Leaf ontogenesis of Trembleya phlogiformis DC. (Microlicieae, Melastomataceae): functional aspects and indumentum characterization

Leaf ontogenesis is determinant for the establishment and regulation of its structural and functional properties, in addition to being an excellent tool for assignment to different groups of angiosperms. Even though the importance of leaf morphology and anatomy for taxonomic use is well known, few studies have addressed the processes of leaf ontogeny in Melastomataceae. Herein, we sought to define the ontogenetic steps of leaf of Trembleya phlogiformis, highlighting the indumentum, to understand the main functional traits. Shoot apex, young and fully expanded leaves were processed by usual light microscopy procedures. At the first node, leaf primordia are densely covered with trichomes and emergences. The adaxial layer of ground meristem gives rise to the palisade parenchyma, the procambium originates from median layers of ground meristem and the spongy parenchyma develops from abaxial layers of ground meristem. The differentiation of isobilateral mesophyll on leaves of T. phlogiformis, a common feature in Microlicieae, comes from ground meristem stratification. However, T. phlogiformis stands out by showing in the leaf mature spongy parenchyma cells with irregular shapes. The leaf ontogeny reveals distinct mechanisms of cell differentiation and may be important for the establishment of functional adaptive traits.

Defensive Traits during White Spruce (Picea glauca) Leaf Ontogeny

Changes during leaf ontogeny affect palatability to herbivores, such that many insects, including the eastern spruce budworm (Choristoneura fumiferana (Clem.)), are specialist feeders on growing conifer leaves and buds. Developmental constraints imply lower toughness in developing foliage, and optimal defense theory predicts higher investment in chemical defense in these vulnerable yet valuable developing leaves. We summarize the literature on the time course of defensive compounds in developing white spruce (Picea glauca (Moench) Voss) needles and report original research findings on the ontogeny of white spruce needle toughness. Our results show the predicted pattern of buds decreasing in toughness followed by leaves increasing in toughness during expansion, accompanied by opposite trends in water content. Toughness of mature foliage decreased slightly during the growing season, with no significant relationship with water content. Toughness of sun-grown leaves was slightly higher than that of shade-grown leaves. However, the literature review did not support the expected pattern of higher defensive compounds in expanding leaves than in mature leaves, suggesting that white spruce might instead exhibit a fast-growth low-defense strategy.

Use eco-physiological modelling to investigate Potassium limitation of wood productivity in tropical eucalypt plantations

<p>Potassium (K) is essential for a wide range of physiological functions in plants, and a limiting element for wood productivity in numerous forest ecosystems. However, the contribution of each of the K-sensitive physiological processes to the limitation of wood productivity is poorly known. In trees, K deficiency acts both on the source and the sinks of carbon making it difficult to disentangle its effects on wood productivity. The literature dealing with the influence of K-limitation on tree physiologywhile disparate, shows some converging results. Furthermore, K-limited tropical <em>Eucalyptus</em> plantations have been studied extensively over the last 2 decades. Large scale fertilization experiments, run over multiple rotations, allow us to gain insight into the ecosystem’s K-cycle as a whole and the physiological processes that are impacted the most by K deficiency. Mechanistic modeling of this system should allow us to quantify the relative contribution of each process when it comes to wood productivity limitation by K. We have thus adapted an eco-physiological model (CASTANEA-CNP), previously used in temperate forest settings, to use in tropical eucalypt plantations. This has led us to adapt existing nutrient (N and P) eco-physiological modeling frameworks specifically for K as well as focus on processes that are little impacted by N and P availability but greatly by K availability. The biological K-cycle model was calibrated using the comprehensive experimental data. Carbon and water fluxes were calibrated using data from a flux tower site (Eucflux) with the same environmental conditions as the experimental plots. The development of a new canopy generation model was mandated by both the continuous nature of leaf generation in <em>Eucalyptus grandis </em>and the major interaction between leaf ontogeny and the K-cycle. At first we focus mainly on carbon assimilation at the canopy level. Here we present the preliminary results obtained by this model.</p>

Sweet Immunity: The Effect of Exogenous Fructans on the Susceptibility of Apple (Malus × domestica Borkh.) to Venturia inaequalis

There is an urgent need for novel, efficient and environmentally friendly strategies to control apple scab (Venturia inaequalis), for the purpose of reducing overall pesticide use. Fructans are recently emerging as promising “priming” compounds, standing out for their safety and low production costs. The objective of this work was to test a fructan-triggered defense in the leaves of apple seedlings. It was demonstrated that exogenous leaf spraying can reduce the development of apple scab disease symptoms. When evaluated macroscopically and by V. inaequalis-specific qPCR, levan-treated leaves showed a significant reduction of sporulation and V. inaequalis DNA in comparison to mock- and inulin-treated leaves, comparable to the levels in fosetyl-aluminum-treated leaves. Furthermore, we observed a significant reduction of in vitro mycelial growth of V. inaequalis on plates supplemented with levans when compared to controls, indicating a direct inhibition of fungal growth. Variations in endogenous sugar contents in the leaves were followed during priming and subsequent infection, revealing complex dynamics as a function of time and leaf ontogeny. Our data are discussed in view of the present theories on sugar signaling and fructan-based immunity, identifying areas for future research and highlighting the potential use of fructans in apple scab management in orchards.

