This paper describes research on the application of tissue culture techniques to the micropropagation of interspecific rootstock ʹDocera 6ʹ. The experimental work was carried out in the period 2017-2018, in the in vitro propagation laboratory of the Fruit Growing Institute Plovdiv. Аxillary buds were employed as initial explantеs in two different seasons (March-May; September-October). The action of the mineral medium was studied in the multiplication stage. The best result was obtained on LS medium included BAP 0.5 mg/l and IAA 0.05 mg/l. Тhe obtained average multiplication rate is 3.08. The concentration of auxin applied to the basal medium influence the quality of the root system Treatment with high concentrations of IBA added to the rooting medium gives the best results (V5). Тhe influence of the season on growth and development of micropropagated ʹDocera 6ʹ rootstock during ex vitro acclimatization is also part of our research. The spring acclimatization gives better results than the autumn.

Abstract We present the data on proliferation corresponding to 10 years of continuous incubation in vitro of papaya shoots, and propose a reliable method for long-term micropropagation for papaya, using two types of explants: Microshoots from somatic embryos, and from axillary buds of papaya. Three different media were assayed. The proliferation medium (PPRM) allowed to maintain papaya shoots under continuous proliferation during 20 years, maintaining a consistent behaviour. Most of the shoots developed in PPRM rooted during the incubation, and after acclimated easily, maintaining the morphological characteristics of the parental plants, flowering and setting fruits normally. The PPRM medium consist in MS medium supplemented with NAA (0.1 mg l-1), BA (0.5 mg l-1), GA3 (0.5 mg l-1) and Adenine sulphate (40 mg l-1). The average multiplication rate was higher than 20 shoots per explant along the long-term assay. The elongation medium (PELM), was designed to recover shoots with a poor growth, and allowed the development of high quality shoots ready for rooting, and consist in a MS basal medium supplemented with NAA (0.1 mg l-1), Kin (0.5 mg l-1) and GA3 (1 mg l-1). The rooting medium (PROM) was designed to induce high quality roots from non-rooted shoots and consist in a half strength MS medium plus IBA (1mg l-1). On PROM, agar can be exchanged for expanded vermiculite. Acclimation took place inside an acclimatization tunnel under progressive hydric stress. After 4 weeks, the plant recovery rate was 90% for plants maintained under continuous proliferation during ten years.

A basal medium consisting of inorganic salts, 100.0 mg/L myo-inositol, 0.4 mg/L thiamine HCl, 100.0 mg/L casein hydrolysate, 30.0 g/L sucrose and 7.0 g/L agar, supplemented with IAA (1.0 mg/L), BAP (4.0 mg/L) and 2-iP (4.0 mg/L) will induce the regeneration of shoots from rootpieces of rutabaga after a 30-day culture period. Newly regenerated shoots were transferred onto a rooting medium consisting of basal medium supplemented with NAA at 0.5 mg/L. Roots were differentiated in 2–3 wk after which time the plantlets were transplanted into a soil mix.

Pyracantha coccinea is a thorny evergreen shrub native to southeast Europe to southeast Asia. It is a popular ornamental plant because of its showy bright red fruits and small white flowers. However, in vitro vegetative propagation of P. coccinea has not been studied. Nodal segments with one or two axillary buds (1 to 1.5 cm in length) were cut and disinfected in a solution of 0.1% (v/v) mercuric chloride (HgCl2) for 5 minutes, and proliferated on Murashige and Skoog (MS) basal medium supplemented with various concentrations 6-benzylaminopurine (6-BA). After 4 weeks, newly formed shoots were transferred to proliferation and rooting media containing various concentrations of indole-3-butyric acid (IBA). Establishment of axillary buds was significantly better with an establishing rate of 67% on basal MS medium augmented with 6.6 µm 6-BA. The best medium for proliferation of shoots was three-fourth basal MS supplemented with 1.5 µm IBA, with a proliferation rate of 3.4 axillary bud. The optimum rooting medium was one-fourth MS basal medium containing 93 µm IBA. Rooting of shoots was as much as 77%. Rooted plantlets were transferred to pots containing vermiculite:perlite:peat (6:1:2) and acclimatized to ambient greenhouse conditions with a 95% survival rate. This protocol can be used for in vitro propagation of P. coccinea.

