ABSTRACT Th e aim of this study was to examine the phenolic and flavonoid contents and antioxidant activities of methanol and chloroform extracts of leaves and twigs of Daphne blagayana. Th e total phenolic content in the chloroform extract of plant twigs (90.26 ± 0.69 mg GA/g) was higher than that of the other extracts (from 76.56±0.89 to 77.45±0.43 mg GA/g). In the case of flavonoids, a greater value was also obtained for the chloroform extract of twigs (35.24±0.55 mg RU/g). Several diff erent methods were used to determine the antioxidant activity of the tested extracts, including total antioxidant capacity, metal chelating activity, hydroxyl radical scavenging activity and inhibitory activity against lipid peroxidation. Our results showed that although secondary metabolites of the plants may contribute significantly to their antioxidant activities, those antioxidant activities were not directly related to the phenolic and flavonoid amounts. Th e results of the present analysis demonstrated, for the first time, that Daphne blagayana leaves and twigs possess high phenolic and flavonoid contents, as well as potential antioxidant activity. Th is study suggests that Daphne blagayana twigs and leaves may potentially be used as an accessible source of natural antioxidants.

Antioxidant Activity of Milling Fractions of Selected CerealsThe aim of this study was to evaluate antioxidant potential of four milling fractions of selected cereals grew in the year 2009 and 2010. Free radical scavenging activity of samples was measured using DPPH assay and reducing power was determined using FRAP assay. Secondary, total phenolic and flavonoid content of cereal extracts was evaluated. We found that flour fractions (break flour and reduction flour) showed the lower proportion of the total antioxidant potential than bran fractions (fine bran and coarse bran), which was balanced in observed years. Extract from barley had the highest values of antioxidant activity and polyphenol content.

Antioxidant activities of wampee peel extracts using five different solvents (ethanol, hexane, ethyl acetate, butanol and water) were determined by using in-vitro antioxidant models including total antioxidant capability, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity, reducing power, and superoxide scavenging activity. Ethyl acetate fraction (EAF) exhibited the highest antioxidant activity compared to other fractions, even higher than synthetic antioxidant butylated hydroxyl toluene (BHT). In addition, the EAF exhibited strong anticancer activities against human gastric carcinoma (SGC-7901), human hepatocellular liver carcinoma (HepG-2) and human lung adenocarcinoma (A-549) cancer cell lines, higher than cisplatin, a conventional anticancer drug. The total phenolic content of wampee fraction was positively correlated with the antioxidant activity. This is the first report on the antioxidant and anticancer activities of the wampee peel extract. Thus, wampee peel can be used potentially as a readily accessible source of natural antioxidants and a possible pharmaceutical supplement.

The polyphenolic contents and the antioxidant activity of the skins and pulps of different grape cultivars were estimated using HPLC and DPPH antioxidant assay, respectively. The phenolics and flavonoids identified were quercetin, kaempferol, caffeic acid, p-coumaric acid, cinnamic acid, and (−)-epicatechin. The total phenolic contents were found to be the highest in the grape skin of Flouxa (>400 mg/100 g), followed by Campbell Early and Tamnara (>300 mg/100 g), and then by Red Globe and Ruby Seedless (>250 mg/100 g), and the total phenolic content was the lowest in Italia and Delaware (<60 mg/100 g). The antioxidant activities of the grape extracts varied from 12.5% (Ruby Seedless) to 60.2% (Hongiseul) for skins, whereas the antioxidant activities of the grape extracts varied from 35.4% (Campbell Early) to 84.5% (Hongiseul) for pulps. The grape pulps have stronger antioxidant activities than those of the grape skins. Our results suggest that the phenolic and flavonoid contents in extracts of grape skins and pulps showed statistically significant correlations with the free radical scavenging activity.

The present study examined the effect of Lactobacillus plantarum ATCC14917 fermentation on the chemical composition and antioxidant activity of apple juice. Apple juice was fermented and examined of its antioxidant activity using chemical models and cellular antioxidant assay. Furthermore, the chemical composition of fermented apple juice was characterized by LC-MS/MS. Lactobacillus plantarum ATCC14917 fermentation showed an increase in DPPH and ABTS radical scavenging activity as well as cellular antioxidant activity of apple juice. However, fermentation decreased the total phenolic and flavonoid content. Subsequent LC-MS/MS analysis of the phenolic profile indicated that the content of 5-O-caffeoylquinic acid (5-CQA), quercetin, and phloretin with strong antioxidant activity was increased significantly after fermentation. The modified phenolic composition may contribute to the increased antioxidant activity of fermented apple juice. Our findings showed that Lactobacillus plantarum ATCC14917 fermentation may be an efficient way to enhance the bioavailability of phenolic compounds and to protect cells from oxidative stress.

