Abstract The productivity of horticultural crops in an artificial light condition are highly influenced by the structure of plant and the area coverage. Accurate measurement of leaf area is very important for predicting plant water demand and optimal growth. In this paper, we proposed an image processing algorithm to estimate the ice-plant leaf area from the RGB images under the artificial light condition. The images were taken using a digital camera and the RGB images were transformed to grayscale images. A binary masking was applied from a grayscale image by classifying each pixel, belonging to the region of interest from the background. Then the masked images were segmented and the leaf region was filled using region filling technique. Finally, the leaf area was calculated from the number of pixel and using known object area. The experiment was carried out in three different light conditions with same plant variety (Ice-plant, Mesembryanthemum crystallinum). The results showed that the correlation between the actual and measured leaf area was found over 0.97 (R2:0.973) by our proposed method. Different light condition also showed significant impact on plant growth. Our results inspired further research and development of algorithms for the specific applications.

Mesembryanthemum crystallinum L. (common ice plant) is an edible halophyte. However, if ice plants are used to phytoremediate salinity soil, there are problems of slow initial growth, and a long period before active NaCl uptake occurs under higher salinity conditions. Application of endophytic bacteria may improve the problem, but there remain gaps in our understanding of how endophytic bacteria affect the growth and the biochemical and physiological characteristics of ice plants. The aims of this study were to identify growth-promoting endophytic bacteria from the roots of ice plants and to document the metabolomic response of ice plants after application of selected endophytic bacteria. Two plant growth-promoting endophytic bacteria were selected on the basis of their ability to promote ice plant growth. The two strains putatively identified as Microbacterium spp. and Streptomyces spp. significantly promoted ice plant growth, at 2-times and 2.5-times, respectively, compared with the control and also affected the metabolome of ice plants. The strain of Microbacterium spp. resulted in increased contents of metabolites related to the tricarboxylic acid cycle and photosynthesis. The effects of salt stress were alleviated in ice plants inoculated with the endobacterial strains, compared with uninoculated plants. A deeper understanding of the complex interplay among plant metabolites will be useful for developing microbe-assisted soil phytoremediation strategies, using Mesembryanthemum species.

This research presents a design optimization of a solar adsorption ice plant using activated carbon and methanol as working pairs in the climate of Makkah to produce a ton of ice per day. The plant consists of six adsorption refrigeration units. Each unit has 72 separate cylindrical adsorbent beds connected with its own condenser, valve, and evaporator. The unit is heated by seven evacuated-tube collectors connected in parallel. Then the total number of collectors are 42 collectors. The beds are filled by estimated amount of 540 kg of activated carbon with 178 kg of methanol. Selection and dimensioning of each component were carried out based on previous recommended values. A mathematical model and simulation were developed to validate the system performance along the year. The results showed that plant could produce up to ton of ice daily along the year. The coefficient of performance can reach 0.9 with condenser and evaporator temperatures of 35°C and -5°C respectively. The performance of the system is greatly affected by ambient temperature than solar radiation. Therefore, the best performance and largest amount of ice was found in the winter season.

The common ice plant (Mesembryanthemum crystallinum L.) is a facultative crassulacean acid metabolism (CAM) plant, and its ability to recover from stress-induced CAM has been confirmed. We analysed the photosynthetic metabolism of this plant during the 72-h response period following salinity stress removal from three perspectives. In plants under salinity stress (CAM) we found a decline of the quantum efficiencies of PSII (Y(II)) and PSI (Y(I)) by 17% and 15%, respectively, and an increase in nonphotochemical quenching (NPQ) by almost 25% in comparison to untreated control. However, 48 h after salinity stress removal, the PSII and PSI efficiencies, specifically Y(II) and Y(I), elevated nonphotochemical quenching (NPQ) and donor side limitation of PSI (YND), were restored to the level observed in control (C3 plants). Swelling of the thylakoid membranes, as well as changes in starch grain quantity and size, have been found to be components of the salinity stress response in CAM plants. Salinity stress induced an over 3-fold increase in average starch area and over 50% decline of average seed number in comparison to untreated control. However, in plants withdrawn from salinity stress, during the first 24 h of recovery, we observed chloroplast ultrastructures closely resembling those found in intact (control) ice plants. Rapid changes in photosystem functionality and chloroplast ultrastructure were accompanied by the induction of the expression (within 24 h) of structural genes related to the PSI and PSII reaction centres, including PSAA, PSAB, PSBA (D1), PSBD (D2) and cp43. Our findings describe one of the most flexible photosynthetic metabolic pathways among facultative CAM plants and reveal the extent of the plasticity of the photosynthetic metabolism and related structures in the common ice plant.

Mesembryanthemum crystallinum (common ice plant) is a halophyte species that has adapted to extreme conditions. In this study, we cloned a McHB7 transcription factor gene from the ice plant. The expression of McHB7 was significantly induced by 500 mM NaCl and it reached the peak under salt treatment for 7 days. The McHB7 protein was targeted to the nucleus. McHB7-overexpressing in ice plant leaves through Agrobacterium-mediated transformation led to 25 times more McHB7 transcripts than the non-transformed wild type (WT). After 500 mM NaCl treatment for 7 days, the activities of superoxide dismutase (SOD) and peroxidase (POD) and water content of the transgenic plants were higher than the WT, while malondialdehyde (MDA) was decreased in the transgenic plants. A total of 1082 and 1072 proteins were profiled by proteomics under control and salt treatment, respectively, with 22 and 11 proteins uniquely identified under control and salt stress, respectively. Among the 11 proteins, 7 were increased and 4 were decreased after salt treatment. Most of the proteins whose expression increased in the McHB7 overexpression (OE) ice plants under high salinity were involved in transport regulation, catalytic activities, biosynthesis of secondary metabolites, and response to stimulus. The results demonstrate that the McHB7 transcription factor plays a positive role in improving plant salt tolerance.