Abstract The aim of the study was to develop an efficient micropropagation system for Rosa ‘Konstancin’, an interspecific hybrid between R. rugosa and R. beggeriana, whose fruits have high pro-health value. Shoot cultures were initiated from shoot buds collected in May and August from 15-year-old field-grown Rosa ‘Konstancin’ shrubs. The effect and interaction of different concentrations of phytohormones, sucrose and iron sources on in vitro initiation, multiplication and rooting of shoots were studied. The time of collecting explants from donor plants significantly affected the initiation of shoot culture of Rosa ‘Konstancin’. Considerably higher frequency of bud break (100%) was obtained in explants isolated in August as compared to those collected at the end of May (30%). All buds developed into single shoots after 2-4 weeks of growing on the basal Murashige and Skoog medium containing 2.2 µM BAP, 0.3 µM GA3 and 88 mM of sucrose. The highest multiplication rate (4.8 shoots/explant) in a 5-week period was obtained on MS medium containing 50% of nitrogen salts, 3.1 µM BAP, 0.9 µM GA3 and 58 mM sucrose. High rooting frequency (100%) and quality of rooted plantlets was obtained on a medium containing 0.5 µM IBA, 138 µM Fe-EDDHA and 88 mM sucrose. Fe-EDDHA had a beneficial effect on the growth and photosynthetic activity of Rosa ‘Konstancin’ plantlets, which were successfully acclimatized ex vitro, with a more than 90% survival rate.

AbstractKnautia sarajevensisis an endemic plant of the Dinaric Alps and is mainly distributed on Bosnian Mountains. Due to the quite large flower heads and easy maintenance, this plant has a potential use as a substitute ornamental plant forK. arvensisin perennial beds. The current study evaluated the germination process in different treatments in an attempt to suppress dormancy and increase germination rate, and to develop a successful protocol for micropropagation. An over 60% germination rate was achieved through cultivation of seeds on MS basal medium with reduced mineral nutrient composition and the absence of sucrose. On the other hand, a below 10% germination rate was achieved with untreated seeds. Suppression of apical dominance was achieved through application of high concentrations of kinetin, apical shoot decapitation or cultivation of shoots in liquid media. Overall, liquid cultures were more successful as a micropropagation system for this plant. Shoots spontaneously developed roots on multiplication treatments and were successfully acclimatized. Moreover, phenolic compound profile was analysed in the light of the possible medicinal potential of this plant. Variable amounts of total phenolic compounds as well as individual phenolics were recorded, according to treatment and solidification of media. An increase in rosmarinic acid content was reported for kinetin treatments and acclimatized plants comparing to mother plants in natural habitat. The present study shows that choice of cytokinin concentration, explant type as well as culture type influences not only shoot proliferation and apical dominance suppression but alsoin vitroproduction of phenolics.

Laurus nobilis L. is among the species which are most threatened by massive degradation in Morocco. The multiplication by seed or by cuttings gives very low percentages of recovery that is insufficient to meet the demand of growing market. In vitro culture proves to be a tremendous asset to solve this problem. Our work has focused on the study of seed germination of this species and its multiplication from microcuttings. Finally, we studied the ac-climatization ability of the plantlets resulting from this germination. The study of the germination, via the further measurement of the length of the aerial part and the roots and the number of axillary buds for nine weeks, showed that the MS basal medium was more efficient than media 1/2M.S and WPM. Among the eight tested hormones, IAA yielded the best growth of the plantlets. Hormonal combination of NAA and kinetin resulted into a per-centage of the greatest success in reaching 67 % micropropagation. The study also revealed that the MS basal medium in the presence of the IAA plants can acclimate most easily in two types of substrates with improved development in the peat alone.

