Bacteria and viruses may enter our bodies through mucous membranes of the airways or the gut. To prevent infections, one defense mechanism of our immune system is antimicrobial peptides (AMPs). Most AMPs are composed of 10–50 amino acids and insert into bacterial cell membranes to destroy the cell. Some AMPs are also active against viruses and fungi. AMPs can be generated by chopping up bigger proteins like hemoglobin. The hemoglobin fragments can inactivate bacteria and viruses, while the whole hemoglobin protein cannot. To identify new AMPs, peptide libraries consisting of thousands of different peptides can be generated from human body fluids and organs. These libraries are tested for antibacterial or antiviral activity and can be further purified to identify the responsible peptide. This method may lead to the development of new antimicrobial substances with a potential for treating infections.

In recent years various Microbiomes (Skin, Gut Lumen) of the human body have attracted the attention of different research groups. In the meantime it has been shown that the conventional therapy of different diseases by making use of antibiotics and similar antibacterial treatments may disturb the harmony of the Skin Microbiome, resulting in dysbiosis. There are efforts of using “live” or “tyndallized (lysed)” probiotics in order to treat different diseases of the skin. It is also known that amino acids are one of the important key elements of the skin. In this paper, a hypothesis for the utilization of yogurt as an excipient for various topical dermatological products will be proposed. Yogurt contains significant amounts of; Probiotics (starter cultures), Amino Acids, Vitamins, Minerals and various Fatty Acids (saturated, monounsaturated and polyunsaturated). Besides, it has been shown that Antimicrobial Peptides (Bacteriocins) are also present in yogurt. Yogurt could eventually be used as an excipient for the production of various topical dermatological products in order to deliver some of the above-mentioned constituents to the Stratum Corneum (Skin) locally.

The increasing antimicrobial-resistant prevalence has become a severe health problem. It has led to the invention of a new antimicrobial agent such as antimicrobial peptides. Heteroscorpine-1 is an antimicrobial peptide that has the ability to kill many bacterial strains. It consists of 76 amino acid residues with a cecropin-like region in N-terminal and a defensin-like region in the C-terminal. The cecropin-like region from heteroscorpine-1 (CeHS-1) is similar to cecropin B, but it lost its glycine-proline hinge region. The bioinformatics prediction was used to help the designing of mutant peptides. The addition of glycine-proline hinge and positively charged amino acids, the deletion of negatively charged amino acids, and the optimization of the hydrophobicity of the peptide resulted in two mutant peptides, namely, CeHS-1 GP and CeHS-1 GPK. The new mutant peptide showed higher antimicrobial activity than the native peptide without increasing toxicity. The interaction of the peptides with the membrane showed that the peptides were capable of disrupting both the inner and outer bacterial cell membrane. Furthermore, the SEM analysis showed that the peptides created the pore in the bacterial cell membrane resulted in cell membrane disruption. In conclusion, the mutants of CeHS-1 had the potential to develop as novel antimicrobial peptides.

Antimicrobial peptides (AMPs) are host defense peptides that are well conserved throughout the course of evolution. Histones are classical DNA-binding proteins, rich in cationic amino acids, and recently appreciated as precursors for various histone-derived AMPs. The present study deals with identification of the potential antimicrobial peptide sequence of teleostin from the histone H2A of marine teleost fishes, Cynoglossus semifasciatus and Tachysurus jella. A 245 bp amplicon coding for 81 amino acids was obtained from the cDNA transcripts of these fishes. The first 52 amino acids from the N terminal of the peptide were identical to previously characterized histone-derived antimicrobial peptides. Molecular and physicochemical characterizations of the sequence were found to be in agreement with previously reported histone H2A-derived AMPs, suggesting the possible role of histone H2A in innate defense mechanism in fishes.

