Abstract Many patients with fibromyalgia (FM) experience fatigue, but the associated biological mechanisms have not been delineated. We aimed to investigate the neural signatures associated with fatigue severity in patients with FM using MRI. We consecutively recruited 138 patients with FM and collected their clinical profiles and brain-MRI data. We categorized the patients into 3 groups based on their fatigue severity. Using voxel-based morphometry analysis and trend analysis, we first identified neural structures showing volumetric changes associated with fatigue severity, and further explored their seed-to-voxel structural covariance networks (SCNs). Results showed decreased bilateral thalamic volumes were associated with higher severity of fatigue. There was a more widespread distribution of the thalamic SCNs to the frontal, parietal, subcortical, and limbic regions in patients with higher fatigue severity. In addition, increased right inferior temporal cortex volumes were associated with higher severity of fatigue. The right inferior temporal seed showed more SCNs distributions over the temporal cortex and a higher strength of SCNs to the bilateral occipital cortex in patients with higher fatigue severity. The thalamus and the right inferior temporal cortex as well as their altered interactions with cortical and subcortical regions comprise the neural signatures of fatigue in FM.

The Shillong Plateau is characterized by multiple phases of deformation and number of prominent sets joints/fracture system. The Southern Shillong Plateau unlike the northern part is highly dissected by the scarp faces which are mostly erosional. The area is characterized by deep incising river networks which cuts across many lithological units. To understand the factors influencing the drainage characteristic of the area and landscape development, the present study has been done in context of morphometry, geomorphology and geology of the drainage basin. Fifteen sub basins of 4th order have been selected from the Um Sohrynkew River basin. Drainage morphometry, indices of active tectonism, geology and tectonics and landform features have been worked out for better evaluation of drainage characteristics. The study area forms part of the Meghalaya Precambrian province of upper Proterozoic age. The southern border of the Shillong Plateau is demarcated by Dauki (Also known as Dawki) fault which is a prominent structural lineament. It consists of at least four E-W trending normal faults with occasional reversal. The segment of the Dauki fault in the study area is believed to be active. The present study focuses on the drainage network and the landscape development of the study area where a very strong relationship has been observed between the lithology and structure together with the tectonic activity influencing the drainage pattern in the area.

AbstractThe endogenous mu-opioid receptor (MOR) system modulates a multitude of social and reward-related functions, and exogenous opiates also influence sex drive in humans and animals. Sex drive shows substantial variation across humans, and it is possible that individual differences in MOR availability underlie interindividual of variation in human sex drive. We measured healthy male subjects’ (n = 52) brain’s MOR availability with positron emission tomography (PET) using an agonist radioligand, [11C]carfentanil, that has high affinity for MORs. Sex drive was measured using self-reports of engaging in sexual behaviour (sex with partner and masturbating). Bayesian hierarchical regression analysis revealed that sex drive was positively associated with MOR availability in cortical and subcortical areas, notably in caudate nucleus, hippocampus, and cingulate cortices. These results were replicated in full-volume GLM analysis. These widespread effects are in line with high spatial autocorrelation in MOR expression in human brain. Complementary voxel-based morphometry analysis (n = 108) of anatomical MR images provided limited evidence for positive association between sex drive and cortical density in the midcingulate cortex. We conclude that endogenous MOR tone is associated with individual differences in sex drive in human males.

AbstractRho-associated kinase isoform 2 (ROCK2) is an attractive drug target for several neurologic disorders. A critical barrier to ROCK2-based research and therapeutics is the lack of a mouse model that enables investigation of ROCK2 with spatial and temporal control of gene expression. To overcome this, we generated ROCK2fl/fl mice. Mice expressing Cre recombinase in forebrain excitatory neurons (CaMKII-Cre) were crossed with ROCK2fl/fl mice (Cre/ROCK2fl/fl), and the contribution of ROCK2 in behavior as well as dendritic spine morphology in the hippocampus, medial prefrontal cortex (mPFC), and basolateral amygdala (BLA) was examined. Cre/ROCK2fl/fl mice spent reduced time in the open arms of the elevated plus maze and increased time in the dark of the light–dark box test compared to littermate controls. These results indicated that Cre/ROCK2fl/fl mice exhibited anxiety-like behaviors. To examine dendritic spine morphology, individual pyramidal neurons in CA1 hippocampus, mPFC, and the BLA were targeted for iontophoretic microinjection of fluorescent dye, followed by high-resolution confocal microscopy and neuronal 3D reconstructions for morphometry analysis. In dorsal CA1, Cre/ROCK2fl/fl mice displayed significantly increased thin spine density on basal dendrites and reduced mean spine head volume across all spine types on apical dendrites. In ventral CA1, Cre/ROCK2fl/fl mice exhibited significantly increased spine length on apical dendrites. Spine density and morphology were comparable in the mPFC and BLA between both genotypes. These findings suggest that neuronal ROCK2 mediates spine density and morphology in a compartmentalized manner among CA1 pyramidal cells, and that in the absence of ROCK2 these mechanisms may contribute to anxiety-like behaviors.