The aim of this study was to determine the procedure parameters, including drying, extracting and pH on the antioxidant activity and antioxidative compounds in Perilla frutescens leaves. The extraction of antioxidants from Perilla frutescens leaves is studied considering different drying and extracting procedures to investigate the selectivity of the process. The radical-scavenging activity (RSA) of 1, 1-diphenyl-2-picrylhydrazyl (DPPH) and the determination of the total phenolic content are applied to evaluate the antioxidant activity and antioxidative compounds of the Perilla frutescens leave extracts. The highest antioxidant activity is observed for the extract obtained by freeze drying, extracting by ultrasound with 80% methanol adjusted at pH 4.The antioxidant compound extraction yield was also the highest at these extraction conditions. The total phenol contents among the Perilla frutescens leave extracts produced by different drying procedures were similar, while the antioxidant activities were different This concluding that the amount of phenolic compounds extracted was similar but the type and probable structure of the phenolic changed during heat-drying procedure providing in this way. Our study showed that proper process technology could ultimately extract compounds potentially effective against diseases related to free radical oxidation.

Background Amaranthus hybridus L. is an annual, erect or less commonly ascending herb that is a member of the Amaranthaceae family. Polysaccharides extracted from traditional Chinese medicines may be effective substances with antioxidant activity. Methods In this study, we isolated crude polysaccharides from A. hybridus (AHP-M) using microwave-assisted extraction. Then, the AHP-M was purified by chromatography with DEAE-32 cellulose, and two fractions, AHP-M-1 and AHP-M-2, were obtained. The structural characteristics of AHP-M-1 and AHP-M-2 were investigated, and their antioxidant activities were analyzed in vitro. Results We found that the monosaccharide composition of AHP-M-1 was different from that of AHP-M-2. The molecular weights of AHP-M-1 and AHP-M-2 were 77.625 kDa and 93.325 kDa, respectively. The results showed that the antioxidant activity of AHP-M-2 was better than that of AHP-M-1. For AHP-M-2, the DPPH radical scavenging rate at a concentration of 2 mg/mL was 78.87%, the hydroxyl radical scavenging rate was 39.34%, the superoxide anion radical scavenging rate was 80.2%, and the reduction ability of Fe3+ was approximately 0.90. The total antioxidant capacity per milligram of AHP-M-2 was 6.42, which was higher than that of Vitamin C (Vc). Conclusion The in vitro test indicated that AHP-M-1 and AHP-M-2 have good antioxidant activity, demonstrating that A. hybridus L. polysaccharide has immense potential as a natural antioxidants.

