The aim of this work was to study the ultrastructure of the internal pyramidal neurons of the neocortex of 5-day-old rat pups after antenatal alcoholization. The studies were carried out on female outbred white rats with an initial weight of 230 ± 20 g and their offspring. Prenatal alcoholization causes deep and varied ultrastructural changes in pyramidal neurons in the neocortex of 5-day-old rats. Moreover, these violations of direct action not only as a consequence of the damaging effect of alcohol, its metabolite acetehyde or the oxidative stress they cause on the membranes and organelles of neurons during embryogenesis, but also as a violation of the normal "program" of development" of neurons in the cortex.

In modern theoretical and practical biology and medicine, the key problem of research is to reveal the patterns of structural and functional organization of the human and animal body at different stages of development. Literature sources provide very limited data on the organization of the intermediate lobe of the adenohypophysis in humans, leaving insufficiently studied the ultrastructural state and activity of melanotropic cells of the intermediate lobe of the adenohypophysis in animals of different ages, including rats. The issues of the intermediate pituitary gland functioning remain important and little studied, especially in the period of the beginning of the optic-thalamic system. The purpose of the research was to study the ultrastructural changes of melanotropic cells of the intermediate lobe of the adenohypophysis in rats of different ages in the norm. Material and methods. In accordance with the purpose of the study we conducted the experiment on 30 nonlinear white male rats of different ages: 14-, 45- and 90-day-old. The animals were kept in the vivarium in equivalent conditions. The keeping and using of animals was carried out in accordance with the provisions of the "General Ethical Principles of Animal Experiments", approved by the IV National Congress of Bioethics. While examining the intermediate pituitary gland of intact rats on the electron microscope, the material was fixed in 2.5% solution of glutaraldehyde on phosphate buffer with fixation in 1% solution of osmium tetroxide according to Caulfield. It was dehydrated in alcohols of increasing concentration (70%, 80%, 90%, 100%) and acetone, poured into a mixture of epon-araldite. Semi-thin sections were made from the obtained blocks, which were stained with toluidine blue. Results and discussion. At the ultrastructural level in the intermediate lobe of the adenohypophysis of 14-day-old intact rats, several cell types could be identified that differed in the number and size of secretory granules. The ultrastructure of glandular cells of 45-day-old rats had no significant differences compared with 14-day-old animals. Secretory granules of different sizes and electron densities were observed in the cytoplasm of melanotropic cells. It was often possible to see a glandular cell with numerous secretory granules in one part of the cytoplasm, while in another they were virtually absent. Most melanotropocytes were characterized by slightly compacted mitochondria, but their numbers were slightly higher than in 45-day-old rats. The nuclei of most cells were large, oval in shape with a clear structure of nucleoli and their ribosomal component. The latter represented groups of ribosomes that were collected in osmophilic complexes. The amount of heterochromatin exceeded euchromatin and it was located mainly in the membrane with areas of rarefaction in the pore area Conclusion. In 14-day-old intact male rats, the intermediate lobe of the adenohypophysis was presented as a formed functionally active organ. In the ultrastructure of the cytoplasm of melanotropes there was a moderate development of organelles, and judging by the number and size of secretory granules, as well as the density of their content, we can assume that all these cells differed from each other in their functional activity. In 45-day-old intact rats, accumulation of secretory granules was observed in the cytoplasm of melanotropes, especially near the nucleus, which indicated an increase in melanocyte-stimulating hormones synthesis with age. The ultrastructural state of the cytoplasm and nucleus also indicated an increase in functional activity. The ultrastructure of the intermediate lobe cells of the adenohypophysis of rats at the age of 90 days differed from that of younger animals by signs of different functional activity of individual melanotropic cells

Disruption of the metabolism of the essential amino acid methionine causes the syndrome of hyperhomocysteinemia. This pathological condition is associated with the risk of developing a number of diseases, including chronic liver disease. The mechanisms of liver tissue damage in hyperhomocysteinemia remain poorly understood and require more detailed study. The aim of the study is to establish the features of submicroscopic changes in the liver structure of old rats with hyperhomocysteinemia. The experimental study was performed on 22 white nonlinear old (24-26 months) male rats, which were divided into a control group and an experimental group. A model of persistent hyperhomocysteinemia was created by administering to rats of experimental group of thiolactone homocysteine at a dose of 200 mg/kg body weight intragastrically for 60 days. The study of ultrastructural changes in the lungs of rats was performed using an electron microscope PEM-125K. At experimental hyperhomocysteinemia in a liver of old rats there are changes in all structural components. Mitochondrial destruction and edema were observed in the vascular endothelium. Organelles have an enlightened matrix, a reduced number of cristae. A significant content of destructively altered mitochondria in endothelial cells indicates a failure of adaptation mechanisms. Erythrocyte sludges are observed in the lumens of the sinusoids. The number of fat-accumulating cells decreases, which indicates their transformation into fibroblasts and leads to the growth of collagen fibers, expansion of the sinusoidal spaces and the development of stromal fibrosis.

