EFFECTS OF MOLYBDENUM AND SULFUR ON MINERALS IN THE DIGESTIVE TRACT OF STEERS

Four Holstein steers with cannulae in the rumen, proximal duodenum (PD) and terminal ileum (TI) were fed diets containing: (1) no added Mo or S; (2) Mo added at 10 mg kg−1; (3) S added at 3.0 g kg−1; and (4) Mo and S added at 10 mg and 3.0 g kg−1, respectively, in a Latin square design. Effects of Mo and S on minerals in the digestive tract of steers were evaluated. The solubility of Cu was lower with Mo (P < 0.01) and S (P < 0.01) at the PD and TI, and with Mo plus S (P < 0.05) at the TI. With Mo more (P = 0.07) Cu tended to be excreted in feces. Effects of Mo: Mn, reduced (P < 0.05) input to stomach region and higher (P < 0.01) absorption distal to PD; Zn, lower (P < 0.01) solubility at TI with less (P < 0.05) absorbed from large intestine (LI); Fe, more (P = 0.06) absorbed from LI with lower (P < 0.01) fecal excretion; P, lower (P < 0.05) input to stomach region with a tendency for less (P = 0.08) absorbed from small intestine (SI); Na, less (P < 0.05) absorbed from LI; K, lower (P < 0.05) apparent digestibility. Effects of S: Mn, slight tendency for higher (P = 0.10) absorption distal to PD; Zn, lower (P < 0.05) solubility at PD; P, less (P < 0.05) absorbed from SI and LI; Mg, tendency for less (P = 0.07) absorbed from stomach region and trend for more (P = 0.08) absorbed distal to PD. 1. Sheep fitted with a rumen fistula and either a re-entrant cannula at the proximal duodenum or a re-entrant cannula at the terminal ileum were given twice daily 480–520 g dry matter as fresh perennial ryegrass, or white or red clover. Flows of digesta were corrected to 100% recovery of chromic oxide.2. The quantities (g/24 h) of zinc and cobalt leaving the stomach were significantly greater than those in the food. No significant change was found in the quantities of copper and manganese. Significantly smaller quantities of Co (all three diets) and Zn (all diets except red clover) left the small intestine than those which entered this region. No significant differences in the quantities of Cu and Mn entering and leaving the small intestine were found. Significantly smaller quantities of Zn, Co, Cu and Mn were excreted in the faeces than entered the large intestine.3. From the flow results it was determined that there was a significant net secretion of Zn and Co in the stomach, and a significant net absorption of Zn (except with the red-clover diet) and Co from the small intestine, and of Zn, Co, Cu and Mn from the large intestine.

Excessive hunting and poaching for commercial purpose of Varanus salvator in Indonesia can cause a decline in this animal population. However, the scientific information of this animal especially about the biologic of organ system is rarely reported. Therefore, this case opens up opportunities for researching, which aims to study the anatomy of digestive tract of water monitor macroscopically. This research has been conducted in Biology Laboratory, University of Muhammadiyah Kupang for 5 months from March to August 2016. The digestive organ of this animal that has been preserved in alcohol 70% was obtained before from two males of water monitors. Preservation process: the animal were anesthetized, exsanguinated, and fixated in 4 paraformaldehyde by tissue perfusion method. Observations were performed to the visceral site and morphometrical of digestive tract. The resulted data was analysed descriptively and presented in tables and figures. The digestive tract of water monitor consist of esophagus, stomach, small intestine, large intestine and cloaca. The dimension of each organ is different based on its structures and functions. The esophagus of water monitor connects the mouth cavity and the stomach and also as the entrance of food to the stomach. Water monitor stomach were found in cranial part of abdomen, in left side of liver. The small intestine was longer than stomach and it is a winding muscular tube in abdomen in posterior side of liver. The large intestine consist of colon and cloaca, while cecum was not found. This channel was extend lateromedially in abdomen to cloaca between left and right kidneys. The cloaca was the end of digestive tract which excreted feces and urine. From this research, we can conclude that the digestive tract of water monitor consists of esophagus, stomach, small intestine, and large intestine. It’s difficult to differentiate small intestine and large intestine because there are no cecum.

1. The effect of drying and ensiling ryegrass on the site of digestion of the energy andcarbohydrate fractions was studied in sheep fitted with rumen cannulas and re-entrant can-nulas in the proximal duodenum and terminal ileum.2. The sheep were given fresh (frozen) grass, dried grass, wilted and unwilted silage pre-pared from herbage harvested from the same sward. The grass diets were offered twice dailyto each animal and paper impregnated with chromium sesquioxide was administered twicedaily into the rumen. Twenty-four hour collections of duodenal and ileal digesta, adjusted togive 100 yo recovery of Cr2O3, were analysed to determine the extent of digestion in the fore-stomachs, the small intestine and the caecum and colon.3. Total digestibility of the gross energy was similar for the fresh grass, dried grass andwilted silage diets (67·4,68·1 and67·5 %)but higher for the unwilted silage (72·0 %, P < 0·01).There was an increased flow of energy into the small intestine when the sheep were given driedgrass and unwilted silage. The proportion of the apparently digested energy lost within thesmall intestine was greater when the dried grass was given (302 yo) than when the fresh grasswas given (23·6 yo).4. Drying or ensiling of wilted material affected digestion neither in the entire alimentarytract nor in the different sections of the tract, of some carbohydrate fractions. About 97 yo ofthe digested water-soluble carbohydrate, over 90 yo of the digested cellulose and over 70 yo ofthe digested hemicellulose were digested before reaching the small intestine. The increasedamount of energy entering the duodenum of the sheep given the dried grass was notaccounted for by changes in the fate of these carbohydrate fractions in the digestive tract. Withunwilted silage, digestibilities of the cellulose and hemicellulose fractions were higher, andlower proportions of the digested carbohydrates were lost before the small intestine.

