The values of heritability coefficients of parental genotypes were determined for such characters as "frequency of callus formation", "frequency of somatic embryogenesis","frequency of plant regeneration" under normal and stressful in vitro conditions in simple maize F1 hybrids. The highest value of the coefficient of heritability of maternal genotypes in traits studied in vitro was found for “frequency of somatic embryogenesis” at low temperatures. The variability and heritability of characters during the initial stages of plant development under the actionof low temperatures, as well as quantitative characteristics of the plant and male gametophyte, were evaluated. Hybrids in which the inbred lines A239 and 092 were used as maternal genotypes showed under stress higher indices of the studied traits both invitro conditions and at the seed level. For the characters “number of grains per row” and “plant height”, the highest heritability coefficients of the interaction of maternal and paternal genotypes were noted. Hybrids A239xP502 and Co125xMK159 were selected by the higher values of the studied quantitative traits. Hybrid combinations with significantly high meanings of the most of the studied characters can be used to create cold-resistant and productive corn hybrids

In the conditions of the south of the Nonchernozem Zone, intraspecific hybridization of winter soft wheat was carried out. When selecting parental forms used in crossing, special attention was paid to such economically valuable traits as productive tilling capacity, the number of grains and the mass of grain per spike. The true (Htru) and hypothetical heterosis (Hhyp) on the basis of “plant height”, “length of the lower internode”, “length of the upper internode”, “general tilling capacity”, “productive tilling capacity”, “spike length”, “number of spikelets”, “number of grains” and “grain weight per spike” were determined. It was established that the inheritance of quantitative traits in hybrids of the first generation was distinguished by the complex nature of distribution by types. Studies have made possible to identify 5 hybrid combinations F1, in which the effect of heterosis manifests itself simultaneously on five quantitative characteristics (ear length, number of spikelets per ear, number of grains per ear, grain weight per ear, mass grain from a plant).

<div><table cellspacing="0" cellpadding="0" align="center"><tbody><tr><td align="left" valign="top"><p>Since coconut is   one of the most recalcitrant species to generate <em>in vitro</em>, it is   necessary to study in detail about the cellular changes that occur during   somatic embryogenesis to enhance our knowledge about this phenomenon. In the   present study, coconut plumular tissues, the shoot meristem including leaf   primordia, were used as explants for <em>in vitro </em>regeneration studies.   Histological studies were carried out in different stages of plumule culture.   No noticeable growth was observed in 15 days old cultures. After 30 days,   meristematic cells could be identified. Abundance of meristematic cells,   foremost to the development of callus structures, was observed after 45 days.   After 75 days, globular friable calli were formed and histological studies   revealed the presence of meristematic centers which eventually formed somatic   embryos. The histological study of matured somatic embryos formed after 120   days of callus initiation showed a clear meristematic zone of parenchyma   cells, surrounded by vascular bundles. Histological studies, carried out for   certain abnormalities like compact calli, abnormal somatic embryoids with   rudimentary shoots and multiplied roots, revealed the presence of intact   cotyledonary leaves which seemed to inhibit the apical meristem development   of somatic embryoids. The presence of vascular bundles in the early stages of   callus formation might lead to the direct formation of meristemoids. These   results could aid future studies leading to enhanced control of the somatic   embryogenic process and greater efficiency of somatic embryo and plantlet   formation in coconut.</p></td></tr></tbody></table></div>

