Plants have a large number of bioactive compounds with high antioxidant activity. Studies for the determination of the antioxidant activity of different plant species could contribute to revealing the value of these species as a source of new antioxidant compounds. There is a large variety of in vitro methods to quantify antioxidant activity, and it is important to select the proper method to determine which species have the highest antioxidant activity. The aim of this work was to verify whether different methods show the same sensitivity and/or capacity to discriminate the antioxidant activity of the extract of different plant species. To that end, we selected 12 species with different content of phenolic compounds. Their extracts were analyzed using the following methods: 2,2-di-phenyl-1-picrylhydrazyl (DPPH) radical scavenging capacity assay, ferric reducing (FRAP) assay, Trolox equivalent antioxidant capacity (ABTS) assay, and reducing power (RP) assay. The four methods selected could quantify the antioxidant capacity of the 12 study species, although there were differences between them. The antioxidant activity values quantified through DPPH and RP were higher than the ones obtained by ABTS and FRAP, and these values varied among species. Thus, the hierarchization or categorization of these species was different depending on the method used. Another difference established between these methods was the sensitivity obtained with each of them. A cluster revealed that RP established the largest number of groups at the shortest distance from the root. Therefore, as it showed the best discrimination of differences and/or similarities between species, RP is considered in this study as the one with the highest sensitivity among the four studied methods. On the other hand, ABTS showed the lowest sensitivity. These results show the importance of selecting the proper antioxidant activity quantification method for establishing a ranking of species based on this parameter.

The flora of Latin America attracts gaining interest as it provides a plethora of still unexplored or underutilized fruits that can contribute to human well-being due to their nutritional value and their content of bioactive compounds. Antioxidant compounds are now of considerable interest due to their effect of preventing or delaying aging and their apparent involvement in prevention of numerous human diseases, including cancer, atherosclerosis, Alzheimer’s disease, inflammation, and rheumatoid arthritis. In this work, the fruit ofCoccoloba uvifera(L.) was investigated for theirin vitroantioxidant capacity using two assays based on reactions with a relatively stable single reagent radical (Trolox equivalent antioxidant capacity, TEAC; and DPPH free radical scavenging assay, DPPH), two assays based on chelating of metallic cations, and one based on the reduction of oxidized species. The TEAC value on ABTS radical, DPPH scavenging activity, ion chelation, and reducing power were found to be 897.6 μM of Trolox/100 g of sample, 22.8% of DPPH free radical scavenging, 11.3% of Cu2+-chelating activity, 23.9% of Fe2+-chelating activity, and a Fe2+-reducing power of 0.76 mg/mL, respectively. The free radical scavenging and antioxidant characteristics ofC. uviferamay be due to the presence of diverse phytochemicals in the fruit as anthocyanins, ascorbic acid, phenolic compounds, and flavonoids.

Dibenz[b,f]azepine and its five derivatives bearing different functional groups were synthesized by known methods. The compounds thus synthesized were evaluated for antioxidant potential through different in vitro models such as (DPPH) free radical scavenging activity,ß-carotene-linoleic acid model system, reducing power assay and phosphomolybdenum method. Under our experimental condition among the synthesized compounds dibenz[b,f]azepine (a) and 10-methoxy-5H-dibenz[b,f]azepine (d) exhibited potent antioxidant activity in concentration dependent manner in all the above four methods. Butylated hydroxyl anisole (BHA) and ascorbic acid (AA) were used as the reference antioxidant compounds. The most active compounds like dibenz[b,f]azepine and its methoxy group substituent have shown more promising antioxidant and radical scavengers compared to the standards like BHA and ascorbic acid. It is conceivable from the studies that the tricyclic amines,i.e. dibenz[b, f]azepine and some of its derivatives are effective in their antioxidant activity properties.

