In this work we investigated the antidiabetic and antibacterial effect of Cissus sicyoides (CS) from Brazil. Diabetic rats that received water (A group) or extracts from the aerial parts of the plant (Cs group) during four weeks were employed. After this period, serum levels of glucose, cholesterol and triglycerides were measured. Glycemia was not affected by treatment with CS. However, there was an increased cholesterol and triglyceride level in Cs group. In addition, bioassay-guided fractionation of methanolic extract from aerial parts of CS was performed for isolation of antibacterial compounds.beta-Sitosterol and sitosterol-beta-D-glucopyranoside isolated showed antibacterial activity against Bacillus subtilis with minimal inhibitory concentrations (MICs) of 50 mug/ml and 100 mug/ml, respectively. In spite of popular belief, CS did not show antidiabetic activity. However, two compounds isolated from aerial parts of the plant (beta-sitosterol and sitosterol-beta-D-glucopyranoside) showed antibacterial activity.

The aim of this study is to evaluate the hypoglycemic activity of the methanolic extract of the aerial parts (leaf + stem) of Vitex on an animal model. Rats of theWistar strain of both sexes, N = 20 females for the short-term test and N = 28 males for the medium-term test, weighing between 125 g and 240 g are rendered diabetic by intraperitoneal injection of streptozotocin (55 mg/kg body weight) and divided into four groups. Triglyceride, total cholesterol, high-density lipoprotein (HDL), and low-density lipoprotein (LDL) cholesterol were determined by the enzymatic method using spectrophotometry, then for the blood glucose is measured by the Accu Chek sensor meter. The oral administration of methanolic extract of Vitex agnus castus at a dose of 300 mg/kg decreased blood glucose after 2 h for the short-term test; with regard to the medium-term test, there was a decrease in the glycemia and an improvement in the lipid profile for 21 days. These results confirm the antidiabetic effects of this plant.

Three new 5-deoxyflavonoid and dihydroflavonoids 2, 3 and 4 have been isolated from the methanolic extract of Abutioln pakistanicum aerial parts, for which structures were elucidated explicitly by extensive MS- and NMR-experiments. In addition to these, 3,7,4′-trihydroxy-3′-methoxy flavonol (1) is reported for the first time from Abutioln pakistanicum. Compound 2 and 4 are p-coumaric acid esters while compounds 2–4 exhibited α-glucosidase inhibitory activity. Docking studies indicated that the ability of flavonoids 2, 3 and 4 to form multiple hydrogen bonds with catalytically important residues is decisive hence is responsible for the inhibition activity. The docking results signified the observed in-vitro activity quite well which is in accordance with previously obtained conclusion that phenol moiety and hydroxyl group are critical for the inhibition of α-glucosidase enzyme.

The objective of the present study was to characterize chemical composition of hitherto unexamined aerial parts of Lactuca sativa var. angustana cv. Grüner Stern. In contrast to leafy and head varieties of the lettuces, asparagus lettuce grown in Europe is much less studied. Fractionation of a methanolic extract from leaves of L. sativa cv. Grüner Stern, supported with HPLC/DAD and 1H NMR analysis, led to the isolation and/or identification of numerous terpenoid and phenolic compounds, including five apocarotenoids—(-)-loliolide, (+)-dehydrovomifoliol, blumenol A, (6S,9S)-vomifoliol, and corchoionoside C; three sesquiterpene lactones; two lignans—((+)-syringaresinol and its 4-O-β-glucoside); five caffeic acid derivatives; and three flavonoids. Some of the compounds, to the best of our knowledge, have never been isolated from L. sativa before. Moreover, monolignols, phenolic acids and a tryptophan-derived alkaloid were found in the analyzed plant material. Stems, leaves and shoot tips of the asparagus lettuce were examined to assess their phenolics and sesquiterpene lactone content as well as DPPH scavenging activity. Another stem lettuce—L. sativa var. angustana cv. Karola, two cultivars of leafy lettuces and one species of wild lettuce—L. serriola, were also examined as a reference material using HPLC/DAD. The results have been discussed regarding our previous studies and the literature data available.

