Phlomis bruguieri is a native plant in Iran, which deserves phytochemical study. The aim of this study was evaluation of its iridoid contents along with its antioxidant capacity. Aerial parts of the plant material were extracted with methanol and applied on repeated normal column chromatography using hexane: chloroform (70:30), chloroform: methanol (90:10), and methanol (100%). The fraction eluted with methanol (100%) containing polar glycosides was selected and medium pressure liquid chromatography (MPLC) on a RP-18 cartridge. Fraction elute by MeOH: water 30:70 was loaded on HPLC on RP-18 column for final purification. Structure elucidation was done using 1D and 2D NMR, and mass spectra. Antioxidant activity including total antioxidant capacity, DPPH and FRAP methods were designated to assess the in vitro antioxidant capacities. This study indicated the presence of lamiide as iridoid compound in the aerial parts of Phlomis bruguieri for the first time in this plant [350 mg/2 kg; 0.0175 % (w/w)]. Lamiide showed moderate antioxidant activity using TAC (EC50: 55 μg/mL; ascorbic acid EC50: 7.5 μg/mL), DPPH (EC50: 116.2 ± 3.51 μg/mL; ascorbic acid:34.7 ± 0.97 μg/mL), and FRAP (EC50:>200 μg/mL; quercetin: 7.5 ± 0.68 μg/mL) methods.

The difference in the scavenging potential of the extracts can be due to variation in the percentage of bioactive compounds present in different solvents. Invitroantioxidant studiesobviouslyshow the methanolic concentrates of Cassia absushave better antioxidant activity. This result indicates that aerial parts of methanolic concentratesCassia absuscould serve as a natural antioxidant, which may be useful in preventfree radical-induced diseases.

AbstractSerum contains various antioxidant molecules that may provide important protection against free radical attack. The aim of this work was to assess the total antioxidant capacity of plasma and a marker of lipid per oxidation [(thiobarbituric acid-reactive substances (TBARS)] in plasma of healthy smoking and non-smoking young and elderly subjects. In addition, we investigated plasma concentrations of α-tocopherol, β-carotene, and ascorbic acid. In in vitro experiments, the effects of exogenous compounds (ascorbic acid, uric acid, Trolox) on total ferric-reducing activity of plasma (FRAP) were also tested. We demonstrated that total antioxidant capacity of plasma obtained from healthy non-smoking young subjects was significantly higher than plasma antioxidant capacity of smoking elderly subjects. The concentration of TBARS in young non-smoking volunteers was lower than that in young smokers. The concentration of TBARS in elderly non-smoking volunteers was lower than in elderly smokers. Plasma concentrations of alpha-tocopherol, beta-carotene and ascorbic acid were significantly lower in elderly smoker than in elderly non-smokers of the same age. No difference in the plasma levels of alpha-tocopherol, beta-carotene and ascorbic acid were found in 22-year-old smoking and non-smoking subjects. In vitro addition of ascorbic acid, uric acid, or Trolox to plasma samples significantly increased their total antioxidant capacity. Decrease of FRAP values and increase of TBARS concentrations is a significant physiologic condition of the aging process. Supplementation of antioxidants could be useful for the enhancement of antioxidant screen in plasma.

Cadaba farinosa (family Capparidaceae) is generally known as “Indian cadaba” in the traditional ayurvedic system. The current study, aerial parts of different concentrates (Pet.ether, ethyl acetate and methanol) of Cadaba farinose was evaluated for its in-vitro antioxidant potential by Diphenylpicrylhydrazyl radical, and total antioxidant activity taking ascorbate as standard and superoxide radical activity taking Quercetin as the standard for the in-vitro methods. The methanolic concentrates of Cadaba farinose& ascorbic acid exhibited antioxidant potential possessing IC50 208µg/ml & 66µg/ml (Diphenylpicrylhydrazyl radical) , 188µg/ml & 57µg/ml (total antioxidant). The methanolic concentrates of Cadaba farinose & quercetin exhibited antioxidant potential possessing IC50 252µg/ml & 60µg/ml (superoxide radical). The IC50 value was originated that methanolic concentrates of Cadaba farinose more efficient in Diphenylpicrylhydrazyl radical, superoxide radical activity, total antioxidant activity compared EA & PE concentrates. The difference in scavenging potential of the extracts can be due to variation in the percentage of bioactive compound present in different solvents. Invitro antioxidant studies show methanolic concentrates of Cadaba farinose have better antioxidant activity. This paper reports a phytochemical study of the aerial parts of Astragalus monspessulanus L. growing in Algeria. It deals with the isolation and structure elucidation of 13 known compounds. From the n-butanol extract, seven flavonoids 1?7, two saponins and one lignan were isolated. In addition, two phytosterols and one triterpenoid were isolated from the ethyl acetate extract. The structures of all the isolated products were determined by using 1D and 2D-NMR techniques, measurement of optical rotation, mass spectrometry ESI-MS and by comparison with literature data. Furthermore, the antioxidant activity of the n-butanol extract of the aerial parts of Astragalus monspessulanus L. was investigated using the DPPH radical scavenging and ferrous ion chelating assays. The n-butanol extract showed low (EC50 = 2.09?0.434 mg mL-1) to moderate (IC50 = 63.60?0.01 ?g mL-1) antioxidant activities depending on the test method.

