Abstract Introduction: AAzathioprine and Methotrexate are both used in the treatment of cancer; and are classified as cytotoxic drugs with reported adverse effects such as oxidative damage to the DNA/RNA, the testes and sperm cells. This study, therefore, tested the hypothesis that AAzathioprine and Methotrexate administrations impair the morphology and functions of the testes in adult male wistar rats. Methods: AAzathioprine (50-150mg per day) and Methotrexate (2.5mg per week) are used in the treatment of cancer in adult Man. We tested the hypothesis that AAzathioprine and Methotrexate impair the morphology and functions of testes in rats. Forty adult male wistar rats (150-230g) were employed in the study: Control Group I received physiological saline while Experimental Groups II - V received oral administrations of 5mg/kg/bodyweight of AAzathioprine per day, 15mg/kg/bodyweight of AAzathioprine per day, 8mg/kg/bodyweight of Methotrexate per week and 20mg/kg/bodyweight of Methotrexate per week respectively for 35 days. Results: Histological examinations of the testes of rats of Groups II - V showed dose-dependent morphological anomalies such as fewer collagen ibers of connective tissues, disrupted seminiferous tubules and scanty spermatozoa when compared to rats of Group I. Statistical analyses showed dose-dependent elevated levels (P≤0.05) of superoxide dismutase and malondialdehyde in testes homogenates of rats of Groups II - V when compared to rats of Group I. This implied increased oxidative stress in rats of Groups II - V. Evaluations of Follicle Stimulating Hormone and Testosterone showed dose-dependent significantly elevated levels (P≤0.05) in rats of Groups II - V when compared to rats of Group I. Conclusions: Our findings are consistent with the stated hypothesis.

Amlodipine is a long-acting calcium channel blocker used in the treatment of hypertension and angina. In adult man, the treatment regimen is 5 or 10 mg daily. This study evaluated the effects of prolonged oral administration of Amlodipine Besylate on the morphology of bone marrow in adult male Wistar rats. Sixteen rats (140 - 190 g) comprising of four groups were employed in the study. Rats of Control Group I received physiological saline orally while rats of Experimental Groups II - IV received oral administrations of 5, 10 and 15 mg/kg bodyweight of Amlodipine Besylate respectively for nine weeks. Histo-pathological examinations of the bone marrow showed normal cytoarchitecture of erythrocytes and leukocytes in rats of Control Group I. However, dose-dependent degeneration and lyses of erythrocytes and leukocytes were observed in amlodipine-treated rats. Cottonseed oil is an extract from the seeds of cotton plant, that is rich in protein, carbohydrate and fiber.          The objective of the research was to study the effect of cottonseed oil on the histology of the testis.          Materials and methods. Twelve adult male Wistar rats weighing 200 – 250g, were randomly sorted into four groups (I - IV), each containing three animals. Cottonseed oil was administered orally to the experimental animals by means of an orogastric tube. The rats in Group I (the control group) received standard rat pellet and oral normal saline ad libitum only. The animals in Groups II, III, and IV received 0.2 ml, 0.6 ml and 1.0 ml of cotton seed oil per body weight respectively. On the 21st day, the animals were sacrificed by cervical dislocation. The testes were excised and, then, fixed with appropriate fixatives for routine tissue processing.          Results. The results revealed an increase in the luminal diameter of seminiferous tubules, a decrease in the Leydig cell population, the disorientation of spermatogenic series and their detachment from the germinal epithelium.          Conclusions. The present study demonstrated that oral administration of cottonseed oil to male rats displayed a deleterious effect on their testis by disrupting spermatogenesis and the histological architecture of the testis.

