Latar Belakang: Ekstrak etanol 70% daun Justicia gendarussa Burm.f, bebas dan tidak bebas alkaloid dipercaya memiliki aktivitas inhibisi yang kuat terhadap enzim Reverse Transcriptase HIV, sebagai upaya menemukan solusi dalam menghadapi prevalensi HIV AIDS yang masih tinggi dengan kendala pengobatan HIV AIDS seperti efeks samping dan resistensi. Metode: Kedua jenis ekstrak diujikan secara in vitro menggunakan teknik Elisa dan dianalisa kandungan kimia yang Gendarusin A yang memiliki efek antiHIV menggunakan kromatografi cair kinerja tinggi. Hasil: Hasil uji Elisa diperoleh persen penghambatan berturut-turut sebesar , 254,2, 254,2, 235,6 dan 279,7 untuk konsentrasi 5 ppm, 10 ppm, 15 ppm dan 20 ppm ekstrak bebas alkaloid dan sebesar169,0, 164,0, 130,5 dan 369,5 untuk konsentrasi 5 ppm, 10 ppm, 15 ppm dan 20 ppm ekstrak tidak bebas alkaloid. Hasil kandungan kimia diperoleh Gendarusin A pada ekstrak bebas bebas alkaloid pada waktu retensi 8,402 menit dan ekstrak tidak bebas alkaloid pada waktu retensi 8,381. Kesimpulan: Justicia gendarussa Burm.f dapat menjadi sumber yang bermanfaat untuk isolasi dan pengembangan inhibitor baru antiHIV.

HIV-AIDS is a global problem and the deadliest disease in the world. One of HIV and AIDS prevention strategy can be done with traditional medicine research program from natural resource that has anti-HIV AIDS activity. It has been found that 70% ethanol extract of Justicia gendarussa Burm.f leaves, alkaloid free and alkaloid non-free, has a strong inhibitory activity against HIV reverse transcriptase enzyme, as an effort to find a solution in the face of HIV AIDS prevalence that is still high with problem of HIV-AIDS treatment such as side effects and resistances. Justicia gendarussa had already known for having an effect anti-HIV and therefore we were looking at the mechanism of inhibition of HIV Reverse Transcriptase enzyme. Both types of extracts were tested in vitro using ELISA technique and analysed chemical content of Gendarusin A as anti-HIV using high performance liquid chromatography. ELISA test results obtained percent inhibition, respectively for 254.2, 254.2, 235.6, and 279.7 for the concentration of 5 ppm, 10 ppm, 15 ppm and 20 ppm of free alkaloid extract and 169.0, 164.0, 130.5 and 369.5 for the concentration of 5 ppm, 10 ppm, 15 ppm and 20 ppm of non-free-alkaloid extract. The results of high performance liquid chromatography obtained Gendarusin A in the free-alkaloid extract at retention time 8.402 minutes and non-free alkaloid extract at retention time 8.381. Therefore, these results concluded that the Justicia gendarussa Burm.f can be a useful resource for the isolation and development of new anti-HIV.

The use of Antiretroviral drugs in treating HIV/ AIDS patients has enormously increased their life spans with serious disadvantages. The virus infection still remains a public health problem worldwide with no cure and vaccine for the viral agent until now. The use of nanoparticles (NPs) for the treatment and prevention of HIV/AIDS is an emerging technology of the 21st century. NPs are solid and colloid particles with 10 nm to <1000 nm size range; although, less than 200 nm is the recommended size for nanomedical usage. There are NPs with therapeutic capabilities such as liposomes, micelles, dendrimers and nanocapsules. The particle enters the body mainly via oral intake, direct injection and inhalation. It has been proven to have potentials of advancing the prevention and treatment of the viral agent. Certain NPs have been shown to have selftherapeutic activity for the virus in vitro. Strategies that are novel are emerging which can be used to improve nanotechnology, such as genetic treatment and immunotherapy. In this review, nanoparticles, the types and its characteristics in drug delivery were discussed. The light was furthermore shed on its implications in the prevention and treatment of HIV/AIDS.

ABSTRACT Ty1 reverse transcriptase/RNase H (RT/RH) is exquisitely sensitive to manganese concentrations. Elevated intracellular free Mn2+ inhibits Ty1 retrotransposition and in vitro Ty1 RT-polymerizing activity. Furthermore, Mn2+ inhibition is not limited to the Ty1 RT, as this ion similarly inhibits the activities of both avian myeloblastosis virus and human immunodeficiency virus type 1 RTs. To further characterize Mn2+ inhibition, we generated RT/RH suppressor mutants capable of increased Ty1 transposition in pmr1Δ cells. PMR1 codes for a P-type ATPase that regulates intracellular calcium and manganese ion homeostasis, and pmr1 mutants accumulate elevated intracellular manganese levels and display 100-fold less transposition than PMR1 + cells. Mapping of these suppressor mutations revealed, surprisingly, that suppressor point mutations localize not to the RT itself but to the RH domain of the protein. Furthermore, Mn2+ inhibition of in vitro RT activity is greatly reduced in all the suppressor mutants, whereas RH activity and cleavage specificity remain largely unchanged. These intriguing results reveal that the effect of these suppressor mutations is transmitted to the polymerase domain and suggest biochemical communication between these two domains during reverse transcription.

The Mauriceville and Varkud plasmids are retroid elements that propagate in the mitochondria of some Neurospora spp. strains. Previous studies of endogenous reactions in ribonucleoprotein particle preparations suggested that the plasmids use a novel mechanism of reverse transcription that involves synthesis of a full-length minus-strand DNA beginning at the 3' end of the plasmid transcript, which has a 3' tRNA-like structure (M. T. R. Kuiper and A. M. Lambowitz, Cell 55:693-704, 1988). In this study, we developed procedures for releasing the Mauriceville plasmid reverse transcriptase from mitochondrial ribonucleoprotein particles and partially purifying it by heparin-Sepharose chromatography. By using these soluble preparations, we show directly that the Mauriceville plasmid reverse transcriptase synthesizes full-length cDNA copies of in vitro transcripts beginning at the 3' end and has a preference for transcripts having the 3' tRNA-like structure. Further, unlike retroviral reverse transcriptases, the Mauriceville plasmid reverse transcriptase begins cDNA synthesis directly opposite the 3'-terminal nucleotide of the template RNA. The ability to initiate cDNA synthesis directly at the 3' end of template RNAs may also be relevant to the mechanisms of reverse transcription used by LINEs, group II introns, and other non-long terminal repeat retroid elements.
