Abstract The ingestion of excess lipids often produces the accumulation of liver fat. The modulation of diet energy partition affects this process and other metabolic responses, and oestrogens and androgens are implied in this process. Ten-week-old male and female rats were fed with either standard rat chow (SD), SD enriched with coconut oil (high-fat diet, HF), SD enriched with protein (high-protein diet, HP) or a “cafeteria” diet (CAF) for one month. HF and CAF diets provided the same lipid-derived percentage of energy (40%), HP diet protein-energy derived was twice (40%) that of the SD. Animals were sacrificed under anaesthesia and samples of blood and liver were obtained. Hepatic lipid content showed sex-related differences: triacylglycerol accumulation tended to increase in HF and CAF fed males. Cholesterol content was higher only in the CAF males. Plasma oestradiol in HF and HP males was higher than in CAF. Circulating cholesterol inversely correlated with plasma oestradiol, which levels were proportional to lactate. These changes agreed with the differences in the expression of some enzymes related with lipid and energy metabolism, such as fatty acid synthetase or phosphoglycolate phosphatase. Oestrogen protective effects extend to males with ‘normal’ diets, i.e. not unbalanced by either lipid or protein, but this protection was not enough against the CAF diet. Oestradiol seems to actively modulate the liver core of 2C-3C partition of energy substrates, regulating cholesterol deposition and lactate production.

Aldabra giant tortoises (Aldabrachelys gigantea) are currently listed as Vulnerable on the IUCN Red List of Threatened Species. However, negative impacts of sea-level rise are predicted to result in an overall population decline of 40-65 % over the next 100 years, rendering the species Endangered. Captive propagation is an important tool for in- and ex-situ species conservation, but breeding success outside the tortoises’ native island habitats has been very limited. Until now, the reproductive cycle of Aldabra tortoises has only been described in anatomic and behavioural studies. During a one-year period, plasma of four female and four male wild tortoises on Aldabra Atoll were examined monthly for levels of gonadal steroid hormones (oestradiol, testosterone, progesterone). Plasma oestradiol and testosterone values as well as meteorological data of the sampling period corresponded to previously published reports on seasonal changes in anatomy, behaviour and climate on the Aldabra Atoll. Seasonal changes in plasma testosterone were evident in males, with high values from January through April, reflecting previously described testicular growth and breeding season, followed by a nadir in August and September. In females, plasma oestradiol levels displayed seasonal changes, coinciding with reported ovarian growth from January to May. The obtained data provide prerequisite knowledge for endocrinological monitoring of reproductive processes and management of breeding programs, both ex-situ as in-situ, to establish reserve- and rewilded populations.

Paracetamol (PAR) is one of the most commonly used drugs by pregnant women because it is considered safe for the mother and fetus. However, PAR is transferred into breast milk and crosses the blood–placental barrier, being present in the progeny during important stages of development. Intrauterine exposure to PAR may decrease the anogenital distance and follicle reserve in female rodent offspring. Therefore, the aim of the present study was to evaluate whether maternal PAR treatment altered the reproductive behaviour of dams and the sexual development of female rat offspring. Pregnant Wistar rats were gavaged daily with 350mg kg−1 day−1 PAR or water during gestation (from Gestation Day (GD) 6 until delivery) or during gestation and lactation (from GD6 until weaning). Maternal PAR treatment had maternal effects (increased grooming behaviour), and resulted in impaired sexual behaviour, decreased follicle reserve and increased plasma oestradiol concentrations in female offspring.

Objective This study aimed to investigate the association between low plasma Platelet-derived growth factor-BB (PDGF-BB) levels and oestradiol in Postmenopausal osteoporosis (PMOP). Methods This prospective study measured plasma PDGF-BB and oestradiol levels in outpatients who were admitted to our hospital. Participants were screened and then allocated to three groups: normal young women, postmenopausal control, and PMOP. Additionally, Sprague–Dawley rats underwent either sham surgery or bilateral ovariectomy (OVX), and were divided into the following groups: sham, OVX, OVX + oestradiol, and OVX + PDGF-BB. Plasma oestradiol and PDGF-BB levels were measured using commercially available ELISA kits. Results A total of 121 participants, including 69 normal young women, 28 patients with primary PMOP, and 24 age-matched postmenopausal women were enrolled. Plasma oestradiol and PDGF-BB levels were lower in postmenopausal women, especially in PMOP ( P < 0.01). Pearson correlations analysis showed that PDGF-BB levels were positively correlated with oestradiol levels and inversely correlated with age ( P < 0.01). The OVX rat model showed that oestradiol replacement increased plasma PDGF-BB levels, while PDGF-BB systematic treatment had no effect on plasma oestradiol levels. Conclusions Plasma PDGF-BB levels are maintained by oestrogen in normal young women and play a major role in PMOP.

