TheStaphylococcus intermediusgroup (SIG) includes zoonotic pathogens traditionally associated with dog bites. We describe a simple scheme for improved detection of SIG using routine laboratory methods, report its effect on isolation rates, and use sequencing to confirm that, apart from one atypical SIG strain, most isolates areStaphylococcus pseudintermedius.

Successful treatment of canine pyoderma has become compromised owing to the development of antimicrobial resistance with accompanying recurrence of infection. Canine skin samples submitted to a veterinary diagnostic laboratory for microbiological culture and sensitivity between January 2007 and June 2010, from which Staphylococcus intermedius was isolated, were selected for this investigation. Antimicrobial resistance of S. intermedius was most prevalent with reference to ampicillin followed by resistance to tetracycline and then potentiated sulphonamides. In general, antimicrobial resistance was low and very few methicillin-resistant isolates were detected. Temporal trends were not noted, except for ampicillin, with isolates becoming more susceptible, and potentiated sulphonamides (co-trimoxazole), with isolates becoming more resistant. In general, both the Kirby–Bauer disc diffusion and broth dilution minimum inhibitory concentration tests yielded similar results for the antimicrobial agents tested. The main difference was evident in the over-estimation of resistance by the Kirby–Bauer test for ampicillin, co-trimoxazole, penicillin and doxycycline. Knowledge of trends in bacterial resistance is important for veterinarians when presented with canine pyoderma. Analysis of antimicrobial susceptibility profiles of S. intermedius isolated from canine pyodermas will guide veterinarians’ use of the most appropriate agent and encourage prudent use of antimicrobials in companion animals.

Antibiotic resistance of canineStaphylococcus intermediusgroup (SIG) - practical implicationsA total of 221 SIG strains were isolated from clinical samples of canine origin submitted to the Diagnostic Laboratory of the Division of Bacteriology and Molecular Biology at the Warsaw University of Life Sciences in Warsaw during the period 2006-2010. The aim of the study was to investigate the frequency of prevalence of methicillin-resistant SIG strains and to determine the MIC values of cephalotin, amoxicillin/clavulanic acid, ciprofloxacin, clindamycin, gentamicin, chloramphenicol, mupirocin for a collection of randomly selected 79 strains belonging toStaphylococcus intermediusgroup (SIG), including 23mecA-positive and 56mecA-negative strains. All isolates were identified as belonging to SIG based on their phenotypic properties and PCR amplification ofS. intermedius-specific fragment of the 16S rRNA gene. ThemecAgene was detected in 26 (12%) of 221 SIG strains. All testedmecA-negative SIG strains were susceptible to amoxicillin/clavulanic acid and cephalotin. One of the 56mecA-negative SIG strains was resistant to ciprofloxacin, six (11%) to gentamicin. It was found that sixteen (29%) of 56mecA-negative SIG strains were resistant to clindamycin. Most of themecA-positive SIG strains were resistant to ciprofloxacin (96%), clindamycin (96%), and gentamicin (96%). Only one MRSIG strain was resistant to chloramphenicol. All examinedmecA-positive SIG strains were found to be susceptible to mupirocin. Our results imply that staphylococcal multidrug resistance has become more prevalent, which could lead to difficulties in effective treatment. With some resistant strains the only therapeutic possibility are antimicrobial agents important in human medicine. New regulations for veterinary medicine concerning appropriate therapy of infections caused by multidrug-resistat staphylococci are needed.

Bovine respiratory diseases cause serious economic loses and present diagnostic difficulties due to the variety of etiologic agents, predisposing conditions, parasites, viruses, bacteria and mycoplasma, and may be multiple or complicated. Several agents which have been isolated from the abnormal lungs are still the subject of controversy and uncertainty. These include adenoviruses, rhinoviruses, syncytial viruses, herpesviruses, picornaviruses, mycoplasma, chlamydiae and Haemophilus somnus.Previously, we have studied four typical cases of bovine pneumonia obtained from the Michigan State University Veterinary Diagnostic Laboratory to elucidate this complex syndrome by electron microscopy. More recently, additional cases examined reveal electron opaque immune deposits which were demonstrable on the alveolar capillary walls, laminae of alveolar capillaries, subenthothelium and interstitium in four out of 10 cases. In other tissue collected, unlike other previous studies, bacterial organisms have been found in association with acute suppurative bronchopneumonia.