Background: In recent years, attention of the scientific community has been drawn to the study of the role of homocysteine in the pathology of diseases in general and the respiratory system in particular. Violations of the synthesis and disposal of homocysteine are the cause of its excessive accumulation in the body, which subsequently leads to damage to all organs and systems. Methods: We conducted this study on 64 white non-linear rats of 6-8 and 24-26 months old rats, which were divided into two control and experimental groups during the experiment. The study of ultrastructural changes in the lungs of rats was performed using an electron microscope. Results: The results of the conducted study show that all the animals of the experimental group, corresponding to 32 (50%) cases, possess conspicuous morphological changes in the structure of the lungs. : The specific features of mature animals were the narrowing of the alveolar space of the part of the alveoli and the increase in the number of macrophages in them. Partial desquamation of capillary endothelial lining and a slight increase of collagen fibers in interalveolar membranes were observed compared to control animals. A characteristic feature for older animals was the growth of connective tissue, predominantly collagen fibers, which led to pneumosclerosis. The thickening of the aerogemic barrier was also detected, and the endothelial lining was intermittent or desquamated. Conclusion: Mature animals of the experimental group were characterized by activation of inflammatory processes, oxidative stress, due to increased production of macrophages and, as a consequence, the launch of the humorous link of immunity, while in old rats were determined by fibrosis, disorders of the trophic and gas metabolism, as well as damage to the endothelium part of lungs.

Two month, eight month and two year old rats were treated with 10 or 20 mg/kg of E. Coli endotoxin I. P. The eight month old rats proved most resistant to the endotoxin. During fixation the aorta, carotid artery, basil arartery of the brain, coronary vessels of the heart, inner surfaces of the heart chambers, heart and skeletal muscle, lung, liver, kidney, spleen, brain, retina, trachae, intestine, salivary gland, adrenal gland and gingiva were treated with ruthenium red or alcian blue to preserve the mucopolysaccharide (MPS) coating. Five, 8 and 24 hrs of endotoxin treatment produced increasingly marked capillary damage, disappearance of the MPS coating, edema, destruction of endothelial cells and damage to the basement membrane in the liver, kidney and lung.

The importance of radiation damage to the skin and its vasculature was recognized by the early radiologists. In more recent studies, vascular effects were shown to involve the endothelium as well as the surrounding connective tissue. Microvascular changes in the mouse pinna were studied in vivo and recorded photographically over a period of 12-18 months. Radiation treatment at 110 days of age was total body exposure to either 240 rad fission neutrons or 855 rad 60Co gamma rays. After in vivo observations in control and irradiated mice, animals were sacrificed for examination of changes in vascular fine structure. Vessels were selected from regions of specific interest that had been identified on photomicrographs. Prominent ultrastructural changes can be attributed to aging as well as to radiation treatment. Of principal concern were determinations of ultrastructural changes associated with venous dilatations, segmental arterial stenosis and tortuosities of both veins and arteries, effects that had been identified on the basis of light microscopic observations. Tortuosities and irregularly dilated vein segments were related to both aging and radiation changes but arterial stenosis was observed only in irradiated animals.

It has been previously demonstrated that DMBA-induced rat mammary carcinoma originates in the terminal end bud (TEB) of the mammary gland by proliferation of intermediate type cells (1). The earliest lesion identified is the intraductal proliferation (IDP), which gives rise to intraductal carcinomas. These evolve to cribriform, papillary and comedo types (2). In the present work, we report the ultrastructural changes that take place in the IDP for the formation of a cribriform pattern.Fifty-five-day-old Sprague Dawley virgin female rats were inoculated intra- gastrically with 20 mg 7,12-dimethylbenz(a)anthracene (DMBA) in 1 ml sesame oil. Non-inoculated, age-matched females were used as controls. Mammary glands from both control and experimental rats were removed weekly from the time of inoculation until 86 days post-inoculation. The glands were fixed and processed for electron microscopy (2).The first change observed in IDP's was the widening of intercellular spaces and the secretion of an electron dense material into these spaces (Fig. 1).

The physiological, biochemical, and ultrastructural aspects of the symbiotic relationship between the Chlorella-like algae and the hydra have been intensively investigated. Reciprocal cross-transfer of the Chlorellalike algae between different strains of green hydra provide a system for the study of cell recognition. However, our attempts to culture the algae free of the host hydra of the Florida strain, Hydra viridis, have been consistently unsuccessful. We were, therefore, prompted to examine the isolated algae at the ultrastructural level on a time course.

The effects of heat stroke on hepatic structure were studied in 32 Bantu patients who worked underground in the Transvaal and Orange Free State Gold Mines.Judging from biochemical and morphologic findings, liver damage is an invariable complication of heat stroke. In the milder cases (90 per cent) raised enzyme levels, bromsulphalein retention, and increased prothrombin times were the most common abnormalities.

The widespread contamination of the environment with PCB, a compound used extensively by industry in hydraulic and heat transfer fluids as well as plasticizers and solvents in adhesives and sealants, has resulted in detectable tissue levels in a large portion of the human population, domestic animals, and wildlife. Intoxication with PCB produces severe hepatic necrosis, degeneration of lymphoid tissues and kidney, skin lesions, decreased reproductive performance, reduced feed efficiency, and decreased weight gain. PCB also has been reported to reduce the binding of thyroid hormone to serum proteins and enhance the peripheral metabolism of thyroxine with increased excretion of thyroxine-glucuronide in the bile (Bastomsky, Endocrinology 95: 1150-1155, 1974).The objectives of this investigation were (1) to investigate the histopathologic, histochemical, and ultrastructural changes in thyroid FC produced by the acute (4 week) and chronic (12 week) administration of low (50 ppm) and high (500 ppm) doses of PCB to rats, (2) to correlate these alterations to changes in serum immunoreactive thyroxine concentration, and (3) to investigate the persistence of the effects of PCB on the thyroid gland.
