Antioxidant, antibacterial and cytotoxic activity of Caulerpa racemosa (Forsskål) J. Agardh and Ulva (Enteromorpha) intestinalis L.

Seaweeds are fresh sources of phytochemical compounds with immense medicinal potential, which have attracted the attention of agriculture, aquaculture and the pharmaceutical industry. The aim of this study was to test the antioxidant, antibacterial and cytotoxic activity and screening the phytochemical properties of methanol crude extract from two green seaweeds Caulerpa racemosa (Forsskål) J.Agardh and Ulva (Enteromorpha) intestinalis L. The antioxidant and cytotoxic activity of these seaweeds were assessed by 2,2diphenyl, 1-picryl hydrazyl (DPPH) free radical scavenging technique and brine shrimp lethality bioassay method, respectively. The antibacterial activities against Vibrio fluvialis was determined using the standard disc diffusion method. The U. intestinalis showed higher total phenolic content with the value of 149.87 ± 18.17 mg of GAE/g than 73.95 ± 16.09 mg of GAE/g of C. racemosa. However, the recorded inhibition concentration (IC50) to corresponding standards ascorbic acid of C. racemosa and U. intestinalis were 119.62 and 34.274 μg/mL, respectively. The values were statistically significant (p <0.05) compared to the reference antioxidative agent ascorbic acid. In antibacterial assay, C. racemosa extract showed higher inhibition zone (22.65 ± 0.58 mm) than U. intestinalis (17± 0.28 mm). Moreover, U. intestinalis exhibited a lower LC50 value than C. racemosa in cytotoxic activity. Phytochemical analysis revealed that these two seaweeds have alkaloids, flavonoids, glycosides, phenols, saponins, steroids and tannins. Bangladesh J. Sci. Ind. Res.55(4), 237-244, 2020

In vitro antibacterial activity of 21 plant methanolic extracts were investigated by disc diffusion method against gram-negative bacteria Escherichia coli. Amongst the extracts tested, the Datura stramonium L. and Ocimum basilicum L. showed significant antibacterial activity against the bacterial pathogen. D. stramonium showed the highest antibacterial activity followed by O. basilicum extract. Phytochemical analysis of extracts showed the presence of alkaloids, tannins, saponins, flavonoids, and phenols. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and ABTS analysis of potent extract D. stramonium showed appreciable antioxidant activity. For DPPH and ABTS assay the highest antioxidant activity was observed at 60 mg ml-1 concentration with a percent inhibition of 71.66 and 77.19 with IC50 value 9.71 and 22.16 μg ml-1 respectively. Moreover, the radical scavenging activity of the extract was lower than that observed for the synthetic antioxidants butylated hydroxyanisole (BHA )and butylated hydroxytoluene (BHT). The total phenolic content of D. stramonium extract was found to be 0.592 mg GAE/gm which was assayed using Folin ciocalteau reagent. The results provide evidence that the extract of D. stramonium and O. basilicum can be further recommended in the treatment of the infections caused by E. coli; further D. stramonium is also a potential source of natural antioxidants.

Abstract Background: as possible sources of natural bioactive molecules, the plant essential oils and extracts have been used globally in new antimicrobial compounds, food preservatives, and alternatives to treat infectious disease.Methods: In this research, the antimicrobial activities of chloroformic and methanolic extracts of Sophora flavescens, Rhaponticum repens, Alhagi maurorum, Melia azedarach, Peganum harmala, and Juncus conglomeratus were evaluated against 8 bacteria (S. aureus, B. subtilis, R. toxicus, P. aeruginosa, E. coli, P. syringae, X. campestris, P. viridiflava) and 3 fungi (Pyricularia oryzae, Fusarium oxysporum and Botrytis cinerea), through disc diffusion method. Furthermore, the essential oils of plants with the highest antibacterial activity were analyzed utilizing GC/MS. Moreover, the tested plants were exposed to screening for possible antioxidant effect utilizing DPPH test, guaiacol peroxidas, and catalase enzymes. Besides, the amount of total phenol and flavonoid of these plants was measured. Results: Among the tested plants, methanolic and chloroformic extracts of P. harmala fruits showed the highest antibacterial activity against the tested bacteria. Besides, the investigation of free radical scavenging effects of the tested plants indicated the highest DPPH, protein, guaiacol peroxidase, and catalase in P. harmala, M. azedarach, J. conglomeratus fruits, and J. conglomeratus fruits, respectively. In addition, the phytochemical analysis demonstrated the greatest amounts of total phenolic and flavonoid compositions in J. conglomeratus and P. harmala, respectively.Conclusion: The results indicated that these plants could act as a promising antimicrobial agent, due to their short killing time.

