Partial quantification of pigments extracted from the zooxanthellate octocoral Sinularia flexibilis at varying irradiances

AbstractChlorophyll-a (chl-a) and carotenoid pigments of the zooxanthellate octocoral Sinularia flexibilis were analyzed using high performance liquid chromatography following exposure to three light intensities for over 30 days. From the coral fragments located at different light intensities, a total carotenoid of >41 μg g−1 dry weight, including peridinin, xanthophylls (likely diadinoxanthin + diatoxanthin), and chl-a as the most abundant pigments, with minor contents of astaxantin and β-carotene were detected. The whole content of chl-a weighed 5 μg g−1 dry weight in all coral colonies. Chl-a and carotenoids contributed 11.2% and 88.2%, respectively, to all pigments detected, and together accounted for 99.4% of the total pigments present. The highest contents of carotenoids and chl-a was observed in the coral grafts placed in an irradiance of 100 μmol quanta m−2 s−1; they showed lower ratios of total carotenoids: chl-a compared to those exposed to 400 μmol quanta m−2 s−1 after >30 days of incubation. The ratios of peridinin and xanthophylls with respect to chl-a from the colonies at 400 μmol quanta m−2 s−1 were approximately double those observed at irradiances of 100 and 200 μmol quanta m−2 s−1. Partial quantification of pigments in this study showed that the carotenoids of S. flexibilis showed a decrease at irradiances above 100 μmol quanta m−2 s−1, with the exception of an increase in β-carotene at 200 μmol quanta m−2 s−1.

The β-carotene and total carotenoid content of either fresh or dried tissue of fruits of a total of 57 cultivars of six Capsicum species were analyzed using high performance liquid chromatography (HPLC). β-Carotene levels in ripe fruit varied from 0 to 166 μg·g-1 fresh weight, and carotenoid levels were from 1 to 896 μg·g-1 in ripe fruit in 1996. The range of values for β-carotene was similar in 1997, but that for total carotenoids was wider (4 to 1173 μg·g-1 fresh weight). Fresh fruit of the cultivars Greenleaf Tabasco, Pulla, Guajillo, NuMex Conquistador, Ring-O-Fire, and Thai Dragon contained greater amounts of β-carotene per 100 g fresh weight than the recommended dietary allowance (RDA) for vitamin A for the average adult. For dried Capsicum entries, New Mexican, aji, pasilla, and ancho types had the highest levels of β-carotene. In 1996, β-carotene levels among the dried Capsicum germplasm ranged from 2 to 739 μg·g-1 dry weight, and carotenoid levels from 111 to 6226 μg·g-1. Values were higher in 1997, ranging from 24 to 1198 μg·g-1 dry weight for β-carotene and from 187 to 10,121 μg·g-1 for total carotenoids. A pasilla type (C. annuum L.) had the highest total carotenoid content among the dried entries in both years.

Cucurbita species ( Cucurbita moschata and Cucurbita maxima) are rich sources of nutrients, especially carotenoids and carbohydrates. Metabolites in pulps of C. moschata and C. maxima lines were examined by high-performance liquid chromatography and gas chromatography–mass spectrometry. Our results revealed that glucose and sucrose were the dominant sugars in C. maxima and C. moschata, respectively. A correlation was found between Brix percentage and sucrose levels as compared with glucose and fructose. A greater amount of myo-inositol had accumulated in C. moschata lines compared with C. maxima. Conversely, total carotenoids and antioxidant activity were found to be higher in C. maxima lines than in C. moschata. A strong correlation of glucose, fructose, and sucrose with β-carotene and violaxanthin revealed that it will be difficult to develop pumpkin cultivars with both high inositol and carotenoid levels. In conclusion, the composition of carbohydrates and carotenoids was more diverse in C. moschata lines than in C. maxima lines. Our results will contribute to a better understanding of metabolite changes in the fruits of these as well as other pumpkin species.

The aim of this research is to extract carotenoids from apricot fruits using a food solvent (ethanol) and a non-food solvent (propanol). In addition, it aims to study the effect of different extraction conditions, i.e., the extraction temperature (20-40-60)°C and the extraction times (4-8-12) hours, on the carotenoids yield in order to improve and develop extraction methods. The extracted carotenoids were determined using high-performance liquid chromatography (HPLC), and the studied carotenoids are α-carotene and β-carotene. It was found that β-carotene constitutes about 80% of the total carotenoids in apricots. The results indicated that the extraction yield using ethanol was low compared to the extraction yield using propanol. Propanol is the most suitable solvent for carotenoids extraction in comparison with ethanol. The temperature also had a significant effect on the extraction yield; at 20°C the extraction yield was very low, and 60°C was the optimum temperature for the extraction of carotene. The extraction yield significantly increased with time in the first hours of extraction, and there was no significant effect from increasing the extraction time for a period of 6-8 hours. Moreover, the pretreatment of fruits by freezing accelerated the extraction process and increased the extraction yield. The optimum conditions for extracting carotenoids were determined in the conditions of food extraction; the optimum conditions for extracting carotenoids are: extraction at 60°C for three hours with pretreatment by freezing as the extraction rate reached up to 6.36 mg/100 g using ethanol as a food solvent.

The study involved the determination of β-carotene and α-tocopherol content in selected fresh and dry vegetables (amaranthus, cowpea leaves, nightshade, slender leaf, pumpkin leaves and frying spider) using High Performance Liquid Chromatography (HPLC) procedure. Fresh blanched vegetables contained high levels of β-carotene; 4000-9700µg/100g and α-tocopherol levels; 3000-7360µg/100g (WW). The solar dried vegetables contained β- carotene levels ranging from 572 to 854µg g-1 dry weight (DW) and α-tocopherol levels ranging from 281 to 673µg g-1 (DW). Solar dried vegetables contained significantly lower (P<0.05) amounts of β-carotene and α-tocopherol which were moderately bioavailable when mixed in good proportion to meet Recommended Dietary Allowance (RDA) of vitamins A and E; which are 750µg retinol equivalent/day and 8mg/day respectively. The results will provide nutritional information on the indigenous vegetables grown in Butula in Busia County.