Flemingia macrophylla locally known as ‘Charchara’ in Bangladesh is a medicinal herb. Being a member of the Fabaceae family it claims a number of medicinal uses including hypoglycemic activity, neuroprotective effect and gynecological remedies. Methanolic extract of F. macrophylla was investigated to evaluate its antioxidant and general toxic properties in this study. Antioxidant potential was evaluated using total antioxidant capacity, total phenol contents, total flavonoid contents and DPPH (1,1-diphenyl-2-picrylhydrazyl) assays. Both leaf and stem extracts of F. macrophylla were found to possess significant amount of phenolics and flavonoids, expressed as gallic acid equivalents (GAE) and quercetin equivalent (QE), respectively. Interestingly total phenol content was equal for both leaf and stem extract and it was 43.8 mg GAE/g. Total flavonoid content was found to be 64.4 and 39.1 mg QE/g for leaf and stem extract, respectively. The total antioxidant capacity was expressed as ascorbic acid equivalents (AAE) and for leaf it was 5.067 mg/gm AAE and for stem 0.8167 mg/gm AAE. DPPH scavenging activity was determined by comparing with ascorbic acid. The IC50 values were 19.95, 125.89 and 707.94 ?g/ml for ascorbic acid, leaf and stem extract, respectively. The results of the present study on methanolic extracts of F. macrophylla revealed the presence of moderate antioxidant activity and extract of leaves produced better results than stem of the plant. In brine shrimp lethality bioassay, methanolic extract of F. macrophylla exhibited insignificant cytotoxicity. Ipomoea pestigridis (Linn) (family Convolvulaceae) is commonly known as “Tiger Foot Morning Glory” in English and locally known as ‘Pulichuvadi’ or ‘Pulichuvadu’ in Malayalam. The current study, aerial parts of different concentrates(Pet.ether, ethyl acetate, and methanol) of I.pestigridis, was evaluated for its in-vitro antioxidant potential by nitric oxide activity, total antioxidant activity, iron chelating activity taking ascorbate & Ethylenediamine tetraacetate as the standard correspondingly. An IC50 value was originated that EA concentrates of I.pestigridis  more efficient in nitric oxide activity, total antioxidant activity, Iron chelating capacity compared methanolic & PE concentrates. The ethyl acetate concentrates of I.pestigridis  & ascorbic acid exhibited antioxidant potential possessing IC50 226µg/ml & 66µg/ml (Nitric oxide). 185µg/ml & 60µg/ml (total antioxidant) , 287µg/ml & 65µg/ml (iron-chelating Activity) respectively. The difference in the scavenging potential of the extracts can be due to variation in the percentage of bioactive compounds present in different solvents.  Invitro antioxidant studies obviously show  EA concentrates of I.pestigridis have better antioxidant activity. These results indicate that aerial parts of methanolic concentrate I.pestigridis  could serve as a natural antioxidant, which may be useful in preventing free radical-induced diseases.