To investigate the effect of Lippia multiflora (Verbanaceae) on the reproductive activity of male Wistar rats, three groups (A,B and C) of rats were treated with 400, 800 and 1600 mg/kg/day of Lippia multiflora leaves extract respectively for 28 days while 5ml/kg of distilled water was given to the control group D. At the end of the experimental period, animals were sacrificed and sperm characteristics, histology of the testes and epididymis were assessed. Lippia multiflora leaf extract caused a significant reduction (P<0.05) in sperm motility, a statistically significant dose dependent reduction in the sperm count and a significant (P<0.05) dose dependent increase in morphological abnormalities of the spermatozoa of the treated rats. Histopathological evaluation of the testis and epididymis revealed varying degrees of degeneration and necrosis of the germinal epithelia cell of the seminiferous tubules, spermiostasis and complete epididymal aspermia. Lippia multiflora leaf extract has significant anti-spermatogenic effects on adult male Wistar rats which could impair reproductive activities in these rats

The present investigation was carried out to evaluate the hepatoprotective effect of probiotic fermented milk (FM) containing Lactobacillus rhamnosus GG and Lactobacillus casei strain Shirota, alone as well as in combination with chlorophyllin (CHL) as an antioxidant agent in male Wistar rats administered aflatoxin-B1 (AFB1). AFB1 was injected intraperitoneally at the rate of 450 μg/kg body weight per animal twice a week for 6 weeks, maintaining an equal time interval between the two consecutive AFB1 administrations. A total of 125 male Wistar rats were randomly allocated to five groups, each group having twenty-five animals. Group I was offered FM containing L. rhamnosus GG and L. casei strain Shirota. Group II was administered AFB1 and served as the control group; group III was administered FM-AFB1, in which besides administering AFB1, FM was also offered. Group IV was offered CHL and AFB1, and group V was offered both FM and CHL along with AFB1. The rats were euthanised at the 15th and 25th week of the experiment and examined for the biochemical and hepatopathological profile. A significant reduction in thiobarbituric acid-reactive substances (TBARS) was observed in the FM–CHL–AFB1 group compared with the AFB1 control group. FM alone or in combination with CHL was found to show a significant (P < 0·05) hepatoprotective effect by lowering the levels of TBARS and by enhancing the activities of antioxidant enzymes such as glutathione peroxidase, superoxide dismutase, catalase and glutathione-S-transferase, indicating that probiotic FM alone or in combination with CHL possesses a potent protective effect against AFB1-induced hepatic damage.

This study evaluated the ameliorative and prophylactic effects of 2 different doses of polyphenolic-rich fraction of Garcinia kola (PPRF Gk) seeds on the histology and hormones of pituitary–testicular axis of male Wistar rats. Thirty-five male Wistar rats (150-200 g) were divided into 7 groups of 5 rats each. Groups I and II were given distilled water (0.5 mL/day) for 8 days followed by propylene glycol (0.2 mL/d) and 600 mg/kg of PPRF Gk, respectively, for 21 days. Group III received sodium arsenate (8 days), left untreated for 21 days. Groups IV and V received sodium arsenate (20 mg/kg) for 8 days followed by PPRF Gk (300 and 600 mg/kg, respectively) for 21 days. Groups VI and VII received PPRF Gk (300 and 600 mg/kg, respectively) for 21 days followed by sodium arsenate (20 mg/kg) for 8 days. Rats were killed by cervical dislocation 24 hours after the last dose and their blood collected through cardiac puncture. Blood sera were assayed for the levels of follicle-stimulating hormone (FSH), luteinizing hormone (LH), and testosterone using immunoassay techniques. Histology of the pituitary gland and testes was carried out. A significant reduction was observed in the concentration of FSH in groups IV, V, VI, and VII in comparison with groups I and II. The concentrations of both LH and testosterone showed significant decreases in groups IV, V, VI, and VII in comparison with group I. Group III presented with the lowest serum hormonal concentrations. Photomicrographs of the pituitary gland revealed greatly reduced basophils in group III and mildly reduced basophils in groups IV, VI, and VII in comparison with groups I and II. Group V revealed hypercellularized and distorted basophils. Photomicrographs of the testes showed detachment of the seminiferous tubules from the basement membrane and disruption of the interstitial space which was worse in group III, moderate in groups V and VI, and mild in group VII. In conclusion, PPRF Gk effected a dose-dependent reversal and prevention of the perturbations caused by arsenate in rats.

