Several 6- and 7-alkoxy-2-oxo-2H-chromene-4-carbaldehydes were prepared from corresponding alkyl ethers of 6- and 7-hydroxy-4-methyl-2-oxo-2H-chromen-2-ones by oxidation using selenium dioxide. 6- and 7-Alkoxy-4-methyl-2H-chromenes were obtained with yields of 57-85%. Corresponding 4-carbaldehyde derivatives were prepared with yields of 41-67%. Thiosemicarbazones of these aldehydes with D-galactose moiety were synthesized by reaction of these aldehydes with N-(2,3,4,6-tetra-O-acetyl-β-Dgalactopyranosyl) thiosemicarbazide with yields of 62-74%. These thiosemicarbazones were screened for their antibacterial and antifungal activities in vitro against bacteria, such as Staphylococcus aureus, Escherichia coli, and fungi, such as Aspergillus niger, Candida albicans. Several compounds exhibited strong inhibitory activity with MIC values of 0.78- 1.56 μM, including 8a (against S. aureus, E. coli, and C. albicans), 8d (against E. coli and A. niger), 9a (against S. aureus), and 9c (against S. aureus and C. albicans).

Objectif : L’objectif de ce travail était d’évaluer in vitro l’activité antimicrobienne des extraits de feuilles et tige de Jatropha multifida sur la croissance de Candida albicans, Escherichia coli et Staphylococcus aureus, puis d’évaluer in vivo la toxicité de cette plante. Méthodologie et résultats : Les méthodes de diffusion en milieu gélosé et de microdilution en milieu liquide ont été utilisées pour évaluer l’effet antimicrobien. Une étude en subaigüe était réalisée afin d’explorer les effets toxiques de l’extrait aqueux des feuilles. Les résultats des tests antimicrobiens montrent une activité des extraits de feuilles et tige de J. multifida sur la croissance des souches utilisées avec des diamètres de zones d’inhibition allant de 8 à 25 mm et des concentrations minimales inhibitrices (CMI) variant de 0,039 mg/mL à 1,25 mg/mL à l’exception des souches de E. coli qui sont résistantes aux extraits de la tige. L’administration en subaigüe de l’extrait aqueux des feuilles de J. multifida à la dose de 600 mg/kg entraîne une perte significative de poids chez les souris. Conclusion et applications des résultats : Les extraits aqueux, éthanolique et hydroéthanolique des feuilles et tige de J. multifida possèdent d’activité antimicrobienne et pourraient être utilisés dans le traitement des Candidoses à C. albicans et des infections à S. aureus. Mais l’essai de toxicité subaigüe montre que l’extrait aqueux de la plante serait toxique. Des études toxicologiques approfondies restent donc nécessaires sur ces extraits afin de mieux élucider leur inocuité. Conclusion and application of findings : The aqueous, ethanolic and hydroethanolic extracts of the leaves and stem of J. multifida have antimicrobial activity and could be used in the treatment of Candidiasis and bacterial infections due respectively to C. albicans and S. aureus. But the subacute toxicity test shows that the aqueous extract of the plant would be toxic. Extensive toxicological studies therefore remain necessary on these extracts in order to better elucidate their safety. ABSTRACT Candida albicans and Staphylococcus aureus are common causes of nosocomial infections with severe morbidity and mortality. Murine polymicrobial intra-abdominal infection (IAI) with C. albicans and S. aureus results in acute mortality dependent on the secreted cytolytic effector alpha-toxin. Here, we confirmed that alpha-toxin is elevated during polymicrobial growth compared to monomicrobial growth in vitro. Therefore, this study sought to unravel the mechanism by which C. albicans drives enhanced staphylococcal alpha-toxin production. Using a combination of functional and genetic approaches, we determined that an intact agr quorum sensing regulon is necessary for enhanced alpha-toxin production during coculture and that a secreted candidal factor likely is not implicated in elevating agr activation. As the agr system is pH sensitive, we observed that C. albicans raises the pH during polymicrobial growth and that this correlates with increased agr activity and alpha-toxin production. Modulation of the pH could predictably attenuate or activate agr activity during coculture. By using a C. albicans mutant deficient in alkalinization (stp2Δ/Δ), we confirmed that modulation of the extracellular pH by C. albicans can drive agr expression and toxin production. Additionally, the use of various Candida species (C. glabrata, C. dubliniensis, C. tropicalis, C. parapsilosis, and C. krusei) demonstrated that those capable of raising the extracellular pH correlated with elevated agr activity and alpha-toxin production during coculture. Overall, we demonstrate that alkalinization of the extracellular pH by the Candida species leads to sustained activation of the staphylococcal agr system. IMPORTANCE Candida albicans and Staphylococcus aureus are commonly coisolated from central venous catheters and deep-seated infections, including intra-abdominal sepsis. Thus, they represent a significant cause of nosocomial morbidity and mortality. Yet how these organisms behave in the context of polymicrobial growth remains poorly understood. In this work, we set out to determine the mechanism by which activation of the staphylococcal agr quorum sensing system and production of its major virulence effector alpha-toxin is enhanced during coculture with C. albicans. Surprisingly, we likely ruled out that a secreted candidal factor drives this process. Instead, we demonstrated that alkalinization of the extracellular milieu by C. albicans and other Candida species correlated with elevated agr activity. Thus, we propose a mechanism where modulation of the extracellular pH by fungal opportunists can indirectly alter virulence of a bacterial pathogen. Uncovering molecular events that drive interkingdom pathogenicity mechanisms may enhance surveillance and treatment for devastating polymicrobial infections.