Cave-dwelling ground beetles in China represent the most impressive specific diversity and morphological adaptations of the cavernicolous ground beetles in the world, but they have not been systematically examined in quantitative terms. The present study focuses on the application of geometric morphological methods to address the morphological adaptations of the tribe Trechini, the most representative group in China. We have employed a geometric morphometry analysis of the head, pronotum, and elytra of 53 genera of Trechini, including 132 hypogean and 8 epigean species. Our results showed that the overall morphological variation of cave carabids has gradually specialized from an anophthalmic to semi-aphaenopsian to aphaenopsian type. There were extremely significant differences (p < 0.01) among four different adaptive types including aphaenopsian, semi-aphaenopsian, anophthalmic, and surface-dwelling Trechini when their adaptability to a cave environment was used as the basis for grouping. Furthermore, there were differences in the phenotypic tree of the head, pronotum, and elytra, and an integrated morphology. To the best of our knowledge, this is the first report on the analysis of the head, pronotum, and elytra of four different adaptive types of ground beetles in order to clarify the morphological adaptations of cavernicolous carabids to the cave environment.

The Caryophyllaceae family is complex. Several attempts have been carried out in the past to study Caryophyllaceae members. This study mainly focused on Allochrusa Bunge to determine its genetic structure and used ISSR markers, ITS, and rps16 data to classify and differentiate Allochrusa species. We collected 122 Allochrusa specimens. Our analysis included morphological and molecular method approaches. Morphometry analysis indicated that floral characters could assist in the identification of Allochrusa species. A. persica (Boiss.) Boiss. and A. versicolor Fisch. & C.A.Mey. showed affinity to each other. A. bungei Boiss. formed a separate group. Analysis of molecular variance showed significant genetic differentiation in Allochrusa (p= 0.001). The majority of genetic variation was among the Allochrusa population. We recorded minimum gene flow (Nm=0.176) between Allochrusa species. Besides this, isolation by distance occurs in Allochrusa members, as shown in the Mantel test result (r = 0.01, p = 0.0002). STRUCTURE analysis revealed three genetic groups. It is evident that A. persica, A. versicolor, and A. bungei differ genetically from each other. Our current findings have implications in plant systematics and biodiversity management.

Neurodegenerative disease syndromes often affect personality and interpersonal behavior in addition to cognition, but there are few structured observational measures of altered social demeanor validated for this population. We developed the Social Behavior Observer Checklist (SBOCL), a 3-min checklist tool, to facilitate identification of patterns of interpersonal behavior that are diagnostically relevant to different neurodegenerative syndromes. Research assistants without formal clinical training in dementia used the SBOCL to describe participants' behavior, including 125 healthy older adults and 357 patients diagnosed with one of five neurodegenerative disease syndromes: 135 behavioral variant frontotemporal dementia (bvFTD), 57 semantic variant primary progressive aphasia (svPPA), 51 non-fluent variant PPA (nfvPPA), 65 progressive supranuclear palsy (PSP), and 49 amyloid-positive Alzheimer's disease syndrome (AD), all of whom had concurrent 3D T1 MRI scans available for voxel-based morphometry analysis. SBOCL item interrater reliability ranged from moderate to very high, and score elevations showed syndrome-specific patterns. Subscale scores derived from a degree*frequency product of the items had excellent positive predictive value for identifying patients. Specifically, scores above 2 on the Disorganized subscale, and above 3 on the Reactive and Insensitive subscales, were not seen in any healthy controls but were found in many patients with bvFTD, svPPA, nfvPPA, PSP, and AD syndromes. Both the Disorganized and Reactive subscale scores showed significant linear relationships with frontal and temporal gray matter volume that generalized across syndromes. With these initial psychometric characteristics, the SBOCL may be a useful measure to help non-experts identify patients who are appropriate for additional specialized dementia evaluation, without adding time to patient encounters or requiring the presence of an informant.

This study assessed the effect of the combination of microencapsulated sodium butyrate (SB) and phytase (PH) on the performance, intestinal integrity, and bone development of broilers. The experiment comprised 736 chicks distributed in a completely randomised design in a 2 × 2 factorial scheme (with and without the inclusion of SB and with the inclusion of PH at the recommended dose and superdosing) totalling four treatments and eight repetitions of 23 birds each. SB was added at 0.750 kg/tonne and PH was included at 750 phytase units (FTU)/kg and 1 500 FTU/kg of diet. The live weight (LW), feed intake (FI) and feed conversion (FC) were evaluated weekly. Bone measurements of the tibia and femur, densitometry, length, the Seedor index, and diameter were performed at seven and 28 days. In the same ages, fragments of the jejunum and ileum segments were collected and subjected to a morphometry analysis. At seven days of age, the birds supplemented with the phytase superdosing showed a better FI and LW, and the microencapsulated SB in the diet showed a better LW and FC. In the period from one to 28 days, the treatment supplemented with SB provided a higher FI and LW. The SB supplementation resulted in greater bone measurements at seven and 28 days. The use of the superdosing phytase increased the villus length and width of the jejunum at seven days and at 28 days. There was an increase in the villus length, villus-to-crypt ratio, absorption area, and muscular layer of the jejunum and a decrease in the crypt width of the jejunum. The SB supplementation increased the ileum crypt width at seven days and there was no effect on any intestinal segment at 28 days. The supplementation of SB or PH 1 500 FTU/kg in the broiler diets’ growth promoter or antibiotic-free resulted in a better performance and bone measurements, while the association of the additional PH 1 500 FTU/kg and SB supplementation resulted in the greater depth and width of the jejunum crypt and villus : ileum crypt at seven days and greater villus length and ileum absorption area at 28 days. The association of SB and PH may be a strategy to improve the performance and bone quality and intestinal integrity in broilers.