The objectives of this study are to screen the phytochemicals, estimate the content of flavonoid and alkaloids compounds and determine the antioxidant capacity of the Rauwolfia serpentina stem. Qualitative analysis of various phytochemical constituents and quantitative analysis of total phenol and alkaloids were determined by the well-known test protocol available in the literature. The hydro alcoholic extract of stem of Rauwolfia serpentina was studied for antioxidant activity on different in vitro models namely 1,1-diphenyl, 2-picryl hydrazyl (DPPH) method. Phytochemical analysis revealed the presence of phenols and flavonoids. The total flavonoids and alkaloids content of Rauwolfia serpentina stem of hydroalcoholic extract was 1.086 and 2.364mg/100mg respectively. Ascorbic acid used as standards was also evaluated for comparison. The extract showed dose dependent free radical scavenging property in the tested models. Rauwolfia serpentina stem extract showed IC50 value 68.10?g/ml for DPPH method, which was comparable to that of ascorbic acid (IC50=17.68?g/ml). The present study describes the phytochemical profile and antioxidant activity of Rauwolfia serpentina which will further used for medicinal applications.

<p><em>Myristica fragrans</em> Houtt (nutmeg) is used as a spice and flavour for food and beverages. It has been traditionally used to treat a number of medical conditions, including diabetes mellitus. The study was undertaken to scientifically validate the traditional use of mace from <em>M.</em><em> </em><em>Fragrans</em><strong>. </strong>The objectives of this study were to evaluate α-glucosidase inhibition, antioxidant and antibacterial activities of water extract (WE) and essential oil (EO) from <em>M.</em><em> </em><em>fragrans</em> mace.  Both WE and EO were evaluated for their α-glucosidase inhibitory activities <em>in vitro</em> and their antioxidant activities based on DPPH radical scavenging assay. Standard compounds were used for every test. Total phenolic and flavonoid contents of both extracts were also determined. The extracts were also tested for their antibacterial activities against six different bacteria by a well diffusion method. Both extracts showed inhibition activities against α-glucosidase, with WE showed stronger activity than EO (IC<sub>50</sub> = 1.86 and 8.15 mg/ml). Good radical scavenging activities were observed for both extracts, with WE showed stronger activity than EO (IC<sub>50</sub> = 1.51 and 4.59 mg/ml). WE showed higher content in phenolic than EO (47.84 and 37.21 mg GAE/100 g DW). Flavonoid content in WE was also higher than EO (215.36 and 30.12 mg RE/ml). Based on the well diffusion method, only EO exhibited antibacterial activities, with inhibition zone in the range 1.03 – 1.30 mm.  The strongest activity was observed against<em> Staphylococcus mutans</em>. The results indicate WE and EO can be exploited further for pharmacological uses, in particular for their antidiabetic and antioxidant activities. </p>

Medicaments, plants and plant-based are the basis of many of the modern pharmaceuticals we use today for our various purposes. The aim of the present study was to evaluate the antioxidant, phytochemical and antibacterial and antifungal activities of the Achyranthes aspera plant extract in different organic solvents. The radical scavenging activity of the different extracts of root, stem, leaf, and seed were evaluated by DPPH assay and the antibacterial activity against Staphylococcus aureus and Pseudomonas aeruginosa, and antifungal activity against Fusarium sp. and Aspergillus niger was studied by Agar well cut diffusion method. All of the extracts exhibited different antioxidant and antibacterial activities and the activities varied from solvent to solvent, and the activities are concentrated. The antioxidant and antimicrobial activities were compared with the positive control Ascorbic acid and Cefuroxime. A qualitative phytochemical analysis was carried out and found to possess bioactive compounds like alkaloids, glycosides, terpenoids, steroids, flavonoids, tannins.

