O presente estudo teve por objetivo realizar a identificação dos fungos encontrados na própolis produzido por Apis mellifera L. da Baía do Iguape, Brasil. Para tanto, foram utilizadas técnicas morfológicas, bioquímicas e moleculares, sendo averiguado o perfil de restrição gerado por espaçador interno transcrito (ITS1 e ITS4). O tamanho dos produtos de PCR foi analisado quanto ao perfil de restrição obtidos com endonuclease (HhaI, HaeIII e HinfI) por espécie. Foram identificadas dezesseis  espécies de fungos filamentosos: Flavodon flavus,  Aspergillus nomius, Aspergillus versicolor, Cladosporium sp., Coniothyrium sidae, Didymella sp., Paecilomyces variotii, Cladosporium cladosporioides, Penicillium citrinum, Fusarium incarnatum, Penicillium chermesinum, Phoma sp., Stagonosporopsis valerianellae, Phoma medicaginis, Paraphoma fimeti e Stagonosporopsis cucurbitacearum; e seis  espécies de leveduras: Candida tropicalis, Candida guiliermondii, Candida famata, Kodomala ohmeri, Trichosporon asahiu e Cryptococcus laurentii. Stagonosporopsis cucurbitacearum e leveduras pertencentes ao gênero Candida foram os microrganismos de maior ocorrência nas amostras da própolis provenientes da Baía do Iguape, Brasil.

Fungi is known to produce a wide range of biologically active metabolites and enzymes. Enzymes produced by fungi are utilized in food and pharmaceutical industries because of their rich enzymatic profile. Filamentous fungi are particularly interesting due to their high production of extracellular enzymes which has a large industrial potential. The aim of this study is to isolate potential soil fungi species that are able to produce functional enzymes for industries. Five Aspergillus species were successfully isolated from antibiotic overexposed soil (GPS coordinate of N3.093219 E101.40269) by standard microbiological method. The isolated fungi were identified via morphological observations and molecular tools; polymerase chain reactions, ITS 1 (5’- TCC GTA GGT GAA CCT GCG G3’) forward primer and ITS 4 (5’-TCC TCC GCT TAT TGA TAT GC-3’) reverse primer. The isolated fungi were identified as Aspergillus sydowii strain SCAU066, Aspergillus tamarii isolate TN-7, Aspergillus candidus strain KUFA 0062, Aspergillus versicolor isolate BAB-6580, and Aspergillus protuberus strain KAS 6024. Supernatant obtained via submerged fermentation of the isolated fungi in potato dextrose broth (PDB) and extracted via centrifugation was loaded onto specific media to screen for the production of xylanolytic, cellulolytic and amylolytic enzymes. The present findings indicate that Aspergillus sydowii strain SCAU066 and Aspergillus versicolor isolate BAB-6580 have great potential as an alternative source of xylanolytic, cellulolytic and amylolytic enzymes.

Background: Aspergillus Versicolor is a marine-derived fungus isolated from Hyrtios Erectus Red Sea sponge. Methods: The aim of this study was to carry out a pharmacological screening and investigation for the in vitro biological activity (antioxidant, cholinergic, antidiabetic and anticancer) of Aspergillus Versicolor crude extract’s active compounds by using different qualitative and quantitative methods. Results: The present study results showed that Aspergillus Versicolor crude extracts contain 0.6 mg total phenolic/mg crude extract. Aspergillus Versicolor also showed a potent antioxidative capacity by decreasing the oxidation of ABTS. The anticancer and inhibitory effects of Aspergillus Versicolor crude extracts on PTK and SHKI were found to be 75.29 % and 80.76%; respectively. The AChE inhibitory assay revealed that Aspergillus Versicolor extracts had an inhibitory percentage of 86.67%. Furthermore, the anti-inflammatory activity using COX1, COX2, TNF, and IL6 was 77.32, 85.21 %, 59.83%, and 56.15%; respectively. Additionally, the anti-viral effect using reverse transcriptase enzyme showed high antiviral activity with 92.10 %. Conclusion: The current study confirmed that the Aspergillus versicolor crude extract and its active constituents showed strong effects on diminishing the oxidative stress, neurodegenerative damage, antiinflammatory, anti-cancer and anti-viral, suggesting their beneficial role as a promising fermented product in the treatment of cancer, oxidative stress, Alzheimer's, anti-inflammatory and anti-viral diseases.

AbstractCladosporium cladosporioides causes asthma and superficial and deep infections, mostly in immunodeficient individuals and animals. This study aimed to investigate whether C. cladosporioides spores can enter the lungs through pulmonary circulation and influence pulmonary immune response. We intravenously injected mice with C. cladosporioides spore suspension and conducted several assays on the lungs. Pulmonary hemorrhage symptoms and congestion were most severe on days 1, 2, and 3 post-inoculation (PI). Extensive inflammatory cell infiltration occurred throughout the period of infection. More spores and hyphae colonizing the lungs were detected on days 1, 2, and 3 PI, and fewer spores and hyphae were observed within 21 d of infection. Numerous macrophages, dendritic cells, and neutrophils were observed on day 5 PI, along with upregulation of CD54, an intercellular adhesion molecule. Th1 and Th2 cells increased after infection; specifically, Th2 cells increased considerably on day 5 PI. These results suggest that days 2 and 5 PI represent the inflammatory peak in the lungs and that the Th2 and Th1 signaling pathways are potentially involved in pulmonary immune responses. In conclusion, the further adaptive immune responses played important roles in establishing effective pulmonary immunity against C. cladosporioides systemic infections based on innate immune responses.

The paper presents the influence of oregano extract (Origanum vulgare L.) on growth of Aspergillus versicolor, Emericella nidulans, Eurotium herbariorum, E. amstelodami, E. chevalieri and E. rubrum - producers of sterigmatocystin (STC) isolated from salads. Antifungal tests were performed by agar plates method. The composition of the active component of extract was determined by GC-MS method and the major components were: carvacrol (34.20%), triacetin (22.91%), carvone (18.05%), p-cymene (8.05%) and thymol (3.74%). The examined extract showed the ability to reduce mold growth at all applied concentrations. Minimum inhibitory concentrations (MIC) for E. nidulans, E. chevalieri and E. amstelodami were 2.5% (v/v) and over 2.5% (v/v) for A. versicolor. At 1.5% (v/v) concentration the extract completely inhibited the growth of E. rubrum, whereas higher dose of 2.5% (v/v) was fungicidal against E. herbariorum. Besides its sensory role in food products, the examined oregano extract also exhibits antifungal activities against producers of STC.

Storability of properly harvested, mature hulless oats (Avena nuda L. ’Terra’) of 9.7–10.5% moisture content was determined from a 24-mo field trial of two 11.5-t lots of oats stored in two wooden farm bins in Manitoba during 1976–78. Oats were stored safely with very low levels of infestation by preharvest fungi, mites and insects and negligible quality loss for up to about 20 mo. Afterwards the grain moisture contents at the bottom layers of both bins rose from about 10 to 13–16%; this led to heavy floor level infestations by the fungi Penicillium and Aspergillus versicolor, and mites Caloglyphus berlesei and Tarsonemus granarius. Fat acidity values rose from the first year levels of about 30 to 50–78 mg KOH/100 g of dry grain. Populations of Cryptolestes ferrugineus and Tribolium castaneum, introduced to one bin, did poorly during the first summer and failed to survive during the second year. A third beetle, Oryzaephilus surinamensis, continued to multiply in small numbers after 22 mo. Low storage temperature levels during most of the years seemed to have affected adversely the multiplication of insects and to a lesser extent mites in farm-stored oats.
