The aim of this study was to evaluate the content and profile of the phenolic compounds (PCs) and antioxidant properties of field-grown leaves, in vitro leaves and in vitro callus cultures of the blackberry ‘Čačanska Bestrna’ and blueberry ‘Toro’. In vitro shoots of the selected genotypes were grown either on original Murashige and Skoog (MS) medium containing 1 mg/L BA, 0.1 mg/l IBA and 0.1 mg/l GA3 (‘Čačanska Bestrna’) or on MS medium with macroelements reduced to ½, 2 mg/L zeatin and 0.2 mg/L IAA (‘Toro’). Callus cultures were induced from in vitro leaves and established on MS medium with 2 mg/L BA and 2 mg/L 2,4-D (‘Čačanska Bestrna’) or MS medium with half strength macroelements, 2 mg/L BA, 2 mg/L 2,4-D and 1 mg/L NAA (‘Toro’). Total phenolic (TPC) and flavonoid content (TFC) were the highest in blueberry leaves, whereas low TPC and TFC values were obtained in callus cultures of both cultivars. A higher content of PCs in blueberry leaves compared to blackberry leaves was determined by the UHPLC-DAD MS/MS technique. Quercetin derivatives and phenolic acids were the dominant PCs in the leaves of both berries, whereas gallocatechin was present in a significant amount in blueberry leaves. Callus cultures of both berries had a specific PC profile, with none detected in the leaves except quercetin-3-O-glucoside and quercetin-3-O-rutinoside. Blackberry leaves showed the best antioxidant properties as estimated by ferric reducing power (FRP), ABTS•+ and DPPH• scavenging activity assays. Callus cultures of both berries exhibited three to five times lower ABTS•+ and ten to seventeen times lower DPPH• scavenging activity compared to corresponding leaves. The analyzed leaves and callus cultures can be a good source of PCs with good antioxidant properties and specific phenolics, respectively, for applications in the food and pharmaceutical industries.

Herein, the antioxidant peptides from a Thai traditional semi-dried fermented farmed hybrid catfish (Clarias macrocephalus × Clarias gariepinus) catfish, Pla Duk Ra, were characterized. After extraction and deproteinization, Pla Duk Ra crude peptide extract (CPE) was fractioned using 2 connected Hitrap Sephadex-G25 columns, yielding two significant fractions, F1 with higher browning intensity (A420) and F2. CPE, F1, and F2 had different amino acid profiles, contents, and sequences evaluated by LC-MS/MS, which could be responsible for their antioxidant properties. F2 contained the highest numbers of hydrophobic amino acid (HBA) (47.45%) and aromatic amino acid (27.31%), followed by F1, and CPE. The peptides with 8–24 amino acid residues were detected in CPE and its fractions. In CPE, F1, and F2, there were 69, 68, and 85 peptides with varied HBA content, respectively. ARHSYGMLYCSCPPND (50% HBA), ALRKMGRK (37.5% HBA), and ANWMIPLM (87.5% HBA) were the most prevalent peptides found in CPE, F1, and F2. Overall, F2 was the most effective at inhibiting free radicals (DPPH● and ABTS●+) and reactive oxygen species (hydroxyl radical, singlet oxygen, and hydrogen peroxide), followed by F1 and CPE. The metal chelation of F1 was, however, superior to that of F2 and CPE. For the stability test, the effects of pH, heating temperature, and in vitro digestion on the DPPH● scavenging activity of F2 were investigated. The activity was boosted by lowering the pH and raising the heating temperature. In the gastrointestinal tract model system, however, roughly 50% of DPPH● scavenging activity reduced after digesting.

Objectives: The goals of this research were to observe antioxidant properties from different parts of super red dragon fruit (Hylocereus costaricensis) using two antioxidant testing methods which were 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2’-azino-bis (3-ethylbenzthiazoline-6-sulfonic acid) (ABTS).Methods: Antioxidant activities were determined using DPPH and ABTS assays, total phenolic content (TPC) using Folin–Ciocalteu reagent, flavonoid content by Chang’s method.Results: Inhibitory concentration 50% (IC50) of DPPH scavenging activity of all of the extracts in the range of 2.69 μg/ml was −94.17 μg/ml. The ethyl acetate peel extract of super red dragon fruit expressed the highest TPC (4.56 g GAE/100 g) and the highest total flavonoid content (12.63 g QE/100 g). TPC in flesh extract of super red dragon fruit had a negative and significant correlation with their IC50 of ABTS. The IC50 of DPPH and IC50 of ABTS of flesh extract of super red dragon fruit showed positive and significant correlation.Conclusion: All different parts extracts of super red dragon fruit (except n-hexane flesh extract) were categorized as a very strong antioxidant by DPPH method. Phenolic compounds in flesh extract of super red dragon fruit were the major contributor in antioxidant activities by ABTS method. DPPH and ABTS showed linear results in antioxidant activities of super red dragon fruit flesh extract.

