The tetrazolium test can be an alternative to obtain fast results of the physiological potential of wheat seeds. In this context, the objective of the present study was to evaluate the efficiency of the tetrazolium test through the evaluation by analysis of digitized images in the determination of the physiological quality of wheat seeds.The experiment was carried out with 22 lots of wheat seeds, submitted to the determination of the physiological potential, including the tetrazolium test evaluated by means of digitized image analysis.For digitization, after longitudinal bisection, and staining in 0.075% tetrazolium solution, the seeds were grouped on tabletop scanner glass, stored and classified into four vigor classes. Data were submitted to analysis of variance, with means grouped by the Scott-Knott test, at 5% of significance; we also performed an analysis of the simple correlation coefficients between tetrazolium test results and other tests.The evaluation of the tetrazolium test through the analysis of digitized images grouped the lots in three levels of vigor.The method is effective in determining the vigor and viability of wheat seeds, because the possibility of expanding the images that allows precise analysis of the embryonic axis structures.

The aim of this work was to correlate macronutrient content of soybean seeds to physiological quality of different cultivars. The work was developed in the Federal University of Pelotas, in the facilities of the Seed Science and Technology Graduate Program. The experimental design was randomized blocks in arranged in four replicates. The following soybean cultivars were used: BMX Apolo RR (12 lots), BMX Ativa RR (13 lots), BMX Energia RR (26 lots), BMX Força RR (24 lots), BMX Impacto RR (35 lots), BMX Magna RR (16 lots), BMX Turbo RR (44 lots), BMX Potência RR (82 lots) and NA 5909 RR (28 lots), with seeds produced in northwest Rio Grande do Sul. The determination of nutritional contents found in plant tissues of soybean seeds, were measured: Nitrogen (N), Phosphorus (P), Potassium (K), Magnesium (Mg), Calcium (Ca), Sulfur (S). The macronutrient contents of the seeds vary according to genetic characteristics of the cultivars, with higher oscillations of nitrogen, phosphorus, magnesium and sulfur contents present in soybean seeds. Potassium and calcium are defined as the most stable nutrients for the cultivars and seed lots analyzed. High concentrations of nitrogen, phosphorus and calcium are determinants for the physiological quality of soybean seeds.

ABSTRACT: The aim of this study was to evaluate the effect of the sowing periods in production, productivity component and physiological quality of seeds of three soybean cultivars (TMG133RR, P98Y70RR and NS7670RR) in Orthic Quartzarenic Neosoil in the cerrado region. The design used was a randomized block a factorial scheme design with four repetitions, and each plot with useful space consisted of three rows of four meters length, spaced at 0.45 meters. Field evaluations were the final stand, the height of the plants, height of the first pod insertion, the mass of 100 seeds and productivity. Already in the lab seeds were evaluated for germination and vigor (first germination count, emergency, emergence speed index, length and dry mass of the aerial part of the plant and roots, electrical conductivity, accelerated aging and tetrazolium test). It concludes that it is possible to use Orthic Quartzarenic Neosoils, located in the Cerrado region at altitudes higher than 600 m, in years and places with good water distribution in the spring-summer seasons, for grain and soybeans production, but as a first goal, the crop should be sown in the first 20 days of November otherwise it should take place in early December.

ABSTRACT: Crambe is a rapeseed with high oil content and can be used as a winter cover or as a source of raw material for the production of biodiesel, however espite the growing interest in the culture, research on the subject is still incipient, especially concerning the seed production and analysis technology. The purpose of this study is to evaluate the physiological quality of crambe seeds, 'FMS Brilhante' cultivar, by testing the pH of exudate. Five seed lots were submitted to the determination of water content and the tests of germination and vigor (first count, emergence and tetrazolium). In the conduction of pH exudate test, temperatures (25 and 30oC), and periods of seed imbibition in water (15, 30 and 45 minutes) were tested. The experiment was conducted in a completely randomized manner, with four replicates, and the mean values were compared by the Tukey test at 5% probability; Pearson correlation between the pH of the exudate and initial tests was also made. Testing the pH of exudate is promising for separating lots of crambe seeds and the following combinations of 25°C/30 minutes or 30°C/45 minutes can be used.

Abstract: Initial seed vigor and sowing density interact in establishment of plants in the field, and can thus affect expression of the plasticity of the wheat crop and the yield and physiological quality of the seeds produced. The aim of this study was to evaluate the effect of seed vigor levels in combination with sowing densities on the yield and physiological potential of wheat seeds. The cultivars BRS Gralha-Azul and BRS Sabiá were used in a randomized block experimental design in a 2 × 4 factorial arrangement, with four replications. The treatments consisted of seeds of high and low vigor and four sowing densities (150, 250, 350, and 450 seeds.m-2). The following evaluations were made: seed yield, germination, first germination count, seedling length, seedling dry matter, accelerated aging, emergence speed index, and seedling emergence in sand. An increase in sowing density favors the seed yield of both cultivars; however, it reduces the vigor of the seeds produced by the cultivar BRS Gralha-Azul, especially of the seeds produced by plants originating from high vigor seeds. For the cultivar BRS Sabiá, an increase in sowing density decreases the germination performance of seeds produced by plants originating from low vigor seeds.