With the increased incidence of antibacterial resistance in microorganisms, combining natural products from plants with antibiotics may be considered interesting alternatives for synergy to attain multitarget effects. In this study, the antioxidant activity of the methanol extract of Ziziphus mucronata and its interactions with antibiotics against bacteria of clinical importance were investigated. While its phytochemicals and antioxidant activities were determined by free radical scavenging assays, the antibacterial activities of the extract and its interactions with the antibiotics were determined by macrobroth dilution and the checkerboard methods. From the results, total phenolic content was 29.67 ± 1.90 mg GAE/100 g, total flavonoid content was 8.72 ± 0.08 mg QE/100 g, and total proanthocyanidin content was 1.94 ± 0.00 mg CE/100 g of dry plant material. The inhibition concentration 50% (IC50) of DPPH, BHT, and ascorbic acid was equal to 0.04 ± 0.02 mg/ml, respectively. Those of the ABTS, BHT, and ascorbic acid were equal to 0.02 ± 0.02, 0.04 ± 0.03, and 0.04 ± 0.02 mg/ml, respectively. The checkerboard assay showed that combining the extract with different antibiotics resulted in synergistic (38.75%), indifferent (30%), additive (28.75%), and antagonistic (2.5%) interactions. The interactions between the extract and antibiotics resulting in enhanced antibacterial activities could have resulted from the antioxidant activities of the extract mopping up the ROS generated by the antibiotics or the ability of both extract and antibiotics simultaneously producing reactive oxygen species with deleterious effects resulting in synergistic antibacterial effects.

Phytochemical analysis of C. alatavicus revealed the presence of phenols, flavonoids, anthocyanins, carotenoids, amino acids and carbohydrates. The flavonoid, amino acids and carotenoid contents were higher in aerial part (1.50%, 7.49% and 9.78mg%, respectively) than in bulb (0.43%, 3.88% and 0.91 mg%, respectively). Total phenolic content (TPC), total antioxidant (TAA), 2.2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and antibacterial activities of water, methanol, ethanol and dichloromethane extracts from aerial part and bulb were tested. TPC ranged from 13.63 to 72.29 mg gallic acid equivalents (GAE)/g extract. The maximum TAA were observed in ethanol (61.34%) and methanol extracts (46.13%) from aerial part with a high TPC (72.29 and 62.37 mgGAE/g extract, respectively). Ethanol extracts from aerial part and bulb had good scavenger of DPPH radicals (65.5% and 54.08%, respectively) with an IC50 387 and 447 µg/ml. Ethanol extract from aerial part was most effective against gram-positive bacterial strains S. aureus, B. subtilis and B. cereus. Biological activities of the extracts were correlated with the TPC. It can be deduced that ethanol and methnol extracts of C. alatavicus contains useful potent bioactive compounds with antioxidant and antimicrobial activities.

Crassocephalum crepidioides belongs to the Asteraceae family and the leaves of the plant were used in the phytochemical screening. The objective of this study was phytochemical analysis of C. crepidioides in order to evaluate its antioxidant, thrombolytic and cytotoxic effects. In this study, DPPH assay and total phenolic content (TPC) were used to evaluate the antioxidant property. Brine shrimp lethality test was performed to find the cytotoxicity. The TPC of methanolic extract of C. crepidioides was 637.22 mg/gm and gallic acid was standard in this procedure. The IC50 value of the plant extract of DPPH assay was 136.016 μg/mL obtained by using ascorbic acid as reference standard. Furthermore, the obtained IC50 value of ascorbic acid was 94.12 μg/mL. In this study, lethal concentrations of vincristine sulfate and the plant extract were 3.064 μg/mL and 69.245 μg/mL respectively. Indeed, the plant exhibited significant antioxidant, cytotoxic and thrombolytic activity.
