Pterostilbene (PTE) (3-5 dimethoxy-4-hydroxy-trans-stilbenes) is an analogue of resveratrol. It is extracted and isolated from a natural source of the heartwood of Pterocarpus marsupium Roxb., red grape skin, and blueberries (Vaccinium spp.). Substantial evidence suggested that PTE displayed numerous preventive and therapeutic properties in many metabolic disorders such as diabetes and obesity. Metabolic diseases result in Insulin resistance (IR) which advances to impaired sensitivity to insulin-mediated glucose disposal. The prominent role of SIRT (silent information regulator proteins) is now getting emphasized in metabolic disorders. SIRT1 represses Uncoupling protein 2 (UCP2) expressions which are further responsible for improving synthesis of ATP from glucose. This results in improving glucose utilization and insulin secretion, thus preventing IR. SIRT1 also exhibits prominent role in facilitating fatty acid mobilization thereby inhibiting adiposity. Metabolic disorders are therefore the consequences of SIRT1 downregulation. Pterostilbene, being a SIRT1 activator, increases insulin sensitivity reduces adiposity, therefore can prove to be beneficial in diabetes as well as obesity. The review summarizes therapeutic effects portrayed by Pterostilbene via the SIRT1 pathway in metabolic diseases.

The present study focuses on heat-induced structural changes and the degradation kinetics of phytochemicals and antioxidant activity of red grape skin extract. The thermal degradation of anthocyanins, flavonoids, polyphenols, and antioxidant activity followed a first-order kinetic model, increasing with temperature due to the intensification of the degradation process. The activation energy (Ea) highlighted this phenomenon. Likewise, the kinetic and thermodynamic parameters certified the irreversible degradation of the bioactive compounds from the skin of the Băbească neagră grape variety. Both temperature and duration of heating had a significant impact on the content of bioactive compounds. In addition, the red grape skin extract inhibited certain enzymes such as α-amylase, α-glucosidase, lipase, and lipoxygenase, which are associated with metabolic syndrome and inflammation. Further knowledge on the possible inhibition mechanisms exerted by the major anthocyanins found in red grape skin extract on the metabolic syndrome-associated enzymes was gathered upon running molecular docking tests. Detailed analysis of the resulting molecular models revealed that malvidin 3-O-glucoside binds in the vicinity of the catalytic site of α-amylase and lipase, whereas no direct contact with catalytic amino acids was identified in the case of α-glucosidase and lipoxygenase.

O desperdício de alimentos é preocupante, pois pelo menos 1,4 bilhão de toneladas de alimentos são desperdiçados no mundo anualmente.  Diversos aspectos ambientais, sociais e econômicos contribuem para o aumento desses números, todavia metade do desperdício de alimentos no mundo poderia ser evitado por medidas preventivas. Desse modo, o presente trabalho teve como objetivo avaliar através de revisão da literatura, o potencial uso de subprodutos da uva, como por exemplo o bagaço, no desenvolvimento de produtos funcionais. Para tanto, foram utilizados os descritores “uva”, “uva qualidade inferior”, “uva exportação”, “uva alimento funcional”, “alimentos funcionais bagaço de uva”, “casca de uva alimento funcional”, “uva de refugo”, "grape", "low quality grape", "export grape", "functional food grape", "functional food grape pomace", "grape skin functional food", "junk grape". Como base de dados foram utilizadas a PubMed (National Library of Medicine), Portal de Periódicos Capes, LILACS (Literatura latino-americana e do caribe em ciências da saúde), Science Direct, Scorpus e Crossref, utilizando descritores da língua inglesa e portuguesa, e selecionando os artigos publicados entre os anos de 2000 a 2022.  Foram encontrados 942 artigos (PubMed= 546 ScienceDirect= 373; Scielo= 25), dos quais 80 artigos foram selecionados, e desses 68 foram excluídos por não se enquadrarem nos critérios de inclusão que eram, produtos funcionais desenvolvidos através dos subprodutos da uva que seriam descartados, e tendo, como também ano de publicação do estudo ter ocorrido a partir do ano de 2000. E assim foram utilizados 15 artigos. Os trabalhos em sua maioria aplicaram a uva para o desenvolvimento de farinhas para a produção de bolos e o enriquecimento de iogurtes, além de produzirem cookies, geleias e barras de cereais. Através da revisão realizada foi possível evidenciar que a utilização de um resíduo vitivinícola que tem potencial para conferir benefícios à saúde através do desenvolvimento de produto funcionais, reduzindo a quantidade destes resíduos na indústria e no meio ambiente, com possibilidade também de reduzir custos na produção dos alimentos.

