine the anti-staphylococcal potential of ethanol extracts of propolis (EEPs). A total of 20 samples of propolis collected from apiaries located in different regions of Poland were used in the study. The two-fold broth microdilution method revealed some important differences in the antimicrobial activity of investigated EEPs. Up to the concentration of 4096 µg/mL no activity was observed against Gram-negative bacteria (E. coli and P. aeruginosa). Staphylococci exhibited much higher susceptibility. The highest efficiency observed for EEP12 and EEP20 (MIC values ranged between 32 and 256 µg/mL). However, the achievement of bactericidal effect usually required higher concentrations. In the case of clinical isolates of S. aureus MBC values for EEP12 and EEP20 ranged from 512 to 1024 µg/mL. The HPLC analysis revealed that these two products contained a higher concentration of flavonoids (flavonols, flavones, and flavanones) compared to other investigated EEPs. In checkerboard test, a synergistic anti-staphylococcal effect was observed for the action of EEP20 in combination with amikacin, kanamycin, gentamycin, tetracycline, and fusidic acid (all these antibiotics inhibit protein synthesis). Moreover, the investigated EEPs effectively eradicated staphylococcal biofilm. The obtained results clearly confirm the high anti-staphylococcal potential of propolis harvested in Polish apiaries.

Essential oils from Calamintha umbrosa and Nepeta species viz. N. leucophylla; N. hindostana; N. ciliaris and N. clarkei (family Lamiaceae), was tested against six bacterial strains. To evaluate the correlation between the antimicrobial activity and the essential oils, PCA and HCA analysis was done. PCA and HCA analysis of the antibacterial activity revealed that essential oils of Nepeta species had a strong and broad spectrum antibacterial effect against bacterial strains of P. aeruginosa and S. scandidus. The N. leucophylla oil showed higher activity against Gram-negative bacteria P. aeruginosa (10.5 mm, MIC 10 µL/mL) and K. pneumonia (9.1 mm, MIC 45 µL/mL) among all Nepeta oils which may be due to presence of active antimicrobial iridoids compounds.

Abstract Background Colistin resistance is considered a serious problem due to a lack of alternative antibiotics. The Rapid ResaPolymyxin Acinetobacter/Pseudomonas NP test is a resazurin reduction-based technique that relies on the visual detection of bacterial growth in the presence of a defined concentration of colistin. The aim of this study was to evaluate the performance of the Rapid ResaPolymyxin Acinetobacter/Pseudomonas NP test in the detection of colistin susceptibility in common clinical Gram-negative bacteria. Results A total of 253 clinical isolates from a teaching hospital, including Acinetobacter baumanii (n = 58, 8 colistin-resistant), Pseudomonas aeruginosa (n = 61, 11 colistin-resistant), Klebsiella pneumoniae (n = 70, 20 colistin-resistant) and Escherichia coli (n = 64, 14 colistin-resistant) were tested in this study. The sensitivity and specificity of the Rapid ResaPolymyxin Acinetobacter/Pseudomonas NP test compared to Broth microdilution method was 100 and 99%, respectively. Conclusions Our results suggest that Rapid ResaPolymyxin Acinetobacter/Pseudomonas NP test could be used as an accurate detection method for colistin resistance.

AbstractCrude methanol extracts from callus cultures of Nigella arvensis, N. damascena, N. hispanica, N. integrifolia, and N. sativa were investigated for their anti-microbial activity. Growth inhibition was determined in Gram-positive and Gram-negative bacterial strains as well as in yeast by using a broth-microdilution method. The results showed that the extracts of all calli tested exhibited significant anti-microbial activity, especially against Bacillus cereus, Staphylococcus aureus and Staphylococcus epidermidis. Compared with other Nigella species, a callus culture of N. hispanica was the most effective against the microorganisms used in this study.

The antibacterial effects of essential oils from Serbian cultivated plants, Thymus vulgaris L. (Lamiace) and Lavandula angustifolia L. (Lamiace) on different bacteria were investigated, with an emphasis on an antibacterial activity-chemical composition relationship. Essential oil was obtained from airdried aerial parts of the plants by hydrodistillation for 3 h using a Clevenger-type apparatus. The essential oil analyses were performed simultaneously by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS) systems. The main constituents of thyme oil were thymol (59.95%) and p-cymene (18.34%). Linalyl acetate (38.23%) and linalool (35.01%) were main compounds in lavender oil. The antibacterial activity of the essential oils samples was tested towards 5 different bacteria: laboratory control strain obtained from the American Type Culture Collection and clinical isolates from different pathogenic media. Gram negative bacteria were represented by Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 43895 and Salmonella enteretidis ATCC 9027 while researched Gram positive strains were Bacillus cereus ATCC 8739 and Staphylococcus aureus ATCC 25923. A broth microdilution method was used to determine the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). Essential oils from thyme have been found to have antimicrobial activity against all microorganisms tested, with a range of MIC values from 0.025 to 0.10 l/ml and MBC values from 0.05 to 0.78 l/ml. Lavender oils demonstrated MIC values from 0.025 to 0.20 l/ml and MBC values from 0.05 and 0.78 l/ml. Reference antibiotic tetracycline was active in concentrations between 0.025 and 0.05 l/ml. The Gram-positive bacteria were more sensitive to the essential oil of thyme, while Gram-negative bacteria were more sensitive to the essential oil of lavender. Essential oils from thyme and lavender may be used at low concentrations for prevention and treatment of infective diseases in animals and humans caused by pathogenic bacterial species.

Antimicrobial resistance (AMR) has become one of the more serious threats to the global health. The emergence of bacteria resistant to antimicrobial substances decreases the potencies of current antibiotics. Consequently, there is an urgent and growing need for the developing of new classes of antibiotics. Three prepared novel iron complexes have a broad-spectrum antimicrobial activity with minimum bactericidal concentration (MBC) values ranging from 3.5 to 10 mM and 3.5 to 40 mM against Gram-positive and Gram-negative bacteria with antimicrobial resistance phenotype, respectively. Time-kill studies and quantification of the extracellular DNA confirmed the bacteriolytic mode of action of the iron-halide compounds. Additionally, the novel complexes showed significant antibiofilm activity against the tested pathogenic bacterial strains at concentrations lower than the MBC. The cytotoxic effect of the complexes on different mammalian cell lines show sub-cytotoxic values at concentrations lower than the minimum bactericidal concentrations.

Novel series of 4,6-disubstituted-1,3,5-triazines containing hydrazone derivatives were synthesized employing ultrasonic irradiation and conventional heating. The ultrasonication gave the target products in higher yields and purity in shorter reaction time compared with the conventional method. IR, NMR (H 1 and C 13), elemental analysis, and LC-MS confirmed the structures of the new products. The antimicrobial and antifungal activities were evaluated for all the prepared compounds against some selected Gram-positive and Gram-negative bacterial strains. The results showed that only two compounds 7i (pyridine derivative) and 7k (4-chlorobenzaldehyde derivative) displayed biological activity against some Gram-positive and Gram-negative bacteria, while the rest of the tested compounds did not display any antifungal activity.
