Abstract Background: Recent research has demonstrated that Type 2 Diabetes (T2D) risk is influenced by a number of common polymorphisms, including rs17782313 MC4R, rs1801282 PPARG, and rs7903146 TCF7L2. Knowledge of the association between these SNPs (single nucleotide polymorphism) and body weight changes in different forms of prediabetes treatment is still limited. The aim of this study was to investigate the association of polymorphisms within the MC4R, PPARG, TCF7L2 genes on the risk of carbohydrate metabolism disorders and body composition changes in overweight or obese patients with early carbohydrate metabolism disorders. Methods and Results: From 327 patients, a subgroup of 81 prediabetic female patients (48.7±14.8 years) of Eastern European descent participated in a 3-month study comprised of diet therapy or diet therapy accompanied with metformin treatment. Bioelectrical impedance analysis and genotyping of rs17782313 MC4R, rs7903146 TCF7L2, rs1801282 PPARG were performed. The MC4R CC and TCF7L2 TT genotypes were associated with increased risk of T2D (OR=1.46, p=0.05 and OR=2.47, p=0.006, respectively). PPARG СС homozygotes experienced increased weight loss; however, no additional improvements were experienced with the addition of metformin. MC4R ТТ homozygotes who took metformin alongside dietary intervention experienced increased weight loss and reductions in fat mass (p<0.05).Conclusions: We confirmed the previous association of the MC4R C and TCF7L2 T alleles with T2D risk. The obesity-protective alleles (MC4R T and PPARG C) were positively associated with weight loss efficiency.

Abstract The study was aimed at comparing pacing adopted by males and females in a 107-km mountain ultramarathon and assessing whether pacing-related variables were associated with intracompetition body weight changes and performance. Forty-seven athletes (29 males; 18 females) were submitted to a cardiopulmonary exercise test before the race. Athletes were also weighted before the start of the race, at three midpoints (33 km, 66 km and 84 km) and after the race. Pacing was analyzed using absolute and relative speeds and accelerometry-derived sedentary time spent during the race. Results showed that females spent less sedentary time (4.72 ± 2.91 vs. 2.62 ± 2.14%; p = 0.035; d = 0.83) and displayed a smaller body weight loss (3.01 ± 1.96 vs. 4.37 ± 1.77%; p = 0.048; d = 0.77) than males. No significant sex differences were revealed for speed variability, absolute and relative speed. In addition, finishing time was correlated with: speed variability (r = 0.45; p = 0.010), index of pacing (r = -0.63; p < 0.001) and sedentary time (r = 0.64; p < 0.001). Meanwhile, intracompetition body weight changes were related with both the absolute and relative speed in the first and the last race section. These results suggest that females, as compared with males, take advantage of shorter time breaks at aid stations. Moreover, performing a more even pacing pattern may be positively associated with performance in mountain ultramarathons. Finally, intracompetition body weight changes in those races should be considered in conjunction with running speed fluctuations.

Home confinement during the COVID-19 pandemic is accompanied by dramatic changes in lifestyle and dietary behaviors that can significantly influence health. We conducted an online cross-sectional survey to assess COVID-19 pandemic-induced dietary and lifestyle changes and their association with perceived health status and self-reported body weight changes among 1000 Indian adults in early 2021. Positive improvements in dietary habits, e.g., eating more nutritious (85% of participants) and home-cooked food (89%) and an increase in overall nutrition intake (79%), were observed. Sixty-five percent of participants self-reported increased oat consumption to support immunity. There were some negative changes, e.g., more binge eating (69%), eating more in between meals (67%), and increasing meal portion size (72%). Two-thirds of participants reported no change in lifestyles, whereas 21 and 23% reported an increase, and 13 and 10% reported a decrease in physical activity and sleep, respectively. Overall, 64 and 65% of participants reported an improvement in perceived health and an increase in body weight during the COVID-19 period compared to pre-COVID-19, respectively. The top motivations for improving dietary habits included improving physical and mental health and building immunity. In conclusion, the overall perceived health was improved and there was an increase in self-reported body weight in most participants during COVID-19. Diet emerged as the most crucial determinant for these changes.

