Evidence indicates that Allium and Brassica species which release bioactive compounds are widely used in bio-fumigation to suppress soil-borne diseases. However, the active molecules of such plant residues are easily volatilized. In this study, we conducted mixed cropping of the apple tree with Allium fistulosum or Brassica juncea; the results demonstrated that such mixed cropping significantly improved the growth of the grafted apple seedlings and alleviated apple replant disease (ARD) for two years. The terminal-restriction fragment length polymorphism profile results showed that the soil fungal community demonstrated distinct variation and diversity in terms of composition. A. fistulosum and B. juncea significantly improved the Margalef, Pielou, and Shannon indices. In addition, the analyses of clone libraries showed that A. fistulosum and B. juncea promoted the proliferation of antagonistic fungi such as Mortierella, Trichoderma, and Penicillium, and inhibited the proliferation of pathogens such as Fusarium. Fusarium. Proliferatum (F. proliferatum) was abundant in replanted soil and proved to be an aggressive pathogen of apple seedlings. Our findings thus indicate that apple tree mixed cropping with A. fistulosum and B. juncea was an effective long-term method for modifying the resident fungal community and alleviating ARD.

In September 2018, severe symptoms and high incidence (about 60%) of an onion anthracnose disease attributable to infection by Colletotrichum spp. was observed in production fields of Dingxi city, Gansu Province, China. The mature onion plants near to harvest (Allium fistulosum L. var. giganteum Makino) expressing necrotic symptoms had oval lesions with dark spots that were made up of stromatic masses that had formed beneath the cuticle of the plant base. Twenty symptomatic plants were sampled. Symptomatic tissues were surface sterilized with sodium hypochlorite, transferred aseptically to 20 mL potato dextrose agar in a petri plate and incubated at 30 ± 2 °C. After seven days, cultures produced acervuli and setae, a characteristic sign of Colletotrichum spp. Acervuli were observed with a black, bulbous base and acicular setae while conidia were elliptical (14-25 x 3-6 µm), unicellular hyaline and slightly falcate (Figure 1). DNA was extracted from a 7-day old culture and the ITS region was amplified using primers D1 (5’-GCATATCAATAAGCGGAGGAAAAG-3’) and D2 (5’-GGTCCGTGTTTCAAGACGG-3’) (Kurtzman and Robnett, 1997). The resulting sequence (548 bp) was deposited with NCBI GeneBank under accession number MW127281. The fungus was confirmed as Colletotrichum circinans after conducting a BLAST search with the ITS sequence that reported a highest homology (99% similarity) with MH81329.1 (C. circinans). The speciation was further confirmed by amplifying regions of the TUB2, GAPDH, and ACT genes using primers given in Table S1 and sequences were also submitted to NCBI GeneBank under accession numbers MZ456033, MZ456032 and MZ456031, respectively. The subsequent BLAST results for these three additional gene regions were consistent with the results of the ITS region and fungus was identified as C. circinans. The isolated pathogen was tested for its pathogenicity on onion plants (Allium fistulosum L. var. giganteum Makino). A conidial suspension 30 ml (5 × 105 conidia/ml) was mixed with 1 kg sterilized potted soil in 15 cm diameter plastic pots. Un-inoculated, sterilized soil was used as control. Three green onion plants per pot were planted (Figure S2). The experiment was repeated three times with 15 replications in each experiment. Plants were maintained for 120 days under greenhouse conditions and were monitored for the development of anthracnose symptoms. Symptoms recorded previously on onion plants in field (i.e. necrosis, sunken oval lesions and dark spots) were observed after 30 days on plants grown in inoculated soil while control plants remained asymptomatic (Figure S1). Three samples from symptomatic tissues of each plant were used for re-isolation of the pathogen on PDA, as described above. Cultures grown on PDA were confirmed both on a morphological and molecular basis as Colletotrichum circinans. These morphological, molecular, and pathogenicity tests of the isolated fungus confirm that the anthracnose symptoms observed on onion in Gansu Province, Beijing, China was caused by Colletotrichum circinans. Six different Colletotrichum spp. have been reported to cause diseases on onion worldwide (Rodriguez et al. 2012). C. circinans, which causes smudge, is an occasional onion pathogen was previously recorded as C. dematium (Pers.) Grove f. circinans Arx, which is specific to Allium spp. (von Arx 1970; von Arx 1981). However, Sutton (1992) described C. circinans as a distinct species from C. dematium. The fungus causes smudge or leaf spot of Allium spp. (Farr et al. 1989) and has been reported from Korea, Japan, Argentina, India, the UK and most other regions of the world (Cho & Shin 2004; Kiehr et al. 2012). Smudge may be a disease of concern post-harvest as fungal growth compromises the onion scales and bulb (Walker 1921). In China, this is the first report of C. circinans causing anthracnose in Allium fistulosum L. var. giganteum Makino in Gansu Province.

