Abstract When produced biologically, especially by photosynthetic organisms, hydrogen gas (H2) is arguably the cleanest fuel available. An important limitation to the discovery or synthesis of better H2-producing enzymes is the absence of methods for the high-throughput screening of H2 production in biological systems. Here, we re-engineered the natural H2 sensing system of Rhodobacter capsulatus to direct the emission of LacZ-dependent fluorescence in response to nitrogenase-produced H2. A lacZ gene was placed under the control of the hupA H2-inducible promoter in a strain lacking the uptake hydrogenase and the nifH nitrogenase gene. This system was then used in combination with fluorescence-activated cell sorting flow cytometry to screen large libraries of nitrogenase Fe protein variants generated by random mutagenesis. Exact correlation between fluorescence emission and H2 production levels was found for all automatically selected strains. One of the selected H2-overproducing Fe protein variants lacked 40% of the wild-type amino acid sequence, a surprising finding for a protein that is highly conserved in nature. We propose that this method has great potential to improve microbial H2 production by allowing powerful approaches such as the directed evolution of nitrogenases and hydrogenases.

UV radiation is a factor affecting the distribution and physiology of photosynthetic organisms in an aquatic ecosystem. Studies with macroalgae indicate diverse biological disturbances in response to UV radiation. This work aimed to study sensitivity of the brown macroalga Sargassum filipendula exposed to UV radiation: PAR (control), PAR+UVA+UVB(++) and PAR+UVA(++)+UVB. Changes in the physiological parameters growth rate, total soluble proteins, photosynthetic pigments and the UV-vis absorbing compounds were analysed after T0, T4, T7 and T10 (days) of UV exposure. Physiological parameters showed little variation between treatments and over time, suggesting that moderate UV radiation doses could regulate resistance responses to re-establish the cellular homoeostasis condition through activation of an antioxidant defence system, such as an overproduction of phenolic compounds. Responses recorded in S. filipendula would be related to acclimation mechanisms against acute UV radiation stress, triggering resistance responses to avoid serious damage to the metabolic machinery, activating control systems to maintain hormesis, and homoeostasis of deleterious actions of reactive species, similar to the phenomenon known as preparation for oxidative stress. Finally, UV-visible absorption spectra showed absorption bands evidencing the presence of mainly UV-absorbing compounds with photoprotective function, such as phlorotannins, flavonoids and carotenoids, which could provide adaptive advantages for organisms exposed to UV radiation.

Abstract. In psychology, thinking is typically studied in terms of a range of behavioral or physiological parameters, focusing, for instance, on the mental contents or the neuronal correlates of the thinking process proper. In the current article, by contrast, we seek to complement this approach with an exploration into the experiential or inner dimensions of thinking. These are subtle and elusive and hence easily escape a mode of inquiry that focuses on externally measurable outcomes. We illustrate how a sufficiently trained introspective approach can become a radar for facets of thinking that have found hardly any recognition in the literature so far. We consider this an important complement to third-person research because these introspective observations not only allow for new insights into the nature of thinking proper but also cast other psychological phenomena in a new light, for instance, attention and the self. We outline and discuss our findings and also present a roadmap for the reader interested in studying these phenomena in detail.