Table grapes are fruits intended for fresh human consumption due to their sensory attributes and nutritional value. The objective of this chapter is to review the existing knowledge about table grapes, including a description of different varieties, with particular emphasis on the new highly appreciated seedless varieties. Following an introductory note on the world distribution and production of table grapes, also considering the impact of climate change, selected varieties of table grapes will be characterized in terms of their physiology, postharvest features, and consumer preferences. A morphological description of each variety, with emphasis on grape skin, grape rachis and grape cluster will be included. A final note on the drying of table grapes into raisins, and the most appropriate varieties for drying, will be given. The major changes occurring throughout the growth, development, and ripening phases of table grapes production will be discussed, regarding both physical (skin color and skin and pulp texture) and chemical (phenolic compounds, sugar content and acidity) parameters, as well as growth regulators.

In this study, we evaluated the effects of ultrasound-assisted extraction (UAE) under different time-temperature conditions on the content of bioactive compounds, antioxidant and antimicrobial activities of Syrah grape skin residue. The application of UAE showed a positive effect on the extraction of total flavonoids, and a negative effect on total polyphenols. The temperature of 40 °C and 60 °C without the UAE caused an increase of 260% and 287% of the total polyphenols, respectively. Nineteen individually bioactive compounds were quantified. The anthocyanin concentration (malvidin-3,5-di-O-glucoside 118.8–324.5 mg/100 g) showed high variation, to a lesser extent for phenolic acids, flavonoids, flavonols, procyanidins and stilbenes due to the UAE process. The Syrah grape skin residue has a high concentration of total phenolic compounds of 196–733.7 mg·GAE/100 g and a total flavonoid content of 9.8–40.0 mg·QE/100 g. The results of free radical scavenging activity (16.0–48.7 mg/100 mL, as EC50) and its inhibition of microbial growth (0.16 mg/mL, as EC50 for S. aureus, and 0.04 mg/mL, as EC50 for E. coli) by grape skin extract (UAE 40:20) indicate high antioxidant and antibacterial activity. It was concluded that the use of ultrasound needs further analysis for its application in this context, as it has shown deleterious effects on some compounds of interest. Syrah grape skin residue has potential as a source of bioactive antioxidants, antimicrobial activity and for use as a functional food ingredient.

Anthocyanins in red grape skin have a positive effect on fruit color and human health. The effect of foliar potassium application on anthocyanin accumulation in grape skin is not well understood. The study aimed to better understand the mechanism of anthocyanin accumulation in grape skin in response to foliar sprays of K2SO4 and KH2PO4. In this study, we investigated the effects of foliar application of KH2PO4 (T2), K2SO4 (T1) and distilled water (CK) on the skin color of ‘Kyoho’ grapes at mid-ripe and mature stages. At 90 and 110 days after full bloom (DAFB), T2 had the greatest total soluble solids (TSS), flavonoid and total anthocyanin contents, followed by T1 and CK. At two stages, the titratable acid content decreased and the juice pH increased under T2 treatment relative to CK. T1 and T2 had lower lightness (L*) than CK, and the color index of red grapes (CIRG) under T1 and T2 increased at two stages compared to CK. KEGG metabolic pathway analysis revealed that flavonoid biosynthesis was the most significantly enriched pathway in CK vs. T2 at 90 and 110 DAFB. At 90 DAFB, T2 had higher expressions of phenylalanine ammonia-lyas (PAL), cytochrome P450 CYP73A100 (CYP73A), 4-coumarate: CoA ligase (4CL), chalcone synthase (CHS), flavanone 3-dioxygenase-like (F3H) and UDP glucose: flavonoid 3-o-glucosyl transferase (UFGT) than CK and T1. Foliar application of potassium fertilizer may accelerate anthocyanin accumulation by altering the transcript levels of PAL, CYP73A, 4CL, CHS, F3H, and UFGT of the flavonoid biosynthesis.

