Objective: The objective of the present study was to evaluate the total phenolic (TPC) and flavonoid content (TFC) of five less utilized fruits such as Aegle marmelos, Spondias pinnata, Limonia acidissima, Averhoa carambola, Crescentia cujete and was compared with Phyllanthus emblica, (Amla) well known for its antioxidant activities. Total phenolic and flavonoid contents of samples were correlated with antioxidant activities like 1,1-Diphenyl-2-Picryl-Hydrazyl (DPPH) free radical scavenging assay, Ferric reducing antioxidant potential (FRAP) assay and total antioxidant capacity (TAC).Methods: The total phenolic of each fruit extract were determined by the Folin-Ciocalteu method with some modifications and the total flavonoids were estimated by Aluminum trichloride colourimetric method. The DPPH antioxidant assay, The FRAP assay and TAC were determined spectrophotometrically.Results: The total phenolics were expressed as mg/100g Gallic acid equivalent (mg GAE/100 gm) and the total flavonoids were expressed as mg/100g Quercetin equivalent (mg QE/100 gm). TPC was found to be maximum in Spondias pinnata with 142.16 mg GAE/100 gm where as TFC was maximum in Phyllanthus emblica with 91.1 mgQE/100 gm. DPPH radical scavenging activity was expressed n percentage(%), FRAP values expressed as mg/100g Ascorbic equivalent (AAE) and the total antioxidant activity was expressed as mg/100g Ascorbic equivalent. Maximum DPPH radical scavenging activity was shown by Spondias pinnata (93.75%), FRAP values were maximum in Phyllanthus emblica with 72.6 mg AAE/100 gm and total antioxidant capacity was found to be highest in Spondias pinnata (50.1 mg AAE/100 gm).Conclusion: Spondias pinnata, an underutilized fruit, was found to be promising with antioxidant activities comparable to Phyllanthus emblica.

Phyllanthus emblicaL. (amla) has been used in Ayurveda as a potent rasayan for treatment of hepatic disorders. Most of the pharmacological studies, however, are largely focused on PE fruit, while the rest of the parts of PE, particularly, bark, remain underinvestigated. Therefore, we aimed to investigate the protective effect of the hydroalcoholic extract ofPhyllanthus emblicabark (PEE) in ethanol-induced hepatotoxicity model in rats. Total phenolic, flavonoid, and tannin content and in vitro antioxidant activities were determined by using H2O2scavenging and ABTS decolorization assays. Our results showed that PEE was rich in total phenols (99.523±1.91 mg GAE/g), total flavonoids (389.33±1.25 mg quercetin hydrate/g), and total tannins (310±0.21 mg catechin/g), which clearly support its strong antioxidant potential. HPTLC-based quantitative analysis revealed the presence of the potent antioxidants gallic acid (25.05 mg/g) and ellagic acid (13.31 mg/g). Moreover, one-month PEE treatment (500 and 1000 mg/kg, p.o.) followed by 30-day 70% ethanol (10 mL/kg) administration showed hepatoprotection as evidenced by significant restoration of ALT (p<0.01), AST (p<0.001), ALP (p<0.05), and TP (p<0.001) and further confirmed by liver histopathology. PEE-mediated hepatoprotection could be due to its free radical scavenging and antioxidant activity that may be ascribed to its antioxidant components, namely, ellagic acid and gallic acid. Thus, the results of the present study support the therapeutic claims made in Ayurveda aboutPhyllanthus emblica.

Abstract. In this study, cytoprotective and antioxidant activities of Rosa canina (RC) and Salix alba (SA), medicinal plants, were studied on mouse primary splenocytes by comparing Controlled Differential Sieving process (CDSp), which is a novel green solvent-free process, versus a conventional technique, employing hydroethanolic extraction (HEE). Thus, preventive antioxidant activity of three plant powders of homogeneous particle sizes, 50–100 µm, 100–180 µm and 180–315 µm, dissolved directly in the cellular buffer, were compared to those of hydroethanolic (HE) extract, at 2 concentrations (250 and 500 µg/mL) in H2O2-treated spleen cells. Overall, compared to HE extract, the superfine powders, i. e., fractions < 180 µm, at the lowest concentration, resulted in greater reactive oxygen species (ROS) elimination, increased glutathione peroxidase (GPx) activity and lower malondialdehyde (MDA) production. Better antioxidant and preventive effects in pre-treated cells were found with the superfine powders for SA (i. e., 50–100 µm and 100–180 µm, both p < 0.001), and with the intermediate powder for RC (i. e., 100–180 µm, p < 0.05) versus HE extract. The activity levels of catalase (CAT) and superoxide dismutase (SOD) in pretreated splenocytes exposed to H2O2, albeit reduced, were near to those in unexposed cells, suggesting that pretreatment with the fine powders has relatively restored the normal levels of antioxidant-related enzymes. These findings supported that CDSp improved the biological activities of plants, avoiding the use of organic solvents and thus it could be a good alternative to conventional extraction techniques.

In the present study, the nutritional quality of four grains including adlay (AD), buckwheat (BW), glutinous barley (GB), and white rice (WR) were evaluated in terms of plasma lipid parameters, gut transit time, and thickness of the aortic wall in rats. The rats were then raised for 4 weeks on the high-fat diet based on the American Institute of Nutrition-93 (AIN-93 G) diets containing 1 % cholesterol and 20 % dietary lipids. Forty male rats were divided into 4 groups and raised for 4 weeks with a diet containing one of the following grains: WR, AD, BW, or WB. The level of thiobarbituric acid-reactive substances (TBARS) in liver was shown to be higher in rats by the order of those fed WR, AD, GB, and BW. This indicates that other grains decreased oxidative stress in vivo more than WR. The superoxide dismutase, glutathione, glutathione peroxidase, and glutathione reductase levels in the AD, BW, and GB groups were significantly higher than those in the WR group (p < 0.05). Plasma lipid profiles differed significantly according to grain combination, and decreased aortic wall thickness was consistent with the finding of decreased plasma low-density lipoprotein cholesterol (LDL-C) (p < 0.05) and increased high-density lipoprotein (HDL-C) in rats fed AD, BW, and GB (p < 0.001). The antioxidant and hypolipidemic capacities of grains are quite high, especially those of adlay, buckwheat, and glutinous barley. In conclusion, this study has demonstrated that the whole grains had a cardioprotective effect. This effect was related to several mechanisms that corresponded to lowering plasma lipids, decreasing TBARS, and increasing antioxidant activities.
