Effect of Some Chemical Mutagens on Nutrient Compositions of Sweetpotato (Ipomoea batatas (L.) Lam)

Effects of sodium azide (SA) and colchicine (COL) (chemical mutagens) on some nutrient compositions of four varieties of sweetpotato (Butter milk, TIS87/0087, UMUSPO/3 and UMUSPO/1) were investigated at the Biotechnology Laboratory of the National Root Crops Research Institute Umudike, Nigeria. The parent plants were treated with sodium azide and colchicine mutagens at concentrations of 0%, 0.03%, 0.05% and 0.07%. Storage roots of the M2V2 generations from the mutant plants were used for the nutrient composition analysis using recommended standard procedures. Nutrient traits studied were: moisture content (%), dry matter content (%), amylose and amylopectin contents (%), starch yield (%), starch content % based on fresh weight and starch content % based on dry weight. Mutants with higher dry matter content than the control were observed in TIS87/0087 treated with SA and COL 0.03%. SA and COL 0.05% produced mutants with higher amylose content (and lower amylopectin) than the control in TIS87/0087. UMUSPO/3 treated with SA 0.03% and SA 0.05% produced mutants with higher values for amylose content (and lower values for amylopectin) than the control; the same variety treated with colchicine (COL 0.05%), recorded higher value for amylose content (and lower value for amylopectin) than the control. These findings showed that sodium azide and colchicine mutagens were potent in inducing variations in nutrient composition of sweetpotato.

Abstract Key message Among the five cassava isoforms (MeAPL1–MeAPL5), MeAPL3 is responsible for determining storage root starch content. Degree of storage root postharvest physiological deterioration (PPD) is directly correlated with starch content. Abstract AGPase is heterotetramer composed of two small and two large subunits each coded by small gene families in higher plants. Studies in cassava (Manihot esculenta) identified and characterized five isoforms of Manihot esculenta ADP-glucose pyrophosphorylase large subunit (MeAPL1–MeAPL5) and employed virus induced gene silencing (VIGS) to show that MeAPL3 is the key isoform responsible for starch and dry matter accumulation in cassava storage roots. Silencing of MeAPL3 in cassava through stable transgenic lines resulted in plants displaying significant reduction in storage root starch and dry matter content (DMC) and induced a distinct phenotype associated with increased petiole/stem angle, resulting in a droopy leaf phenotype. Plants with reduced starch and DMC also displayed significantly reduced or no postharvest physiological deterioration (PPD) compared to controls and lines with high DMC and starch content. This provides strong evidence for direct relationships between starch/dry matter content and its role in PPD and canopy architecture traits in cassava.

The effect of harvest period on the quality of storage roots and leaves of cassava cultivars was determined in an experiment carried out in a randomized complete block design with four replications in a split plot scheme, with five cultivars in the plots and ten harvest times in the subplots. The IAC 13 cultivar had the highest rate of dry matter accumulation in the storage roots and the Mico cultivar the lowest. The period of least dry matter content in the storage roots occurred later for the Fécula Branca, Mico and IAC 14 cultivars, and the minimum starch content in the storage roots occurred later for the Fécula Branca and Mico cultivars. In general, the IAC 13, IAC 14 and Fécula Branca cultivars had higher dry matter content in the storage roots, while higher starch content in the dry and fresh matter were obtained in the Fécula Branca cultivar. The crude protein content in the leaves decreased as the plant aged.

This study was conducted to determine the changes in dry matter (DM), free sugar (FS) and starch contents in the three yam (D. rotundata) tuber regions under two storage environmental conditions (natural light- yam barn, or and darkness). Three varieties of D. rotundata (alaako, Dodoro and Odo) were studied. The dry matter content of the Head region was the highest (28.6%), followed by the Middle (26.9%) and Tail (22.3%) regions. Change in dry matter content of the tuber is mainly due to rapid changes in the dry matter content of the Tail region over time. Storage environment significantly (P< 0.05) affected dry matter content of the three tuber regions at 8 and 16 weeks in storage (WIS). Head regions per variety were lower in dry matter when stored under Light than in the Dark at 8 WIS, while the reverse was the case for the Tail. This may relate to the release of endodormancy. By 16 WIS, when tubers stored under Light had well developed sprouts and those stored in the Dark only showed the first sign of sprouting, all Head regions had higher dry matter content under Light than in Dark storage. This may be due to the presence of sprouts. Storage environment did not significantly affect starch content at the various tuber regions. Head regions had higher percentage free sugar content under Light than Dark storage, and these effects were manifested between 8 and 16 WIS.

The purpose of research is to identify the most adaptive and productive varieties of potatoes for the steppe zone of Southern Urals. The experience was laid by one-factor circuit in 3-fold repetition. The studies were conducted for middle- and middle-early varieties of potatoes domestic and foreign selection. Determined yield, marketability, starch and dry content in potato tubers. At 4 years of data were allocated potato varieties – Artemis (57.8 t/ha), Aerrow (52.0 t/ha) and Romano (49.5 t/ha). Distinguished highest yield of marketable products varieties: Artemis – 97.9%, Tarasov – 97.7%, and Aerrow Romano – 97.4%. The starch content in tubers was measurable by 10.2% (Ed Scarlett) to 16.0% (Kovalenko Memory), and the dry matter content in tubers ranged from 15.6% (Ed Scarlett) to 23.8% (Memory Kovalenko).