The present work was conducted in order to investigate the effect of auxin type (2,4-D and NAA) and concentration (0.00, 0.25, 0.50, 1.00 and 2.00 mg/l) on Balanites aegyptiaca callus cultures growth and production of secondary metabolites. Obtained results demonstrated that supplementation MS medium with 2,4-D at 2.0 mg/l could enhanced and recorded the ultimate values of callus fresh weight, antioxidant activity (%), total flavonoids, total phenolic compounds and total saponins contents and yields of Balanites aegyptiaca L. callus.

To evaluate the antioxidant potential of less valued green leafy vegetables of Bangladesh, the methanolic extracts of Alternanthera sessilis, A. philoxeroides, A. paronychioides, Glinus oppositifolius and Portulaca oleracea were subjected to assay for total phenolic content, total flavonoid content and DPPH free radicle scavenging activity. Among the studied materials, A. sessilis showed the highest phenolic and flavonoid content. However, G. oppositifolius presented the lowest phenolic content (14.53 ± 0.48 mg of GAE/g of sample) and P. oleracea exhibited minimum (7.005 ± 0.25 mg of RUE/g of sample) flavonoid content. DPPH radical scavenging activity was found to increase with increase in the concentration of each individual material. A. philoxiroides (IC50 = 6.07 ± 0.27 μg/ml) was found as the most active in DPPH radical percentage inhibition, which was followed by A. sessilis (IC50 = 7.17 ± 0.67 μg/ml). Interestingly, A. paronychioides, in spite of having good amount of total phenolic and flavonoid content, showed less DPPH scavenging activity. No linear relationship was found to exist in case of the total phenolic, flavonoid content and DPPH scavenging activity among G. oppositifolius, P. oleracea and A. paronychioides. The results from different methods may differ substantially because each complex chemical reaction generates unique values. However, all the five plants exhibited creditable antioxidant activity and thus consumption of these vegetables may play important role in combating oxidative stresses and thereby help in retaining good health.

Due to their antioxidant properties, polyphenolic compounds are considered beneficial for human health. In this work, we investigated the polyphenol profile and antioxidant activity of edible tejocote (Crataegus pubescens) fruit extracts by micellar electrokinetic chromatography (MEKC) and HPLC/UV. The major phenolic compounds in the pulp extracts were (+)-catechin (9.17 ± 0.20 mg/100 mg dry fruit), (−)-epicatechin (4.32 ± 0.11 mg/100 mg dry fruit), and chlorogenic acid (5.60 ± 0.24 mg/100 mg dry fruit). The total phenolic content was 168.6 ± 0.9 mg gallic acid equivalent/g dry fruit; the total proanthocyanidin content was 84.6 ± 1.4 mg cyanidin/100 g dry fruit; and the total flavonoid content was 55.89 ± 1.43 mg quercetin/g dry fruit. Interestingly, procyanidins (dimers, trimers, and tetramers of (−)-epicatechin and (+)-catechin) were detected in the extract. This is the first study reporting the presence of polymeric polyphenols in Crataegus pubescens fruit. Accordingly, these fruits demonstrate great potential as a natural source of antioxidant phenolic compounds and could therefore be used as a nutraceutical and functional food.

The aim of this study was to evaluate the differences in the antioxidant activity and phenolic profile of sunflower (Helianthus annuus L.) extracts obtained from the aerial parts of plants harvested at five growth stages. In vitro assays were used to determine the antioxidant activity, i.e., ABTS•+ and DPPH• scavenging activity, the ferric-reducing antioxidant power (FRAP) and the ability to inhibit β-carotene–linoleic acid emulsion oxidation. Phenolic compounds, such as mono- and dicaffeoylquinic acid isomers and caffeic acid hexose, were identified using the LC–TOF–MS/MS technique. The predominant compound during the growth cycle of the plant was 3,5-di-O-caffeoylquinic acid, whose content was the highest at the mid-flowering stage. The total phenolic content was also the highest in sunflowers at the mid-flowering stage. The main phenolic compound contents were closely correlated with ABTS•+ and DPPH• scavenging activity and FRAP. No significant correlation was found between the total phenolic content and the antioxidant activity in the emulsion system. The highest antiradical activity and FRAP were generally determined in older plants (mid-flowering and late flowering stages). In conclusion, the aerial parts of sunflowers, in particular those harvested at the mid-flowering stage, are a good plant material from which to obtain phenolic compound extracts, albeit mainly of one class (esters of caffeic acid and quinic acid), with high antioxidant activity.