Opuntia is one of the most widespread cacti, primarily due to their edible fruit and vegetable mass used as feed. The high demand for young plants of Opuntia made it necessary to find a rapid method of multiplication of the cactus, the safest method consisting in vitro micropropagation of species belonging to this genus. With aim of large production of plant material, a propagation system of three important prickly pear cactus cultivar (Opuntia ficus-indica) in Morocco was developed. Segments of healthy young cladode (containing one areole) were cultivated in Murashige and Skoog medium (MS) containing adenine sulfate (40 mg/1), monosodium phosphate (50 mg/l), sucrose (50 g/l), phytagel (0.3%) and benzyladenine (BA) at 22.2 μM, to start the process of micropropagation. In vitro-developed shoots from areoles were used as secondary explants to induce shoot development in the MS medium with 5 mg/l of BA. All of the three studied cultivars showed an important multiplication rate in this medium. ‘Sidi Ifni M’ (‘Moussa’) cultivar shows the greatest number of shoots followed by ‘Sidi Ifni A’ (‘Aissa’) and ‘Delahia’ 17.26, 14.12 and 12.13 respectively. Rooting of in vitro-generated shoots was achieved most efficiently on half-strength MS basal medium supplemented with 0.5 mg/l of indole-3-butyric acid (IBA) or IAA. Rooting frequencies were in the range from 95 to 100% and the highest mean number of root (19.1) was obtained with IBA for ‘Delahia’ cultivar. All micropropagated plants were transferred to greenhouse and all of them survived acclimatization process and showed good overall growth.

A method for rapid micropropagation of sea kale (Crambe maritima L.) was developed. Petiole explants placed in vitro on a medium containing 0.5 mg/L indoleacetic acid (IAA), 6.0 mg/L kinetin, and 1.5 mg/L benzylaminopurine developed callus within 15 days and shoots within 28 days. Nearly four adventitious shoots could be developed within 3 weeks by placing the initial shoot on media without IAA. To develop roots, the shoots were then transferred to the basal medium containing 0.1 to 1.0 mg/L indolbutyric or α-naphthaleneacetic acid. Rooted plantlets were obtained within 2 or 3 weeks. After an acclimatization period of 6 weeks in a greenhouse in unsterilized medium, the plantlets could be set outdoors.

The in vitro shoot multiplication of apple cv. Jonagold was tested on media containing benzyladenine, benzyladenine riboside or meta-topolin in different concentrations (from 0.0 to 5.0 mg l-1). The optimal concentration for the best multiplication varied according to the type of cytokinin. The highest multiplication rate (on average 6.9 and 5.9 new shoots per explant) was achieved using 5.0 mg l-1 meta-topolin or 2.0 mg l-1 benzyladenine riboside. The longest shoots were formed on media containing benzyladenine riboside at a concentration of 0.5 mg l-1. The length of newly developed shoots was strongly suppressed by high concentrations of different cytokinins, but the suppression effect of a high concentration of meta-topolin on shoot length was less than that of benzyladenine or benzyladenine riboside. In this study meta-topolin and benzyladenine riboside proved to be effective cytokinins to induce adequate shoot proliferation, while benzyladenine was the least active cytokinin

Brassinosteroids (BRs) are new class of hormones noted to perform multiplephysiological functions in plant growth and development and have the potentialof influencing cell and tissue growth in vitro. Many naturally occurring BRs,including brassinolide, have been discovered, their mode of action and their growthpromoting activities on plants. The use of brassinolide in in vitro propagation isnew. The Murashige and Skoog (1962) medium was used as basal medium. Plantgrowth regulators - IAA, BA and BR - were added to the medium. The study usedthe Completely Randomized Design (CRD) in factorial with three replications.The cultivar of banana and plant growth substances affected the number of budsproduced, shoot length, root length, and stem girth. The interaction of thesetwo factors (cultivar x PGR) influenced the number of buds produced in vitroand the shoot length of the meriplants.
