A brine shrimp toxic compound has been isolated from Mangifera foetida L. steam bark. Based on spectroscopic data (IR,1H- NMR, 13C-NMR 1D and 2D), the structure of this compound was identified as mangiferenes B, C30H47O2. In this study, the n-hexane, ethyl acetate, methanol extracts and isolated compound showed toxic effects (LC50 471.9543; 534.5209, 678.4421, and 22.1615 μg/mL, respectively). The toxicity degree among the three extracts and isolated compound is isolated compound > n-hexane extract > ethyl acetate extract > methanol extract.

Mahkota dewa plant (Phaleria macrocarpa (Scheff.) Boerl.) which is included into family of Thymelaeaceae is one of Indonesia's traditional medicines. Chemical constituent has been isolated from ethyl acetate extract of leaves of mahkota dewa. Sample was extracted with methanol, concentrated then extracted by n-hexane, chloroform and ethyl acetate. The ethyl acetate extract was separated and fractionated by column chromatography. The first fraction was purified by TLC preparative and recrystalization. Compound was isolated as red-brown spherical crystal in 8 mg (m.p. 129-131 °C). Its spot gave dark fluoroscence at TLC plate (UV366) with Rf of 0.3 at TLC chromatogram with eluent of n-hexane : ethyl acetate (7:3); 0.6 with n-hexane : ethyl acetate (1:1); 0.9 with -hexane : ethyl acetate (4:6). This compound was dissolved in methanol. Compound was identified by UV, IR, 1H NMR, 13C NMR and NMR 2 dimension (HMQC, COSY, HMBC and DEPT-135) spectroscopic as 2,6,4'-trihydroxy-4-methoxybenzophenon. This compound as well as the ethyl acetate extract showed antioxidant activity on DPPH with IC50 was 10.57 and 101.06 μg/mL, respectively. This compound showed strong antioxidant activity on DPPH, almost to the standard antioxidant activity of quercetin (IC50 of 2.93 μg/mL)

Two triterpenoids, phragmalin triacetate (1) and lupeol (2) were isolated from an ethyl acetate extract of the stem bark of Crataeva nurvala (Capparidaceae) by repeated chromatography over silica gel. The structures of these compounds were determined by spectroscopic analyses (UV, IR, 1H NMR, 13C NMR and EIMS). This is the first report of the systematic phytochemical investigation and the presence of these compounds 1 and 2 from this plant. Coumarin has been isolated and characteritated from rind of  Citrus nobilis Lour by maseration methode with methanol. After fractionation by n-hexane and ethyl acetate, collected the phase ethyl acetate which positive of coumarin. From ethyl acetate extract coumarin have been isolated by column chromatography. The isolation results was obtain 2,159 g of pure white needle-shape crystalline with the melting point of 126,2-127,60C. Structure of the isolated coumarine was elucidated by spectroscopic methodes, UV-Vis,13C-NMR, 1H-NMR, 2D-NMR (DEPT/HSQC, COSY, NOESY, HMBC) and IR spectra. Based on the spectra data the isolated coumarine is marmin.