DPPH• scavenging peptides (<3kDa) from underutilized Dunaliella salina protein were obtained by the following successive treatment, i.e., ultrasound extraction, simulated in vitro gastrointestinal digestion hydrolyzation, and membrane ultrafiltration classification. The optimal condition for ultrasound-assisted extraction was an ultrasound wave with 800 W of power treating a mixture of 60 mL of 1.0 mol L−1 NaOH and 2 g algae powder for 15 min. A high correlation (r=0.8146) between DPPH• scavenging activity and yield of the intact peptides showed their antioxidant capacity. Simulated in vitro digestion assay resulted in excellent DPPH• scavenging activity of the total peptide, amounting to (86.5 ± 10.1)%, comparing with the nondigestion samples at (46.8 ± 6.5)%. After fractionation, the 500-1000 Da fraction exhibited the highest DPPH• scavenging activity (81.2 ± 4.0)%, increasing 1.5 times due to digestion. Then, the 500-1000 Da fraction was analyzed by RPLC-Q Exactive HF mass spectrometer, and 4 novel peptides, i.e., Ile-Leu-Thr-Lys-Ala-Ala-Ile-Glu-Gly-Lys, Ile-Ile-Tyr-Phe-Gln-Gly-Lys, Asn-Asp-Pro-Ser-Thr-Val-Lys, and Thr-Val-Arg-Pro-Pro-Gln-Arg, were identified. From these amino acid sequences, hydrophobic residues accounted for 56%, which indicated their high antioxidant property. The results indicated that underutilized protein of Dunaliella salina could be a potential source of antioxidative peptides through simulated in vitro gastrointestinal digestion.

Antioxidant properties of crude extract, partition extract, and fermented medium from Dendrobium sabin (DS) flower were investigated. The oven-dried DS flower was extracted using 100% methanol (w/v), 100% ethanol (w/v), and 100% water (w/v). The 100% methanolic crude extract showed the highest total phenolic content (40.33 ± mg GAE/g extract) and the best antioxidant properties as shown by DPPH, ABTS, and FRAP assays. A correlation relationship between antioxidant activity and total phenolic content showed that phenolic compounds were the dominant antioxidant components in this flower extract. The microbial fermentation on DS flower medium showed a potential in increasing the phenolic content and DPPH scavenging activity. The TPC of final fermented medium showed approximately 18% increment, while the DPPH of fermented medium increased significantly to approximately 80% at the end of the fermentation. Dendrobium sabin (DS) flower showed very good potential properties of antioxidant in crude extract and partition extract as well as better antioxidant activity in the flower fermented medium.

Hippophae commonly known as sea-buckthorn, is a versatile plant with multipurpose uses including the control of soil erosion, biological nitrogen fixation and medicinal properties. In India, two species of sea-buckthorn; namely, Hippophae salicifolia D. Don and Hippophae rhamnoides L., are very common. H. salicifolia has a shrub-to-tree habit and is restricted to the Himalayan region, whereas H. rhamnoides is bushy, growing at higher altitude in India, and widely distributed in Europe and Asia. In this study, aqueous, methanol and acetone extracts of various parts of female and male plants of H. salicifolia were evaluated for total antioxidant activity, phenolic and flavonoid contents, and correlations were made. The total phenolic and flavonoid content in all the samples ranged from 99 ± 32 to 1459 ± 53 mg gallic acid equivalent g–1, 135 ± 12 to 707 ± 62 mg quercetin equivalent g–1 of dry extract, respectively. The highest free radical (DPPH) scavenging activity was found in methanol extracts of female bark and male leaf. Methanol extracts of male bark and male leaf showed maximum H2O2 activity at a concentration of 1 mg mL–1. Antioxidant activity (FRAP method) ranged from 0.021 to 1.041 at concentrations ranging from 20 to 200 µg mL–1 with a reducing capacity of extracts as water > methanol > acetone. Strong correlation (P < 0.05) was observed between DPPH scavenging activity and total phenolic content from all the extracts under study.

Water lily (Nymphaeaceae antares) contains a vast array of phenolic compounds with important natural antioxidative properties that could inhibit the adverse effects of the reactive oxygen species produced in living things. These substances are complex in composition which requires a suitable extraction medium to exploit the phenolic compounds. This study was designed to determine the antioxidative properties in petal and stamen of water lily extracted using different extraction solvents namely ethyl acetate and 60% methanol. Antioxidant properties of the samples were determined using 2,2-diphenyl -1-picrylhydrazyl (DPPH) radical scavenging activity, ferric thiocyanate (FTC) method, thiobarbituric acid (TBA) test, total phenolic content (TPC), total flavonoid content (TFC) and total tannin content (TTC) along with individual flavonoids content using High Performance Liquid Chromatography (HPLC). The yield of extract was found to be the highest in petals extracted with 60% methanol (44.55±1.63%) while stamen extracted with ethyl acetate yielded the lowest (24.50±1.09%). In the DPPH inhibition method, petal extracted with ethyl acetate obtained the highest scavenging activity (89.15±3.62%). FTC analysis revealed that ethyl acetate extract of petal experienced the highest percentage inhibition (76.70±6.48%) while in TBA analysis ethyl acetate extract of stamen exhibited the highest percentage inhibition (70.78±1.39%). Flavonoids such as quercetin, kaempferol, rutin, gallic acid, catechin, epigallocatechin, p-coumaric acid and myricetin were found to be present in ethyl acetate and 60% methanol extracts of water lilies stamen and petal. The information from this study indicates that water lily petals and stamens may be integrated as a good source of nutrients in the food industry, the pharmaceutical industry. If the advantages of water lily are not used and integrated into everyday life for health benefits, it will be a massive waste of natural resources.
