This study is the first approach to in vitro asymbiotic germination of two species of Sicilian threatened terrestrial orchids, Anacamptis longicornu and Ophrys panormitana. Seeds were collected in the wild and cultured in two different media—Orchimax medium (OM) and Murashige and Skoog (MS)—and exposed to different photoperiods and temperatures to evaluate the best conditions for the specific stages of development. The germination of A. longicornu was very high on OM (95.5%) and lower on MS medium (21.4%), whereas O. panormitana germinated only on OM medium, with significantly lower percentages (12.0%), compared with A. longicornu. This difference is caused by variation in quality and quantity of nutrients used, primarily by nitrogen source. The results show that temperature and photoperiod widely affect seed germination and development. Although further investigations on asymbiotic and symbiotic germination are needed for the improvement of conservation of Mediterranean terrestrial orchids, our results contribute to the conservation of this group of plants.

The effect of various pretreatments, culture conditions, and storage time on in vitro germination of seeds, as well as the effect of explant origin and plant growth regulators on in vitro propagation of Teucrium capitatum L. (Teucrium polium sp. capitatum Arcang., Lamiaceae) were examined. Seeds, collected from native plants and stored at room temperature for 3, 7, and 12 months, were cultured for germination in vitro in petri dishes with solid half-strength (½) Murashige and Skoog (1962) growth medium (MS) at 5, 10, 15, 20, 25, and 30 °C, and 16 hours light or continuous darkness. Pretreatments, such as cold stratification, scarification with sandpaper, dipping in concentrated sulfuric acid (H2SO4 > 95%), or dipping in boiling water were tested. Seeds without any pretreatment germinated at lower than 10%. Dipping in concentrated H2SO4 for 15 or 20 minutes was the most effective pretreatment, but still seed germination achieved was low (36%). Seeds preserved their germination capacity for at least 1 year, and germinated satisfactorily at a wide temperature range, from 15 to 25 °C (optimum), while photoperiod did not affect seed germination. Explants excised from in vitro-grown seedlings were established in vitro on MS medium with 1.0 mg·L−1 6-benzyladenine (BA) at much higher rates (≈90%) compared with those collected from plants grown from cuttings in a greenhouse (25%), while explants collected from adult wild plants failed to do so. Explants from seedlings showed strong variability in their response; those excised from branched seedlings formed shoots at significantly higher percentage (90%) at the establishment stage (cultured on MS medium with 0.5–2.0 mg·L−1 BA) than explants excised from unbranched seedlings (36% to 43%), while during subcultures on MS medium with 1.0 mg·L−1 BA, explants from branched seedlings also showed higher multiplication rates than those from unbranched ones. BA at 0.5–2.0 mg·L−1 induced shoot multiplication during both establishment and multiplication stages (7–8 and 14–15 shoots per explant, respectively), while kinetin and 6-γ-γ-(dimethylallylamino)-purine (2iP) were less effective than BA, and zeatin the least appropriate. Microshoot rooting was enhanced by 1-week culture on (½) MS medium with 1.0–4.0 mg·L−1 indole-3-butyric acid (IBA), followed by transfer to auxin-free, (½) MS medium (93% to 98%, 7–8 roots per microshoot), compared with culture on the same medium continuously for 5 weeks (69% to 80%, 5–6 roots per microshoot) or at lower IBA concentrations. Plantlets were acclimatized to ex vitro conditions at 98% on a peat–perlite (1:1, v/v) mixture.

Asymbiotic seed germination of Eulophia nuda Lindl. was observed on Knudson C medium. About 90% seeds germinated within 8-10 weeks and formed green protocorms in 11-12 weeks. Effect of BA and IBA was studied on plantlet development from protocorms. BA shows the best results with respect to number and length of shoots. Maximum number (6.45±1.36) and length (3.90±0.99) was observed on MS medium supplemented with 4.44μM.BA. Maximum root growth was also observed on same medium (4.8±0.99 number of roots and 1.43±0.13cm length). The regenerated plantlets were successfully acclimatized and transferred to earthen pots. The results presented here show that in vitro seed germination and plantlet development in Eulophia nuda Lindl., an endangered orchid, can be achieved at a higher rate by this method.Int J Appl Sci Biotechnol, Vol 3(2): 243-247 DOI: http://dx.doi.org/10.3126/ijasbt.v3i2.12476