Background: Consumption of traditional herbal beverages has been generally increased in the last decades, Terebinth coffee, known as ‘‘menengic coffee’’ in Turkish, is one of the most consumed herbal coffees in Turkey, turpentine tree is one of the components of the Mediterranean bush, particularly in Algeria, known as Betoum el Kiffan is largely used as food and in traditional medicine. Aims: In this study, Total phenol, flavonoid content, and antioxidant activity of three extracts of Pistacia trebinthus fruit growing in Algeria was measured using radical scavenging activity tests and metal-related tests including, ferric-reducing antioxidant power (FRAP). The chemical composition profile of the fruits and the coffee brands was identified by thin-layer chromatography, the effects of roasting method of this fruit was rivaled also. Materials and Methods: The total phenolic content of the extracts was determined using the Folin-Ciocalteu method. All extracts of the terebinth fruits and coffee brands displayed a high DPPH scavenging effect. Results: The results of the ferric-reducing antioxidant power show that the reduction capacity is proportional to the increase in the concentration of the samples. All the extracts of the plant exhibit antioxidant activities lower than those of the reference product besides the infusion extract of the P. terebinthus roasted coffee, which is the most active with an optical density of 1.68 nm at a concentration of 400 μg/mL. The chromatography results show that the various extracts of Pistacia terebinthus fruit carry a large number of polyphenols, in particular the carboxylic acids phenols. Conclusions: The plant can be considered as a coffee substitute and opens up promising avenues for the food and pharmaceutical industry in Algeria. This study was carried out to evaluate the antioxidant activity of Iraqi sumac seeds (Rhus coriaria. L) (Anacardiaceae). Total phenolic compounds and flavoniods were determined in three different sumac seed extracts (SSE) (aqueous,ethanolic and methanolic extract). For extraction Antioxidant activity of SSE were evaluated by various antioxidant assays, including total antioxidant capacity, reducing power,by using 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging, nitric oxide scavenging, Hydroxyl radical scavenging, and metal ion chelating activities. These various antioxidant activities were compared with ascorbic acid as a standard antioxidant.The results showed that the three(SSE), contained large amounts of phenolic and flavoniods compounds. The results also showed that the three( SSEs ) have antioxidant activities , the methanolic (SSEs) contain larger amounts of phenolic and flavoniod compounds and have a higher antioxidant activities than that of aqueous or ethanolic (SSE).These results indicated that Iraqi aqueous and alcoholic extracts of Iraqi sumac in especial methanolic) SSE( are good scavengers for reactive oxygen species(ROS) and are a potential source of natural antioxidant, that may be used in pharmaceutical or food industry .

Stephania glabra is a wild medicinal plant possessing multiple uses as anti-cancer, antimicrobial and antioxidant activities. Different extracts and fractions were prepared from S. glabra tubers and were analyzed for different activities. Evaluation of the total phenolic and flavonoid content revealed that chloroform extract of S. glabra contain maximum phenolic content and maximum flavonoid content was found in methanolic extract. Dry powder analysis of the tubers revealed the presence of saponins, alkaloids and proteins. Ethyl acetate extract of S. glabra tubers showed highest antioxidant potential as revealed by ABTS radical scavenging activity. Methanolic extract has the highest anti-cancer activity while chloroform extracts and fractions have highest antimicrobial activity. These activities can be attributed to the high amount of the phytoconstituents present in these extracts

Abstract Objectives Inhibition of carbohydrate absorption from intestine is an efficient way to prevent type 2 diabetes. Some edible plants have shown the potential as anti-diabetic agents by inhibiting carbohydrate-digestive enzymes in intestine. In terms of edible plant's health effect, bioactive compounds’ stability during the process of digestive enzymes has been commonly questioned. Therefore, the objectives of this study were to 1) examine inhibitory effect of garlic scape (GS), the green flower head of garlic on carbohydrate-degrading enzymes and 2) investigate antioxidant activity and total phenolic content (TPC) of GS treated with intestinal digestive enzymes. Methods GS extract (GSE) was prepared with 80% methanol (v/v) by homogenization, sonication, and filtration, and evaporation. α-glucosidase, maltase, glucoamylase, sucrase, α-amylase inhibitory activities were determined by measuring glucose hydrolyzed from nitrophenyl-glucopyranoside, maltose, starch, sucrose, and starch, respectively. To prepare gastrointestinal enzyme-digested GSE, GSE was digested with pepsin and a pancreatin-bile solution. GSE and gastrointestinal enzyme-digested GSE were assessed for TPC using a Folin-Ciocalteu reagent and for total antioxidant and radical scavenging activities by the following methods, phosphomolybdeum, 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), and 2,2-diphenyl-1-picrylhydrazyl (DPPH) methods, respectively. Results GSE (20 mg/ml) showed 30%, 50%, and 42% inhibition on α-glucosidase, maltase, and glucoamylase enzyme activities, respectively. GSE (50 mg/ml) showed 55.1% DPPH radical and 5782 mg vitamin C equivalent (VCE)/g ABTS radical scavenging activities. Total antioxidant activity of the GSE (1.5 mg/ml) exhibited 2.76 mg VCE/g. Consistent with its antioxidant property, the GSE showed a dose-dependent increase in TPC. Gastrointestinal enzymatic treatment resulted in approximately 46% reduction in DPPH and ABTS radical scavenging activity, total antioxidant activity, and TPC of the GSE. Conclusions Garlic scape has the potential as a natural anti-diabetic agent. Garlic scape keeps beneficial effects after the process of intestinal digestive system.