Fruit waste can be a good source of liquid organic fertilizer (LOF) because it contains macro and micronutrients needed by plants. This study aims to determine the type and concentration of LOF for the growth and yield of leek (Allium fistulosum, L.). This research was conducted from June to August 2019, in Pematang Gubernur, Muara Bangkahulu, Bengkulu City, Indonesia. Treatment consist of types of Liquid Organic Fertilizer (LOF) which were cow manure (CM) and guava waste (GW) at concentrations of 25%, 50%, and 75%. Experimental treatment included; without LOF, CM LOF (25%), CM LOF (50%), CM LOF (75%), GW LOF (25%), GW LOF (50%), GW LOF (75%), CM LOF + GW LOF (25%), CM LOF + GW LOF (50%) and CM LOF + GW LOF (75%). The study used a completely randomized design (CRD), one factor, repeated five times. Differences in LOF concentration and source significantly affected the number of leaves and tiller diameter. Nonetheless, it did not affect the number of tillers, plant height, and fresh weight. Cow manure liquid organic fertilizer at a 50 % concentration raised the number of leaves by 35 %, whereas guava waste increased the leaves numbers by 25%. With the application of guava waste liquid organic fertilizer at a concentration of 75%, so the number of tillers increased by 23%.

Pesticide dissipation on foods like vegetables, fruits, and cereals is one of the most concerning aspects in pesticide toxicology as it involves risks to human health and food safety. In light if this, a laboratory-scale study was conducted for two separate systems, green onions (Allium fistulosum) and mustard greens (Brassica juncea),in order to identify the dissipation of fipronil and cypermethrin, two commonly used insecticides, under a tropical climate. After the pesticide application, vegetable samples from these microcosms were collected each day for 7 days to measure fipronil and cypermethrin (n=3) residues. The analytical method was validated and showed repeatability and trueness. The decay equations fit well to first-order kinetics with a correlation coefficient of R2>0.93 and p<0.0005. The calculated half-life values of fipronil were 2.9 d for green onions and 3.2 d for mustard greens while those of cypermethrin were, respectively, 4.5 d and 3.2 d. To meet the maximum residue levels (MRL) of fipronil (0.02 mg/kg) and cypermethrin (0.7 mg/kg) on vegetables, the estimated pre-harvest intervals should be updated to 23 d and 7.5 d, respectively.

Allium fistulosum is a perennial plant species grown worldwide belonging to the family Liliaceae. In Korean medicine, it is referred to as Chongbaek (CB), and it is prescribed for symptoms associated with the common cold due to its antipyretic properties. This study examined the effects of aqueous (CBW) and 30% ethanol (CBE) extracts on bone growth using a calcium- and vitamin D-deficient animal model. In an in vitro experiment, the alkaline phosphate activities of the extracts were examined using MC3T3-E1 and MG63 cells, and both the aqueous and ethanolic extracts had significant alkaline phosphate activities. In vivo, a serum analysis indicated that the CB extracts promoted bone growth based on the osteogenic markers ALP, calcium, osteocalcin, and collagen type 1 and increased the bone mineral content (BMC), bone mineral density (BMD), and growth plate length. Overall, our results indicate that both CBW and CBE of A. fistulosum can be utilized to facilitate bone growth and increase BMD in children and adolescents by lengthening the growth plate without adverse side effects, such as metabolic disorders or the release of obesity-inducing hormones.
