Oxyria sinensis adopts a tolerant strategy as a metal excluder to survive toxic metal concentrations. Biodiversity and the endophytic fungal community colonizing the O. sinensis roots were assessed from a mining area (MA) and a neighboring non-mining area (nMA) in southwestern China. All O. sinensis roots formed fully developed dark septate endophytes (DSEs) and arbuscular mycorrhizal fungi (AMF). Total DSE colonization was higher for the MA versus nMA, in contrast to the total AMF colonization in the two sites. The DSE colonization was higher than AMF colonization regardless of the site. Pure-culture data showed that the fungi closely related to Exophiala, Cadophora and Phialophora dominantly colonized the O. sinensis roots. A total of 450 operational taxonomic units (OTUs) were identified showing the presence of a distinct fungal community in MA and nMA, which was shaped by soil physiochemical properties, including soil Zn concentrations and organic matter. We found that O. sinensis accumulates and adapts efficiently to local endophytic fungi to achieve the expansion of its community, including the spontaneously reclaimed DSE. This property may be targeted to achieve its colonization with a pioneer plant for phytoremediation in the restoration of a vegetation cover in a metal-contaminated area.

Millions of people have inadequate Se and Zn intakes, but agronomic biofortification could prevent this. This study evaluated the effect of the combined Zn and Se biofortification on the quality parameters of grain, and on the composition of minerals (Zn, Se, Mg, Ca and Fe) and their availability in bread-making wheat (Triticum aestivum L.) products, white flour, wholemeal bread and white bread were evaluated. The studied treatments were soil Zn (no Zn, and 50 kg Zn ha−1) and foliar applications (0, 10 g Se ha−1, 8 kg Zn ha−1, and 10 g Se ha−1 + 8 kg Zn ha−1) and were tested in a two-year field experiment (2017–2018, 2018–2019). The foliar combined biofortification increased the concentration of both minerals in white flour, wholemeal bread and white bread by about 33%, 24% and 51%, respectively for Zn, and 3.3-fold, 3.4-fold and 2.7-fold for Se, showing a synergistic effect on Se concentration with the Se and Zn combination. While the loss of Zn and Se during the milling process was41% and 18%, respectively, baking caused a loss of 15% and 19%, respectively, for wholemeal bread, and up to 61% and 29% for Zn and Se for white bread. Hence, although the consumption of wholemeal bread instead of white bread may enhance Zn and Se intake more than biofortification, until consumption habits change, the biofortification of wheat can help to mitigate inadequate Zn and Se intakes in the general population.

Abstract This study compared N HNO3 to other methods to determine plant available heavy metals in heavy-metal polluted soils. Soil samples were obtained from an experimental field treated with industrial waste after 22 years of the amendment and employed to conduct the comparative and correlation study. Soil samples were analyzed for Cu using various methods, planted in a glass house with several plants, and analyzed for soil and plant Cu and Zn. The relative strength of the chemical extractants followed the order of N HNO3 ≈ N HCl > Buffered DTPA ≈ Unbuffered DTPA > M CaCl2 ≈ N NH4OAc pH 7. A high correlation was observed for soil extracted Cu by M CaCl2 or N NH4OAc pH 7 or N HCl vs. Buffered DTPA and N HNO3 or N NH4OAc pH 7 vs. N HCl. High correlations of plant and soil Cu extracted by N HNO3 were shown by caisim, water spinach, land spinach, and corn, while plant and soil Zn were shown by caisim, water spinach, land spinach, and lettuce.