Resveratrol, a phenolic compound possessing vital biological activities such as anti-cancer, is present abundantly in grape skin, a waste produced during the processing of grape juice. The objectives of this study were to prepare resveratrol-gold nanoparticles and a resveratrol nanoemulsion from grape skin and study their inhibition effects on pancreatic cancer cells BxPC-3. The spherical-shaped citrate gold nanoparticles (GNPs) and resveratrol-gold nanoparticles (R-GNPs) were, respectively, prepared with a surface plasmon resonance peak at 528 and 538 nm, mean particle size of 20.8 and 11.9 nm, and zeta-potential at −32.7 and −66.7 mV, by controlling an appropriate concentration of citrate/resveratrol and gold chloride as well as stirring time and temperature. The resveratrol nanoemulsion, composed of soybean oil, Tween 80, and sucrose fatty acid ester in glycerol and water, possessed a high storage stability with a mean particle size of 14.1 nm, zeta-potential of −49.7 mV, and encapsulation efficiency of 95.5%. An antiproliferation study revealed that both R-GNPs and resveratrol nanoemulsion could effectively inhibit the growth of pancreatic cancer cells BxPC-3, with the latter showing a higher inhibition effect. Western blot analysis implied that both can down-regulate expressions of cyclin A, cyclin B, CDK1, and CDK2 and up-regulate expressions of p53 and p21, accompanied by enhancing cytochrome C expression, decreasing BcL-2 expression, increasing Bax expression, and leading to the elevation of caspase-8, caspase-9, and caspase-3 activities for cell apoptosis execution. Future research is needed to study the inhibition of pancreatic tumors in vivo by R-GNPs and resveratrol nanoemulsions.

Grape pomace is commonly considered a waste product of monovarietal red wine production. Methods: HPLC-DAD analysis was performed to determine the polyphenol and flavonoid contents of all the extracts obtained from Montepulciano d’Abruzzo red wine and grape skins whereas, GC-MS was applied to the determination of fatty acid composition in grape seeds oil. Biological characterization involves antioxidant and antimicrobial assays for all the extracts and seeds oil; Their ability to inhibit α-glucosidase, α-amylase, α-tyrosinase, and ChE enzymes was also detected, together with anti-inflammatory activity on wine, grape skin extracts, and seeds oil by lipoxygenase (5-LOX) and LPS-stimulated macrophage release assays. Data indicate significative polyphenols content (199.31 ± 7.21 mgGAE/g), antioxidant (CUPRAC assay (1036.98 mgTE/g)), enzymatic inhibition (α-tyrosinase: 151.30 ± 1.20 mgKAE/g) and anti-inflammatory activities for wine-organic extract 2, while the antimicrobial activity of grape skin decoction is higher than those reported by wine extracts on three bacterial strains. Interestingly only dealcoholized wine and wine-aqueous extract exerts inhibitory effects on α-glucosidase (20.62 ± 0.23 mmolACAE/g and 19.81 ± 0.03 mmolACAE/g, respectively), while seeds oil is rich in oleic and linoleic acids. These results confirm the strong antioxidant properties of Montepulciano d’Abruzzo grape pomace, suggesting the potential use of this waste product as functional food supplements in the human diet and in cosmeceutics.

This work investigated the structural, biochemical, and molecular characteristics of grape skin cell wall during ripening, related to susceptibility to Botrytis cinerea. The comparative study between the two main grape cultivars in Champagne region, Pinot noir and Chardonnay, quantified: (1) the maturity and physical profile of grape skin; (2) the morphological characteristics; (3) soluble pectic polysaccharides located in grape skin cell walls; and (4) the gene expression of the two main degrading enzymes (VvPME1 and VvPG1) and PME activity. During the maturation period, the grape skins of the two cultivars appear different in their structure and composition. Chardonnay is characterized by higher relative humidity (RH) and level of VvPG1 expression, lower disease incidence and penetrometry values, and thicker cell walls than Pinot noir skins. Thus, the cell wall composition is sufficiently different between grape varieties from the same area to allow their discrimination and could be used to better manage the harvest date.