Summary The effect of aqueous matured stem extract of Opuntia dillenii on selected biochemical parameters in Male Wistar rats was explored. Standard analytical methods were applied. Forty Wistar rats (80-100g) were used in the animal studies, separated into four groups. The control group was solely administered normal feed and saline, group I was administered 100mgkg−1 of the extract, group II received 300mgkg−1 of the extract and group III received 500 mg/kg−1 of the extract. A significant increase (p<0.05) in the activities of alanine aminotransferase (ALT) and alkaline phosphatase was observed in group II and III rats, as compared with the controls. A significant decrease in urea and creatinine concentrations was found only in group III rats against the controls. Also, a significant (p<0.05) decrease in triglyceride, total cholesterol, and low density lipoprotein (LDL)-cholesterol was seen in group II and group III rats when compared with the control. The hematological evaluation revealed a significant (p<0.05) decrease in red blood cell and hemoglobin levels in group III rats when compared with the control. The findings showed both beneficial and toxicological effects of the plant. Hence, for optimal therapeutic benefits, a further toxicological survey could still be carried out perhaps at higher doses.

Background: Hyperlipidemia is a well known risk factor for cardiovascular disease, especially atherosclerotic coronary artery disease. Peroxisome proliferator activated receptor α (PPARα), a member of this nuclear receptor family, has emerged as an important player in this scenario, with evidence supporting a central co-ordinated role in the regulation of fatty acid oxidation, lipid and lipoprotein metabolism and inflammatory and vascular responses, all of which would be predicted to reduce atherosclerotic risk. The low-density lipoprotein (LDL) receptor (LDLR) is the primary pathway for removal of cholesterol from the circulation, and its activity is meticulously governed by intracellular cholesterol levels. Hence in this study we investigated the effect of Lutein on PPARα and LDLR expression in liver of wistar rats.Methods: Male Wistar rats were divided into 6 groups of 6 each. Group I served as control. Group II III, IV, V and VI rats were received high cholesterol diet. Group III was treated with Atorvastatin 5mg/kg. Group IV, V and VI rats were treated with 25mg/kg, 50mg/kg and 100mg/kg of Lutein. After 16 weeks, liver tissue samples were collected from all the groups of animals to evaluate the expression of PPARα and LDLR.Results: The expression of Peroxisome proliferator activated receptor α and low-density lipoprotein (LDL) receptor (LDLR) was significantly increased in Lutein treated hypercholesterolemic male wistar rats.Conclusions: The results of this study indicate that Lutein activates LDL receptor and PPARα in hypercholesterolemic male wistar rats.

Despite its nutritional content and huge application in the food, pharmaceutical and cosmetic industries, the use of anthocyanin remains restricted due to the difficulty in its extraction process. In this study, we examined the antioxidant effects of anthocyanin (one of the numerous flavonoids known in modern enthno-medicinal practice) on some serum free radicals in wistar rats. Twenty (20) adult wistar rats of between 100 – 250 g were procured and housed in the animal house of the Ambrose Alli University, Ekpoma, Edo State. After two (2) weeks of acclimatization, the animals were then grouped into five groups of four (4) rats each (n=4). Control (Group I) rats were fed normal rat chow and water ad libitum, whereas, group II rats received 100 mg/kg body weight (bw.) of anthocyanin extract. Groups III, IV and V rats also received 200 mg/kg bw., 300 mg/kg bw., and 400 mg/kg bw of anthocyanin extract respectively for a period of four weeks, using the oro-gastric cannula post prandial. For each week, body weights were obtained, and serum assayed for changes in the levels of anti-oxidant enzyme activities [Catalase (CAT), Malondialdehyde (MDA), Glutathione Peroxidase (GPx) and superoxide Dismutase (SOD)] using appropriate biochemical procedures. Obtained results were subjected to statistical comparisons using the one way analysis of variance (ANOVA). Tukey post hoc tests were also performed (where necessary) to ascertain the source(s) of mean differences between groups. We found a statistically significant decrease (at p < 0.05) by week in serum CAT, MDA, GPx and SOD levels with the administration of test substance. This also proved to be dose-dependent as increasing dose cause an increase in the enzyme activities over the period of administration. Body weight changes were also duration and dose dependent. Anthocyanin therefore showed potency in improving serum antioxidant enzyme activities. We recommend similar studies on other body tissues than blood.
