Effects of Methanol Extract of Telfairia occidentalis Seed on Serum Lipid Profile, Biochemical and Antioxidant Activity in Female Wistar Rats

Objective: The study was aimed at evaluating the folkloric use of the Catunaregam spinosa (Thunb) bark for its hypoglycaemic and antioxidant activity against dexamethasone-induced diabetes in wistar rats. Methods: Diabetes was induced in wistar rats by dexamethasone administration 10 mg/kg, b. wt., s. c for 11 consecutive days to all group animals (except Group I). After confirmation of diabetes, the animals were divided into 5 groups (n=6). Group I: Vehicle control treated with normal saline only, Group II: Diabetic control treated with dexamethasone only, Group III: Standard control treated with dexamethasone plus glibenclamide (5 mg/kg, b. wt., p. o), Group IV and V: Test control received dexamethasone plus ethanolic extract of Catunaregam spinosa (EECS) at graded doses of 200 mg/kg, b. wt. and 400 mg/kg, b. wt., p. o for 21days respectively. On the last day of the experiment, the effect of Catunaregam spinosa bark was measured by estimating the biochemical and antioxidant parameters. Results: There was a significant alteration in the serum glucose, HbA1c, total proteins, serum lipid profile and tissue antioxidant parameters in dexamethasone-treated animals when compared to the normal control rats. In test groups treated with glibenclamide and EECS, it was significantly ameliorated the altered parameters such as serum glucose, HbA1c, total proteins, serum lipid profile and the antioxidant parameters like SOD, CAT, GSH and MDA when compared to dexamethasone-treated animals with evidence of histopathological studies of the pancreas. Conclusion: The results indicated that the ethanolic extract of Catunaregam Spinosa (Thunb) bark has shown the hypoglycemic and antioxidant property.

Objective: This study investigated the effects of Vernonia amygdalina supplementation on Vigna subterrenea (bambara groundnut) pudding on serum lipid profile and electrolyte levels in Wistar rats. Materials and Methods: Twenty five weanling rats were randomly selected into 5 groups of 5 rats each. Group 1 received normal rat pellet. Groups 2 and 3 received 10% and 5% (w/w) non supplemented bambara pudding, respectively. Groups 4 and 5 were fed with 10% and 5% (w/w) Vernonia supplemented bambara pudding, respectively, for 21 days. The animals were sacrificed using anaesthesia at the end of the experiment and blood sample collected for serum lipid profile and serum electrolyte determinations using standard methods. Results: There was a significant increase (p<0.05) in total cholesterol level in groups 2 (1.64 ± 0.01 mmol/L) and 4 (1.66 ± 0.01 mmol/L) compared to normal control (1.31 ± 0.02 mmol/L). Triacylglycerol level increased significantly in groups 2, 3, and 4 compared to normal control. The high density lipoprotein cholesterol (HDL-c) and Serum sodium level were significantly increased (p < 0.05) in group 2 and 4 respectively compared to the normal control. Potassium, chloride, bicarbonate and calcium levels showed significant increases in group 2 relative to normal control. Phosphate level decreased significantly in group 2 (1.37 ± 0.03 mmol/L), 3 (1.43 ± 0.03 mmol/L) and 5 (1.37 ± 0.03 mmol/L) when compared to normal control (1.75 ± 0.03 mmol/L) respectively. Conclusion: Consumption of bambara groundnut pudding supplemented with Vernonia leaves increases HDL-c level as well as supply appreciable amounts of electrolytes in the body.

Abstract This study evaluated the hypolipidemic, hepatoprotective, nephroprotective and anti-lipid peroxidation properties of a methanol extract of Paullinia pinnata root-bark, in alloxan-induced hyperglycemic rats. The extract of P. pinnata root-bark was prepared using a cold maceration method with 80% methanol and concentrated at 40°C in hot air oven. The extract was administered once daily per os at 50, 100 and 200 mg/kg for 21 consecutive days. Distilled water (5 mL/kg) and glibenclamide (2 mg/kg) were used as the vehicle and reference standard, respectively. The serum lipid profile, markers of liver and kidney functions, antioxidant status (malondialdehyde level, superoxide dismutase and catalase activities), histopathological changes in liver and kidney were examined 24h after the last treatment on day 21. The extract reduced serum lipid profile, markers of liver and kidney functions of treated rats relative to vehicle-treated rats. The superoxide dismutase and catalase activities of the extract treated rats were also elevated relative to the vehicle-treated rats. The extract reversed liver and kidney injuries induced by alloxan in the treated rats. This study provides some basic information which suggest that P. pinnata could be effective in managing diabetic complications.