Biochemical and Molecular Characterization of PvNTD2, a Nucleotidase Highly Expressed in Nodules from Phaseolus vulgaris

Nucleotides are molecules of great importance in plant physiology. In addition to being elementary units of the genetic material, nucleotides are involved in bio-energetic processes, play a role as cofactors, and are also components of secondary metabolites and the hormone cytokinin. The common bean (Phaseolus vulgaris) is a legume that transports the nitrogen fixed in nodules as ureides, compounds synthetized from purine nucleotides. The first step in this pathway is the removal of the 5’-phosphate group by a phosphatase. In this study, a gene that codes for a putative nucleotidase (PvNTD2) has been identified in P. vulgaris. The predicted peptide contains the conserved domains for haloacid dehalogenase-like hydrolase superfamily. The protein has been overexpressed in Escherichia coli, and the purified protein showed molybdate-resistant phosphatase activity with nucleoside monophosphates as substrates, confirming that the identified gene codes for a nucleotidase. The optimum pH for the activity was 7–7.5. The recombinant enzyme did not show special affinity for any particular nucleotide, although the behaviour with AMP was different from that with the other nucleotides. The activity was inhibited by adenosine, and a regulatory role for this nucleoside was proposed. The expression pattern of PvNTD2 shows that it is ubiquitously expressed in all the tissues analysed, with higher expression in nodules of adult plants. The expression was maintained during leaf ontogeny, and it was induced during seedling development. Unlike PvNTD1, another NTD previously described in common bean, the high expression of PvNTD2 was maintained during nodule development, and its possible role in this organ is discussed.

Leaf metabolic signatures induced by real and simulated herbivory in black mustard (Brassica nigra)

Introduction The oxylipin methyl jasmonate (MeJA) is a plant hormone active in response signalling and defence against herbivores. Although MeJA is applied experimentally to mimic herbivory and induce plant defences, its downstream effects on the plant metabolome are largely uncharacterized, especially in the context of primary growth and tissue-specificity of the response. Objectives We investigated the effects of MeJA-simulated and real caterpillar herbivory on the foliar metabolome of the wild plant Brassica nigra and monitored the herbivore-induced responses in relation to leaf ontogeny. Methods As single or multiple herbivory treatments, MeJA- and mock-sprayed plants were consecutively exposed to caterpillars or left untreated. Gas chromatography (GC) and liquid chromatography (LC) time-of-flight mass-spectrometry (TOF-MS) were combined to analyse foliar compounds, including central primary and specialized defensive plant metabolites. Results Plant responses were stronger in young leaves, which simultaneously induced higher chlorophyll levels. Both MeJA and caterpillar herbivory induced similar, but not identical, accumulation of tricarboxylic acids (TCAs), glucosinolates (GSLs) and phenylpropanoids (PPs), but only caterpillar feeding led to depletion of amino acids. MeJA followed by caterpillars caused higher induction of defence compounds, including a three-fold increase in the major defence compound allyl-GSL (sinigrin). When feeding on MeJA-treated plants, caterpillars gained less weight indicative of the reduced host-plant quality and enhanced resistance. Conclusions The metabolomics approach showed that plant responses induced by herbivory extend beyond the regulation of defence metabolism and are tightly modulated throughout leaf development. This leads to a new understanding of the plant metabolic potential that can be exploited for future plant protection strategies.

A comparison of inducible, ontogenetic, and interspecific sources of variation in the foliar metabolome in tropical trees

Plant interactions with other organisms are mediated by chemistry, yet chemistry varies among conspecific and within individual plants. The foliar metabolome—the suite of small-molecule metabolites found in the leaf—changes during leaf ontogeny and is influenced by the signaling molecule jasmonic acid. Species differences in secondary metabolites are thought to play an important ecological role by limiting the host ranges of herbivores and pathogens, and hence facilitating competitive coexistence among plant species in species-rich plant communities such as tropical forests. Yet it remains unclear how inducible and ontogenetic variation compare with interspecific variation, particularly in tropical trees. Here, we take advantage of novel methods to assemble mass spectra of all compounds in leaf extracts into molecular networks that quantify their chemical structural similarity in order to compare inducible and ontogenetic chemical variation to among-species variation in species-rich tropical tree genera. We ask (i) whether young and mature leaves differ chemically, (ii) whether jasmonic acid-inducible chemical variation differs between young and mature leaves, and (iii) whether interspecific exceeds intraspecific chemical variation for four species from four hyperdiverse tropical tree genera. We observed significant effects of the jasmonic acid treatment for three of eight combinations of species and ontogenetic stage evaluated. Three of the four species also exhibited large metabolomic differences with leaf ontogenetic stage. The profound effect of leaf ontogenetic stage on the foliar metabolome suggests a qualitative turnover in secondary chemistry with leaf ontogeny. We also quantified foliar metabolomes for 45 congeners of the four focal species. Chemical similarity was much greater within than between species for all four genera, even when within-species comparisons included leaves that differed in age and jasmonic acid treatment. Despite ontogenetic and inducible variation within species, chemical differences among congeneric species may be sufficient to partition niche space with respect to chemical defense.