AbstrakGarcinia mangostana L. dikenal dengan sebutan queen of the tropical fruits. Buah manggis terbentuk secara apomiksis yang bersifat rekalsitran. Salah satu cara perbanyakan tanaman manggis adalah dengan teknik kultur in vitro melalui embriogenesis somatik. Embriogenesis somatik manggis dilakukan dengan pembentukan kalus terlebih dahulu. Penelitian ini bertujuan untuk menentukan konsentrasi terbaik BAP dan madu secara tunggal serta kombinasinya dalam pembentukan embriogenesis somatik pada kalus biji manggis asal Bengkalis. Penelitian ini menggunakan rancangan acak kelompok (RAK) dengan pemberian konsentrasi BAP (3 dan 7 mg/L) dan madu (3, 6, dan 9 mL/L), secara baik tunggal maupun kombinasi, pada media Murashige-Skoog (MS) dengan 3 ulangan. Hasil penelitian menunjukkan bahwa pemberian BAP dan madu dalam seluruh perlakuan tersebut berpengaruh terhadap pembentukan fase-fase embriogenesis somatik kalus manggis. Konsentrasi terbaik dalam pembentukan fase embriogenesis somatik diperoleh dari perlakuan 3 mg/L BAP + 9 mL/L madu dengan presentase pembentukan kalus 100%, waktu muncul kalus 10,67 hst, volume kalus 1,33 dan adanya fase embriogenesis somatik berupa globular, hati, dan torpedo.Abstract Garcinia mangostana L. was known as the queen of the tropical fruits. Mangosteen was formed by apomixis which is recalcitrant. One of the methods of mangosteen propagation is by using a tissue culture technique through somatic embryogenesis. Mangosteen somatic embryogenesis occurs preceded by callus formation. This study aimed to determine the best concentration of BAP and honey in single as well as in combination for the formation phase of somatic embryogenesis in the callus of mangosteen from Bengkalis. The study used a randomized block design with the addition of BAP (3 and 7 mg/L) and honey (3; 6; and 9 mL/L) either single or combination in Murashige-Skoog (MS) medium with 3 replications. The results of this study indicated that the addition of BAP and honey in all treatments affected the phases of somatic embryogenesis of  mangosteen callus. The best concentration in the formation of somatic embryogenesis was obtained from, the treatment of 3 mg/L BAP + 9 mL/L which produced 100% of callus formation, with callus emergence time of 10.67 days after plantation, callus volume of 1.33 and the presence of somatic embryogenesis in the form of globular, heart, and torpedo.

The embryogenesis (SE) has important role for genetic engineering of cassava (Manihot esculenta Crantz). However, the success of SE induction depend on plant growth regulator s (PGR)s and treatment enriched in induction media.  This experiment tried to induce callus formation of cassava from several in vitro explants: immature leaf, apical bud, and internode; and to develop somatic embryogenesis of cassava in several media enriched with tyrosine and copper sulphate (CuSO4) added into media enrich with picloram as treatment.  Different response of explants source to callus induction treatment from those three varieties in callus induction as well as friable callus formation were found in this experiment. The best medium to induce varied with variety; MS media supplemented 12 mg/L picloram + 0.5 mg/L CuSO4 was the best for “Adira 4” and  half MS and half GD media supplemented 12 mg/L picloram + 100 mg/L tyrosine for “Malang 6”.  All treatments resulted somatic embryo which developed indirectly and in morphologically normal somatic embryos

In Vitro Callus and Plant Regeneration Rate of Tarabas Rice on Several Concentrations of 2,4-D The Agricultural Research and Development Agency and the West Java Provincial Government are developing new superior varieties with Japonica rice standards, namely the Tarabas variety. However, the equivalence of somatic embryogenesis ability of Tarabas rice with original Japonica variety has not been reported. In this study, the frequency of callus regeneration of Tarabas vs Hwayoung rice varieties was compared. Induction of callus from mature embryos with several concentrations of 2,4-D showed the same extent of callus formation in both rice varieties. Callus induced by 1 ppm of 2,4-D showed the higher rate of shoot formation. On the other hand, percentage of callus formation of Tarabas rice was not affected by the increase of 2,4-D concentrations and was able to show 100% regeneration rate  at the fourth week in the regeneration medium, although the shoot growth was not as fast as those from medium with 1 ppm 2,4-D. Therefore, these results suggest that Tarabas variety has a somatic embryogenesis capacity equivalent to that of japonica rice and has the potential as research objects in the field of biotechnology. Badan Penelitian dan Pengembangan Pertanian serta Pemerintah Provinsi Jawa Barat mengembangkan varietas unggul baru dengan standar padi Japonica yaitu varietas Tarabas. Namun, kesetaraan varietas Tarabas dengan varietas Japonica asli dalam kemampuan embriogenesis somatik belum dilaporkan. Penelitian ini membandingkan respons kultur jaringan antara beras Tarabas dan padi Japonica varietas Hwayoung. Induksi kalus dari embrio matang dengan beberapa konsentrasi 2,4-D menunjukkan respons pembentukan kalus yang sama pada kedua varietas padi. Kalus yang diinduksi 1 ppm 2,4-D menunjukkan laju pembentukan tunas yang lebih tinggi. Di sisi lain, kalus Hwayoung yang diinduksi konsentrasi 2,4-D yang lebih tinggi menunjukkan penghambatan dalam pembentukan tunas. Di lain pihak, pembentukan kalus padi Tarabas tidak terpengaruh oleh peningkatan konsentrasi 2,4-D dan mampu menunjukkan 100% laju regenerasi tanaman pada minggu keempat di media regenerasi walaupun pertumbuhan tunas tidak secepat pada perlakuan 1 ppm 2,4-D. Karena itu, hasil penelitian ini menunjukkan bahwa varietas Tarabas memiliki kapasitas embriogenesis somatik yang setara dengan padi japonica dan padi Tarabas mempunyai potensi sebagai obyek penelitian di bidang bioteknologi.

