Bulchhozia coriacea (Capparaceae) seeds are used in managing snake bite in Western Nigeria were investigated against Naja nigricollis (Spitting cobra) venom-induced hemolytic, hemorrhagic and fibrinolytic effects. This study was aimed at determining the antihaemolytic, antihaemorrhagic as well as antifibrinolytic effects of B. coriacea on N. nigricollis crude venom. Microwave-assisted extraction with hexane, chloroform, ethyl acetate, and methanol was carried out. Naja nigricollis venom-induced erythrocyte lysis (100 %) was significantly reduced to 18% by the chloroform fraction at 0.625 mg/mL. At the concentration of 0.625 mg/mL, the hexane, chloroform, ethyl acetate and methanol fractions administered in combination with the venom reduced percentage hemorrhagic activity to 23%, 17%, 49%, and 87%, respectively. In conclusion, Bulchhozia coriacea seed fractions exhibited significant antihaemolytic, antihaemorrhagic and antifibrinolytic activities against N. nigricollis crude venom and may beneficial as a pre-treatment the while victim is transferred to a healthcare facility to receive the definite treatment to ensure speedy recovery. The present study proposes microwave-assisted extraction as a sustainable technique for the biosynthesis of bioactive compounds from rice fermented with Aspergillus flavus (koji). First, fermentation conditions (i.e., pH from 3–12, five temperatures from 20–40 °C, and four culture-fermentation media viz. wheat, wheat bran, malt and rice) were optimized for producing microbial bioactive compounds. Microwave extraction was performed at 2450 MHz and 500 W for 20, 30, and 40 s with seven solvents (distilled water, ethyl acetate, hexane, ethanol, chloroform, diethyl ether, and methanol). The obtained results revealed that ethyl acetate is the most appropriate solvent for extraction. Effects of this ethyl acetate extract were compared with a commercial synthetic antioxidant. Antioxidant properties were enhanced by preventing the oxidation of the linoleic acid (C18H32O2) with an inhibition rate (antioxidant efficacy) of 73.13%. Notably, the ferrous ion binding ability was marginally lower when compared to the disodium salt of ethylenediaminetetraacetic acid (EDTA). Additionally, the obtained total content of phenolic compounds in the ethyl acetate extract of fermented rice (koji) by Aspergillus flavus was 232.11 mg based on gallic acid/mL. Antioxidant compounds in the ethyl acetate extract of fermented rice showed stability under neutral conditions, as well as at high temperatures reaching 185 °C during 2 h, but were unstable under acidic and alkaline conditions. The results demonstrate the efficacy of novel microwave-assisted extraction technique for accelerating antioxidant production during rice fermentation.

Ultrasonic-microwave assisted extraction (UMAE) of Trametes orientalis polysaccharides was optimized by response surface methodology. Hepatoprotective effects of a purified T. orientalis polysaccharide (TOP-2) were evaluated by alcohol-induced liver injury model mice. The optimal UMAE parameters were indicated as below: ratio of water to raw material 28 mL/g, microwave power 114 W, extraction time 11 min. The polysaccharides yield was 7.52 ± 0.12%, which was well consistent with the predicted value of 7.54%. Pre-treatment with TOP-2 effectively increased the liver index and spleen index in alcohol-treated mice. The elevated serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels of mice after alcohol exposure were inhibited by TOP-2 administration. The liver tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) levels have decreased significantly as a result of alcohol exposure, while pre-treatment with TOP-2 could mitigate these consequences. Furthermore, pre-treatment with TOP-2 could efficiently boost the superoxidase dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities, and observably constrain the malondialdehyde (MDA) level. The findings suggest that TOP-2 might be useful for alleviating the alcohol-induced hepatotoxicity via its antioxidant and anti-inflammatory potential.

Tannins are phenolic compounds with considerable abundance in nature. In this study, the extraction of Indian gooseberry fruit was performed using Microwave Assisted Extraction (MAE) method with ethyl acetate as solvent. Two grams of Indian gooseberry fruit were extracted for 1 minute with variable microwave power operation (100, 180, 300, 450 and 600) W. The experiment is continued at the power that gave the highest yield of tannin with  variable ratio sample to solvent (1/10, 1/20, 1/30, 1/40 and 1/50) g/mL. Total tannins in the extract were analyzed using UV-Vis Spectroscopy method to find the yield of tannin. Functional groups that were contained in the extract were analyzed using Fourier-Transform Infrared Spectroscopy (FTIR) method. The results of this study indicate that the extract contains tannins in qualitative tests with gelatin and FeCl3 as well as in FTIR analysis. The highest yield of tannin was 36.86 mg/g which was obtained under conditions of power 100 W and ratio sample with ethyl acetate 1/50 g/mL.

AbstractHigh performance liquid chromatography is the most common technique for analysing isoflavones in soy; however, the appropriate sample pre-treatment is required, including extraction and, often, hydrolysis. In the present work, the kinetics of extraction was investigated with regard to the number of extraction steps and their duration for the exhaustive extraction of bonded forms of isoflavones and aglycones. In addition, a systematic experimental study of hydrolysis depending on the temperature and acidity of the medium was conducted. The results showed that the assisted methods were more effective for the isolation of isoflavones. Moreover, the differences between the results obtained using ultrasound-assisted extraction (UAE), microwave-assisted extraction and pressurised liquid extraction were not statistically significant; however, the appropriate number of extraction steps and their duration for UAE were required. The investigation also revealed that, due to the varying susceptibility of glycosides on conversion to free forms and the stability of the aglycones obtained, two parallel hydrolysis procedures should be conducted to obtain the highest hydrolysis efficiency. The optimised conditions, taking into account the shortest time combined with the highest efficiency, were as follows: 4 M HCl and 60 min for genistein; 8 M HCl and 30 min for glycitein and daidzein.

Background: Adherence to subcutaneous biologic agents for the treatment of psoriasis can be negatively influenced by injection pain.Objective: To explore the differences in injection site pain when patients are pre-treated with heat or cold, versus no pre-treatment prior to administration of a subcutaneous biologic agent.Methods: In an observational cohort study, patients receiving subcutaneous injections of ustekinumab were randomly assigned to receive pretreatment with ice, heat, or no intervention over three visits. Post-dose, patients rated pain on a 100 mm visual analogue scale (VAS).Results: There was an increase in the VAS score for both heat (2.51, P=0.30) and ice (3.33, P=0.16), compared to no intervention. No differences were found between the two intervention groups (-0.83, P=0.73). On average, females had the same VAS scores with ice compared to that of no intervention (-0.12, P=0.97) and a non–significant decrease of 3.29 points (P=0.38) with heat. Males had increased pain scores by 5.65 points (P=0.07) with ice and by 6.39 points (P=0.04) with heat.Limitations: Pain is a subjective measurement and objective quantification is difficult.Conclusions: On average, neither heat nor cold application reliably reduced pain. Our results do not support the application of heat or cold prior to ustekinumab injection.