1. Six 45 kg pigs with re-entrant ileal cannulas were used in two 3 × 3 Latin-square design experiments to study the site of absorption of protein and amino acids. Semi-purified diets containing soya-bean meal (SBM), rapeseed meal (RSM) or no protein source (protein-free) were offered at the rate of 1 kg dry matter/d.2. Flow-rates of ileal contents for 24 h collection periods, corrected for recovery of marker, were 3135, 3127 and 1243 ml (SE 390) for SBM, RSM and protein-free diets respectively.3. Amounts of dry matter digested in the small intestine were 730, 669 and 809 g/d for SBM, RSM and protein-free diets respectively, all values being significantly different (P < 0·001).4. Nitrogen intakes were 32·6, 29·9 and 5·9 g/d, and amounts digested in the small intestine were 25·7, 20·2 and 1·6 g/d for SBM, RSM and protein-free diets respectively, all values being significantly different (P < 0·001). Amounts digested in the large intestine were 2·6, 3·7 and 0·7 g/d.5. Total amino acid intakes and amounts collected at the ileum and in the faeces were (g/d): SBM, 177, 24 and 18; RSM, 149, 28 and 22; protein-free 3, 9 and 12. Digestibility in the small intestine was higher for SBM than RSM for seventeen of the eighteen amino acids estimated. Greater quantities of arginine, methionine, cystine and tyrosine were voided in the faeces than passed through the ileal cannulas for pigs receiving the SBM and RSM diets. For those receiving the protein-free diet this was true for each amino acid except proline.6. Significant differences were found between all diets in the concentration of some amino acids in ileal and faecal amino-N, and endogenous protein secretions did not mask the differences between diets.7. Differences in digestibility between SBM and RSM were greater at the ileum than in the faeces. Amino acid fermentation in the large intestine obscured or reduced differences between SBM and RSM.

This study used a digesta transfer protocol to determine the site of absorption of lysine synthesized by the gastrointestinal microflora of pigs. Eight pigs were used, four with reentrant cannulas in the terminal ileum, two with simple T cannulas in the terminal ileum, and two intact. All pigs were given, for 5 days, the same low-protein diet that included fermentable carbohydrates. The diet of two pigs with reentrant cannulas (donor) and of the two intact (control) pigs was supplemented with15NH4Cl. The two other pigs with reentrant cannulas (acceptor pigs) and those with simple cannulas (used to supply unlabeled digesta) were given the same diet but unlabeled NH4Cl. Ileal digesta were collected continuously from all of the reentrant cannulas and kept on ice. All digesta from each donor pig were reheated and returned to the distal cannula of its companion acceptor, whose ileal digesta were discarded. Unlabeled ileal digesta from the pigs with simple cannulas were instilled into the distal cannulas of the donor pigs. At the end of the experiment, the average15N enrichment in the plasma free lysine of control pigs was 0.0407 atom % excess (APE); that of donor pigs was 0.0322 APE (79% of controls), whereas that of acceptor pigs was only 0.0096 APE (24% of controls). Due to nitrogen recycling, acceptor pigs had labeled lysine in the digesta of the stomach and small intestine, and donor pigs had labeled lysine in the digesta of the large intestine. If account is taken of the higher 15N enrichment of microbial lysine in the large compared with the small intestine, it can be estimated that >90% of the absorption of microbial lysine took place in the small intestine.

Anin saccomobile bag technique was used to determine the rate and extent of disappearance of unmolassed sugar-beet pulp, soyabean hulls, hay cubes and an oat hull–naked oats mixture (67:33, w/w) in the foregut and total digestive tract of ponies. Ponies were administered naso-gastrically polyester mesh bags (60×10×10 mm) containing 350 mg feed, in a 3×4 Latin square design. Bags were collected at the ileo-caecal junction (small intestine bag, SIB) and in the faeces (faecal bag, FB) and their residues analysed for proximate constituents and NSP composition and content. DM disappearances from individual bags were fitted to degradation profiles () and effective degradability values determined. Significant differences (P<0·05) in DM, organic matter and crude protein (N×6·25) losses from SIB were noted between feeds with hay cubes and oat hulls–naked oats > sugar-beet pulp and soyabean hulls. Acid-detergent fibre, neutral-detergent fibre and NSP disappearances were small and varied little between feeds. In contrast, FB losses showed significant (P<0·05) differences for all constituents measured with sugar-beet pulp > soyabean hulls > hay cubes > oat hulls–naked oats. Crude protein losses from sugar-beet pulp and soyabean hulls in FB were significantly higher (P<0·05) than from SIB. FB degradation curves showed degradation to be affected by bag residence time with sugar-beet pulp > soyabean hulls and the effective degradability showed that significantly more (P<0·05) sugar-beet pulp was lost at 40.0 and 60.0 h than for the other three feeds. These results show that sugar-beet pulp and soyabean hulls are rapidly degraded by ponies and could be used as alternatives to hay in equid rations. However, the foregut availability of crude protein from sugar-beet pulp is poor, so a readily digestible source of crude protein should be offered to animals with high protein demands when diets are based on sugar-beet pulp.

