Identification of main chemical constituents of diterpene lactone effective fraction of Andrographis panniculata by HPLC-DAD/ ESI-MS and their preliminary pharmacodynamics research

A phytochemical study on the bark of Walsura pinnata has led to the isolation of a new oleanane triterpene acid, 3-oxo-olean-9(11),12-dien-28-oic acid (1), together with nine known compounds (2–10). Their structures were established on the basis of the detailed spectroscopic analysis, including one- and two-dimensional NMR, ESI-MS and HR-ESI-MS techniques. Compounds 2, 3, 5, 6 and 8 were isolated from W. pinnata for the first time. Compounds 3 and 4 showed in vitro growth inhibitory activity against two human cancer cell lines MCF-7 and SK-OV-3 with IC50 values within the range of 8.85 - 18.28 μg/mL. To the best of our knowledge, this is the first report on the cytotoxic activity of compound 3 towards both cancer cell lines.

Agarwood is the resinous wood produced in some Aquilaria species and is highly valued for wide usages in medicine, incense, and perfume. To protect the threatened Aquilaria species, the cultivation of Aquilaria sinensis and artificial agarwood induction techniques have been effectively established in China. To evaluate the quality of agarwood induced by different techniques, patterns of chemical constituents in artificial agarwood by four methods (wounding using an axe, burning-chisel-drilling, chemical inducer, and biological inoculation) were analyzed and compared by UPLC-ESI-MS/MS and GC-EI-MS in this study. Results of GC-MS gave a panorama of chemical constituents in agarwood, including aromatic compounds, steroids, fatty acids, sesquiterpenoids, and 2-(2-phenlyethyl)-chromones (PECs). Sesquiterpenoids were dominant in agarwood induced by wounding using an axe. PEC comprised over 60% of components in agarwood produced by biological inoculation and chemical inducers. PECs were identified by UPLC-ESI-MS/MS in all artificial agarwood and the relative contents varied in different groups. Tetrahydro-2-(2-phenylethyl)-chromones (THPECs) in wounding by axes induced agarwood were lower while 2-(2-phenylethyl)-chromones (FPECs) were higher than other groups. The results showed that methods used for inducing agarwood formation in Aquilaria sinensis affect the chemical constituents of agarwood.

A new ergosterol, 15β-hydroxyl-(22 E,24 R)-ergosta-3,5,8,22-tetraen-one (1), along with three known ergosterols, two known cytochalasins, and two known azapholines were isolated from Chaetomium globosum Z1. The structures of these compounds were elucidated on the basis of spectroscopic methods (HR-ESI-MS, 1D NMR, and 2D NMR). Compound 6 showed significant cytotoxic activity against A-549 and MG-63 cell lines with IC50 values of 6.96 and 1.73 μg/mL, respectively.

Several extracts of Syzygium species have been shown to inhibit the growth of some fungal microorganisms implicated in skin diseases, such as Candida albicans included S. samarangense. However, an antifungal test of C. Albicans on this plant's stem bark had not been reported. This study aimed to identify chemical constituents of the selected fraction from dichloromethane extracts of the stem bark of S. samarangense and determine the antifungal activity of the selected fraction at various concentrations on C. Albicans. The identification of the chemical constituent of the selected fraction has been performed by the LC-ESI-MS technique. An antifungal test of the selected fraction was carried out using the disk diffusion method. The samples used included the selected fraction with 5 variations of concentration (2.5; 2.0; 1.5; 1.0; and 0.5%), positive control (ketoconazole 1%), and negative control (DMSO). The results showed that the selected fraction has antifungal activity on C. Albicans. The results showed that the selected fraction contains four flavonoids: pinocembrin, uvangoletin, stercurensin, and aurentiacin. Due to antifungal activity on C. Albicans, it has moderate activity at a concentration of 2.5%, while concentrations of 2.0, 1.5, 1.0, and 0.5% have weak activity.

 From the roots of Myxopyrum smilacifolium Blume, five known compounds, verbascoside (1), arenarioside (2), myxopyroside (3), 3-formylindole (4), and 5-hydroxymethyl furfural (5) were isolated. Their chemical structures were determined by  ESI-MS, 1D, 2D-NMR spectrascopic data analysis as well as in comparison with those reported in the literatures.
