Sheet pipe is a type of perforated pipe used for drainage designed initially for drainage but has the potential for sub-surface irrigation. The objectives of this study were to experiment and observe the performance of the sub-surface irrigation control system with sheet pipe. This investigation covered the observation of water table control and its effect on soil moisture. The detailed process of water flow during the setting of the water table was numerically modeled in 2 dimensions to observe the distribution of soil moisture, soil pressure, and flux. The results showed that the system successfully controlled the water table at the desired level in the experiment. The developed two-dimensional numerical simulation showed the distribution of soil moisture in the model center as a response to the water table increase, represented by the variable head. The soil wetting advances toward soil surface driven by the water table, which was increased gradually and reached saturation at the height of water table setpoint.

Abstract The effects of adjuvants for increasing the immunogenicity of influenza vaccines are well known. However, the effect of adjuvants on increasing the breadth of cross-reactivity is less well understood. In this study we have performed a systematic screen of different toll-like receptor (TLR) agonists, with and without a squalene-in-water emulsion on the immunogenicity of a recombinant trimerized hemagglutinin (HA) vaccine in mice after single-dose administration. Antibody (Ab) cross-reactivity for other variants within and outside the immunizing subtype (homosubtypic and heterosubtypic cross-reactivity, respectively) was assessed using a protein microarray approach. Of all the formulations tested, a combination of CpG, MPLA and AddaVAX (termed “IVAX-1”) yielded the greatest breadth and magnitude of Ab responses, particularly against the HA1 region (which includes the variable head domain) of HA. Antigen-specific plasma cell labeling experiments show the components of IVAX-1 are synergistic. This adjuvant preferentially stimulates CD4 T cells to produce Th1>Th2 type (IgG2c>IgG1) antibodies and cytokine responses. Moreover, IVAX-1 induces identical homo- and heterosubtypic IgG and IgA cross-reactivity profiles when administered intranasally. Consistent with these observations, a single-cell transcriptomics analysis demonstrated significant increases in expression of IgG1, IgG2b and IgG2c genes of B cells in H5/IVAX-1 immunized mice relative to naïve mice, as well as significant increases in expression of the IFNg gene of both CD4 and CD8 T cells. These data support the use of adjuvants for enhancing the breath and durability of antibody responses of influenza virus vaccines.

Crossflow turbines represent a valuable choice for energy recovery in aqueducts, due to their constructive simplicity and good efficiency under variable head jump conditions. Several experimental and numerical studies concerning the optimal design of crossflow hydraulic turbines have already been proposed, but all of them assume that structural safety is fully compatible with the sought after geometry. We show first, with reference to a specific study case, that the geometry of the most efficient impeller would lead shortly, using blades with a traditional circular profile made with standard material, to their mechanical failure. A methodology for fully coupled fluid dynamic and mechanical optimization of the blade cross-section is then proposed. The methodology assumes a linear variation of the curvature of the blade external surface, along with an iterative use of two-dimensional (2D) computational fluid dynamic (CFD) and 3D structural finite element method (FEM) simulations. The proposed methodology was applied to the design of a power recovery system (PRS) turbine already installed in an operating water transport network and was finally validated with a fully 3D CFD simulation coupled with a 3D FEM structural analysis of the entire impeller.

Influenza viruses remain a constant burden in humans, causing millions of infections and hundreds of thousands of deaths each year. Current influenza virus vaccine modalities primarily induce antibodies directed towards the highly variable head domain of the hemagglutinin protein on the virus surface. Such antibodies are often strain-specific, meaning limited cross-protection against divergent influenza viruses is induced, resulting in poor vaccine efficacy. To attempt to counteract this, yearly influenza vaccination with updated formulations containing antigens from more recently circulating viruses is required. This is an expensive and time-consuming exercise, and the constant arms race between host immunity and virus evolution presents an ongoing challenge for effective vaccine development. Furthermore, there exists the constant pandemic threat of highly pathogenic avian influenza viruses with high fatality rates (~30–50%) or the emergence of new, pathogenic reassortants. Current vaccines would likely offer little to no protection from such viruses in the event of an epidemic or pandemic. This highlights the urgent need for improved influenza virus vaccines capable of providing long-lasting, robust protection from both seasonal influenza virus infections as well as potential pandemic threats. In this narrative review, we examine the next generation of influenza virus vaccines for human use and the steps being taken to achieve universal protection.