SummaryCircadian variation in the apparent digestibility of diets having different physical characteristics was measured in samples taken from the terminal ileum using simple cannulae and marker ratios. Six pigs were used in three latin squares involving three iso-nitrogenous diets (30 g N/kg D.M.). Diet A was based on barley, weatings, soyabean meal and fish meal, diet B included barley, weatings and oats and diet C was purified. The allocation of diet provided 100 g D.M./kg Weg0·75/24 h and in each latin square a different pattern of feed intake was used; diets were given at intervals of either 1 or 12 h, or ad libitum.Differences in the digestibility of the diets were consistently distinguished by the technique. The circadian variation in digestibility was related to the type of diet and could be modified by changing the number and distribution of feeds per day. The results show that an understanding of the variation associated with different diets and feeding methods is necessary for optimizing the strategy for sampling from the terminal ileum.

This study aimed to determine the cellulase activity in broilers digestive tract after the treatment of soya wastes and palm kernel fermented by A.niger (AKBIS Prob) in the ration. This study used a complete randomized factorial design, which consists of two factors, namely the AKBIS Prob composition factor and the duration of AKBIS Prob. Twenty four broilers were divided into four groups by randomized and three replications of each. The feed supplement combinations were P0 (commercial), P1 (commercial and AKBIS Prob 2%), P2 (commercial and AKBIS Prob 4%), and P3 (commercial and AKBIS Prob 6%). The intestine preparation was taken at the 22 and 36 days to extraction and detection the cellulase activity concentration. Data were analyzed using variance analysis of factorial patterns. AKBIS Prob 2,4 and 6% was no effect (P 0.05) to the cellulase activity in the small and large intestine, while the treatment duration was significant (P0,05) in the small intestine but neither in the large intestine (P0,05) to the cellulase activity. The addition of AKBIS Prob 2.4 and 6% had no effect (P 0.05) on the concentration of cellulase activity in the small intestine and large intestine, while the duration the treatment was very significant (P 0.05) in the small intestine and neither in the large intestine (P 0.05) on the concentration of cellulase activity. The AKBIS administration in the ration for 22 days can increase the activity and concentration of cellulase activity in the small intestine of the digestive tract of broiler chickens.

A latin square design was used with four sheep, each equipped with a rumen fistula and re-entrant cannula in the proximal duodenum. The four treatments were intraruminal dosings with different amounts (0, 450, 900 and 1800 mg/day) of nitrilotriacetic acid (NTA). The sheep were fed a pelleted diet with excessive concentrations of manganese, iron, zinc and copper. Fecal excretion of iron and zinc increased, and apparent absorption decreased, with increasing dose of NTA. However, only the differences between means for zinc were statistically significant. Fecal excretion and apparent absorption of manganese and copper were not affected by NTA. Flow of soluble iron through the duodenum increased with increasing dose of NTA. There was no effect of NTA on concentrations of manganese, iron, zinc and copper in the rumen bacteria or on microbial activity in the rumen as measured by the concentration of volatile fatty acids in the rumen and by flow of nitrogen into the small intestine.

The effect of heat treatment on canola meal (CM) dry matter (DM) and nitrogen (N) degradability in the rumen and digestion in the small and large intestines and total digestive tract of steers was determined. CM was moist-heat-treated at 127 °C for 15 and 45 min. The degradation of DM and N was estimated using small nylon bags in the rumen for 0.1, 4, 8, 12, 16, 24, 48 and 72 h. The mobile bag technique with an in vitro incubation in an acid-pepsin solution was used with a separate set of bags to estimate the digestion of each nutrient in the small and large intestines as well as the total digestive tract for rumen incubation times of 8, 16 and 24 h. Chemical analysis of heated CM showed a significant reduction in soluble N, sucrose and glucosinolate content while the concentration of ADIN increased. Treatment significantly reduced the degradation of DM and N in the rumen while the digestibility of DM and N in the small intestine increased with both treatments proportional to the decreased rumen degradation. Heat treatment had no effect on DM and N digestibility at the large intestine site. Total tract disappearance of DM and N were not affected by 15 min heating; however, after 45 min heating, DM and N disappearances were reduced. Moist heat treatment was effective in reducing rumen disappearance of CM nitrogen while it increased the N digestion in the small intestine.