Zusammenfassung: Gegenstand und Ziel: Im Gegensatz zur Hündin liegen bei der Katze bisher wenige Studien über die genitale Keimflora geschlechtsgesunder Tiere vor. Ziel der Untersuchung war daher, physiologische Daten über die aerobe Vaginalflora bei dieser Spezies zu gewinnen. Material und Methoden: Für die vorliegende Studie standen 26 gesunde, anöstrische Katzen zur Verfügung, die zu einer Ovariohysterektomie vorgestellt wurden. Nach einer klinischen Untersuchung wurden von allen Probanden unter sterilen Bedingungen Vaginaltupfer entnommen. Ergebnisse: In allen Proben konnte ein Bakterienwachstum mit durchschnittlich zwei verschiedenen Bakterienspezies nachgewiesen werden. Die Gesamtkeimgehalte wurden bei 50% der Vaginaltupferproben als gering-, bei 15% als mittel- und bei 35% als hochgradig beurteilt. Vorherrschend waren Mischkulturen aus zwei bis vier verschiedenen Keimarten. Monokulturen wurden aus 38% der Tupferproben isoliert. Am häufigsten gelang der Nachweis von E. coli variatio haemolytica (E. coli var. haem.) (58%) und Staphylococcus epidermidis (42%). Als weitere Spezies wurden E. coli, α-, β-hämolysierende Streptokokken, anhämolysierende Streptokokken, aerobe Bazillen, Staphylococcus aureus, Staphylococcus intermedius, Pasteurella multocida sowie Klebsiellen isoliert. Auffällig ist die hohe Nachweisrate von E. coli var. haem. mit 35% in Mischkulturen und 23% in Reinkultur. Schlussfolgerungen: Die physiologische Mikroflora der felinen Vaginalschleimhaut differiert deutlich von der der anöstrischen Hündin. Besonders die Dominanz von E. coli var. haem. in 38% der Mischkulturen und 23% der Monokulturen bei der Katze ist hervorzuheben. Klinische Relevanz: Die vorliegenden Ergebnisse geben eine erste Grundlage für die Interpretation mikrobiologischer Befunde feliner Vaginaltupfer.

Human parechoviruses (HPeV) belonging to the family Picornaviridae are widespread enteric pathogens and are associated with various clinical syndromes in human. At present, 16 HPeV genotypes (HPeV1–16) are known. There is no report on the detection of HPeVs in Central Europe. Aims: The aim of the retrospective study was to detect and characterize HPeVs using molecular methods in cell cultures with “enterovirus-like” cytophatic effect (CPE) archived between 1990 and 2004, in two virology laboratories, in Hungary. Materials and methods: In Laboratory I, fecal samples from children with symptoms of gastroenteritis under the age of 10 years were cultured as a previous routine diagnostic laboratory protocol for “enterovirus”. Cell cultures indicating CPE were archived between 1990 and 2000. In Laboratory II, 2 fecal samples, a liquor and a nasopharyngeal aspirate were re-tested which contained an “enterovirus-like” virus in cell cultures and were positive by HPeV1 neutralization immunosera between 2000 and 2004. Specimens were tested retrospectively for HPeV by reverse transcription–PCR (RT-PCR) method using 5’UTR conserved primers. Specific primers were designed to determine the HPeV structural region (VP0-VP3-VP1). Results: 9 of the 66 archived samples (9.1%) from Laboratory I and all the 4 samples from Laboratory II were found to be HPeV-positive. 10 samples were identified as HPeV1, 2 were HPeV4 and 1 could not be determined. 3 HPeV1 clusters were identified in Laboratory I according to the isolation date originated from years 1990/1991, 1992/1995 and 1998. HPeV1 was detected in clinical syndromes: gastroenteritis (in a 24-years-old adult), recurrent stomatitis aphtosa (in a 42-years-old adult), encephalitis and ataxia cerebellaris acuta in infants and children in Laboratory II. Conclusions: This is the first detection of HPeVs in Central Europe. Detection and genetic characterization of HPeV in available historical samples infected with previously unidentifiable agents with “enterovirus-like” cytopathogenic effect may help to understand the clinical importance and spectrum of the infections and the genetic diversity and evolution of these viruses.