Daun sukun (Artocarpus altilis) sudah lama dijadikan sebagai obat tradisional untuk mengatasi berbagai penyakit seperti liver cirrhosis, hipertensi, dan diabetes. Ekstrak etil asetat dari daun sukun juga berpotensi menghambat pertumbuhan sel kanker. Beberapa senyawa yang telah diketahui sebagai agen antikanker di dalam ekstrak etil asetat daun sukun adalah golongan geranil flavonoid. Akan tetapi, senyawa pyranoflavoid belum dilaporkan dapat ditemukan pada daun sukun. Untuk itu, penelitian ini bertujuan untuk mengisolasi senyawa pyranoflavonoid dari ekstrak etil asetat daun sukun dan menguji aktivitasnya sebagai antikanker. Daun sukun diekstraksi dengan etanol 70% kemudian dilakukan partisi dengan petroleum eter dan etil asetat. Ekstrak etil asetat yang diperoleh difraksinasi dengan kromatografi kolom silica gel dengan gradient elusi heksan-etil asetat sehingga diperoleh senyawa kristal berwarna kuning, AA3. Senyawa AA3 kemudian diidentifikasi dengan spektrofotometer UV-Vis, LC-MS, 1H-NMR dan 13C-NMR dan diperoleh hasil bahwa senyawa AA3 tersebut adalah siklokomunol yang merupakan golongan pyranoflavonoid. Uji antikanker siklokomunol dengan MCF-7 dan T47D menunjukkan adanya aktivitas antikanker dengan nilai IC50 masing-masing adalah 75.46 µM dan 36.20 µM selama 48 jam.Kata kunci:Artocarpus altilis, siklokomunol, antikanker, MCF-7, T47D. Leaves of breadfruit Artocarpus altilis (Parkinson) Fosberg has long been used as traditional medicine to overcome a variety of diseases such as liver cirrhosis, hypertension, and diabetes. Ethyl acetate extract of the leaves of breadfruit also potentially inhibit the growth of cancer cells. Several compounds have been known as an anticancer agent in the ethyl acetate extract of leaves of breadfruit is belong to geranyl flavonoid group. However, the compound belong to pyranoflavoid group has not been reported can be found on the leaves of breadfruit. Therefore, this study aims to isolate the pyranoflavonoid compound from ethyl acetate extract of leaves of breadfruit and tested as an anticancer agent. Breadfruit leaves were extracted with 70% ethanol and then made a partition with petroleum ether and ethyl acetate. Ethyl acetate extract obtained was fractionated by silica gel column chromatography with gradient elution of hexane-ethyl acetate to obtain a yellow crystalline compound, AA3. AA3 compound was identified by UV-Vis spectrophotometer, LC-MS, 1H-NMR and 13C-NMR and obtained results that AA3 compound is siklokomunol which is belong to pyranoflavonoid group. Anticancer test of siklokomunol with MCF-7 and T47D showed anticancer activity with IC50 values of each are 75.46 µM and 36.20 µM, respectively, for 48 hours.Keywords: Artocarpus altilis, siklokomunol, anticancer, MCF-7, T47D.

Research on medicinal plants are important to Nepal because most of its rural population relies on it as mode of medicine. Medicinal plants namely Swertia chirayita and Dendrobium amoenum were collected from mid hills of Nepal. The present study was undertaken to find the antimicrobial activity, phytochemical presence and their cytotoxicity in different extraction medium. The percentage yield from the plants were highest in warm methanol extraction with 12.6%, followed by ethyl acetate and lowest was for cold methanol. Plant extract showed the presence of antioxidants like alkaloid, terpenoids, flavonoids, tannin, glycosides. The Brine Shrimp Bioassay of methanol and ethyl acetate extract showed cytotoxicity. Chiraito extract showed LC50 of 199 ppm for Dhunche sample, 128.82 ppm for Daman sample and 131.82 ppm of Illam sample. The antibacterial activity of methanol extract of Chiraito and Dendrobium amoenum showed significant bioactivity by inhibiting growth of microbial species selected for the test. The zone of inhibition shown by the extracts was comparable to the standard antibiotics. Similarly, methanol extract of Chiraito also showed significant antifungal activity with the zone of inhibition comparable to amphotericin.Nepal Journal of Biotechnology. A phytochemical investigation was conducted on the flowers of Nyctanthesarbor-tristis Linn. For isolation of compounds, the dried flower’s powder was successively extracted with n-hexane, dichloromethane, ethyl acetate and methanol. The extracts were fractionated using different chromatographic techniques and four compounds were isolated. Stigmasterol (1) from n-hexane, rengyolone (2) from dichloromethane and two other compounds namely, 2-phenylethyl β-D-glucopyranoside (3) and n-tetradecyl-β-D-glucopyranoside (4) from ethyl acetate extract, were isolated. These compounds (1-4) were characterized on the basis of IR, 1H NMR, 13C NMR, DEPT-135 NMR. Compounds 1 and 3 were isolated for the first time from this plant while compound 4 has been isolated and completely characterized from this plant as well as from the natural sources. Bangladesh J. Phytochemical study of the roots of Erythrina senegalensis led to the isolation of a new α-sophoradiol glycoside, erythrinoside (1), together with four known compounds, lupeol (2), α-sophoradiol (3), isoneorautenol (4) and D-mannitol (5). The structures of the compounds were elucidated using spectroscopic data including 1D and 2D NMR, mass spectrometry and by comparison made with some data reported previously; the samples (extracts and compounds) were also subjected to antidiabetic assay. Erythrinoside and isoneorautenol exhibited good α-amylase inhibitory potential of 54.6% and 53.3%, respectively, compared to acarbose (72.5%) at 400 µg/mL. With α-glucosidase, all samples showed promising inhibition percentages above 50% at 200 µg/mL. In the α-glucosidase assay, the ethyl acetate extract (65.5%), methanol extract (72.1%), erythrinoside (63.3%) and isoneorautenol (66.0%) had percentage inhibitions closer to that of acarbose (69.0%) at 200 µg/mL. The methanol extract (IC50 = 81.2 ± 0.9 µg/mL) was more active than acarbose (IC50 = 94.5 ± 0.7 µg.mL) in the α-glucosidase assay. The inhibition of α-amylase and α-glucosidase indicates that E. senegalensis extracts and compounds could be used to manage diabetic conditions.