This is the first report of an efficient and effective procedure to optimize the biosynthesis of huperzine A (HupA) and huperzine B (HupB) in vitro from Huperzia selago gametophytes. Axenic tissue cultures were established using spores collected from the sporophytes growing in the wild. The prothalia were obtained after 7–18 months. Approximately 90 up to 100% of the gametophytes were viable and grew rapidly after each transfer on to a fresh medium every 3 months. The best biomass growth index for prothallus calculated on a fresh (FW) and dry weight (DW) basis, at 24 weeks of culture, was 2500% (FW) and 2200% (DW), respectively. The huperzine A content in the gametophytes was very high and ranged from 0.74 mg/g to 4.73 mg/g DW. The highest yield HupA biosynthesis at >4 mg/g DW was observed on W/S medium without growth regulators at 8 to 24 weeks of culture. The highest HupB content ranged from 0.10 mg/g to 0.52 mg/g DW and was obtained on the same medium. The results demonstrate the superiority of H. selago gametophyte cultures, with the level of HupA biosynthesis approximately 42% higher compared to sporophyte cultures and 35-fold higher than when the alkaloid was isolated from H. serrata, its current source for the pharmaceutical industry. Moreover, the biosynthesis of HupB was several-fold more efficient than in H. selago sporophytes growing in the wild. HPLC-HR-MS analyses of the extracts identified eight new alkaloids previously unreported in H. selago: deacetylfawcettine, fawcettimine, 16-hydroxyhuperzine B, deacetyllycoclavine, annopodine, lycopecurine, des-N-methylfastigiatine and flabelline.

Blepharis maderaspatensis (L.) Heyne ex. Roth is a short-term plant which contains many important secondarycompounds with high medicinal value. Currently, most of the researches focus on chemical composition and pharmacological activity, but the source of raw materials is very limited. In this study, the first step is transferring the samples from nature into in vitro culture conditions to understand the effects of the factors related to shooting and callus morphogenesis was performed, the first node from shoots apical meristem was isolated and sterilized with 1.5% NaOCl for 20 minutes to achieve high efficiency with 86.11% sterile samples and 85.56% shoot growth rate after 2 weeks of culture on MS medium. The shoot generation from axillary shoots was continued to be investigated with the highest number of shoots formed on MS medium supplemented with BA (1 mg / l) showed 1.53 shoots/implant which the height and the number of leavesare 3.65cm and 6.67, respectively. Besides, the formation of callus from leaves of MS medium supplemented with 2.4 - D (0.25 mg / l) achieved the rate of 66.67% of cultured samples, forming good callus after 4 weeks of culture. The results of the study not only contribute importantly to understanding morphogenesis for micropropagation purposes but also serve as the scientific database for further studies at the cellular and molecular levels of this plant.

In vitro seed germination and embryo culture have been achieved in Nothapodytes foetida, this plant is known for its rich source of anticancer drug i. e., Camptothecin. In present study both normal and decoated seeds were subjected to different treatments viz., H2O, GA3, H2O2, H2SO4, chlorine water and mechanical scarification, further these were germinated on water agar medium (WA), filter paper bridge (FB), half strength MS (HMS) and full strength MS (FMS) medium. The highest percentage (69%) of germination was achieved from decoated seeds treated with 10mg/L GA3 and germinated on Filter Paper Bridge. And for embryo culture mature embryos were inoculated on MS medium containing various combination and concentrations of cytokinins (BAP, Kn and TDZ) and auxin (IAA and NAA) for rapid conversion into a plantlet. Among the different combinations of growth regulators; highest frequency (100%) of plantlet conversion was obtained on MS medium containing Kn (1.0mg/L) and NAA (0.2mg/L).