Objective To investigate the role of the medial preoptic area (MPOA) of the hypothalamus in electroacupuncture (EA) treatment of reproductive dysfunction in ovariectomised (OVX) rats and to examine the underlying mechanisms of action. Methods 62 female Sprague-Dawley rats (200–220 g) underwent bilateral ovariectomy (OVX). Three weeks later, 32 rats assigned to experiment 1 were randomly divided into four groups: two groups underwent MPOA lesioning (n=16) followed by EA at CV4, CV3, SP6 and bilateral Zigong (30 min per day) for 3 days (OVX+MPOA+EA group, n=8) or no treatment (OVX+MPOA group, n=8); and two groups underwent sham (S) surgery (n=16) followed by EA (OVX+SMPOA+EA group, n=8) or no treatment (OVX+SMPOA group, n=8). Plasma oestradiol and luteinising hormone (LH) were measured by radioimmunoassay and mature vaginal epithelial cells stained and quantified post-treatment. 30 rats assigned to experiment 2 were randomised to receive EA (OVX+EA group, n=15) or remain untreated (OVX group, n=15), following which push-pull perfusion in combination with high pressure liquid chromatography/electrochemical (HPLC-EC) detection was used to measure neurotransmitter release in the MPOA. Results EA significantly increased the percentage of mature vaginal epithelial cells and plasma oestradiol levels in sham MPOA-lesioned rats compared with MPOA-lesioned OVX rats (p<0.05). In sham MPOA-lesioned OVX rats, EA significantly decreased plasma LH levels compared with no treatment (p<0.01). HPLC-EC results showed that γ-aminobutyric acid (GABA) and β-endorphin levels in the MPAO were significantly increased in OVX rats following EA (p<0.01). Conclusions The present study suggests that the MPOA plays a crucial role in EA treatment of abnormal reproductive function in OVX rats.

This study investigated the efficacy of a synthetic progestogen, levonorgestrel (LNG), to control koala ovarian activity for the purposes of oestrous synchronisation. Captive koalas were administered either saline control or a 70-mg LNG implant on Day 2 of oestrus. Urogenital cytology, oestrous behaviour and plasma oestradiol-17β and LH concentrations were monitored over a 6-week period. After LNG implant removal females were monitored to determine if the return to oestrus was synchronised. LNG-treated koalas immediately ceased displaying oestrous behaviour, showed no evidence of cornified epithelial cells in smears of urogenital cytology and exhibited low plasma oestradiol-17β concentrations throughout the implantation period. In contrast, oestradiol-17β levels in control koalas showed evidence of continued cyclic activity associated with behavioural oestrus and increased cornified epithelial cells in urogenital smears on Days 33 to 35 after saline injection. After implant removal, LNG-treated koalas exhibited oestrus at 13, 14, 17 and 30 days after implant removal. Plasma LH concentrations varied throughout the study period with no significant time (P = 0.49) or treatment (P = 0.13) effect. Overall results from this study suggest that LNG implants in koalas can inhibit oestrous behaviour and reduce circulating oestradiol-17β levels before oestrus, most likely by preventing development of the pre-ovulatory follicle. However, there was no evidence of LH suppression by the LNG implants. Removal of LNG implants resulted in the synchronous return to oestrus in three of the four treated koalas. Further studies on a larger population are required to validate these findings.

The study was conducted to develop a simulated heat stress model for sheep depicting natural environmental conditions and to observe its impact on growth, reproductive performance, physiological adaptability, blood metabolites and endocrine responses in Malpura ewes. The experiment was conducted on 16 adult Malpura ewes for a period of 35 days. The animals were randomly divided into two groups of eight animals each (GI – control, GII – heat stress). The GII ewes were exposed to different temperature at different hours of the day i.e. 38°C at 1000–1100 hours; 40°C at 1100–1200 hours; 42°C at 1200–1300 hours; 43°C at 1300–1400 hours; 44°C at 1400–1500 hours and 42°C at 1500–1600 hours in a climatic chamber. Feed intake (P < 0.01), bodyweight (P < 0.05) and body condition score (P < 0.01) reduced significantly in GII as compared with GI ewes while water intake increased significantly (P < 0.01). All physiological responses also showed significant variation between the groups for treatment. In addition, all endocrine parameters showed highly significant (P < 0.01) variation for the treatment. Plasma cortisol increased significantly (P < 0.01) while T3 (P < 0.01) and T4 (P < 0.01) decreased significantly in GII as compared with GI ewes. Further, plasma oestradiol decreased significantly (P < 0.01) while plasma progesterone increased significantly (P < 0.01) in GII as compared with GI. Among the biochemical parameters, hemoglobin, packed cell volume, plasma glucose, total cholesterol, urea and blood urea nitrogen differed significantly between the groups. In addition, oestrus duration also significantly (P < 0.05) reduced in GII as compared with GI. Based on the results obtained from the study it can be concluded that simulated heat stress influenced the adaptive behaviour of sheep in terms of changes in physiological, blood biochemical and endocrine responses. Ultimately, their growth and reproductive performance are compromised during the process of adaptation to the simulated heat stress.

The objective of this study was to determine the effects of vehicle and route of administration of letrozole on ovarian function in sexually mature beef heifers. On Day 3 (Day 0 = ovulation), heifers were assigned randomly to four treatment groups and given 1 mg kg–1 letrozole intravenously (iv, n = 10) or intramuscularly (im, n = 10) or given a placebo iv (control iv, n = 5) or im (control im, n = 5). The interwave interval was longer in heifers treated with letrozole im than in im and iv controls (11.7 ± 0.30 vs 9.5 ± 0.50 and 10 ± 0.43, respectively; P < 0.05). Corpus luteum diameter profiles and plasma progesterone concentrations were greater (P < 0.03 and P < 0.05, respectively) in heifers treated with letrozole im compared with control im. Plasma oestradiol concentrations were lower in both letrozole-treated groups compared with controls (P ≤ 0.03). Plasma LH concentrations tended to be elevated at the time of wave emergence in heifers treated with letrozole im compared with other groups (group-by-day interaction, P = 0.06) and plasma FSH concentrations tended to be greater (P < 0.09) in heifers treated with letrozole by either route compared with a single control group. We conclude that intramuscular administration of letrozole in oil is a feasible route and vehicle for the development of a letrozole-based treatment protocol for herd synchronisation in cattle.
