O presente estudo teve por objetivo realizar a identificação dos fungos encontrados na própolis produzido por Apis mellifera L. da Baía do Iguape, Brasil. Para tanto, foram utilizadas técnicas morfológicas, bioquímicas e moleculares, sendo averiguado o perfil de restrição gerado por espaçador interno transcrito (ITS1 e ITS4). O tamanho dos produtos de PCR foi analisado quanto ao perfil de restrição obtidos com endonuclease (HhaI, HaeIII e HinfI) por espécie. Foram identificadas dezesseis  espécies de fungos filamentosos: Flavodon flavus,  Aspergillus nomius, Aspergillus versicolor, Cladosporium sp., Coniothyrium sidae, Didymella sp., Paecilomyces variotii, Cladosporium cladosporioides, Penicillium citrinum, Fusarium incarnatum, Penicillium chermesinum, Phoma sp., Stagonosporopsis valerianellae, Phoma medicaginis, Paraphoma fimeti e Stagonosporopsis cucurbitacearum; e seis  espécies de leveduras: Candida tropicalis, Candida guiliermondii, Candida famata, Kodomala ohmeri, Trichosporon asahiu e Cryptococcus laurentii. Stagonosporopsis cucurbitacearum e leveduras pertencentes ao gênero Candida foram os microrganismos de maior ocorrência nas amostras da própolis provenientes da Baía do Iguape, Brasil.

Abstract: Aflatoxins, which are produced by Aspergillus flavus, Aspergillus nomius, and Aspergillus parasiticus, are a group of pentacyclic natural products with difuran and coumarin skeletons. They mainly include aflatoxin B1, B2, G1, G2, M1, and M2. Biologically, aflatoxins are of concern to human health as they can be present as contaminants in food products. The unique skeletons of aflatoxins and their risk to human health have led to the publication of nine remarkable total syntheses (including three asymmetric syntheses) and ten formal total syntheses (including four asymmetric formal syntheses) of aflatoxins in the past 55 years. To better understand the mechanism of the biological activity of aflatoxins and their presence in samples from the food industry, this review summarizes progress in the total synthesis of aflatoxins.

Sponge-derived fungi have recently attracted attention as an important source of interesting bioactive compounds. Aspergillus nomius NC06 was isolated from the marine sponge Neopetrosia chaliniformis. This fungus was cultured on rice medium and yielded four compounds including three new oxisterigmatocystins, namely, J, K, and L (1, 2, and 3), and one known compound, aspergillicin A (4). Structures of the compounds were elucidated by 1D and 2D NMR spectroscopy and by high-resolution mass spectrometry. The isolated compounds were tested for cytotoxic activity against HT 29 colon cancer cells, where compounds 1, 2, and 4 exhibited IC50 values of 1.21, 2.23, and 5.62 µg/mL, respectively. Under the fluorescence microscope by using a double staining method, HT 29 cells were observed to be viable, apoptotic, and necrotic after treatment with the cytotoxic compounds 1, 2, and 4. The result shows that compounds 1 and 2 were able to induce apoptosis and cell death in HT 29 cells.

Palmarosa essential oil (PEO) is an alternative to synthetic fungicides to control the contamination by food-deteriorating fungi, such as Aspergillus nomius. Nonetheless, the low long-term stability and volatility hamper its utilization. Thus, this study aimed to develop nanostructured lipid carriers (NLCs) containing PEO to improve its stability and consequently prolong the activity against A. nomius. A mixture design was applied to find the best preparation conditions for antifungal activity. The characterization analyses included size measurements, zeta potential (ζ-potential), entrapment efficiency (EE), and antifungal activity (by inhibition of mycelial growth (IMG) and/or in situ test (pre-contaminated Brazil nuts) tests). The nanocarriers presented particle sizes smaller than 300 nm, homogeneous size distribution, ζ-potential of −25.19 to −41.81 mV, and EE between 73.6 and 100%. The formulations F5 and F10 showed the highest IMG value (98.75%). Based on the regression model, three optimized formulations (OFs) were tested for antifungal activity (IMG and in situ test), which showed 100% of inhibition and prevented the deterioration of Brazil nuts by A. nomius. The preliminary stability test showed the maintenance of antifungal activity and physicochemical characteristics for 90 days. These results suggest a promising system as a biofungicide against A. nomius.

Isolation of marine fungi symbiont of green algae Bornetella sp as a producer of antibacterial compounds has been carried out. This study aims to obtain symbiont fungi from green algae Bornetella sp which produces antibacterial compounds. The symbiont fungus was isolated using the direct planting method. Screening for the antibacterial activity of pure symbiont fungi isolates against Escheriscia coli and Staphylococcus aureus, and was carried out by the agar diffusion method. The isolation results obtained 4 isolates, but the one with the most potential to inhibit the growth of the tested bacteria was MFALM2. Molecular characterization showed that the MFALM2 isolate was identified as Aspergillus nomius with a 100% closeness level. Keywords: isolation; marine fungi; Aspergillus nomius; green alga; Bornetella sp. AbstrakIsolasi jamur laut yang bersimbion dengan alga hijau Bornetella sp sebagai penghasil senyawa antibakteri telah dilakukan. Penelitian ini bertujuan untuk memperoleh jamur simbion dari alga hijau Bornetella sp yang menghasilkan senyawa antibakteri. Isolasi jamur simbion dilakukan dengan metode direct planting. Skrining aktivitas antibakteri isolat murni jamur simbion terhadap bakteri uji Escheriscia coli dan Staphylococcus aureus dilakukan dengan metode difusi agar.  Hasil isolasi diperoleh 4 isolat, namun yang paling berpotensi menghambat pertumbuhan bakteri uji adalah MFALM2. Karakterisasi molekuler menunjukkan bahwa isolat MFALM2 teridentifikasi sebagai Aspergillus nomius dengan tingkat keeratan sebesar 100%.Kata kunci: isolasi; jamur lau; Aspergillus nomius; alga hijau; Bornetella sp.

Endophytic fungi from medicinal plants were hardly ever reported as compared to those from soil and marine sources. The present results associate the presence of endophytic fungi in medicinal plants by isolating them from three medicinal plants i.e. Bombax ceiba, Aloe vera, and Ximenia americana. In the present research investigation, surface sterilization method and media were standardized, and 32 endophytic fungi were isolated from three medicinal plants. We assessed the competence of three different surface sterilization methods and four media for isolation of endophytic fungi. The RoVe method used was more effective in eliminating epiphytic microorganisms. Therefore by using new method we have isolated total of 32 fungal endophytes and those were belongs to Aspergillus nomius (63.20%), Aspergillus niger (41.60%), Thielaviopsis basicola (38.33%), Fusarium oxysporum (33.20%), Pestalotiopsis inflexa (27.20%), Nigrospora sphaerica (20%), Alternaria alternata (15.30%) and Phomopsis archeri (20%). This is the first report of successful isolation of endophytic fungi from the said medicinal plants, for using newly formulated surface sterilization method.
