Stress of Nutrients Deficiency on Growth and Lipid Accumulation in Oil-Rich Navicula sp.

Based on optimized aquatic diatom medium, treatment of nutrient deficiency was carried out to Navicula sp. and the analysis results of comprehensive biomass and total lipid content indicated that the optimal Tmass concentrations of TFeClB3B·3HB2BO,T TCdSOB4 Band Citric acid were 1.2×10P-5P mol/L, 0.8μg/L and 12 mg/L, respectively. Maximum biomass of Navicula sp. on this culture medium at 680 spectrophotometrically could reach 1.201. Final lipid content of this microalgae could be improved from 26.60% to 37.68% after the stress of nutrient deficiency treatment.

Chlorella ellipsoidea and Chlorococcum infusionum, promising microalgae for biodiesel feedstock production, were treated with ethylenediaminetetraacetate (EDTA) and phosphorous to induce stress which was then followed by flow cytometry to study the enhanced intracellular neutral lipid content. Treatment resulted in up to a threefold increase in total lipid content of Chlorella (41.8±1.9% at 16 days of incubation period) and more than twofold increases in Chlorococcum (31.3±1.0% at 18 days of incubation period) under phosphorous starvation in the culture. It was observed that maximum biomass yields in Chlorella and Chlorococcum were 1.56±0.06 and 2.17±0.12 g/L at 1.5 g/L of phosphorous after 20 and 18 days of incubation periods, respectively. The qualitative analyses of neutral lipid bodies under stress conditions were performed by confocal microscopy and revealed bright golden-yellow lipid droplets in stress exposed cells. Significant increase of monounsaturated fatty acids under the nutrient limited conditions was suitable to produce biodiesel. The maximum biomass (g/L) and lipid content (% dry cell weight) at different stresses showed significant results (p<0.05) by single-factor Analysis of Variance (ANOVA) followed by Duncan’s Multiple Range Test (DMRT).

Objective: Lipids are secreted into milk as bilayer-coated structures: milk lipid globules (MLGs). Adipophilin (ADRP) and perilipin 3 (TIP47) are associated with MLGs in human breast milk; however, the role of these proteins in milk lipid secretion is not fully understood. The aim of the study was to investigate levels of ADRP, TIP47 and total lipid content in human breast milk, their mutual correlations and dynamics during lactation. Research Methods &amp; Procedures: Milk samples from 22 healthy lactating women (Caucasian, Central European) were collected at five time points during lactation (1&ndash;3, 12&ndash;14, 29&ndash;30, 88&ndash;90 and 178&ndash;180 days postpartum). Mass spectrometry-based method was used for quantification of ADRP and TIP47 in the samples. The gravimetric method was used to determine milk total lipid content. Results: We observed distinctive trends in ADRP, TIP47 levels and lipid content in human breast milk during the first 6 months of lactation. We also found a significant association between lipid content and ADRP, lipid content and TIP47, and ADRP and TIP47 concentrations in breast milk at all sampling points. Moreover, we derived an equation for estimating the mean lipid content of milk based on ADRP concentration in human breast milk. Conclusions: A mass spectrometry-based method was developed for quantifying ADRP and TIP47 in human breast milk. Strong mutual correlations were found between ADRP, TIP47 and total lipid content in human breast milk.