Abstract Background Dietary zinc (Zn) deficiency is widespread in sub-Saharan Africa (SSA) with adverse impacts on human health. Agronomic biofortification with Zn fertilizers and improved soil fertility management, using mineral and organic nutrient resources, has previously been shown to increase Zn concentration of staple grain crops, including maize. Here, we show the potential of different soil fertility management options on maize crops to reduce dietary Zn deficiency in Zimbabwe using secondary data from a set of surveys and field experiments. Methods An ex-ante approach was used, informed by published evidence from studies in three contrasting smallholder production systems in Zimbabwe. To estimate current Zn deficiency in Zimbabwe, data on dietary Zn supply from non-maize sources from the Global Expanded Nutrient Supply (GENuS) data set were linked to maize grain Zn composition observed under typical current soil fertility management scenarios. Results A baseline dietary Zn deficiency prevalence of 68% was estimated from a reference maize grain Zn composition value of 16.6 mg kg−1 and an estimated dietary Zn intake of 9.3 mg capita−1 day−1 from all food sources. The potential health benefits of reducing Zn deficiency using different soil fertility management scenarios were quantified within a Disability Adjusted Life Years (DALYs) framework. A scenario using optimal mineral NPK fertilizers and locally available organic nutrient resources (i.e. cattle manure and woodland leaf litter), but without additional soil Zn fertilizer applications, is estimated to increase maize grain Zn concentration to 19.3 mg kg−1. This would reduce the estimated prevalence of dietary Zn deficiency to 55%, potentially saving 2238 DALYs year−1. Universal adoption of optimal fertilizers, to include soil Zn applications and locally available organic leaf litter, is estimated to increase maize grain Zn concentration to 32.4 mg kg−1 and reduce dietary Zn deficiency to 16.7%, potentially saving 9119 DALYs year−1. Potential monetized yield gains from adopting improved soil fertility management range from 49- to 158-fold larger than the potential reduction in DALYs, if the latter are monetized using standard methods. Conclusion Farmers should be incentivized to adopt improved soil fertility management to improve both crop yield and quality.

Purple rice is recognized as a staple food for humans and as a source of anthocyanins and micronutrients such as zinc (Zn). This study examined how nitrogen (N) and Zn fertilizers affected grain yield and grain N, Zn, and anthocyanin concentration among purple rice genotypes. Six purple rice genotypes (PIZ, KAK, KS, KH-CMU, KDK, and HN) were grown under two levels of N, the optimum N60 (60 kg/ha) and high N180 (180 kg/ha) rates, along with three Zn application methods (no Zn application (Zn0), soil Zn application (ZnS; 50 kg ZnSO4/ha), and foliar Zn spray (ZnF; 0.5% ZnSO4 at the rate of 900 L/ha three times at heading, flowering, and early milk stages). Grain yield of the five purple rice landraces increased by 21–40% when increasing N from N60 to N180, although no response was found with HN. The higher N rate increased grain N concentration by 10–50% among the genotypes, while anthocyanin concentration increased by 100–110% in KAK and KS, and grain Zn was increased in KS. Applying ZnS increased grain yield by 16–94% but decreased anthocyanin and N concentrations compared to the control Zn0. Applying ZnF effectively increased grain Zn concentration by 40–140% in the genotypes without adversely impacting grain anthocyanin or N concentration. This study demonstrated that the appropriate management of N and Zn fertilizers for specific purple rice genotypes would be one way to increase productivity and grain N, Zn, and anthocyanin concentration.

Abstract Purpose Green manuring can increase the plant available fraction of zinc (Zn) in soil, making it a potential approach to increase wheat Zn concentrations and fight human Zn deficiency. We tested whether green manure increases the ability of both the native soil bacteria and inoculated Zn solubilizing bacteria (ZSB) to mobilize Zn. Methods Wheat was grown in a pot experiment with the following three factors (with or without); (i) clover addition; (ii) soil x-ray irradiation (i.e. elimination of the whole soil biota followed by re-inoculation with the native soil bacteria); and (iii) ZSB inoculation. The incorporation of clover in both the irradiated and the ZSB treatments allowed us to test green manure effects on the mobilization of Zn by indigenous soil bacteria as well as by inoculated strains. Results Inoculation with ZSB did neither increase soil Zn availability nor wheat Zn uptake. The highest soil Zn availabilities were found when clover was incorporated, particularly in the irradiated soils (containing only soil bacteria). This was partly associated with the stimulation of bacterial activity during the decomposition of the incorporated green manure. Conclusion The results support that the activity of soil bacteria is intimately involved in the mobilization of Zn following the incorporation of green manure.