The detailed investigation of a methanolic extract of aerial parts of Achillea pannonica SCHEELE. within a chemotaxonomic study led to the isolation of 6 flavonoid glycosides. Besides rutin, apigenin-7-O-glucopyranoside, luteolin-7-O-glucopyranoside, apigenin-7-O-rutinoside and acacetin-7-O-rutinoside, an unusual flavondiglucoside was isolated. Its structure was established by UV, 1HNMR and 13C NMR spectroscopic methods including 2D-NMR techniques and ESI-MS as luteolin-7,4′-O-β-diglucoside. This substance is reported for the first time in the genus Achillea. Chemotaxonomic aspects are discussed briefly

Seven flavonoids were isolated from the butanol fraction of the methanolic extract of the aerial parts of Cynanchum acutum L. (Asclepiadaceae). All of which have been isolated for the first time from the genus Cynanchum. Their structures were established as quercetin 3-O-β-galacturonopyranoside (1), quercetin 7-O-β-glucopyranoside (2), tamarixtin 3-O-β-galacturonopyranoside (3), kaempferol 3-O-β-galacturonopyranoside (4), 8-hydroxyquercetin 3-O-β-galactopyranoside (5), tamarixtin 3-O-α-rhamnopyranoside (6), and tamarixtin 7-O-α-arabinopyranoside (7) on the basis of their chromatographic properties, chemical and spectroscopic data. The major isolated flavonoids 1, 2 and 3 were found to exhibit significant antioxidant and antidiabetic activities (by measuring blood glucose and insulin levels). This is the first report about the antioxidant and antidiabetic activities of compounds 1 - 3.

  Objective: The present study was aimed to evaluate antihyperglycemic and antihyperlipidemic activities of methanolic extract of Aristolochia bracteolata (MEAB) against dexamethasone-induced diabetic rat model.Methods: Methanolic extract was prepared by soxhlet extraction and was evaluated for antihyperglycemic and antihyperlipidemic activity using dexamethasone-induced model. The MEAB was administered orally at a dose of 200 and 400 mg/kg body weight glibenclamide was used as standard drug. On 0th and 11th day, blood was collected by retro-orbit plexus.Results: In this model blood glucose levels were determined on 0th and 11th days and MEAB significantly reduced the blood glucose levels in diabetic rats. The effect of MEAB on serum lipid profile such as total cholesterol (TC), triglycerides (TGs), low-density lipoprotein (LDL), very LDL (VLDL), and high-density lipoprotein (HDL) was also measured on the 11th day in the diabetic rats. Significant reduction in TC, TGs, LDL, and VLDL levels and improvement in HDL level were observed in diabetic rats.Conclusion: From the results, it was found that the MEAB possess antihyperglycemic and antihyperlipidemic activities.

Abstract Chemical investigation of the aerial parts of Ammania aegyptiaca ethanol extract (AEEE) revealed significant high concentrations of polyphenols and flavonoids content with notable antioxidant activity in DPPH, ORAC, and reducing power assay. New acylated diglucoside flavonol myricetin 3-O-β-4­C1-(6"-O-galloyl glucopyranoside) 7-O-β-4C1-glucopyranoside (MGGG) was isolated from aerial parts of AEEE along with four additional known phenolics, not characterized previously from AEEE. Moreover, powerful inhibitory effects of MGGG, AEEE, and all isolates against α-amylase, pancreatic lipase and β-glucosidase, were assessed. In addition, flexible molecular docking was used to reveal the inhibition towards digestive enzymes and confirmed that the MGGG interacted strongly with the active site residues of these enzymes with the highest binding free energy against β-glucosidase (DG=-8.98 kcal/mol) compared to the commercial drug Acarbose, thus justifying its dual management of diabetes and obesity. In streptozotocin (STZ) induced diabetic rats, AEEE significantly decreased high serum glucose, α-amylase activity, liver and kidney function markers and increased insulin level. Moreover, it improved lipid profile due to diabetes with increased SOD activity and inhibited of TBARS formation. Consequently, AEEE and MGGG are found useful in controlling the secondary complications associated with type 2 diabetes mellitus. Histopathological studies proved the decrease in the pancreas damage and agreed with the biochemical findings. These results provide evidence that AEEE and MGGG have potent antidiabetic activity, which warrants additional investigations.