Background: Stilbene phytalexis (1,2-diphenyloethylen) and benzamide are beneficial for human health. To increase the stilbene ring activity, a new series of its derivatives containing benzamide structure was synthesized and evaluated for their in vitro antioxidant power. Methods: 1H nuclear magnetic resonance, mass spectroscopy, and chromatographic analyses were used to confirm the successful synthesis. The antioxidant properties were determined by the elimination of , HO , DPPH , ABTS+ radicals, total antioxidant status (TAS) and the ferric reducing antioxidant activities (TAC) measurements. Results: Stilbenebenzamide compounds showed a wide spectrum of antioxidant ability, however their total antioxidant power was weaker than those of butylated hydroxytoluene (BHT), ascorbic acid, and resveratrol. The highest antiradical activity towards and HO was shown by the compounds with structures containing amine group (SBEBA, SBA) ( : 37.7 – 38.0% and 40.8 – 43.5%, HO : 29.8%, 28.7% inhibition, respectively) at1.25 mM concentration. The antiradical power of SBEBA (0.29) in DPPH assay was lower than those of resveratrol (1.83), ascorbic acid (3.63) and BHT (4.09). The TAS values of the synthesized compounds ranged from 152.9±5.3 to 240.2±6.7µM trolox equivalent/gram (TE/g) and were much lower than those of BHT (1304±43.0), reservatrol (1360±29.0) and ascorbic acid (2782±39.7) µM TE/g. Similarly, the TAC values ranging from 29.7±0.9 to 41.5±1.6 µM TE were weaker than that of resveratrol (239.2 ±6.7 µM TE/g). Conclusion: The results suggest that the presence of hydroxyl group in stilbene ring should be considered in further design of stilbenebenzamide compounds to enhance their antioxidant activity.

Leaves of Actinodaphne hookeri (AH) Meissn (family, lauraceae, pisa) are used traditionally in the treatment of diabetes, disorders of the urinary tract which are more common in Chhattisgarh and eastern part of India. It contains flavonoids and quercetin-3-rhamnoside as an important constituent. The leaves were extracted with 70% hydro alcoholic solvent. Preliminary phytochemical investigation showed the presence of triterpenoids, alkaloids, tannins, flavonoids, glycosides and carbohydrates. The antioxidant activity of 70% hydro alcoholic extracts of AH was evaluated in vitro by various experimental parameters such as b-carotene-linoleic acid, lipid peroxidation and total antioxidant capacity. Our results showed that Actinodaphne hookeri leaves displayed potent antioxidant properties, supporting the ethnomedical use given to this plant to treat diabetes and urinary disorder. Keywords: Actinodaphne hookeri; Antioxidant; Lipid peroxidation; Total antioxidant capacity. © 2009 JSR Publications. Antioxidants are substances that hamper or inhibit the oxidation of oxidizable substrates in the cells. The aerial parts of Blepharis maderaspatensis dry powder was extracted with various solvents (PE, EA and methanol) through Soxhlet extractor. The aerial parts of different concentrates (Pet.ether, ethyl acetate and methanol) of Blepharis maderaspatensis was evaluated for its in-vitro antioxidant potential by hydroxyl radical, total antioxidant activity taking ascorbate used as standard for the both methods and total flavonoids content was estimated as equivalent to rutin. The methanolic concentrates of Blepharis maderaspatensis & standard exhibited antioxidant potential possessing IC50 210µg/ml & 62µg/ml (hydroxyl radical) 200µg/ml & 57µg/ml (Total antioxidant activity) respectively. An IC50 value was originate that methanolic concentrates of Blepharis maderaspatensis more efficient in hydroxyl radical, total antioxidant activity compared EA & PE concentrates. The methanolic and EA concentrates of Blepharis maderaspatensis showed the total flavonoids content (11.230±0.22 and 2.858±0.56 mg of rutin equivalent/g of powder) respectively. The difference in scavenging potential of the extracts can be due to variation in the percentage of bioactive compound flavonoids present in methanolic extracts. Invitro antioxidant studies obviously show methanolic concentrates of Blepharis maderaspatensis have better antioxidant activity due to the presence of total flavonoids content. The findings from the study reveal that the antioxidative potentials of methanolic concentrates Blepharis maderaspatensis could serve as free radical inhibitors, possibly acting as natural antioxidants.

The antioxidant activity in vitro of methanol extract of Archidendron clypearia was evaluated by in vitro tests on isolated liver cells of mouses with ED50 value ​​ of 2.18 μg/mL compared to that of curcumin of 1.87 µg/mL. Using combined chromatographic methods, four compounds were isolated from chloroform extract of the Archidendron clypearia. Their structures were elucidated to be daucosterol, 1-octacosanol, docosenoic acid, and methyl gallate by 1D- and 2D-NMR spectroscopic methods and in comparison with those reported in the literature. This is the first report of these compounds from the plant. All four compounds showed quite high antioxidant activity, for which methyl gallate was the highest one.
