Effect of Ethanolic Extract of Seeds of Solanum torvum in Acetic Acid induced Ulcerative Colitis in Male Wistar Rats

Abstract Objectives Vanadium has been reported to possess relevant therapeutic properties such as anti-diabetic and anti-tumoral. This study aimed at determining the effects of vanadium on experimentally induced colitis in rats. Methods Forty-five male Wistar rats (103 ± 3.90 g, n=15) were used for this study and were divided into three groups. Group 1 (Untreated control) had nothing added to their drinking, while groups 2 and 3 received sodium metavanadate at a dose of 50 and 200 mg/L respectively in their drinking water for 10 weeks. Colitis was thereafter induced by intra colonic administration of 1.50 mL of 6% acetic acid. Animals were sacrificed on day 0 (pre-induction), three- and seven-days post induction. Blood samples were collected for haematological variables and the distal 8 cm of the colon was collected for macroscopic, histological and biochemical (malondialdehyde-MDA, superoxide dismutase-SOD, catalase-CAT, glutathione peroxidase- GPx and nitrite concentration- NO) assessment. Results Low dose vanadium proved beneficial in ameliorating acetic acid-induced colitis by improving both histopathological and haematological changes. Gross observation showed a faster healing rate in vanadium treated groups (50 and 200 mg/L) compared with untreated control at day 3 (40 and 26.20 vs. 2.50%) and day 7 (80 and 66.70 vs. 42%) respectively. Vanadium also appears to exert its beneficial effects on acetic acid-induced colitis via up regulation of antioxidant enzymes (SOD, CAT, GPx) and NO while decreasing the over production of MDA. Conclusions Vanadium at small concentration functions as an essential trace element and may be able to promote healing process during ulcerative colitis.

Enhydra fluctuations an edible semi-aquatic vegetable plant are widely used in the traditional system of medicine. Ethanolic extract of Enhydra fluctuans was screened for analgesic & anti-inflammatory activity. Analgesic activity was evaluated by the hot plate method and anti-inflammatory activity was evaluated by formalin induce paw edema in Wistar rats (male). Ethanolic extract dose of 300 mg/kg reduce formalin induce paw inflammation and also increased the pain threshold in rats evidenced by hot plate method. The experimental results concluded that Enhydra fluctuans have significant analgesic and anti-inflammatory activity may due to flavonoid and phenolic compound content.

Syzygium campanulatum and Syzygium aromaticum contains antioxidant components suchas flavonoids, phenolic, and terpenoids. May have hepatoprotective properties in reducing SGPT and SGOT activity. This research wants to determine the potency of hepatoprotective of ethanolic extract of Syzygium campanulatum (Korth) and Syzygium aromaticum leaf compared with curcuma tablets. This research uses 24 male Wistar rats divided into 6 groups: I, II, III (as a normal, induction, and compared control), group IV, V, VI were treated 105, 210, and 420 mg/kg BW respectively. The study was conducted for 9 days. After 7 days of treatment, treated groups were exposed by hepatotoxic dose of paracetamol (2000 mg/kg BW). The SGPT and SGOT activity of all groups was measured by enzimatic assay. The result can be concluded that Syzygium campanulatum extract was found to be active as hepatoprotective agent with 210 mg/kg BW dosage (SGPT 21.76 ± 3.98 U/L and SGOT 7.32±6.74U/L) as eff ective as with the curcuma tablets (SGPT 23.91 ± 4.41 U/L and SGOT 14.12±5.37 U/L) and the hepatoprotective activity of Syzygium campanulatum extract at a dosage 420 mg/kg BW better than curcuma tablets (SGPT 12.43 ± 6.51 U/L and SGOT 6.64 ± 5.88 U/L). The study evaluated the pharmacognostic characters and toxicity of the aqueous ethanolic extract of Bambusa vulgaris leaf in male wistar rats. The microscopy of the leaf revealed diagnostic characters such as anomocytic stomata, sinuous epidermal cells, numerous prisms of calcium oxalate crystals and covering trichomes. Histopathological examination revealed no significant adverse effects on the lungs, kidneys and the spleens after fourteen days oral administration of the extract at 250 and 500 mg/kg doses. Haematological evaluation however revealed a significant 31% reduction (p<0.05) in packed cell volume and a significant 31% increase (p<0.05) in white blood cell count at 500 mg/kg. The results suggest that administration of B. vulgaris extract may possess low toxicity when used.Keywords: Bambusa vulgaris, Toxicity, Microscopy, Standardization, Histology

