Objective: To compare paraoxonase activity and changes in oxidative status in asthmatic children and healthy children by determining serum paraoxonase activity and total oxidative status, total antioxidant capacity and lipid hydroperoxidation. Methods: Forty two asthmatic children were compared with 32 healthy children of similar age and sex. To evaluate the paraoxonase and oxidative status, total antioxidant capacity and lipid hydroperoxidation were examined. Serum paraoxonase activity was evaluated by measuring the rate of paraoxon hydrolosis. Oxidative status was evaluated by the method developed by Erel. Lipid hydroperoxide was measured by an iodometric method. Results: In comparison with the healthy control group, the paraoxonase activity of the asthmatic children was found to be low (163.7 ± 73.0 (U/L) and 349.2 ± 153.9 (U/L), P = 0.002) and total oxidant status (9.0 ± 3.5 ?mol H2O2 Eq/L and 13.4 ± 7.0 ?mol H2O2 Eq/L, P =0.002), total antioxidant capacity (5.5 ± 2.5 µmol Trolox Eq/L and 1.0 ± 0.6 µmol Trolox Eq/L, P < 0.001), and lipid hydroperoxidation values (9.9 ± 3.4 ?mol H2O2 Eq/L and 4.4 ± 1.5 ?mol H2O2 Eq/L, P < 0.001) were found to be high. The high density lipoprotein (HDL) concentration of the asthmatic children was lower than that in the control group (40.1 ± 9.2 mg/dl and 54.5 ± 15.9 mg/dl, P < 0.001) Conclusion: In asthmatic children, when total oxidant status, total antioxidant capacity and lipid hydroperoxidation levels increase, paraoxonase activity decreased.

Background: Recently, methods of quantifying total oxidant status (TOS) and total antioxidant capacity (TAOC) were developed to investigate periodontitis. This study was performed to evaluate the salivary TOS and TAOC levels of patients with chronic periodontitis (CP) and investigating the association between periodontal clinical parameters and these oxidative stress biomarkers. Material and methods: 40 participants (23-65 years old) were classified into two groups of 20 each, namely the CP group (participants with CP) and the control group (periodontally healthy controls). Clinical periodontal parameters were monitored, and TOS and TAOC levels were measured using laboratory assays. Results: TOS level increased in the saliva of patients with CP and the salivary TAOC in patients with CP was significantly lower than that of the control group. The TOS had a positive moderate correlation with the plaque index and clinical attachment loss (r=0.32 and 0.37, respectively) while TAOC was negatively and moderately correlated with clinical attachment loss (r=-0.35). Conclusion: Salivary TOS and TAOC were distinguished in healthy and chronic PD patients. Further studies are required to comprehensively evaluate the potential role of these biomarkers in diagnosis and treatment evaluation of CP.

This study aimed to determine the impact of sesame meal, selenium (Se), and vitamin E (VitE) on goats’ oxidative status. Thirty mid-lactation crossbred goats were divided into five homogeneous groups, and were fed 1 kg of alfalfa hay and 1.2 kg of concentrates daily. The control group (C) received a basal diet. In the concentrates of the treated groups, 10% of the soybean meal was replaced by sesame meal and no extra VitE or Se (SM), or an extra 60 mg of VitE (SME), or 0.1 mg organic Se (SMSe), or their combination (60 mg VitE and 0,1 mg organic Se/kg of concentrate (SMESe). In the plasma of the goats, the dietary treatments did not affect glutathione reductase, glutathione peroxidase, glutathione transferase, catalase, superoxide dismutase activities, malondialdehyde (MDA) content, or the total antioxidant capacity. A reduction and a trend for lower protein carbonyls content was found in goats fed SM (p = 0.03) and SME (p = 0.06) compared to SMESe. In the milk, the lactoperoxidase activity decreased with SMSe and SMESe. A numerical decrease in the total antioxidant capacity and an increase in the MDA content in the milk of the SMESe group compared with the other treated groups was found. In mid-lactation goats, SM improves the oxidative status of both the organism and the milk.

The objective of the present study was to determine the plasma total oxidative status (TOS) and total antioxidant capacity (TAC) in patients with endemic fluorosis. A total of 79 (35 males and 44 females; mean age 44.0 ± 11.9 years) patients with endemic fluorosis and 55 (23 males and 32 females; mean age 48.3 ± 8.5 years) age-, sex- and body mass index-matched healthy controls were included in this study. The urine fluoride levels and plasma TOS and TAC levels were measured. The urine fluoride levels of fluorosis patients were significantly higher than control subjects as expected (1.91 ± 0.15 vs. 0.49 ± 0.13 mg/L, respectively; p < 0.001). TOS was significantly higher in fluorosis group than in control group (17.55 ± 3.82 vs. 15.06 ± 4.31 μmol H2O2 Eq/L, respectively; p = 0.001). TAC was significantly lower in fluorosis group than in control group (1.60 ± 0.36 vs. 1.82 ± 0.51 mmol Trolox Eq/L, respectively; p = 0.004). Oxidative stress index (OSI) was significantly higher in fluorosis group than in control group (11.5 ± 3.8 vs. 8.8 ± 3.7, respectively; p < 0.001). Correlation analysis in all the groups indicated that TAC was negatively correlated with urine fluoride ( r = −0.25, p = 0.003), TOS was positively correlated with urine fluoride ( r = 0.34, p < 0.001) and OSI was positively correlated with urine fluoride ( r = 0.36, p < 0.001). The results of our study demonstrate that oxidative stress plays an important role in the pathogenesis of the endemic fluorosis.