Abstract A study was carried out in Central Bohemia to understand the effect of row spacing and stand density on plant morphology, productivity and quality of silage maize in two row spacing treatments (0.70 m and 0.35 m) at two stand densities (92,000 plants/ha and 110,000 plants/ha). The results of the study showed that row spacing and stand density had no effect on plant height or weight; however, significantly higher ear ratio and dry matter content was found in narrow rows at 110,000 plants/ha. It was observed that plant morphology was affected more by the interaction between row spacing and stand density than by a single effect of tested factors. Significantly higher dry matter yield was recorded at higher stand density, but there was no row spacing × stand density interaction. Row spacing had no impact on the whole plant neutral detergent fiber (NDF) content, crude protein of stover and starch content of ear, while narrow rows resulted in almost significantly higher stover NDF content. Our results suggest that narrow rows could be advantageous for maize morphology and quality in cases where higher stand density is applied.

This study was conducted to determine the effects of use organo-mineral fertilizers and poultry manure on yield and quality of seed tubers different potatoes varieties (Solanum tuberosum L.). The study was designed in three replicated completely randomized block factorial design with eight potato cultivars (Madaline, Provento, Marfona, Agata, Arizona, Mustang, Rumba and Saviola) and including three different fertilizer as Hexaferm organo-mineral (%20 organics, 8-21-0+Zn), compost (1 t da-1) and fermented poultry manure (1 t da-1). In order to determine the effect of fertilizer forms on the yield and quality characteristics of different varieties of potatoes were examined features like output time of the tuber, output rates, plant height, stem number per hill, tuber yield per hill, total tuber yield, cracks tuber yield, dry matter content, starch content and specific gravity. Used fertilizers have no significant effect on output time of the tuber, output rates, plant height, stem number per hill, total tuber yield, dry matter content, starch content and specific gravity of varieties. The difference observed in terms of output time of the tuber, output rates, plant height, stem number per hill between the varieties was insignificant. The difference between the average tuber yield per hill, total tuber yield, cracks tuber yield, dry matter content, starch content and specific gravity of varieties were significant. Similarly, the fertilizers had a significant effect on the average tuber yield per hill of varieties . The significant interactions were found on traits average tuber yield per hill and average specific gravity between Fertilizer x Varieties. As a result, considered economically the fermented and compost poultry manure can be used instead of organo-mineral fertilizers in the production of seed potatoes.

Nghiên cứu ảnh hưởng của các nồng độ Na2SO3 (0, 100, 300, 500 ppm) đến 03 giống sắn KM 94, KM 21-12 và KM 444 trong năm 2019 tại vùng sinh thái gò đồi thị xã Hương Trà, tỉnh Thừa Thiên Huế nhằm mục đích xác định ảnh hưởng và nồng độ Na2SO3 phù hợp cho cây sắn. Kết quả đạt được là: 1) Phun Na2SO3 ở các nồng độ thí nghiệm đã tăng sinh trưởng thân - lá - củ, năng suất, chất lượng củ và hiệu quả kinh tế của 03 giống sắn thí nghiệm. 2) Tại nồng độ phun Na2SO3 từ 300 đến 500 ppm, so với đối chứng giống KM 94 tăng năng suất thực thu 19,5 - 20,2%, tỷ lệ sắn lát đạt 40,9 - 41,1%, hàm lượng tinh bột đạt 29,3 - 30,2% và năng suất ethanol tăng 23,6 - 26,6%; Giống KM 21-12 tăng năng suất thực thu 15,1 - 18,6%, tỷ lệ sắn lát đạt 40,5 - 41,0%, hàm lượng tinh bột đạt 29,7 - 29,8% và năng suất ethanol tăng 21,6 - 25,8%; Giống KM 444 tăng năng suất thực thu 18,4 - 19,9%, tỷ lệ sắn lát đạt 40,6 - 40,8%, hàm lượng tinh bột đạt 30,3 - 30,9% và năng suất ethanol tăng 24,9 - 25,7%. ABSTRACT The study on the effects of Na2SO3 concentrations (0, 100, 300, 500 ppm) on 03 cassava varieties (KM 94, KM 21-12 and KM444) was conducted in 2019 in hilly ecological area of Huong Tra town, Thua Thien Hue province to determine the effect and suitable concentration of Na2SO3 for cassava. The results showed that: 1) Spraying Na2SO3 at a rate of 640 liters/ha with experimental concentrations increased stem - leaf - root growth, yield and quality of roots and economic efficiency of all of 03 cassava varieties. 2) Spraying Na2SO3 at concentrations from 300 to 500 ppm, compared with the control, KM 94 increased root yield of 19.5 - 20.2%, dry matter content reached 40.9 - 41.1%, starch content reached 29.3 - 30.2% and ethanol production increased by 23.6 - 26.6%; KM 21 - 12 increased root yield of 15.1 - 18.6%, dry matter content reached 40.5 - 41.0%, starch content reached 29.7 - 29.8% and ethanol production increased by 21.6 - 25.8%; KM 444 increased root yield of 18.4 - 19.9%, dry matter content reached 40.6 - 40.8%, starch content reached 30.3 - 30.9% and ethanol production increased by 24.9 - 25.7%.

The effect of FP (a flat plot without mulch), FPM (a flat plot with film mulching), RM (on-ridge planting with full film mulching), and FM (on-furrow planting with full film mulching) on the tuber and its storage quality of continuous cropping potatoes was investigated. The results showed that with the increase of continuous cropping years, the potato yield was reduced year by year. The furrow-ridge mulching film can significantly increase potato yield and reduce small tubers. Among them, the yield of RM was the highest. From the perspective of tuber quality, RM had the highest dry matter content and starch content, while FP had the lowest. Meanwhile, the reducing sugar content of FP was always higher than that of other treatments. The amino acid content and vitamin C content of FPM, RM, and FM were higher than FP before and after storage. In addition, with the extension of storage time, the dry matter content, starch content, amino acid content, and vitamin C content of all treatments decreased, while the content of reducing sugar increased significantly. Thus, on-ridge planting with full film mulching (RM) can effectively improve potato yield and tuber quality before and after storage.