Abstract Background Patients with type 2 diabetes (T2D) may experience frequent body weight changes over time. The prognostic impact of these weight changes (gains or losses) requires further study. Aims To study the associations between changes in body weight (intentional or unintentional) with subsequent outcomes. Methods The EXAMINE trial included 5380 patients with T2D and a recent acute coronary syndrome, who were randomized to alogliptin or placebo. Time-updated Cox models and mixed effects models were used to test the associations between changes in body weight and subsequent outcomes over a median follow-up of 1.6 (1.0–2.1) years. Results During the post-randomization follow-up period, 1044 patients (19.4%) experienced a weight loss ≥ 5% of baseline weight, 2677 (49.8%) had a stable weight, and 1659 (30.8%) had a ≥ 5 % weight gain. Patients with weight loss were more frequently women and had more co-morbid conditions. In contrast, patients who gained ≥ 5% weight were more frequently men with less co-morbid conditions. A weight loss ≥ 5% was independently associated with a higher risk of subsequent adverse outcomes, including all-cause mortality: adjusted HR (95% CI) = 1.79 (1.33–2.42), P < 0.001. Similar associations were found for cardiovascular mortality, the composite of cardiovascular mortality or heart failure hospitalization, and the primary outcome. A weight gain ≥ 5% was independently associated with an increase in the risk of subsequent cardiovascular mortality or heart failure hospitalization only: adjusted HR (95% CI) = 1.34 (1.02–1.76), P = 0.033. Conclusions In patients with T2D who had a recent ACS/MI, a ≥ 5% loss of body weight was associated with a higher risk of subsequent cardiovascular events and mortality.

This study was designed to investigate the effects of supplementation of high and low energy based concentrate diets (13.77 Vs 12.14 MJ/kg DM metabolizable energy) on the nutrient intake and body weight changes of indigenous growing buffalo bull calves in Bangladesh. The crude protein contents of both the supplemented diets were similar (about 12.50% on DM basis). In this study, a total of fifteen indigenous growing buffalo bull calves (av. Live weight 85.10±0.874 kg) were selected from different villages. It was found that the total dry matter intake was the highest (28.22±0.015 g/kg W0.75/d) in group supplemented with high energy concentrate diet along with farmer’s practices (D1), thereafter, in group supplemented with low energy concentrate diet along with farmer’s practices (D2) (27.79±0.015 g/kg W0.75/d) and the lowest (26.39±0.016 g/kg W0.75/d) in control group (D0) in which the animals were fed only farmers practices diet. Similarly, the crude protein intake was the highest in D1 group (2.86 ±0.029 g/kg W0.75/d), followed by D2 group (2.83±0.010 g/kg W0.75/d) and the lowest in farmer’s practices diet, D0 (2.08b±0.021 g/kg W0.75/d).  However, the final body weight of buffalo bull calves was higher in both groups supplemented with concentrate diets (D1 and D2) compared with control (D0) and differed significantly (p<0.01) while there were no significant differences (p>0.05) found between D1 and D2 where high energy concentrate supplementation group buffalo bull claves, D1 tended to be higher than low energy concentrate supplementation group buffalo bull claves, D2. The highest final (254.6 kg) body weight was recorded in high energy concentrate supplemented diets and the lowest (144.0 kg) was found in control. Additionally, the average final body weight was increased by 153.40 kg (151.58%), 150.40 kg (152.22%) and 45.60 kg (46.34%) in D1, D2 and D0 group buffalo bull calves, respectively. On the other hand, the average final body weight of D1 and D2 was increased by 110.60 kg (76.80%) and 105.20 kg (73.05%), respectively, compared with D0. In addition, the highest (6.46) feed conversion efficiency (FCE) was recorded in D1 followed by D2 (6.64) and the lowest (12.17) was found in D0. The trends of cost benefit ratio was the same like other considering factors, the highest (1:3.60) found in D1 followed by D2 (1:3.51) and the lowest (1:1.97) in D0. Finally, considering all the parameters, it was clearly indicated that supplementation of concentrate diet along with farmers practice is essential for fast growing, higher body weight gain and more benefits by rearing buffalo bull calves at Subornochar Upozila of Noakhali district in Bangladesh.