Sudarshanapowder (SP) is one of the most effective Ayurveda powder preparations for paediatric febrile conditions. The objective of the present study was to evaluate thein vitroandin vivoantioxidant potentials of SP. Thein vitroantioxidant effects were evaluated using ABTS radical cation decolourization assay where the TROLOX equivalent antioxidant capacity (TEAC) was determined. Thein vivoantioxidant activity of SP was determined in Wistar rats using the Lipid Peroxidation (LPO) assay in serum. Thein vitroassay was referred to as the TROLOX equivalent antioxidant capacity (TEAC) assay. For thein vivoassay, animals were dosed for 21 consecutive days and blood was drawn to evaluate the MDA level. Thein vitroantioxidant activity of 0.5 μg of SP was equivalent to 14.45 μg of standard TROLOX. The percentage inhibition against the radical formation was50.93±0.53%. The SP showed a statistically significant (p<0.01) decrease in the serum level of thiobarbituric acid-reactive substance in the test rats when compared with the control group. These findings suggest that the SP possesses potent antioxidant activity which may be responsible for some of its reported bioactivities.

Many plants with high antioxidant activity are great of significant in the pharmaceutical and cosmetic industries. Oxidative stress plays a major part in the development of many diseases including cancer, which is known imbalance free radicals and antioxidants. Herein, new natural antioxidant compounds have great interest in the scientific research. The genus Silene is a major group in the Caryophyllaceae family. In Turkey, Silene species have been used for several medicinal purposes such as skin softening, asthma, bronchitis. In our study, the antioxidant capacity of three Silene species (S. conoidea, S. dichotoma and S. italica) were evaluated by different in vitro assays, including free radical scavenging, reducing power, metal chelating, and phosphomolybdenum. In addition, total phenolic and flavonoid contents were analyzed spectrophotometrically. The water extracts contained higher total phenolic content than ethyl acetate extracts. All extracts showed antioxidant capacity. This data indicated that Silene species could potentially be used as antioxidant sources in pharmaceutical and cosmetic areas.

Objective: This study was examined to in vitro antioxidant activity and Total Phenol and Flavonoids content analysis of methanolic root extract of Eleutherococcus senticosus (Siberian ginseng). Methods: 1,1-dephenyl-2-picryl-hydrazyl free radical scavenging and FRAP assay propose that antioxidant activity of methanol root extract because of reducing capacity of the antioxidant against oxidative effects of reactive oxygen species. Results: Scavenging activity of Siberian ginseng root RC50 value was shown 713.42±11.55 µg/ml and reducing power 0.13±0.01 mmol/g was investigated. In addition, total phenol 12.6±1.13 mg GAE/g DW and total flavonoids 9.8±0.20 mg QE/g DW were recorded. Conclusion: Although all tests were performed in vitro assay, these results recommend that Siberian ginseng root may be a good source of antioxidant ingrediant.

1. A new method has been developed for measuring the total antioxidant capacity of body fluids and drug solutions, based on the absorbance of the ABTS*+ radical cation. 2. An automated method for use on a centrifugal analyser, as well as a manual method, is described. 3. The procedure has been applied to physiological antioxidant compounds and radical-scavenging drugs, and an antioxidant ranking was established based on their reactivity relative to a 1.0 mmol/l Trolox standard. 4. The Trolox equivalent antioxidant capacity of plasma from an adult reference population has been measured, and the method optimized and validated. 5. The method has been applied to investigate the total plasma antioxidant capacity of neonates and how this may be compromised in prematurity.

Phenolic content and antioxidant activity of two common fruits of Bangladesh, namely Phyllunthus emblica and Elaeocarpus floribundus, were measured in water, methanol, ethanol, acetone and hexane extracts. Several in vitro models including phosphomolybdenum assay, DPPH free radical scavenging assay, FRAP assay and reducing power assay were used to assess the antioxidant activity of these extracts in comparison with reference antioxidants. Between the two fruits, P. emblica showed higher phenolic content and antioxidant activity in all the solvents used. In the DPPH scavenging assay, the activity of P. emblica extracts was close to reference antioxidants, ascorbic acid and BHT. Besides, considering the solvents used, extracts of both fruits had the highest phenolic and antioxidant activity in polar solvents. The correlation coefficient between total phenolics and antioxidant activities was found statistically significant. These findings suggest that P. emblica could be an excellent antioxidant resource for industries like food, pharmaceutical, and cosmetics.