An effective solution to global human zinc (Zn) deficiency is Zn biofortification of staple food crops, which has been hindered by the low available Zn in calcareous soils worldwide. Many culturable soil microbes have been reported to increase Zn availability in the laboratory, while the status of these microbes in fields and whether there are unculturable Zn-mobilizing microbes remain unexplored. Here, we use the culture-independent metagenomic sequencing to investigate the rhizosphere microbiome of three high-Zn (HZn) and three low-Zn (LZn) wheat cultivars in a field experiment with calcareous soils. The average grain Zn concentration of HZn was higher than the Zn biofortification target 40 mg kg–1, while that of LZn was lower than 40 mg kg–1. Metagenomic sequencing and analysis showed large microbiome difference between wheat rhizosphere and bulk soil but small difference between HZn and LZn. Most of the rhizosphere-enriched microbes in HZn and LZn were in common, including many of the previously reported soil Zn-mobilizing microbes. Notably, 30 of the 32 rhizosphere-enriched species exhibiting different abundances between HZn and LZn possess the functional genes involved in soil Zn mobilization, especially the synthesis and exudation of organic acids and siderophores. Most of the abundant potential Zn-mobilizing species were positively correlated with grain Zn concentration and formed a module with strong interspecies relations in the co-occurrence network of abundant rhizosphere-enriched microbes. The potential Zn-mobilizing species, especially Massilia and Pseudomonas, may contribute to the cultivars’ variation in grain Zn concentration, and they deserve further investigation in future studies on Zn biofortification.

Millions of people worldwide have an inadequate intake of selenium (Se) and zinc (Zn), and agronomic biofortification may minimise these problems. To evaluate the efficacy of combined foliar Se and Zn fertilisation in bread making wheat (Triticum aestivum L.), a two-year field experiment was established in southern Spain under semi-arid Mediterranean conditions, by following a split-split-plot design. The study year (2017/2018, 2018/2019) was considered as the main-plot factor, soil Zn application (50 kg Zn ha−1, nor Zn) as a subplot factor and foliar application (nor Se, 10 g Se ha−1, 8 kg Zn ha−1, 10 g Se ha−1 + 8 kg Zn ha−1) as a sub-subplot factor. The best treatment to increase both Zn and Se concentration in both straw, 12.3- and 2.7-fold respectively, and grain, 1.3- and 4.3-fold respectively, was the combined foliar application of Zn and Se. This combined Zn and Se application also increased on average the yield of grain, main product of this crop, by almost 7%. Therefore, bread-making wheat seems to be a very suitable crop to be used in biofortification programs with Zn and Se to alleviate their deficiency in both, people when using its grain and livestock when using its straw.

To better understand the source–sink flow and its relationships with zinc (Zn) and other nutrients in wheat (Triticum aestivum L.) plants for biofortification and improving grain nutritional quality, the effects of reducing the photoassimilate source (through the flag leaf removal and spike shading) or sink (through the removal of all spikelets from one side of the spike, i.e., 50% spikelets removal) in the field of the accumulation of Zn and other nutrients in grains of two wheat cultivars (Jimai 22 and Jimai 44) were investigated at two soil Zn application levels. The kernel number per spike (KNPS), single panicle weight (SPW), thousand kernel weight (TKW), total grain weight (TGW) sampled, concentrations and yields of various nutrient elements including Zn, iron (Fe), manganese (Mn), copper (Cu), nitrogen (N), phosphorus (P), potassium (K), calcium (Ca) and magnesium (Mg), phytate phosphorus (phytate-P), phytic acid (PA) and phytohormones (ABA: abscisic acid, and the ethylene precursor ACC: 1-aminocylopropane-1-carboxylic acid), and carbon/N ratios were determined. Soil Zn application significantly increased the concentrations of grain Zn, N and K. Cultivars showing higher grain yields had lower grain protein and micronutrient nutritional quality. SPW, KNPS, TKW (with the exception of TKW in the removal of half of the spikelets), TGW, and nutrient yields in wheat grains were most severely reduced by half spikelet removal, secondly by spike shading, and slightly by flag leaf removal. Grain concentrations of Zn, N and Mg consistently showed negative correlations with SPW, KNPS and TGW, but positive correlations with TKW. There were general positive correlations among grain concentrations of Zn, Fe, Mn, Cu, N and Mg, and the bioavailability of Zn and Fe (estimated by molar ratios of PA/Zn, PA/Fe, PA × Ca/Zn, or PA × Ca/Fe). Although Zn and Fe concentrations were increased and Ca was decreased in treatments of half spikelet removal and spike shading, the treatments simultaneously increased PA and limited the increase in bioavailability of Zn and Fe. In general, different nutrient elements interact with each other and are affected to different degrees by source–sink manipulation. Elevated endogenous ABA levels and ABA/ACC ratios were associated with increased TKW and grain-filling of Zn, Mn, Ca and Mg, and inhibited K in wheat grains. However, the effects of ACC were diametrically opposite. These results provide a basis for wheat grain biofortification to alleviate human malnutrition.
