<p>Lactic acid bacteria are classified ‘Generally Recognized As Safe’ (GRAS) with most effective potential to divert significant amount of fermentable sugars towards the biosynthesis of functional exopolysaccharide. Exopolysaccharides from lactic acid bacteria are receiving a renewed interest due to the claims of human health benefits, such as modulation of immune response system and more importantly in food and pharma industries as a texturizer, viscosifer, emulsifier and syneresis-lowering agent. Its purification methodology involves: a) Extraction of cell-free supernatant from lactic acid bacteria; b) Denature of protein using trichloroacetic acid; c) Ethanol precipitation; d) Dialysis; and e) Freeze drying. Exopolysaccharides (EPS) produced by three Lactic Acid Bacteria strains,Lactococcus lactisSLT10,Lactobacillus plantarumC7, andLeuconostoc mesenteroidesB3, were isolated using two methods: ethanol precipitation (EPS-ETOH) and ultrafiltration (EPS-UF) through a 10 KDa cut-off membrane. EPS recovery by ultrafiltration was higher than ethanol precipitation forLactococcus lactisSLT10 andLactobacillus plantarumC7. However, it was similar with both methods forLeuconostoc mesenteroidesB3. The monomer composition of the EPS fractions revealed differences in structures and molar ratios between the two studied methods. EPS isolated fromLactococcus lactisSLT10 are composed of glucose and mannose for EPS-ETOH against glucose, mannose, and rhamnose for EPS-UF. EPS extracted fromLactobacillus plantarumC7 andLeuconostoc mesenteroidesB3 showed similar composition (glucose and mannose) but different molar ratios. The molecular weights of the different EPS fractions ranged from 11.6±1.83 to 62.4±2.94 kDa. Molecular weights of EPS-ETOH fractions were higher than those of EPS-UF fractions. Fourier transform infrared (FTIR) analysis revealed a similarity in the distribution of the functional groups (O-H, C-H, C=O, -COO, and C-O-C) between the EPS isolated from the three strains.

We have investigated accelerating the ripening of Pecorino Umbro cheese by adding crude cytoplasmic extract from Pseudomonas fluorescens, non-starter lactic acid bacteria (NSLAB) or cheese slurry. Microbiological and biochemical analyses and sensory evaluation were carried out on control and experimental cheeses over 28 d ripening. In the cheeses containing NSLAB or slurry, counts of mesophilic lactobacilli ranged from log 7·6 at day 1 to ∼log 8·6 cfu/g after 28 d ripening, ∼2 log cycles higher than in the control cheese. All the experimental cheeses contained higher levels than the control of total free amino acids and N soluble at pH 4·6 and in 120 g trichloroacetic acid/l. Compared with the control, higher aminopeptidase and dipeptidase activities were found in the cheeses containing NSLAB and slurry, and especially in those containing the Pseudomonas enzyme. The cheeses containing NSLAB or slurry were characterized by an accumulation of short peptides (Mr<2000) detected by FPLC. Although the cheese containing enzyme had an atypical flavour, the addition of mesophilic lactobacilli reduced from 60 to 28 d the ripening period of Pecorino Umbro cheese, without the appearance of off flavour.

The effects of milk supplementation with catfish bone hydrolysate (CBH) at levels of 0, 0.05, 0.15, and 0.5 % on the pH, lactic acid bacteria count (LABC), fermentation time, texture attributes and microstructure of yogurt were investigated. The addition of CBH stimulated acidification and reduced fermentation time (P < 0.05). In comparison with the control yogurt, CBH caused a decrease in product firmness, consistency, cohesiveness and index of viscosity/consistency. The additon of 0.5% CBH produced a higher decrease (P < 0.05). Scanning electron microscopy showed that CBH caused changes in microstructure of yogurt that had a coarser, less cross-linked microstructure than control yogurt.

Sago extraction processing into sago flour is generally still done in a traditional or semi-mechanical way, with less attention to aspects of hygiene, and resulted sago fermentation spontaneously. This study aimed to isolate and characterize indigenous lactic acid bacteria (LAB) from sago wastewater which obtained directly from the traditional sago processing industry in Malangke Barat, South Sulawesi. The sago wastewater was stored for 1, 3 and 7 days of fermentation. The research was conducted on the LAB isolation and purification on MRS agar medium supplemented with 0.5% CaCO3. Two purified putative LAB isolate were each obtained from 1 day fermentation and 3 days fermentation. The LAB isolate from 1 day fermentation has milky white colors and smooth-round colonies, while LAB isolates from 3 days fermentation has creamy colors and smooth-round colonies. Both of the isolate were gram positive-rods, non-motile and negative catalase reaction.

The effect of sourdough inoculated with three novel single strains of lactic acid bacteria (LAB) (Lactobacillus casei jQ412732, Lactobacillus plantarum jQ301799, and Lactobacillus brevis IBRC-M10790) as well as mixed strains was evaluated on the quality characteristics of Toast bread. Antifungal properties of sourdoughs due to organic acid production were measured by HPLC, and storability was evaluated by thermal and textural analysis in days 1, 3, and 6. Despite the impact of sourdough concentration on microbial preservation, no significant effect was observed in the case of enthalpy reduction. Mixed LAB strains showed the best results in reducing the enthalpy and hardness of bread as well as better microbial preservation by producing the highest amount of organic acids, justified by sensory panelists. Among single strains, L. casei gave better results in reducing hardness and staling rate of bread. Scanning Electron Microscopy micrographs of bread also showed the differences.

Abstract This study aims to determine the effect of lactic acid bacteria supplement from sewge force feeding fish to the LDL content of broiler chicken blood. The research samples are 40 of 1 day old broiler chicken from PT. Multi Breeder Adiram, which were divided into 4 treatments, were taken randomly and maintined for 42 days. Data were analyzed by analysis of variance using a completely randomized design unidirectional pattern, followed by a test of Duncan's Multiple Range Test (DMRT).Probiotic Lactic Acid Bacteria Isolates treatment (BAL) were used in this study is the bacterium Streptococcus thermopillus in the form of freeze drying from the Laboratory of Nutritional Biochemistry, Faculty of Animal Husbandry, UGM. I as a control treatment (without BAL) Treatment II BAL cell count was 106 CFU / ml., The third treatment is the number of BAL Cells are 107CFU / ml. , IV treatment BAL cell count was 108 CFU /ml. The results showed that there is no significant effect of BAL isolate addition on the LDL level of the broiler chicken blood. Keywords: lactic acid bacteria, LDL, freeze drying