In recent years polyurethane microstructures (PM) have gained increasing attention in the pharmaceutical field due to the importance of their practical application. Since finding that such a formulation with genistein could improve its applications, we have conducted a preliminary study regarding the in vitro antiproliferative (MCF7, MDA-MB-231 and T47D) and antimicrobial ( Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Salmonella enteritidis (D), Bacillus subtilis, B. cereus, and Candida albicans) activity in order to test whether polyurethane micro structuresre present a good option for further modulation of genistein's bioavailability. It was concluded that the polyurethane micro structures are a bad in vitro partner for the isoflavone genistein.

In experiments with mixed cultures of Staphylococcus aureus and Candida albicans both in the absence and in the presence of 5-fluorocytosine (5-FC), we have observed that (1) there is an inhibition of S. aureus growth in mixed cultures with C. albicans in media supplemented with 1 μg/mL of 5-FC and that 5-FC has no effect on staphylococci in pure cultures; (2) this inhibition occurred with clinically isolated and laboratory strains and could be reversed by specific metabolites; (3) Staphylococcus aureus was inhibited by filtrates of C. albicans cultures treated with 5-FC and this seemed to be favored by some C. albicans filterable product which can affect the cell wall and the permeability of the staphylococcal cells since they become sensitive to 5-FC; (4) nine other commonly used antimicrobials showed an increased inhibitory activity against S. aureus in mixed cultures with C. albicans; and (5) there is a decrease in the number of precipitating antigens of S. aureus and of the activity of alpha toxin when this species was grown with both C. albicans and 5-FC. Our results indicate that the susceptibility of some species to antimicrobials could be significantly modified in the presence of other species. One cannot exclude that a similar phenomenon could happen in hosts under treatment with antibiotics against infection.

ABSTRACTWe here report on thein vitroactivity of toremifene to inhibit biofilm formation of different fungal and bacterial pathogens, includingCandida albicans,Candida glabrata,Candida dubliniensis,Candida krusei,Pseudomonas aeruginosa,Staphylococcus aureus, andStaphylococcus epidermidis. We validated thein vivoefficacy of orally administered toremifene againstC. albicans and S. aureusbiofilm formation in a rat subcutaneous catheter model. Combined, our results demonstrate the potential of toremifene as a broad-spectrum oral antibiofilm compound.