Red beetroot is rich in bioactive compounds such as polyphenols, flavonoids, betaxanthins, betacyanins, among others. According to selected processing methods, the bioaccessibility of these compounds could be either enhanced or decreased. This study evaluated the effect of four different drying conditions: (1) Traditional Drying (TD), (2) Swell Drying (SD), (3) DIC Blanching + Traditional Drying (BTD), and (4) DIC Blanching + Swell Drying (BSD) on the antioxidant content and the antioxidant activity of red beetroots. Obtained results showed that in all the cases, by comparing to Traditional Drying (TD), the coupling of a DIC Blanching pre-treatment to a Swell Drying treatment (BSD) maintained or enhanced the preservation of the Total Phenolic Compounds (TPC), the Total Flavonoids Compounds (TFC), the Betanin Concentration (BC), the Trolox Equivalent Antioxidant Capacity (TEAC), and the Free Radical Scavenging Activity by DPPH (IC50) of red beetroots. Various studies have shown that thanks to the expanded and porous structure triggered by the Swell Drying process, it has been possible to achieve better antioxidants extraction and better whole quality. Hence, by coupling DIC as a blanching–steaming pre-treatment, it was possible to preserve better the antioxidant content and the antioxidant activity of red dried beetroots.

Background. Quezon province has been one of the primary agricultural sources of vegetables and crops in southern Luzon due to its diverse agricultural topography. Having increased dietary awareness, consumption of antioxidant-rich foods has become relevant. Objectives. Thirteen (13) methanolic extracts of endemic Quezon plant foods were evaluated for phytochemical constituents and antioxidant potential.Methods. The plant extracts were subjected to in vitro antioxidant assays, which include DPPH [2,2-diphenyl-1picrylhydrazyl], FRAP [Fluorescence recovery after photobleaching], metal chelation, superoxide, nitric oxide, hydroxyl radical scavenging activities and MTT [ (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide)] reduction. Qualitative phytochemical analysis were also employed. Results. P. umbellatum, locally known as kamamba and C. longa, locally known as luyang dilaw showed high antioxidant activity using DPPH and MTT assays by 82.46±0.75% and 82.35±1.79% and 78.46±0.71% and 60.97±2.60% at 66.67 μg/mL, respectively. FRAP assay revealed a comparable reducing power with BHT (butylated hydroxytoluene) (93.61±0.56%) at 72.15 μg/mL in C. longa (92.49±1.32%), P. umbellatum (92.38±0.64%), and Z. officinale (90.33±2.06%) (luya), but found with low metal chelating activity. The highest activity against hydroxyl radical were observed in S. edule (sayote) (34.30±1.88%), P. edulis (pasyonaryo) (33.48±1.34%) and D. philippinensis (katmon) (34.71±0.85%) at 45.5 μg/mL. On the other hand, Z. officinale, C. esculenta (gabi), V. unguiculata (sitaw) fruit and D. philippinensis showed dual action, as antioxidant and as pro-oxidant, in Superoxide Scavenging and Nitric Oxide assays. Quezon province plant foods contain flavonoids, phenolic compounds, glycosides and coumarins and quinones which may explain their behavior as antioxidant.Conclusion. The study revealed that different plant foods showed different capacity to scavenge particular oxidants. However, P. umbellatum and C. longa may be considered promising sources of natural antioxidants.

Second-grade dates (Phoenix dactyliferaL.), with hard texture, from three selected Tunisian cultivars (Allig, Deglet Nour, and Bejo) were analysed from their antioxidant activities using DPPH radical scavenging activity, FRAP assay, H2O2scavenging activity, and metal chelating activity. Date extracts showed strong and concentration-dependant activity in all tested methods. The results showed that the best antioxidant activity was obtained in Allig, followed by Bejo and Deglet Nour. Total phenolics, total flavonoids, carotenoids, and tannins were determined spectrophotometrically in three date extracts. Results indicated that date contained significantly different amounts of these compounds. In fact, Allig presented the highest antioxidant compounds, followed by Bejo and Deglet Nour. Correlation analyses indicated a positive linear relationship between antioxidant activities and total phenolic and flavonoid content of date. This study demonstrates the potential antioxidant activity with Tunisian date, where we can use these natural extracts as food additives in replacement of synthetic compounds.