        This study aimed to find relationship between thymidine kinase-1 (TK-1) as tumor marker and total antioxidant capacity (TAC) in Iraqi children patients with thrombocytopenia and with thrombocytosis. The present study conducted 60 children patients (30 patients with idiopathic thrombocytopenia purpura (ITP) and 30 patients with thrombocytosis caused by leukemia) attending the Children Fever Hospital in the Medical City / Baghdad, and 30 healthy children as a control group. All study groups were with range ages (1-15) years, and they were diagnosed by assay of platelet count, Prothrombin Time   (PT), and partial Thromboplastin Time (PTT). The results shown elevation  in plasma TK-1 and TAC values in children patients with thrombocytopenia and with thrombocytosis when compared with control group, and there was no significant different in TK-1 level and a significant different in TAC level in two patients groups. There were a highly significant positive correlation between TK-1 and TAC levels in both Iraqi children patients with thrombocytopenia and with thrombocytosis. The current study concluded that TK-1 may be a novel biomarker for platelet count disorder disease and there was a probability of expose these patients for tumor diseases.

<p>Diabetes is the most common metabolic disorder. Idiopathic destruction of pancreatic beta cells will result in progressive loss of insulin, increase in ketone bodies, PH reduction and changes in bicarbonate neutralizing system in all body fluids including saliva and the oral cavity. The aim of this study was to compare the quality and quantity of saliva and oral health in children and adolescents with diabetes compared to healthy children. In this study, 27 diabetic patients (9 males, 18 females, age range 10 ± 5 years) were studied. A control group (27 persons) was selected from health persons with the same condition of gender and age. The amount of saliva was evaluated in 5 minutes, by non-stimulant collecting, in plastic vials. The PH and Total Antioxidant Capacity (TAC) were measured using paper strip and TAC kit. Oral and dental health was measured using DMFT (Decayed, missed, filled teeth) and MGI (Modified Gingival Index) indexes. Saliva in patients had less secretion (1.09 ± 0.13, 5.28 ± 0.23, P &lt; 0.01), PH (5.28 ± 0.09, 7.11 ± 0.10, P &lt; 0.001), and total antioxidant capacity (0.36 ± 0.04, 0.5 ± 0.04, P &lt; 0.001) in compare with controls group. DMF and MGI indicators were more in patients than in control group (P &lt; 0.001). Patients with type 1 diabetes had less secretion, PH and antioxidant defense and as a result had more dental and oral problems in compare with healthy children that with higher DMFT and MGI these patients require further training in this field and regularly examinations.</p>

Background: Nicotinic acid (niacin) is a broad-spectrum lipid-modifying agent that has potent antioxidant properties and reduces the production of lipid peroxidation. Objective: The purpose of the present study was to investigate the maturation, embryo development and cryo-tolerance merit, and levels of malondialdehyde (MDA), total oxidant status, and total antioxidant capacity following the supplementation of bovine oocytes maturation medium with different concentrations of niacin. Materials and Methods: Immature cumulus-oocyte complexes were cultured in tissue culture medium-199 maturation media supplemented with 0, 100, 200, and 400 µM niacin under a standard in vitro culture condition. After 24 hr of culture, the nuclear maturation rate was assessed. Then, two groups of immature cumulus-oocyte complexes were cultured in TCM-199 either with or without 400 µM niacin and evaluated for embryo development. Also, matured cumulus-oocyte complexes in both groups were frozen using a standard vitrification procedure. After vitrification, oocytes were warmed in two steps and evaluated for embryo development. In addition, the level of total antioxidant capacity, total oxidant status, and MDA were measured. Results: The results indicated that although the treatment with 400µM niacin increased in vitro nuclear maturation (87.6 ± 5.3), it did not improved the embryo development to the blastocyst stage. Higher cleavage and blastocyst rates were observed in vitrified oocytes that were cultured with supplemented 400 µM niacin compared to the control group (without niacin) (53.6 ± 2.7 and 10.6 ± 1.6 vs. 46.2 ± 4.1 and 6.3 ± 2.4, respectively). Also, the addition of 400 µM niacin to the maturation media could decrease MDA levels after maturation. Conclusion: Niacin could improve the quality of in vitro embryo production (IVP) embryos and tolerance of bovine oocytes to vitrification.