Indonesian cinnamon (Cinnamomum burmannii) is a native plant of Indonesia that has a lot of potential. The most consumed part is the bark. This study aims to examine the antimicrobial effect of Cinnamomum burmannii bark extract on various types of pathogenic microbes, namely Staphylococcus aureus, Enterobacter spp., Pseudomonas aeruginosa, and Candida albicans. An experimental study using a water extract (infusion) of Cinnamomum burmannii bark and a microbial test obtained from the Faculty of Pharmacy, Widya Mandala Catholic University, Surabaya. The antimicrobial effect test was carried out by the microdilution method in 96-well-microplate to determine the Minimum Inhibitory Level (MIC) and implantation on solid media to determine the Minimum Kill Rate (KBM). The MIC and KBM against Staphylococcus aureus were 625-1,250 ppm and 1,250-2,500 ppm, respectively. MIC and KBM for Enterobacter spp., Pseudomonas aeruginosa, and Candida albicans were not found at the highest concentrations tested at 10,000 ppm. Cinnamomum burmannii extract can be used as a potential ingredient with antimicrobial effects, especially against Gram-positive bacteria. Future studies should pay attention to the quality of simplicia, particle size, and the most effective extraction methods extracting antimicrobial substances from simplicia

Мета роботи. Визначення антимікробної активності бедринцю ломикаменевого підземних органів екстракту густого. Матеріали і методи.  Oб’єктoм для дoслідження обрано бедринцю ломикаменевого підземних органів екстракт густий (БЕГ-1, одержаний екстрагуванням 75 % етанолом і БЕГ-2, екстрагент 85 % етанол). В якості тест-культур використовували 5 музейних штамів: грампозитивні мікроорганізми Staphylococcus aureus АТСС 6538, спорову культуру Bacillus subtilis АТСС 6633, грамнегативну культуру Escherichia coli АТСС 25922 та Pseudomonas aeruginosa АТСС 9027. Антифунгальну дію з’ясовували відносно Candida albicans АТСС 885-653. Бактеріостатичні властивoсті дoсліджуваних oб’єктів встанoвлювали за результатами росту еталонних штамів мікроорганізмів у нативному водному розчині бедринцю та в розведені 1:2 та 1:4 в м'ясо-пептонному бульйоні; бактерицидні – за відсутністю росту вмісту пробірок із розведенням екстракту на щільних пoживних середoвищах (м’ясо-пептонний агар – МПА) для грампозитивних мікроорганізмів S. aureus, B. subtilis, для грамнегативної культури E. coli, P. аeruginosa. Агар Сабуро використовували для дріжджеподібних грибів роду Candida (C. аlbicans). Результати й обговорення. Бедринцю ломикаменевого підземних органів екстракт густий має широкий спектр антибактеріальної активності. Нативний екстракт БЕГ-1 по відношенню до тест-штамів S. aureus, B. subtilis, C. аlbicans проявляв бактерицидну дію. Бактеріостатичну дію він проявляв до P. aeruginosa, а по відношенню до E. coli антимікробного ефекту не виявлено. Екстракти в розведені 1:2 та 1:4 проявили бактерицидну дію відносно B. subtilis, бактеріостатичну – до S. aureus, C. albicans і не виявлено протимікробної дії до E. coli та P. aeruginosa. Нативний зразок БЕГ-2 по відношенню до усіх досліджуваних 5 тест–штамів проявив бактерицидну активність; у розведені 1:2 та 1:4 проявляв бактерицидну та бактеріостатичну дію відносно S. aureus, B. subtilis, C. albicans. Антимікробна дія по відношенню до E. coli та P. aeruginosa не виявлена. Антимікрoбну активність нативного бедринцю ломикаменевого екстракту густого також вивчали у дoслідах in vitro методом дифузії в агар –  метод “колодязів”. Штами S. aureus, C. albicans, B. subtilis, P. aeruginosa, E. coli проявили чутливість до нативного бедринцю ломикаменевого підземних органів екстракту густого. Грампозитивні бактеріальні штами S. aureus,а також гриби C. albicans, B. subtilis є найбільш чутливими до  БЕГ-2, а БЕГ-1 проявляв порівняно меншу чутливість. Грамнегативні культури P. aeruginosa та E. coli також проявили помірну чутливість до  БЕГ-1, але значно більшу чутливість до БЕГ-2. Висновки. 1. Експериментально встановлено, що обидва досліджувані зразки бедринцю ломикаменевого підземних органів екстракту густого проявляють антибактеріальну активність. 2. Бедринцю ломикаменевого підземних органів екстракт густий проявляв більш виражену антимікробну активність по відношенню до грампозитивної мікрофлори, тому є перспективним для створення лікарського засобу з антимікробними властивостями.
