One-pot, Multicomponent Synthesis under Microwave Conditions of New Imidazole and Acridine Benzamido Acetic Acid Derivatives

Starting from 4-aminohippuric acid (AHA), two novel series of heterocyclic compounds of pharmaceutical interest have been synthesised by one-pot, multicomponent reactions under microwave conditions. Reaction of AHA with various aldehydes and benzil in (1:1) EtOH/acetic acid in the presence of ammonium acetate yielded 1,2,3,5-tetra-arylated imidazole derivatives, whereas a solvent-free reaction of AHA with various aldehydes and dimedone (2 equiv.) in the presence of p-TSA yielded 4,9-diarylated 1,8-dioxo-decahydroacridine derivatives. These reactions produced good yields in short reaction times with an easy work-up, and purification of products was achieved by simple recrystallisation.

A combinatorial library of benzylpyrazolyl coumarin derivatives have been synthesized by a green one-pot four-component reaction between aryl hydrazine/hydrazine hydrate (1), ethyl acetoacetate (2), aromatic aldehydes (3) and 4-hydroxycoumarin (4) catalyzed by niobium pentachloride with silver salt under solvent-free conditions has been developed. Experimental simplicity, simple work-up procedure and solvent-free reaction condition at room temperature are important features of the present protocol.

: A green and efficient protocol was developed for the one-pot three-component synthesis of novel 2-(4-hydroxy-2-oxo-2H-chromen-3-yl)-2-(arylamino)-1H-indene-1,3(2H)-dione derivatives by the reaction of 4-hydroxycoumarin, ninhydrin and aromatic amines in the presence of guanidine hydrochloride as an organocatalyst under solvent-free conditions. The present approach offers several advantages such as low cost, simple work-up, short reaction times, chromatography-free purification, high yields and greener conditions.

Silica gel-supported polyphosphoric acid (PPA-SiO2) was found to be highly efficient, eco-friendly and recyclable heterogeneous catalyst for the synthesis of 2,4,6-triarylpyridines through one-pot three-component reaction of acetophenone, aryl aldehydes, and ammonium acetate under solvent-free conditions. This method has several advantages, such as simple procedure with an easy work-up, short reaction times, and high yields. Furthermore, the catalyst could be recycled after a simple work-up, and used at least three times without substantial reduction in its catalytic activity.

Highly efficient, ecofriendly, and improved protocol for the synthesis of 1,8-acridinediones has been developedviaone-pot multicomponent condensation of 1,3-cyclohexanedione/dimedone, aromatic aldehydes, and ammonium acetate utilizing poly(4-vinylpyridinium)hydrogen sulfate as catalyst in aqueous medium. Excellent yields in shorter reaction time, simple work-up procedure, easy recovery, and reusability of the catalyst are attractive features of this green protocol.

A simple one-pot four-component synthetic method was reported for the preparation of tetrasubstituted imidazole derivatives from benzil, aromatic aldehydes, primary amines and ammonium acetate in the presence of Preyssler-type heteropoly acid catalyst. This method was proved to be eco-benign, easy work-up, convenient, relatively short reaction times and the products were isolated with high yields.

Coaled carbon-based solid acid (CCBSAC) is reported as a new, efficient and recyclable catalyst for the regioselective multicomponent synthesis of dihydropyrimidin-2(1H)-ones from aldehydes, β-ketoesters and urea or thiourea under solvent-free conditions. The structure and thermal stability of CCBSAC which resulted from ultra-clean coal resources mainly were characterized by IR, XRD, and TG. Nineteen dihydropyrimidin-2(1H)-ones examples were prepared and this catalyst could recycle up to 4 consecutive runs without losing its efficiency. The high yields of products, short reaction times, ease of work-up and clean procedure will make the present method a useful and important addition to the previous methodologies for the synthesis of dihydropyrimidin-2(1H)-ones.

The microwave-promoted one-pot multicomponent synthesis of substituted pyrrolidinols using a bifunctional sugar-derived hydroxy-[gamma]-lactone component of amine component or cyanide group is reported. Clean reaction profile, easy work-up procedure, excellent yields and short reaction times are some remarkable features of this method.This modular approach features the in situ-generation of iminium ions intermediate that allows the diastereoselective assembly of the diverse pyrrolidinones efficiently under microwave irradiation has been used.

Abstract Silica ammonium acetate (SiO2-NH4OAc) was applied as an inexpensive, practical and heterogeneous catalyst for the preparation of new and known dihydropyrazolo[4',3':5,6]pyrano[2,3-d]pyrimidine-5,7-diones via one-pot four-component reaction of hydrazine hydrate, ethyl acetoacetate aldehydes and barbituric acid/dimethyl barbituric acid under solvent-free conditions. Silica supported ammonium acetate (SiO2-NH4OAc) was prepared according to easy procedure under ambient condition. After completion of the reaction, the catalyst was separated by filtration and reused. So, recycling system, simple work-up, using non-toxic materials, excellent yields and short reaction times makes our research green and convenient for preparation of these classes of organic compounds.

<p>A simple, green, and efficient method for the synthesis of 2,4,5-triaryl-1<em>H</em>-imidazoles using <em>N</em>-bromosuccinimide (NBS) as a catalyst under solvent-free condition is described. The major advantages of the present method are: high yields, less reaction times, solvent-free conditions, easy purification of the products, environmental friendliness, and convenient operation.</p>

Silica supported molybdenum oxide (MoO<sub>3</sub>/SiO<sub>2</sub>) was found to be an efficient, eco-friendly and heterogeneous catalyst for the multicomponent reaction of aromatic aldehydes, dimedone and ammonium acetate or aromatic amines under solvent-free conditions to afford the corresponding 1,8-dioxodecahydroacridines  in high yields. The catalyst can be easily recovered and reused for several times without considerable loss of activity. Furthermore, the present method offers several advantages, such as an easy experimental and work-up procedures, short reaction times and good to excellent yields.
