It was aimed to evaluate the effects of different ingredi-ents on total antioxidant sta-tus (TAS) and total oxidant status (TOS) of the brewed Brazil mild roasted coffee. Sugar, sucralose, butter, co-conut-oil, animal and plant-based milk types were add-ed and milk types&sweeteners were combined separately. TAS and TOS were measured and oxidative stress indice (OSI) was calculated. TAS value of coffee with whole milk was the highest among all coffee types. TOS values of coffee with soy milk, with soy milk+sugar and soy milk+sucralose were lower than all other coffee types. When compared to OSI val-ues, coffee with soy milk, with soy milk+sugar and soy milk+sucralose had the lowest OSI values among all coffee types. Moreover, ad-dition of coconut-oil to cof-fee samples increased the TOS and OSI values of cof-fees. While the TOS and OSI of the sugar and su-cralose addition were found higher in comparison to plain coffee; TOS and OSI of the sucralose addition was lower than those sugar combinations. In conclu-sion, the OSI of the coffee may change depending on the alterations of chemical structures and nutritional matrices by the added in-gredients. The addition of soy milk or sucralose in-stead of sugar or whole cow milk instead of oil-based ingredients could be a promising strategy of reduc-ing the oxidative capacity.

Background and Objectives: Hypertrophic cardiomyopathy (HCM) depends on the primary impairment of sarcomeres, but it can also be associated with secondary alterations in the heart related to oxidative stress. The present study aimed to examine oxidative-antioxidant disturbances in patients with HCM compared with control individuals. Materials and Methods: We enrolled 52 consecutive HCM patients and 97 controls without HCM. The groups were matched for age, body mass index, and sex. Peripheral blood was collected from all patients to determine the total antioxidant capacity (TAC), total oxidant status (TOS), lipid hydroperoxide (LPH), and malondialdehyde (MDA). The oxidative stress index (OSI) was defined as the ratio of the TOS level to the TAC level. Results: The median age was 52 years, and 58.4% were female. The area under the curve (AUC) indicated good predictive power for the TAC and TOS [AUC 0.77 (0.69–0.84) and 0.83 (0.76–0.90), respectively], as well as excellent predictive power for the OSI [AUC 0.87 (0.81–0.93)] for HCM detection. Lipid peroxidation markers also demonstrated good predictive power to detect HCM patients [AUCLPH = 0.73, AUCMDA = 0.79]. Conclusions: The TOS, the TAC, LPH levels, and MDA levels have good predictive power for HCM detection. The holistic assessment of oxidative stress by the OSI had excellent power and could identify patients with HCM.

Rheumatoid arthritis (RA) is a chronic autoimmune disease associated with a significantly increased risk of cardiovascular mortality, mainly attributed to accelerated atherosclerosis. Methods: Thirty-two women (aged more than 18 years) with RA, and 25 age-matched healthy women were included in this study. Biomarkers of inflammation, red blood cells (RBCs) redox balance, estrogen receptor alpha (ER-α) expression as well as ERK 1/2 phosphorylation content were evaluated in RA patients at baseline and six months after treatment with disease modifying anti‐rheumatic drugs (DMARDs). Results: For the first times we demonstrated that in RA patients: i) disease activity score (DAS-28) positively correlated with RBC ER-α expression, and negatively with total antioxidant capacity of plasma; ii) RBC ER-α expression positively correlated with systemic inflammatory biomarkers and oxidative stress parameters as well as ERK 1/2 phosphorylation; and iii) DMARDs treatments improved the clinical condition measured by DAS-28 score decrease, although the RBCs appeared to be more prone to pro-oxidant status associated to the expression of survival molecules. Conclusion: Our data strongly suggest that RBCs could also participate in vascular homeostasis through fine modulation of an intracellular signal linked to the ER-α.

Abstract Objectives This article aims to explain the altered oxidative status and thiol/disulfide homeostasis before and after surgery in children with congenital heart disease (CHD). Methods Blood samples were taken from the patients (n=50) before the operation (baseline), at the 1st hour, and at the 24th hour after the operation. Thiol-disulfide levels, total antioxidant status (TAS), total oxidant status (TOS), oxidative stress index (OSI), myeloperoxidase (MPO), ceruloplasmin, albumin, ischemia-modified albumin (IMA), and prolidase activities of all samples were measured. Pre-operative oxygen saturation (SaO2) values and neutrophil/lymphocyte ratios (NLR) were also measured. Results Before the operation, TOS, OSI, MPO, ceruloplasmin, IMA, NRL, and disulfide levels were higher in the cyanotic group than in the acyanotic group. When the indicated three different time points were compared, the TOS, OSI levels, and MPO activities of the 24th hour were significantly lower than the baseline and 1st hour. In comparison, ceruloplasmin levels of 1st hour were significantly higher than of the baseline and 24th hour. Native thiol and total thiol levels in the baseline group were significantly lower than in the 1st and 24th hours. Disulfide levels of the 24th hour were significantly lower than of the baseline. Conclusions The operation leads to changes in the thiol-disulfide balance and oxidant status in CHD.

Gastroesophageal reflux disease (GERD) is a global health problem in which the prevalence is increasing over periods. This disease is a significant cause of disorders in the upper gastrointestinal tract with very complex pathogenesis. Oxidative stress (OS) due to gastric acid reflux plays a role in the inflammatory response of the esophageal epithelium. Several OS markers have been widely studied and are thought to affect the severity degree of the esophageal mucosa. However, there has been no research on total oxidant status (TOS), total antioxidant status (TAS), and OS index (OSI) in the adult with GERD; hence the aim of this review was to determine the association between TOS, serum TAS, and OSI with the GERD degree. A literature review was conducted by searching articles related to the TOS, TAS, OSI, and its correlation with GERD degree on an online database, particularly Pubmed and Google scholar. We conclude that TAS and OSI might influence the severity of GERD; however, further clinical study is needed to prove this theory.