Periodontitis adalah inflamasi kronis yang disebabkan oleh bakteri periodontopatogen. Pada periodontitis terjadi peningkatan infiltrasi neutrofil yang berfungsi untuk memfagositosis bakteri yang menginfiltrasi jaringan gingiva. Kulit manggis mempunyai bahan aktif yaitu xanton yang mempunyai efek antiinflamasi dengan jalan menghambat sintesis PGE2 sehingga akan menurunkan infiltrasi sel inflamasi. Penelitian ini bertujuan untuk mengetahui infiltrasi sel neutrofil pada periodontitis setelah pemberian ekstrak etanolik kulit manggis. Tikus wistar jantan sebanyak empat puluh delapan ekor diinduksi periodontitis dengan ligasi pada daerah subgingiva gigi anterior rahang bawah selama 7 hari. Setelah hari ke-7, ligasi dilepas selanjutnya tikus dibagi menjadi 4 kelompok perlakuan yaitu ekstrak kulit manggis 30 mg/kg BB dan 60 mg/kg BB, ibuprofen dan saline. Tikus dikorbankan pada hari ke-1, 3, 5 dan 7 setelah perlakuan. Jaringan pada bagian anterior rahang bawah ditanam dalam paraffin dan dilakukan pemotongan serial kemudian diwarnai dengan hematoxylin eosin. Jumlah neutrofil dihitung di bawah mikroskop dengan perbesaran 400x. Data jumlah neutrofil dianalisa dengan Two Way Anova. Hasil Two Way Anova menunjukkan perbedaan yang bermakna rerata sel neutrofil antar kelompok perlakuan (p<0,05) mengindikasikan pemberian ekstrak etanolik kulit manggis berpengaruh terhadap jumlah sel neutrofil. Dapat disimpulkan bahwa pemberian ekstrak etanolik kulit manggis mampu menurunkan infiltrasi sel neutrofil pada tikus yang diinduksi periodontitis.Neutrophil infiltration in rats with periodontitis after the granting of Ethanolic Extract Skin Mangosteen. Periodontitis is a chronic inflammatory disease caused by periodontopathic bacteria. When periodontitis occurs are followed by neutrophil infiltration. Mangosteen rind contains xanthone, an anti-inflammatory substance which inhibits the synthesis of PGE2 and therefore reduces inflammatory cells infiltration. This research aimed to study neutrophil cells infiltration in experimental-induced periodontitis rats after mangosteen rind ethanolic extract administration. Forty-eight male wistar rats were induced the periodontitis by putting silk ligature subgingivally around the neck of the anterior lower teeth for seven days. After the ligation had been taken out, the rats were divided into four groups and treated orally with mangosteen rind extract 60 mg/kg BB, 30 mg/kg BB, ibuprofen, and saline. The rats were sacrificed on the 1st, 3rd, 4th, seventh day after the treatment. Their anterior lower jaws were processed for paraffin embedded tissue, cut serially and stained with hematoxylin-eosin. The neutrophil cells were observed and counted under the microscope (400x). The data were analyzed using Anova. Anova result showed a significant difference among group (p<0,05) indicating that mangosteen rind ethanolic extract affected the number of neutrophils. In conclusion, mangosteen rind ethanolic extract reduced the number of neutrophil infiltration in periodontitis rats.

Aim: The study was aimed at evaluating the effects of ethanolic extract of Sesamum indicum seeds on sperm morphology and viability in ketoconazole induced testicular damage in Wistar rats. Methods: Forty eight adult male Wistar rats were divided into eight Groups of six rats each. Groups 1,2,3,4,5,6,7 and 8 were administered 1ml of normal saline, 100 mg /kg body weight (bwt) of Ketoconazole only, 500 mg/kg bwt of ethanolic extract of sesamum indicum (EESI) seeds then100 mg/kg bwt of Ketoconazole, 100 mg/kg bwt of Ketoconazole then 250 mg /kg body weight of EESI seeds,100 mg/kg bwt of Ketoconazole then 500 mg/kg bwt of EESI, 100 mg/kg body weight of Ketoconazole then 0.36 mg/kg body weight of Mesterolone, 0.36 mg/kg body weight of Mesterolone, 500 mg/kg body weight of EESI respectively. All administrations were carried out orally once per day. The sperm smears were collected for semen analyses. Result: A significant increase in the abnormalities of sperm morphology and viability respectively were noted in the group administered ketoconazole alone (Group 2) compared with other experimental Groups (P ≤ 0.05). Conclusions: The administration of ethanolic extract of Sesamum indicum appeared to have minimized the damages and sperm deaths caused by ketoconazole which may be due to its androgenic properties.