Abstract The oil palm (Elaeis guineensis Jacq.) is a perennial woody oil crop in the Arecaceae family. Oil palm is well known for a long regeneration time, therefore, in vitro propagation received great enthusiasm from oil palm industries. Somatic embryogenesis (SE) has become one of the most promising clonal propagation techniques in recent times. This study was aimed to determine a protocol for micropropagation somatic embryogenesis from female inflorescences explants of oil palm. The explant used was obtained from the female inflorescences of the oil palm 2.5 years old Tenera variety. The basal media used was Y3 with the addition of the hormone 2,4-D with different concentrations (33,66,99 and 132 mg/L). Callus initiation was formed in 99 mg/L and 132 mg/L 2,4 – D concentration with the basal area and the percentage of callus formation is 31,25% and embryogenic callus was formed from primary callus development in the basal area of female florescence. Embryo somatic induction with cell suspension culture, because the liquid medium is more efficiently used in commercial-scale propagation. The embryogenic callus phase which is generally used in liquid medium is the nodular phase because it is still meristematic so that the potential for cell division is still high and can increase the percentage of embryogenic callus.

The induction and control of in vitro somatic embryogenesis is dependent of a number of factors. The genotype of the mother plant donor of explants and the kind of auxin play essential roles in conferring embryogenic competence. Once obtained somatic embryos may be encapsulated in alginate originating synthetic seeds that can be stored at low temperatures. Considering this the present work aimed at the assessment of five different genotypes, five periods of 2,4-D shock (20 muM for 1, 2, 4, 8 and 16 weeks) (), and different substances supplemented to sodium alginate to obtain synthetic seeds. Zygotic embryos of Feijoa sellowiana were inoculated in basal LP medium (von Arnold and Erikson, 1981) supplemented with Morel’s vitamins, sucrose (3%) and phytagel (0.2%). The highest rates of somatic embryogenesis induction were observed in the genotypes 50-4 and 101. Shock of two weeks in 2,4-D conferred embryogenic competence in the same levels obtained with the shock of 8 weeks. Histological evaluations revealed the direct origin of the somatic embryos from the epidermic surface of cotyledons. Synthetic seeds containing MS salts and sucrose resulted in higher rates of contamination than capsules free of these substances. KNO3 (100 mM) resulted in the opening of 81.2% of the capsules as compared to 0% of opening in the treatment with water.

This long term study demonstrates for the first time that it is possible to propagate embryogenic Vigna trilobata and to subsequently initiate the differentiation of embryos into complete plantlets. Initiation of callus was possible on 2,4-D. Somatic embryos differentiated on modified MS basal nutrient medium with 1.0 mg/l  of 2,4-D and 0.5 mg/l  of Kn. Sustained cell division resulted in globular and heart shape stages of somatic embryos. Transfer of embryos on to a fresh modified MS basal medium with 0.5 mg/l of Kn and 0.5 mg/l of GA3 helped them to attain maturation and germination. However, the propagation of cells, as well as the differentiation of embryos, were inhibited by a continuous application of these growth regulators. For this reason, a long period on medium lacking these growth regulators was necessary before the differentiation of embryos occurred again.