ABSTRACT: The objectives of the present project were to evaluate the sanity and germination of tobacco seed samples cultivated in Brazil and to identify potential pathogenic fungi to the culture. Thirty-four representatives samples of tobacco seeds were evaluated through germination and sanity test by the blotter-test. On the germination test, results were expressed in percentage of seedlings considered normal, abnormal and non-germinated seeds. The percentage of seeds germination varied between 54.5 and 90%. According to sanity test, it was verified the incidence of Alternaria alternata, Cladosporium spp., Fusarium spp., and Phoma spp. The incidence of A. alternata varied from 3 to 67%, and Fusarium spp. varied from 0 to 19%. There was significative positive correlation between the incidence of A. alternata and the percentage of seed germination. Species of Alternaria and Fusarium can be potential pathogens to tobacco culture and an alarm for the necessity of seed treatment of the seeds that are transporting these fungi.

Abstract: The germination test for recently harvested wheat seeds requires a period of up to 20 days, because of the presence of dormancy. The possibility of reducing this period provides greater speed to the allocation of seeds, as well as an increased operational capacity of analytical laboratories. This study aimed at evaluating the effectiveness of treatments to stimulate the germination of recently harvested wheat seeds, reducing the five-day pre-cheeling period recommended to run the test. Seeds from various wheat genotypes were used: BRS Guamirim, BRS Tangará, BRS Pardela, CD 108, CD 116, CD 150, IPR Catuara TM, Frontana, IPR Taquari, Safira and Quartzo. They were analyzed immediately after collection and then stored for 30 and 180 days. In each evaluation period (immediately after harvesting, 30 and 180 days of storage), the determination of water content, the germination test (seeds without pre-chilling and pre-chilling between 5 and 10 ° C for three to five days), and also the tetrazolium test to quantify viability and seed dormancy were carried out. Based on these results, it is concluded that there is effectiveness in overcoming wheat seed dormancy, adopting the pre-cheeling procedure between 5 and 10 ° C for three days.

Abstract: Some desiccation techniques for harvesting wheat may affect seed quality. The objective of this study was to evaluate the effect of different desiccant herbicides applied to the wheat crop and their effects on the production and seed quality. The experiment was conducted with cultivar COODETEC 150 (CD 150) in two sites in Palotina (Paraná state, Brazil) with a randomized block design consisting of 7 treatments (desiccation herbicides) and 4 replications. Desiccation was realized when 50% of the plants were in stage 80 with seeds in powdery or mass consistency. The variables analyzed were percentage of moisture content after harvest, Yield, hectoliter weight, mass of 100 seeds, vigor, germination, fresh mass of seedlings, dry mass of seedlings and root length. The herbicides carfentrazone-ethyl and clethodim were the ones that promoted greatest reduction in vigor. The herbicide paraquat caused reduction in seedling length. There was a reduction in the mass of 100 seeds for glufosinate-ammonium, clethodim, diquat and carfentrazone-ethyl. Glufosinate-ammonium, paraquat, glyphosate, clethodim and diquat caused reduction in yield. Desiccation of Cultivar CD 150 at stage 80 is not recommended, since plant Yield and physiological seed parameters were adversely affected by herbicides.

Germination test for papaya seeds demands thirty days to obtain the results, which is a long period for decision-making. Thus, rapid tests become essential tools to evaluate seed viability, and the tetrazolium test is an alternative for this purpose. Hence, this study aimed to establish the adequate procedure to apply this test, in order to reduce the time of evaluation of papaya seeds. For that, four tetrazolium solution concentrations (0.05, 0.1, 0.5 and 1%) and three staining times (3, 6 and 9 hours) were tested at temperatures of 35 and 40 ºC. The experimental design was completely randomized in 4 × 3 + 1 factorial scheme (concentrations × staining times + one control = germination test). It was found that the tetrazolium test allows to evaluate the physiological quality of papaya seeds, thus reducing the time for decision-making. For the test, we recommend using the concentrations of 0.1% for 9 hours or 1% for 6 hours of staining, at temperature of 40 ºC.

Eugenia brasiliensis Lam. ("grumixameira"), Eugenia uniflora L. ("pitangueira") and Eugenia pyriformis Cambess. ("uvaieira") are forest and fruit species of pharmacological and gastronomic potential and have desiccation-sensitive seeds. The development of quick viability tests for the assessment of physiological quality of these seeds is needed. The tetrazolium test is an important method providing fast assessment of the seed physiological quality. Thus, this work aimed to develop a method for the tetrazolium test for determining viability of "grumixameira," "pitangueira" and "uvaieira" seeds. Initially the seeds of these species were soaked in water for 3 h at 25 ºC. Three concentrations of tetrazolium solutions were analyzed (0.100 %, 0.125 % and 0.250 %) for seed staining using three incubation periods (2, 3 and 6 h) at 35 ºC in the dark. After staining, seed viability was determined. Then, the seeds were subjected to different levels of controlled drying and were analyzed for their physiological quality by germination and electrical conductivity tests. The tetrazolium test is a suitable tool for determining viability after staining the seeds at 35 ºC using a 0.250 % concentration solution for 3 hours for "grumixameira" seeds, 0.125 % for 3 hours for "pitangueira" seeds and 0.100 % for 2 hours for "uvaieira" seeds.
