Listeria monocytogenes (Lm) is a ubiquitous bacterium that causes the foodborne illness, listeriosis. Clonal complexes (CC), such as CC121, are overrepresented in the food production industry, and are rarely reported in animals and the environment. Working within a European-wide project, we investigated the routes by which strains are transmitted from environments and animals to food and the food production environment (FPE). In this context, we report, for the first time, the occurrence of a ST121 (CC121) strain isolated from a dolphin brain. The genome was compared with the genomes of 376 CC121 strains. Genomic comparisons showed that 16 strains isolated from food were the closest to the dolphin strain. Like most of the food strains analyzed here, the dolphin strain included genomic features (transposon Tn6188, plasmid pLM6179), both described as being associated with the strain’s adaptation to the FPE. Like all 376 strains, the dolphin strain contained a truncated actA gene and inlA gene, both described as being associated with attenuated virulence. Despite this fact, the strain was able to cross blood-brain barrier in immunosuppressed dolphin exposed polychlorinated biphenyl and invaded by parasites. Our data suggest that the dolphin was infected by a food-related strain released into the Mediterranean Sea.

Listeria monocytogenes is the etiological agent of listeriosis, a foodborne illness associated with high hospitalizations and mortality rates. This bacterium can persist in food associated environments for years with isolates being increasingly linked to outbreaks. This review presents a discussion of genomes of Listeria monocytogenes which are commonly regarded as persisters within food production environments, as well as genes which are involved in mechanisms aiding this phenotype. Although criteria for the detection of persistence remain undefined, the advent of whole genome sequencing (WGS) and the development of bioinformatic tools have revolutionized the ability to find closely related strains. These advancements will facilitate the identification of mechanisms responsible for persistence among indistinguishable genomes. In turn, this will lead to improved assessments of the importance of biofilm formation, adaptation to stressful conditions and tolerance to sterilizers in relation to the persistence of this bacterium, all of which have been previously associated with this phenotype. Despite much research being published around the topic of persistence, more insights are required to further elucidate the nature of true persistence and its implications for public health.

ABSTRACT Listeria monocytogenes is the causative agent of the highly fatal foodborne disease listeriosis and can persist in food production environments. Recent research highlights the involvement of L. monocytogenes plasmids in different stress response mechanisms, which contribute to its survival in food production facilities. Ultraviolet (UV) light in the UVC spectrum (200 to 280 nm) is used in food production to control microbial contamination. Although plasmid-encoded UV resistance mechanisms have been described in other bacteria, no research indicates that L. monocytogenes plasmids contribute to the UV stress response. The plasmids of L. monocytogenes strains 6179, 4KSM, and R479a are genetically distinct and were utilized to study the roles of plasmids in the UV response. Wildtype and plasmid-cured variant cells were grown to logarithmic or late-stationary phase, plated on agar plates, and exposed to UVC for 60 or 90 s, and colony-forming units (CFUs) were determined. CFUs of 6179 and 4KSM, bearing pLM6179 and p4KSM, respectively, were significantly (p-value < 0.05) higher than the plasmid-cured strains in both logarithmic and stationary phases. No difference in survival was observed for the R479a strain. Our data show for the first time that certain L. monocytogenes plasmids contribute to the survival of UVC light stress.

The virulence of 82 Listeria monocytogenes isolates from human cases and cold-smoked salmon, cooked peeled shrimp, and their production environments was assessed using the plaque-forming assay and a subcutaneous inoculation test in mice. These isolates were previously typed using serotyping and pulsed-field gel electrophoresis. The isolates from food-production environments were collected in several surveys over the period of 5 years. Sixty-eight (99.8%) of 69 isolates tested from food and food-processing environments were considered virulent while only one was avirulent. All clinical isolates (13) were highly virulent. The isolates were from raw materials, final products, and the production environment. This stresses the importance of hygiene in the processing environment as well as among personnel to avoid contamination of the final product.

Introduction: Salmonella has been reported from foods and the food production environment, with outbreaks occurring in the human population worldwide. Methodology: A survey on Salmonella in two beef production lines (a beef abattoir line and a processing line) in Addis Ababa, Ethiopia was conducted, with a total of 668 various samples randomly collected from animal-related materials, the environment, and a beef product (mortadella). Results: Overall, a 12.9% prevalence (26.3% from the abattoir line, 5.3% from the processing plant line) was observed. The prevalence in the abattoir line environment (36.6%) was higher than that in animal-related samples (14.7%); the reverse was true for the processing plant line. Out of 86 isolates, 10 serovars were identified, and 8 remained unidentified. The predominant serotypes were S. Saintpaul (32.5%), S. Muenchen (19.8%), and S. Larochelle (12.8%). S. Kastrup and S. London were isolated for the first time in Ethiopia. Conclusions: Data indicate open ports of entry for Salmonella, with possible transfer along the line. Further investigations from farm to fork are recommended in order to identify these positions of entry.

The Strait of Messina is located at the centre of the Mediterranean Sea and is considered a biodiversity hotspot and an obligatory seasonal passage for different pelagic species such as sharks, marine mammals, and billfishes. For the first time, in the Strait of Messina, our research group tagged a Mediterranean spearfish (Tetrapturus belone) using a pop-up satellite archival tag (PSAT). The observation of abiotic parameters (depth, light, and temperature) recorded by the PSAT confirmed that the tagged specimen was predated after about nine hours. The tag was then regurgitated 14 days after the tag deployment date. The analysis of collected data seems to indicate that the predator may be an ectothermic shark, most likely the bluntnose sixgill shark (Hexanchus griseus).

Dinoflagellate infections have been reported for different protistan and animal hosts. We report, for the first time, the association between a dinoflagellate parasite and a rotifer host, tentativelySynchaetasp. (Rotifera), collected from the port of Valencia, NW Mediterranean Sea. The rotifer contained a sporangium with 100–200 thecate dinospores that develop synchronically through palintomic sporogenesis. This undescribed dinoflagellate forms a new and divergent fast-evolved lineage that branches among the dinokaryotic dinoflagellates.
