Effect of Sweet Potato Anthocyanin (Ipomoea Batatas L.) on Levels of Follicle Stimulating Hormone and Folliculogenesis in Rattus Norvegicus Exposed to Cigarette Smoke

Background — Cigarette smoking negatively impacts the health of both active smokers and passive smokers. ROS (Reactive Oxygen Species) from cigarette smoke can cause damage to the ovary and inhibit ovarian follicle growth. Objectives — This study aimed to assess the effects of the anthocyanins from the sweet potato (Ipomoea batatas L.) purple cultivar on ovarian malondialdehyde (MDA) and 17β-estradiol levels in rats (Rattus norvegicus) exposed to cigarette smoke. Material and Methods — The Rattus norvegicus Wistar strain was used in this study. Thirty rats were divided into five groups. Cigarette smoke was given as two cigarettes/day for four minutes, in the morning and afternoon for eight weeks after rats were at the proestrus stage. MDA levelwere measured with the BIOXYTECH MDA-586TM spectrophotometric assay for malondialdehyde, whilst 17β-estradiol was measured by ELISA. Data were then analyzed using one-way ANOVA. Results — MDA level were significantly different between the negative control and positive control groups based on double comparison with the LSD test (p=0.000 <∝). The level of 17β-estradiol also showed asignificant difference between the negative and positive control groups. There was a significant correlation between MDA level and 17β-estradiol levels (p=0.000 <∝) in the group of rats exposed to cigarette smoke and treated with anthocyanins from sweet potato at doses of 40 mg and 80 mg. Conclusions — Anthocyanins from the sweet potato (Ipomoea batatas L.) purple cultivar given at various doses decreased ovarian MDA level and increased 17β-estradiol levels in female rats exposed to cigarette smoke.

Background: Cigarette smoke exposure is one of the source of free radicals that causes oxidative stress and decreases superoxide dismutase (SOD) level. Purple fleshed sweet potato is a type of food that contains antioxidants to neutralize oxidative stress.Objectives: To study the effect of purple fleshed sweet potato on superoxide dismutase (sod) level on rats exposed to cigarette smoke.Methods: This was a true experimental study with a post-test randomized control group design. The rats were randomized into 4 groups (6 rats in each group). The negative control group (K-) was treated with standard diet; the positive control group (K+) was treated with cigarette smoke exposure and standard diet; the treatment 1 (P1) group was treated with standard diet and purple fleshed sweet potato with the dose of 8 g / 200 g bw/day, and the treatment 2 (P2) group was treated with cigarette smoke exposure and purple fleshed sweet potato with the dose of 8 g /200 g bw/day.Results: There was a significant difference of SOD levels in each group (p=0.00) except between group K- and P1. Giving purple fleshed sweet potatoes increased SOD levels as much as 85.81±4.59 (P1). The K+ group had the lowest SOD level 22.34±3.98. The SOD level for K- group and P2 group was 82.27±4.59 and 67.73±6.68 respectively.Conclusion: The highest SOD level is on the treatment 1 group which is administered with purple fleshed sweet potato.

Cardiovascular disease is a disease caused by impaired functions of heart and blood vessel, one of those is coronary heart disease. This study aimed to identify the effect of purple sweet potato (Ipomoea batatas L) extract on MDA (Malondialdehyde) levels of male white rat (Rattus norvegicus Wistar strain) model of atherosclerosis. This research was an experimental study using Post-Test Only Control Group Design method, used 5 groups (positive control group, negative control, treatment 1, treatment 2, and treatment 3), each group contained 5 rats plus 2 reserve rats. ANOVA one way test obtained a significance of 0.00 (p <0.05). The regression test results showed the coefficient of determination R² = 0.71, which means a decrease in serum MDA levels of experimental animals is influenced by the dose of purple sweet potato extract administration as much as 71% while the remaining 29% can be influenced by factors outside the study. It can be concluded that purple sweet potato extract can affect serum MDA levels of experimental animals.

Penelitian ini bertujuan mengetahui efek injeksi inhibin B terhadap penurunan konsentrasi follicle stimulating hormone (FSH) di dalamserum pada tikus putih (Rattus norvegicus). Dalam penelitian ini digunakan 24 ekor tikus putih berjenis kelamin jantan dengan strain Wistar berumur 4 bulan dengan bobot badan 150-200 g. Tikus-tikus dikelompokkan secara acak ke dalam 4 kelompok, yaitu KK0, KP1, KP2, dan KP3, masing-masing kelompok terdiri atas 6 ekor. Kelompok KK0 merupakan kelompok kontrol hanya diinjeksi dengan phosphate buffer saline (PBS), sedangkan kelompok KP1, KP2, dan KP3 diinjeksi dengan inhibin B dengan dosis berturut-turut 25, 50, dan 100 pg/ekor. Injeksi inhibin B dilakukan secara intraperitoneum sebanyak 5 kali selama 48 hari dengan interval waktu 12 hari. Injeksi pertama inhibin B dilarutkan dengan0,05 ml PBS dan 0,05 ml Freud’s complete adjuvant (FCA). Injeksi kedua sampai kelima, inhibin B dilarutkan dengan 0,05 ml PBS dan 0,05 ml Freud’s incomplete adjuvant (FICA). Pada hari ke-6 setelah injeksi inhibin B terakhir, tikus dikorbankan secara dislocatio cervicalis,lalu darah dikoleksi langsung dari jantung dan didiamkan hingga didapatkan serum untuk pemeriksaan konsentrasi FSH dengan menggunakan metode enzyme-linked immunosorbent assay (ELISA). Hasil penelitian menunjukkan bahwa injeksi inhibin B dengan dosis 100 pg/ekor menurunkan konsentrasi FSH secara nyata (P<0,05) bila dibandingkan dengan kelompok kontrol. Berdasarkan hal tersebut, inhibin B berpeluang untuk dikembangkan sebagai kandidat kontrasepsi pria hormon berbasis peptida.