This work is part of a series of chemical investigations of the genus Grevillea. Two new arbutin derivatives, seven new bisresorcinols, including a mixture of two isomers, three known flavonol glycosides, and four known resorcinols, including a mixture of two homologous compounds, were isolated from the ethyl acetate extract of the leaves and methanol extract of the stems of Grevillea banksii. The new compounds were identified, on the basis of spectroscopic data, as 6′- O-(3-(2-(hydroxymethyl)acryloyloxy)-2-methylpropanoyl)arbutin (1), 6′- O-(2-methylacryloyl) arbutin (2), 5,5′-(4( Z)-dodecen-1,12-diyl)bisresorcinol (6), 2′-methyl-5,5′-(4( Z)-tetradecen-1,14-diyl)bisresorcinol (8), 2,2′-di(4-hydroxyprenyl)-5,5′-(6( Z)-tetradecen-1,14-diyl)bisresorcinol (9), 2-(4-acetoxyprenyl)-2′-(4-hydroxyprenyl) 5,5′-(6( Z)-tetradecen-1,14-diyl)bisresorcinol (10), 2-(4-acetoxyprenyl)-2′-(4-hydroxyprenyl)5,5′-(8( Z)-tetradecen-1,14-diyl)bisresorcinol (11), 5,5′-(10( Z)-tetradecen-1-on-diyl)bisresorcinol (12) and 5,5′-(4( Z)-tetradecen-1-on-diyl)bisresorcinol (13).

Penelitian ini melakukan kajian terhadap toksisitas ekstrak etil asetat dari kuli biji jengkol (Archidendron jiringa) Jack. I. C. Nielsen) dengan menggunakan metode Brine Shrimp Lethality Test (BSLT). Nilai LC50 ekstrak diperoleh dari persamaan regresi linear dari log konsentrasi vs probit % mortalitas. Kulit biji jengkol dikeringkan pada temperature ruang selama 1 x 24 jam dan dimaserasi dengan methanol. Filtrat yang diperoleh diuapkan dengan menggunakan rotary evaporator untuk memperoleh ekstrak pekat methanol. Selanjutnya, ekstrak pekat methanol dilarutkan dalam air dan dipartisi dengan etil asetat, diulangi beberapa kali, dan diuapkan kembali untuk memperoleh ekstrak pekat etil asetat. Uji fitokimia untuk penentuan kandung fenolik dilakukan dengan FeCl3. Uji kandungan fenol dengan FeCl3 menunjukkan hasil positif untuk ekstrak etil asetat. Nilai LC50 untuk ekstrak etil asetat adalah 14.19 ppm. Berdasarkan nilai LC50 ekstrak etil asetat kulit biji jengkol dikategorikan sangat toksik. Toksisitas ekstrak etil asetat dapat diasosiasikan oleh kandungan senyawa fenol dalam ekstrak.   This research studied about toxicity ethyl acetate extract of jiringa (Archidendron jiringa) Jack. I. Pods of jiringa dried at room temperature 1 x 24 h and then macerated with methanol. Filtrat were evaporated with rotary evapoarator to obtained methanol extract. Further, methanol extract dissolved with water and partitioned with ethyl acetate for several times, and then evaporated to obtained ethyl acetate extract. Test consitution of phenolic compound performed by using FeCl3. Test phenolic compounds with FeCl3 showed positive test for ethyl acetate. The LC50 values of ethyl acetate was 14.19 ppm. Ethyl acetate extract of jiringa (Archidendron jiringa) Jack. I. C. Nielsen) pods showed high toxicity. The high toxicity cause of phenolic compounds constitution. Ethyl acetate extract of jiringa’s pods has high toxicity because it contains phenolic compounds.

The petroleum ether, ethyl acetate and methanol extracts of the stem bark of Callistemon citrinus were subjected to screenings for antimicrobial and antioxidant activities and brine shrimp lethality. The ethyl acetate  extract and its column fractions XVIII and XIV exhibited moderate antimicrobial activity, while the methanol extract  revealed significant antioxidant activity having IC50 of 3.84 ?g/ml. The methanol extract and fraction-II showed  potent cytotoxic its cytotoxicity with the LC50 of 11.27 and 11.35 ?g/ml, respectively. DOI: http://dx.doi.org/10.3329/dujps.v11i1.12487 Dhaka Univ. J.