Aim: The aim of this study was to assess the antioxidant properties of ethanolic and aqueous extracts of Punica granatum (Pomegranate) seed in testosterone induced benign prostate hyperplastic albino Wistar rats. Study design: This study is an interventional study. Place and Duration of Study: The experimental aspect of this study was conducted at the animal house, Department of Pharmacology, University of Port Harcourt between April and September, 2019. Methodology: Seventy (70) adult albino male wistar rats were used for this study. They were divided into 12 groups of 5 rats each and fed with commercial rat diet and clean drinking water. Aqueous and ethanolic extracts of Punica granatum seed were prepared using the maceration method. Benign Prostate Hyperplasia was induced in rats after they submitted to bilateral orchiectomy by daily injections of testosterone propionate (TP) (4 mg/kg b.wt.sc). Rats were treated with 500 or 1500 mg/kg b.wt. of aqueous or ethanoI extracts of Punica granatum seed, dutasteride or in combination. Administration of extracts was done by gavage. Plasma total oxidant status (TOS), total antioxidant status (TAS), superoxide dismutase (SOD) activity, were analyzed using sandwich ELISA Kits by Shanghai Korain Biotech Co., Ltd, China, while oxidative stress indices (OSI) were calculated. Statistical analysis was done using SPSS version 22.0 of Windows Stat Pac and p values less than 0.05 were considered statistically significant. Results: The results showed that the mean TOS, TAS, SOD and OSI for the rats in the normal control group were 1.66±0.2U/ml, 2.71±0.25U/ml, 41.8±2.9pg/ml and 0.62±0.10 respectively. After BPH induction (group 2), the values were 3.25±0.5U/ml,1.17±0.14U/ml, 23.38±2.09 pg/ml and 2.81±0.60pg/ml respectively. There were significant decreases for TOS and OSI, and significant increases for TAS and SOD when the rats where treated with lower and higher doses of both aqueous and ethanolic extracts of Punica granatum and Dutasteride. Conclusion: In conclusion, both doses of Punica granatum seed for ethanolic and aqueous extracts individually and in combination and with dutasteride markedly reduced total oxidant status, oxidative stress indices and improved the activities of antioxidant parameters like superoxide dismutase and total antioxidant status.

It is known that plants have been used by humans for a very long time in the treatment of diseases. A wide variety of plant species grown in different regions have been very successful in the treatment of many diseases. In this study, Echium italicum L. plant collected from different regions was used as material. The plant was extracted with ethanol in a soxhlet device. Then, antioxidant and oxidant levels of the plant extract were determined using Rel Assay kits. As a result of the analysis, the TAS values of the plant were determined 5.442±0.240 (Gaziantep), 4.896±0.081 (Hakkari) and 6.056±0.139 (Mersin), TOS values were 21.372±0.747 (Gaziantep), 12.255±0.341 (Hakkari) and 19.107±0.438 (Mersin), OSI values were 0.395±0.027 (Gaziantep), 0.250±0.005 (Hakkari), and 0.316±0.005 (Mersin). It was observed that antioxidant and oxidant values changed as the regions where the plant was collected changed. In addition, it was determined that E. italicum could be a natural antioxidant source.

This study was based on the determination of post-thaw semen quality, oxidant–antioxidant status and DNA-damage status of frozen Mesopotamian catfish semen. To this end, the sperm was frozen in diluent containing different inositol concentrations (5, 10, 20 and 40 mg). Increasing levels of inositol linearly increased the spermatozoa motility rate and duration significantly (p < 0.05). High doses of inositol increased antioxidants and decreased oxidants. In addition, lower intracellular DNA damage and percentage of apoptotic spermatozoa occurred in high doses of inositol (p < 0.05).Abstract: In this study, the effects of supplemented inositol on sperm extenders were examined on the spermatozoa motility rate and duration, total antioxidant and oxidant status, apoptotic spermatozoa and DNA damage, during the sperm post-thaw process of Mesopotamian Catfish (Silurus triostegus, H. 1843). The semen was frozen in diluents containing different inositol concentrations (5, 10, 20 and 40 mg). Increasing levels of inositol linearly improved the spermatozoa motility rate and duration significantly (p < 0.05). MDA and TOS were linearly decreased, however, TAS and GSH linearly increased (p < 0.05). The increasing inositol levels resulted in a linear and quadratic decrease in DNA damage in the comet assay, 8-hydroxydeoxyguanosine and the determined percentage of apoptotic spermatozoa (p < 0.05). These results suggest that there are many positive effects of the use of supplemental inositol on enhancing sperm cryopreservation efficiency in Silurus triostegus.

PCOD is a complex heterogenous metabolic disorder affecting 10 % of women at their reproductive age. A well effective treatment for a complete cure for PCOD is still a challenge and evaluation of new strategies to treat this disease is of great priority. This review focuses on two aspects of PCOD, namely types of rodent screening models and diagnosis of major parameters. A fully convincing animal model to study PCOD is still a challenge as they are multi-ovular species. But in many ways, the rodent models are similar to human PCOD. This study explores on the parallels and problems associated with the use of different rodent models. Oligo- or anovulation, clinical or biochemical hyperandrogenism and polycystic ovaries constitute the key clinical features on which the diagnosis should be based. Determination of diagnosis parameters such as estrus cyclicity, insulin sensitivity, lipid profile, anti-oxidant status, weight of reproductive system and histopathological features of ovary are discussed along with the currently available treatment methods for PCOD.