ABSTRACT The current study aims to evaluate the occurrence and concentration of carotenoids in the pulp and peel of 14 banana and one plantain cultivars in two ripening stages. We analyzed the occurrence and content of lutein, α-carotene, β-carotene, β-cryptoxanthin, and lycopene - by high performance liquid chromatography with UV-visible detection, the content of total carotenoids by spectrophotometry. In the unripe pulp, the ‘Ouro’ cultivar stood out with lutein concentration 13 times higher than that of the ‘Marmelo’. As for α-carotene and β-carotene, ‘Terrinha’ plantain stood out with mean concentration of 1195.30 and 1126.11 μg (100 g MF-1), respectively. Total carotenoids ranged from 159.66 to 2553.51 μg (100 g MF-1) in ‘Caipira’ and ‘Terrinha’, respectively. In the ripe pulp there was 36% increase in the lutein content in comparison to the unripe pulp and there was 7.3 and 8.5% reduction in α-carotene and β-carotene levels, respectively. The total carotenoid concentration in the ripe pulp was 17% higher than that found in the unripe pulp. The unripe peel showed lutein predominance, although ‘Terrinha’ stood out with higher α-carotene and β-carotene concentrations than those found in the other cultivars. Lutein concentration hardly changed due to fruit ripening; however, there was slight reduction in α-carotene and β-carotene concentrations.

The β-carotene and total carotenoid content of different Capsicum fruit types and species were analyzed using HPLC. This information is useful for breeding high carotenoid chiles (New Mexican type) for the food industry, and also provides nutritional data for the range of fruit types within the Capsicum genus. Fresh fruit from 25 accessions and dried fruit from 39 accessions were evaluated in 1996 and 1997. β-carotene levels varied from 0 to 16.6 mg/100 g fresh weight, and carotenoid levels were from 0.1 to 89.6 mg/100 g in red ripe fruit in 1996. The range of values for β-carotene was similar in 1997, but a wider range in total carotenoids (0.4 to 117.3 mg/100 g fresh weight) was observed. Fresh fruit (100 g) of the cultivars `Greenleaf Tabasco', `Pulla', `Guajillo', `NuMex Conquistador', `Ring-O-Fire', and `Thai Dragon' contained greater amounts of β-carotene than the RDA for vitamin A for the average adult. For dried Capsicum entries, New Mexican, aji, pasilla, ancho, and guajillo types had the highest levels of β-carotene. In 1996, β-carotene levels among the dried Capsicum germplasm ranged from 0 to 739.2 μg/g dry weight, and carotenoid levels were from 21.3 to 6,225.9 μg/g. Values were higher in 1997, and ranged from 23.7 to 1,198.1 μg/g dry weight for β-carotene and from 76.9 to 10,120.6 μg/g for total carotenoids. A pasilla type (C. annuum) had the highest total carotenoid content among the dried entries in both years.

Our research was aimed at assessing the effect of accumulation of carotenoids, polyphenols, vitamin C and ripening stage in the rosehip fruits of two species—Rosa canina, Rosa rugosa and two cultivar—Rosa rugosa ‘Rubra’ and Rosa rugosa ‘Alba’. The amounts of carotenoids, polyphenols and vitamin C were determined using the high-performance liquid chromatography (HPLC) method. The obtained results showed that the significantly highest amount (107.15 mg 100 g−1) of total carotenoid was determined in the fruits of Rosa canina at ripening Stage V. While results indicated that significant amount of total polyphenols were established at Stages I and II in the Rosa Rugosa ‘Alba’ and Rosa rugosa ‘Rubra’ cultivars (110.34 mg 100 g−1, 107.88 mg 100 g−1 and 103.20 mg 100 g−1 103.39 mg 100 g−1). At ripening Stage I, in the fruits of Rosa rugosa the greatest increases were established in the contents of vitamin C (3036.08 mg 100 g−1).

Broccoli is a source of bioactive compounds that provide an important nutritional value. The content of these compounds can vary depending on agronomic and environmental conditions, as well as on elicitation. In this study, three crop trials were carried out to evaluate the effects of the cultivation season, the application of different dosages of methyl-jasmonate (MeJA) on the overall quality and on the total content of bioactive compounds of ‘Parthenon’ broccoli cultivated under the field conditions of southeastern Spain. Color parameters, chlorophyll content, total phenolic compounds, total flavonoids and antioxidant activity were measured to evaluate the overall quality. Moreover, individual carotenoids, phenolic compounds and glucosinolates were evaluated by high performance liquid chromatography with diode array detection (HPLC-DAD) and high performance liquid chromatography equipped with diode array detector coupled to mass spectrometer using electro spray ionization (HPLC-DAD-ESI/MSn). The content of total carotenoids, phenolic compounds and glucosinolates were higher in autumn compared with spring, showing increases of 2.8-fold, 2-fold and 1.2-fold, respectively. Moreover, a double application of MeJA increased the contents of total carotenoids, phenolic compounds and glucosinolates by 22%, 32% and 39%, respectively, relative to the untreated samples. Considering our results, the controlled and timely application of 250 µM MeJA to the aerial parts of the plants four days before harvest, on two consecutive days, seems to be a valid agronomic strategy to improve the health-promoting capacity of Parthenon broccoli, without compromising its overall quality.
