We previously showed that novel filterable bacteria remain in “sterile” (<0.2 μm filtered) terrestrial environmental samples from Japan, China, and Arctic Norway. Here, we characterized the novel filterable strain IZ6T, a representative strain of a widely distributed lineage. Phylogenetic analysis showed that this strain was affiliated with the Rhizobiales (now proposed as Hyphomicrobiales) of Alphaproteobacteria, but distinct from any other type strains. Strain IZ6T shared the following chemotaxonomic features with the closest (but distantly) related type strain, Flaviflagellibacter deserti SYSU D60017T: ubiquinone-10 as the major quinone; phosphatidylethanolamine, phosphatidylcholine, and phosphatidylglycerol as major polar lipids; and slightly high G+C content of 62.2 mol%. However, the cellular fatty acid composition differed between them, and the unsaturated fatty acid (C18:1ω7c/C18:1ω6c) was predominantly found in our strain. Moreover, unlike methyrotrophs and nitrogen-fixers of the neighboring genera of Hyphomicrobiales (Rhizobiales), strain IZ6T cannot utilize a one-carbon compound (e.g., methanol) and fix atmospheric nitrogen gas. These findings were consistent with the genome-inferred physiological potential. Based on the phylogenetic, physiological, and chemotaxonomic traits, we propose that strain IZ6T represents a novel genus and species with the name Terrihabitans soli gen. nov., sp. nov. (=NBRC 106741T = NCIMB 15058T). The findings will provide deeper insight into the eco-physiology of filterable microorganisms.

Six strains (CB7T, CB18, CB23, CB26, CB28 and CB35T) were isolated from human faeces. Based on phylogenetic analysis, phenotypic characteristics, cellular fatty acid profiles and menaquinone profiles, these strains could be included within the genus Prevotella and made up two clusters. 16S rRNA gene sequence analysis indicated that five strains were most closely related to Prevotella veroralis, sharing about 92 % sequence similarity; the remaining strain was most closely related to Prevotella shahii, sharing about 90 % sequence similarity. All six strains were obligately anaerobic, non-pigmented, non-spore-forming, non-motile, Gram-negative rods. The cellular fatty acid compositions of the six strains differed significantly from those of other Prevotella species. Five strains (CB7T, CB18, CB23, CB26 and CB28) contained dimethyl acetals and the major menaquinones of these strains were MK-11, MK-12 and MK-13. The major menaquinones of CB35T were MK-12 and MK-13. Based on phenotypic and phylogenetic findings, two novel species, Prevotella copri sp. nov. and Prevotella stercorea sp. nov., are proposed, representing the two different strain clusters. The DNA G+C contents of strains CB7T and CB35T were 45.3 and 48.2 mol%, respectively. The type strains of P. copri and P. stercorea are CB7T (=JCM 13464T=DSM 18205T) and CB35T (=JCM 13469T=DSM 18206T), respectively.

Two strains (JA575T and JA576T) of orange- to pink-pigmented, rod-shaped, motile and budding phototrophic bacteria were isolated from paddy soils. Both strains contained bacteriochlorophyll a and carotenoids of spirilloxanthin series. Both strains had C18 : 1ω7c as the major cellular fatty acid, ubiquinone-10 (Q10) as the main quinone, and diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and phosphatidylcholine as polar lipids. Phylogenetic analysis on the basis of 16S rRNA gene sequences showed that both strains clustered with species of the genus Rhodopseudomonas in the class Alphaproteobacteria . Strains JA575T and JA576T were genotypically (<35 % DNA–DNA relatedness) and phenotypically distinct from each other. Further, both strains showed less than 48 % DNA–DNA relatedness with the type strains of all recognized species of the genus Rhodopseudomonas . The molecular evidence is supported by phenotypic evidence. It is proposed that strains JA575T and JA576T be classified as representing two novel species of the genus Rhodopseudomonas with the species names Rhodopseudomonas pentothenatexigens sp. nov. and Rhodopseudomonas thermotolerans sp. nov., respectively. The type strains of the proposed novel species are JA575T ( = NBRC 108862T = KCTC15143T) and JA576T ( = NBRC 108863T = KCTC 15144T), respectively.

One hundred and twenty-seven apyocyanogenic fluorescent Pseudomonas strains capable of growth at 41 degrees C, but differing from Pseudomonas aeruginosa, were typed serologically and tested for pyocin production, antibiotic susceptibility, selected biochemical reactions, and utilization of selected substrates. Results were compared with those from 40 apyocyanogenic and 14 pyocyanin-producing strains of P. aeruginosa. Unidentified fluorescent Pseudomonas (UFP) strains generally were not agglutinated by P. aeruginosa antisera and showed little or no pyocin activity. In contrast to P. aeruginosa strains, UFP strains usually failed to oxidize D-gluconate or reduce nitrate to nitrogen gas. They could not use D-gluconate or D-mannitol as sole carbon source and were susceptible to kanamycin. The cellular fatty acid compositions of major UFP groups resembled those of the alcaligenes-stutzeri groups.

A characterization of the Cu Kα1,2 spectrum is presented, including the 2p satellite line, Kα3,4, the details of which are robust enough to be transferable to other experiments. This is a step in the renewed attempts to resolve inconsistencies in characteristic X-ray spectra between theory, experiment and alternative experimental geometries. The spectrum was measured using a rotating anode, monolithic Si channel-cut double-crystal monochromator and backgammon detector. Three alternative approaches fitted five Voigt profiles to the data: a residual analysis approach; a peak-by-peak fit; and a simultaneous constrained method. The robustness of the fit is displayed across three spectra obtained with different instrumental broadening. Spectra were not well fitted by transfer of any of three prior characterizations from the literature. Integrated intensities, line widths and centroids are compared with previous empirical fits. The novel experimental setup provides insight into the portability of spectral characterizations of X-ray spectra. From the parameterization, an estimated 3d shake probability of 18% and a 2p shake probability of 0.5% are reported.
