Pickering high internal phase emulsions (HIPEs) are gel-like concentrated emulsions that have the potential to be an alternative to partially hydrogenated oil (PHO). In this study, egg yolk granules (EYGs), natural complexes of protein and lipid isolated from egg yolk, were used as an emulsifier to prepare Pickering HIPEs. Gel-like HIPEs with an oil phase volume fraction of 85% and with an emulsifier concentration of only 0.5% could be prepared by using EYGs as an emulsifier. The EYGs were able to form stable HIPEs at NaCl ionic strengths over 0.2 M and at pH over 5.0 with NaCl ionic strength of 0.3 M. The EYGs, which could stabilize HIPEs, were easily to adsorb and cover the oil-water interface to form emulsion droplets with small particle size. In addition, interacting EYGs in the aqueous phase formed a continuous network structure, and the oil droplets packed closely, exhibiting high elasticity and shear thinning behavior. Furthermore, the formed HIPEs had suitable storage stability with no significant changes in appearance and microstructure after storage for 60 days. This work can transform traditional oils from liquid-like to solid-like by using EYGs to enrich food processing diversity and improve the storage stability of oils while reducing the intake of PHO and providing a healthier diet for consumers.

AbstractThe silver pomfret (Pampus argenteus) is one of the most commercially important marine fish species in the Bay of Bengal, Indian Ocean. However, detailed information on the reproductive biology of silver pomfret is limited for the Bay of Bengal, Bangladesh. To understand their reproductive biology, especially the gonadal developmental cycle and spawning season, a total of 373 individuals were collected monthly from February 2016 to January 2018 and examined. The total length (TL) and weight (TW) of P. argenteus ranged from 18.82–35.73 cm and 89.26–617.60 g, respectively. The highest gonadosomatic index (GSI) values were observed from April–June with a second (smaller) peak in October. The highest oocyte diameter was observed in May (630.50 ± 96.70 μm), when the fish reached full maturity, and the lowest was observed in July (35.10 ± 7.12 μm), when new eggs began their development for the next spawning season. The maximum number of eggs in the ovary were found in the yolk granule stage in May with a number of post-ovulatory follicles, which indicated that the ovaries developed to a peak leading up to the spawning season. A number of spermatids were also seen in males in April–June along with females. From these detailed observations, it may be concluded that P. argenteus shows two spawning peaks, in May–June and October. This observation will be helpful for the artificial breeding, sustainable management and conservation of this species in the study area.

The breeding season of fishes plays an important role in fish domestication and conservation of fish diversity. Freshwater goby, Glossogobius giuris is an important food fish of Bangladesh. Therefore, a study was performed to determine the breeding biology through assessing the gonadosomatic index (IG), fecundity and histology of gonad of G. giuris. A total of 1371 females and 1051 male fishes were collected from the Belai beel of Gazipur district. Significantly (P<0.05) higher values of mean IG were observed during March and from June to August for both females and males. The highest mean fecundity was recorded in August and the lowest was recorded in May. The smallest mean oocyte length was 0.037±0.005 mm in May and the largest was 0.071±0.001 mm in August. The early and late perinucleolar stages of oocytes were observed from January to March. The yolk vesicle stage was observed during April to May and yolk granule stage was observed during March and from June to August. Egg yolk granule phosvitin (45 kDa) is a phosphoprotein known for its emulsifying properties. Recently, high hydrostatic pressure (HHP) treatment of granule induced the transfer of phosvitin to the soluble plasma fraction. This project evaluated the performance of the ultrafiltration (UF) used to concentrate phosvitin from the plasma fraction to produce a natural emulsifier. Phosvitin was characterized in plasma from a pressure-treated granule (1.73 ± 0.07% w/w) and in its UF retentate (26.00 ± 4.12% w/w). The emulsifying properties of both retentates were evaluated. The emulsion prepared with phosvitin-enriched retentate was more resistant to flocculation and creaming. Confocal laser scanning microscopy showed a network of aggregated protein similar to a gel, which encapsulated oil droplets in emulsions made with UF-retentate of plasma from pressure-treated granule. However, although sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) showed that β-phosvitin is recovered in the cream, it is difficult to attribute the improved emulsifying properties of the UF-retentate of plasma from pressure-treated granules only to phosvitin.

Fertilization occurs during female meiosis in most animals, which raises the question of what prevents the sperm DNA from interacting with the meiotic spindle. In this study, we find that Caenorhabditis elegans sperm DNA stays in a fixed position at the opposite end of the embryo from the meiotic spindle while yolk granules are transported throughout the embryo by kinesin-1. In the absence of F-actin, the sperm DNA, centrioles, and organelles were transported as a unit with the yolk granules, resulting in sperm DNA within 2 µm of the meiotic spindle. F-actin imaging revealed a cytoplasmic meshwork that might restrict transport in a size-dependent manner. However, increasing yolk granule size did not slow their velocity, and the F-actin moved with the yolk granules. Instead, sperm contents connect to the cortical F-actin to prevent interaction with the meiotic spindle.

The spiny eel, Mastacembelus armatus is one of the most common and economically important inland teleost species of Asia having high market and nutritive values. The demand for the fish is almost always exceeds its supply. Histological analysis of developmental stages of both oocytes and sperrnatocytes maturation were undertaken and dimensions of ovarian oocytes were observed to evaluate the annual reproductive condition of M. armatus during August 2012 to July 2013. Monthly sampling of adult individuals of M. armatus was done from haor region of Kishoreganj district, Bangladesh. Both the gonads were paired, elongated and of unequal in length. The colour of ovaries varied from reddish brown to light yellowish; whereas, testes varied from whitish to creamy or dull white. From the histological study, five stages of oocyte development (oogonia, chromatin nucleolar stage oocyte, perinucleolar stage oocytes, yolk vesicle oocyte, and yolk granule stage) were observed upon their changes in nucleus. Female M. armatus having immature stages of oocytes (early and late perinucleolar stage oocytes) were mostly available in the months of November and December. Yolk vesicle stage oocytes appeared during the months of January and February. Early yolk granule stage oocytes appeared during the months of March and April while late yolk granule stage oocytes were observed during the months of May and June. Oocytes and nuclei diameters were observed ranged from 12–1050 ?m and 10–212 ?m, respectively until ovulation. In male gonads, four stages of spermatogenesis (spermatogonia, spermatocytes, spermatids, and spermatozoa) were distinguished during the study period where spermatids were prominent in January to March and spermatozoa in April to July. From this study, the peak gonadal development of M. armatus is found in June and thus, it is clearly indicated that the fish breed during May to June.J. Bangladesh Agril.