Vasavaleha is one of the best medicine given for respiratory diseases. Corona viruses typically affect the respiratory system, causing symptoms such as coughing, fever and shortness of breath. It also affects host immune system of human body. Spreading rate of this disease is very high. Whole world is seeking for the treatment which can uproots this diseases. There in no vaccine available till date against this pandemic disease. Ayurveda mainly focuses on prevention of diseases alongwith its total cure. Rajyakshma Vyadhi is MadhyamMarga Roga as per Ayurveda. It shows many symptoms such as Kasa, Shwasa etc. By overall view of Covid 19, shows its resemblance with Rajyakshma Vyadhi described in Ayurveda. Vasavaleha is a Kalpa which is described in Rogadhikara of Rajyakshma. It shows Kasahara, Shwashara properties. It consists of Vasa, Pipalli, Madhu and Goghrita. These components shows actions like bronchodilation, antitussive effect and many more other actions. Pipalli shows important Rasayana effect. So in present review, we have tried to focus on role of Vasavaleha in the management of Covid 19. This can be used as preventive as well as adjuvant medication in treating Covid 19. There is need of further clinical research to rule of exact action of Vasavaleha against Covid 19.

The gastrointestinal tract (GIT) of humans and animals is host to a complex community of different microorganisms whose activities significantly influence host nutrition and health through enhanced metabolic capabilities, protection against pathogens, and regulation of the gastrointestinal development and immune system. New molecular technologies and concepts have revealed distinct interactions between the gut microbiota and dietary amino acids (AAs) especially in relation to AA metabolism and utilization in resident bacteria in the digestive tract, and these interactions may play significant roles in host nutrition and health as well as the efficiency of dietary AA supplementation. After the protein is digested and AAs and peptides are absorbed in the small intestine, significant levels of endogenous and exogenous nitrogenous compounds enter the large intestine through the ileocaecal junction. Once they move in the colonic lumen, these compounds are not markedly absorbed by the large intestinal mucosa, but undergo intense proteolysis by colonic microbiota leading to the release of peptides and AAs and result in the production of numerous bacterial metabolites such as ammonia, amines, short-chain fatty acids (SCFAs), branched-chain fatty acids (BCFAs), hydrogen sulfide, organic acids, and phenols. These metabolites influence various signaling pathways in epithelial cells, regulate the mucosal immune system in the host, and modulate gene expression of bacteria which results in the synthesis of enzymes associated with AA metabolism. This review aims to summarize the current literature relating to how the interactions between dietary amino acids and gut microbiota may promote host nutrition and health.

Abstract Background Antimicrobial peptides are very primitive innate defense molecules of almost all organisms, from microbes to mammalians and vascular seed-bearing plants. Antimicrobial peptides of plants categorized into cysteine-rich peptides (CRPs) and others and most of the antimicrobial peptides belong to CRPs group. These peptides reported showing the great extent of protecting property against bacteria, fungi, viruses, insect, nematode, and another kind of microbes. To develop a resistant plant against pathogenic fungi, there have been several studies executed to understand the efficiency of transgenicity of these antimicrobial peptides. Main text Apart from the intrinsic property of the higher organism for identifying and activating microbial attack defense device, it also involves innate defense mechanism and molecules. In the current review article, apart from the structural and functional characterization of peptides defensin and thionin, we have attempted to provide a succinct overview of the transgenic development of these defense peptides, that are expressed in a constitutive and or over-expressive manner when biotic and abiotic stress inflicted. Transgenic of different peptides show different competence in plants. Most of the transgenic studies made for defensin and thionin revealed the effective transgenic capacity of these peptides. Conclusion There have been several studies reported successful development of transgenic plants based on peptides defensin and thionin and observed diverse level of resistance-conferring potency in different plants against phytopathogenic fungi. But due to long regulatory process, there has not been marketed any antimicrobial peptides based transgenic plants yet. However, success report state that possibly in near future transgenic plants of AMPs would be released with devoid of harmful effect, with good efficiency, reproducibility, stability, and least production cost.

AbstractVarious nutrients can change cell structure, cellular metabolism, and cell function which is particularly important for cells of the immune system as nutrient availability is associated with the activation and function of diverse immune subsets. The most important nutrients for immune cell function and fate appear to be glucose, amino acids, fatty acids, and vitamin D. This perspective will describe recently published information describing the mechanism of action of prominent nutritional intervention agents where evidence exists as to their action and potency.