In vitro embryo morphogenesis and micropropagation of Dendrobium aggregatum Roxb. were described. The gradual developmental stages of embryos to seedlings were traced out. Within two weeks of culture the cells of undifferented embryos underwent repeated aniclinal and periclinal division producing a compact, green parenchymatous cell mass called spherule that emerged out by rupturing the testa. The spherules subsequently differentiated into greenish protocorms were considered as typical seed germination. Germination occurred on both (MS and Phytamax (PM) medium but MS medium proved to be more efficient. The primary protocorms underwent profuse proliferation through production of secondary (2º) protocorms when transferred to different plant growth regulators (PGRs) supplemented MS; the medium fortified with 2.0 mg/l BAP and 1.0 mg/l NAA proved to be most effective for induction of 2º protocorms and seedling development. Multiple shoot buds (MSBs) were induced in pseudobulb segments of the in vitro grown seedlings when cultured on different PGRs supplemented media; and the maximum number of MSBs were obtained MS + 2.0 mg/l BAP + 0.5 mg/l picloram. The MSBs underwent elongation and then they rooted when they were transferred to half strength of MS + 0.5 mg/l IAA. The well rooted plantlets were finally transferred to outside natural environment with 80% survival. D. O. Selaginella martensii, an evergreen perennial fern that is native to South America and New Zealand, is named “frosty fern” because of its beautiful white-colored leaves and it is used as an ornamental plant. Efficient propagation methods for this species have not been developed. We aimed to develop an efficient propagation method for S. martensii through in vitro culture. We investigated culture conditions that are suitable for shoot-tip proliferation and growth. The optimum shoot-tip culture conditions were determined while using Murashige and Skoog (MS) medium (quarter, half, full, or double strength) and macronutrients (sucrose and two nitrogen sources) at various concentrations. In MS medium, the shoot tips formed a maximum of 6.77 nodes per explant, and each node formed two new shoot tips (i.e., 26 or 64 shoot tips). When using branching segments containing an angle meristem, the shoot-to-rhizophore formation ratio could be controlled by medium supplementation with plant-growth regulators. Sporophytes that were grown from shoot tips in vitro were acclimated in ex vitro soil conditions and successfully survived in the greenhouse. Numerous shoot tips could be obtained from in vitro-grown sporophytes and be proliferated ex vitro to produce a large number of plants. This method provides a way of shortening the time that is required for producing a large stock of S. martensii planting material.

Seeds of almost all orchids depend on mycorrhizal fungi to induce their germination in the wild. The regulation of this symbiotic germination of orchid seeds involves complex crosstalk interactions between mycorrhizal establishment and the germination process. The aim of this study was to investigate the effect of gibberellins (GAs) on the symbiotic germination of Dendrobium officinale seeds and its functioning in the mutualistic interaction between orchid species and their mycobionts. To do this, we used liquid chromatograph-mass spectrometer to quantify endogenous hormones across different development stages between symbiotic and asymbiotic germination of D. officinale, as well as real-time quantitative PCR to investigate gene expression levels during seed germination under the different treatment concentrations of exogenous gibberellic acids (GA3). Our results showed that the level of endogenous GA3 was not significantly different between the asymbiotic and symbiotic germination groups, but the ratio of GA3 and abscisic acids (ABA) was significantly higher during symbiotic germination than asymbiotic germination. Exogenous GA3 treatment showed that a high concentration of GA3 could inhibit fungal colonization in the embryo cell and decrease the seed germination rate, but did not significantly affect asymbiotic germination or the growth of the free-living fungal mycelium. The expression of genes involved in the common symbiotic pathway (e.g., calcium-binding protein and calcium-dependent protein kinase) responded to the changed concentrations of exogenous GA3. Taken together, our results demonstrate that GA3 is probably a key signal molecule for crosstalk between the seed germination pathway and mycorrhiza symbiosis during the orchid seed symbiotic germination.

An experiment was conducted to study the in vitro culture of pointed gourd. Cotyledon rescued from physiologically matured seeds (PMS) and immatured seeds (IMS) of pointed gourd were used as explants. Cotyledon excised from PMS responded very well in all culture conditions. Plant regenerated from cotyledon of PMS ranged from 38 to 96% in different hormonal formulations of culture media. Highest percentage of shoot regeneration was observed in MS + 1.0 mg/l BAP and lowest in MS + 2.5 mg/l BAP. No plant regeneration was observed in cotyledon from immatured seeds. The highest percentage of root induction (99%) was achieved in half MS medium supplemented with 0.5 mg/l NAA. The regenerated plantlets were successfully established in earthen pot. In vitro seed germination and seedling development of Phaius tancarvilleae (L’Her.) Blume. was carried out on 0.8%(w/v) agar solidified MS Medium (Murashige and Skoog, 1962) without hormones or supplemented with different concentration and combination of Naphthalene acetic acid (NAA) and 6-benzylaminopurine (BAP). MS medium supplemented with 0.5 mg/l of BAP was the most ideal condition for early seed germination, protocorm formation and development of seedlings. Germination started after 7 weeks of culture and complete seedlings were obtained after 24 weeks of culture. This protocol might be helpful for mass propagation of orchids by asymbiotic seed germination.
