AbstractExtracorporeal membrane oxygenation (ECMO) is a cardiopulmonary technology capable of supporting cardiac and respiratory function in the presence of end-stage lung disease. Initial experiences using ECMO as a bridge to lung transplant (ECMO-BTLT) were characterized by high rates of ECMO-associated complications and poor posttransplant outcomes. More recently, ECMO-BTLT has garnered success in preserving patients' physiologic condition and candidacy prior to lung transplant due to technological advances and improved management. Despite recent growth, clinical practice surrounding use of ECMO-BTLT remains variable, with little data to inform optimal patient selection and management. Although many questions remain, the use of ECMO-BTLT has shown promising outcomes suggesting that ECMO-BTLT can be an effective strategy to ensure that complex and rapidly decompensating patients with end-stage lung disease can be safely transplanted with good outcomes. Further studies are needed to refine and inform practice patterns, management, and lung allocation in this high-risk and fragile patient population.

Conditionally activated (“caged”) oligonucleotides provide useful spatiotemporal control for studying dynamic biological processes, e.g., regulating in vivo gene expression or probing specific oligonucleotide targets. This review summarizes recent advances in caging strategies, which involve different stimuli in the activation step. Oligo cyclization is a particularly attractive caging strategy, which simplifies the probe design and affords oligo stabilization. Our laboratory developed an efficient synthesis for circular caged oligos, and a circular caged antisense DNA oligo was successfully applied in gene regulation. A second technology is Transcriptome In Vivo Analysis (TIVA), where caged oligos enable mRNA isolation from single cells in living tissue. We highlight our development of TIVA probes with improved caging stability. Finally, we illustrate the first protease-activated oligo probe, which was designed for caspase-3. This expands the toolkit for investigating the transcriptome under a specific physiologic condition (e.g., apoptosis), particularly in specimens where light activation is impractical.

Abstract Purpose To investigate the characteristics of eyes with dry eye disease (DED) whose lipid layer thickness (LLT) measured 100 nm on a LipiView II interferometer and compare the DED parameters of them to those with LLT below 100 nm. Methods A total of 201 eyes of 102 enrolled DED patients (mean age 56.4 ± 11.8 years) were classified into 3 groups according to their average LLT; < 60 nm as thin-LLT (n = 49), 60–99 nm as normal-LLT (n = 77), and 100 nm as thick-LLT (n = 75). LLT, meiboscore, Schirmer I test, tear film break-up time (TBUT), ocular surface staining (OSS), and ocular surface disease index (OSDI) were assessed. Results The OSS and TBUT were significantly worse in the thick-LLT group than in the normal-LLT group (p = 0.020, and p = 0.028, respectively). The OSDI was significantly higher in the thick-LLT group than in the thin-LLT group (p = 0.006). However, the meiboscore was not different among the three groups (p = 0.33). Age, OSS, and OSDI showed a positive correlation with LLT (r = 0.16, p = 0.023; r = 0.213, p = 0.003; and r = 0.338, p = 0.001, respectively). In sensitivity analyses, eyes with corneal erosions had a significantly higher average LLT (p = 0.015), higher OSDI (p = 0.009), shorter TBUT (p < 0.001), and shorter Schirmer I value (p = 0.024) than those with clear corneas. Conclusion The average LLT of eyes with corneal erosions was thicker than those without erosions, suggesting that the LLT of 100 nm in the eyes with corneal erosions should not be regarded as a stable physiologic condition. Cautious interpretation of LLT along with other dry eye parameters is required.

Background Flat foot, also known as pes planus, is a common problem in daily pediatric practice. For most children, this physiologic condition gradually disappears with age. However, flat foot that persists after the age of four might have developmental coordination disorder in the future and it may relate to the history of delayed walking . Objective To determine the prevalence of delayed walking in children with pes planus compared to children with normal foot curvature and to assess for a possible association between flat foot and history of delayed walking. Methods This cross-sectional study was done in 120 children aged 5-6 years. Foot curvature was evaluated by wet footprint test. Inclusion criteria include children in 3 playgroups in Tangerang from January to August 2018, in children aged 5-6 years. Children with history of neurologic diseases, genetic disorders, chronic diseases, disorders of the lower extremities except pes planus, and obesity were excluded. Grading of pes planus refers to Olivier et al. criteria and evaluation of delayed walking at 18-month-old was done through history taking from their parents. Results Of 120 children, 41 (34.2%) had pes planus while 11 (9.2%) had a history of delayed walking. Of those 11 children, 9 had pes planus. Most children with pes planus had grade I (78%). There was a significant association between pes planus and delayed walking (OR=10.8; 95%CI 2.2 to 52,9; P=0.001;). Conclusion In 5-6-year-old children, there is a significant association between pes planus grade 2 & 3 and history of delayed walking early in life. Wet footprint test screening for children with a history or signs of delayed walking may be used to identify pes planus in order to implement treatment in a timely manner.

Chronic high glucose (HG) exposure increases methylglyoxal (MG)-derived AGEs and is involved in the onset of pathological conditions, such as diabetes, atherosclerosis and chronic‐degenerative diseases. Under physiologic condition the harmful effects of MG are contrasted by glyoxalase system that is involved in the detoxification of Reactive Carbonyl Species (RCS) and maintain the homeostasis of the redox environment of the cell. Polyphenols are the most abundant antioxidants in the diet and present various health benefits. The study aimed at investigating the role of polyphenols extracted from an apple high in polyphenols (Calville White Winter), on glyco-oxidative stress induced by chronic HG-exposure. Intestinal Caco-2 cells were treated in physiological glucose condition (25mM) as a control and in HG condition (50mM) with or without apple extract for one week. Our data demonstrated that HG-treatment triggers glyco-oxidation stress with a significantly increase in ROS, lipid peroxidation, AGEs and Glyoxalase I (GlxI) activity with a significant decrease in total antioxidant intracellular defense. Treatment with polyphenols under HG condition restores to the control levels GlxI activity, decreases Glyoxalase II (GlxII) in relation to the control and induces a drop of glyco-oxidative damage. This paper seeks to highlight the roles of polyphenols in glyco-oxidative stress.

Female wasps such as Polistes versicolor can form aggregates to face weather conditions that are not suitable to sustain their colonies. The interactions between individuals in these aggregates, just as in other associations, are probably facilitated by chemical signals. Of these compounds some of the most efficient during social interactions of insects are those called contact pheromones or superficial pheromones. This special type of pheromones, known as cuticular hydrocarbons, can be found in insects cuticle. They facilitate the differentiation of caste, species and nestmates, and may be important indicators of dominance as well as fertility. Some studies indicate that linear alkanes are important cuticular compounds for intraspecific recognition and discrimination. The aim of this study was to evaluate the relationship between reproductive physiologic condition and the linear alkanes present in the cuticle of females of P. versicolor in aggregates employing Gas Chromatography with Flame Ionization Detector (GC-FID). Females from distinct aggregates were differentiated by the chemical composition of their cuticle. In each aggregate, there was difference in cuticular chemical composition between females with different ovarian development degrees, allowing the distinction between inseminated and non-inseminated females.
